    Enhancing methane production from the invasive macroalga Rugulopteryx okamurae through anaerobic co-digestion with olive mill solid waste: process performance and kinetic analysis

    The biomass valorisation of the invasive brown alga Rugulopteryx okamurae (Dictyotales, Phaeophyceae) is key to curbing the expansion of this invasive macroalga which is generating tonnes of biomass on southern Spain beaches. As a feasible alternative for the biomass management, anaerobic co-digestion is proposed in this study. Although the anaerobic digestion of macroalgae barely produced 177 mL of CH4 g−1 VS, the co-digestion with a C-rich substrate, such as the olive mill solid waste (OMSW, the main waste derived from the two-phase olive oil manufacturing process), improved the anaerobic digestion process. The mixture improved not only the methane yield, but also its biodegradability. The highest biodegradability was found in the mixture 1 R. okamurae—1 OMSW, which improved the biodegradability of the macroalgae by 12.9% and 38.1% for the OMSW. The highest methane yield was observed for the mixture 1 R. okamurae—3 OMSW, improving the methane production of macroalgae alone by 157% and the OMSW methane production by 8.6%. Two mathematical models were used to fit the experimental data of methane production time with the aim of assessing the processes and obtaining the kinetic constants of the anaerobic co-digestion of different combination of R. okamurae and OMSW and both substrates independently. First-order kinetic and the transference function models allowed for appropriately fitting the experimental results of methane production with digestion time. The specific rate constant, k (first-order model) for the mixture 1 R. okamurae- 1.5 OMSW, was 5.1 and 1.3 times higher than that obtained for the mono-digestion of single OMSW and the macroalga, respectively. In the same way, the transference function model revealed that the maximum methane production rate (Rmax) was also found for the mixture 1 R. okamurae—1.5 OMSW (30.4 mL CH4 g−1 VS day−1), which was 1.6 and 2.2 times higher than the corresponding to the mono-digestions of the single OMSW and sole R. okamurae (18.9 and 13.6 mL CH4 g−1 VS day−1), respectively.Ministerio de Ciencia e Innovación PID2020-114975RB-10

    Development, cross-cultural adaptation, and psychometric characteristics of the persian progressive aphasia language scale in patients with primary progressive aphasia: A pilot study

    Introduction: Primary Progressive Aphasia (PPA) is a neurological condition characterized by progressive dissolution of language capabilities. The Progressive Aphasia Language Scale (PALS) is an easy-to-apply bedside clinical scale capable of capturing and grading the key language features essential for the classification of PPA. The objective of the present study was to develop and validate the Persian version of the PALS (PALS-P) as a clinical language assessment test. Methods: In this cross-sectional study, PALS was translated and adapted into Persian according to the international guidelines. A total of 30 subjects (10 subjects with PPA and 20 control subjects without dementia) were recruited to evaluate the intra-rater reliability and discriminant validity of PALS-P. Results: The intra-rater reliability of the PALS-P within a 14-day interval was excellent for each subtest (ICC agreement range=0.81-1.0). PALS-P results were statistically significant among groups, suggesting its discriminative validity. Conclusion: This preliminary study indicates that PALS-P was successfully developed and translated. It seems to be a valid and reliable screening tool to assess language skills in Persian-speaking subjects with progressive aphasia

    Inhibiting cyclin-dependent kinase 5 in the nucleus accumbens enhances the expression of amphetamine-induced locomotor conditioning

    When psychostimulant drugs like amphetamine are administered repeatedly in the presence of a contextual stimulus complex, long-lasting associations form between the unconditioned effects of the drug and the contextual stimuli. Here we assessed the role played by the proline-directed serine/threonine kinase cyclin-dependent kinase 5 (Cdk5) in the nucleus accumbens (NAcc) on the expression of the conditioned locomotion normally observed when rats are returned to a context previously paired with amphetamine. Infusing the Cdk5 inhibitor roscovitine (40 nmol/0.5µl/side) into the NAcc 30-min before the test for conditioning significantly enhanced the conditioned locomotor response observed in rats previously administered amphetamine in the test environment. This effect was specific to the expression of a conditioned response as inhibiting Cdk5 produced no effect in control rats previously administered saline or previously administered amphetamine elsewhere. As inhibiting Cdk5 during exposure to amphetamine has been found to block the accrual of locomotor conditioning, the present results suggest distinct roles for NAcc Cdk5 in the induction and expression of excitatory conditioning by amphetamine

    Modelling \u3ci\u3eLeucaena\u3c/i\u3e Biomass Under Rainfed Production Systems of Semiarid Regions

    The climatic variability of semiarid regions is the main source of uncertainties associated with forage and animal production, indicating a need for tools that accurately estimate forage production in order to construct a forage budgeting plan for livestock. This study simulated the biomass of Leucaena (Leucaena leucocephala) using the PHYGROW model in four locations located in Brazilian Semiarid. The work was carried out based on field data collected from 2019 to 2021. After sowing in 2018, leucaena was harvested when it reached 200 cm and to a target residual height of 70 cm. The biomass (fresh matter) was weighed, sampled and dried to obtain the total forage biomass (BFT) of each sample. The BFT was also estimated using the PHYGROW model, with field data being used to parameterize, calibrate and validate the model. The model performance, in turn, was evaluated based on the mean forecast error (BIAS %), root mean square error (RMSE) and Willmott index. Afterwards, a BFT time series was downloaded for each location, with the highest biomass simulated for each year being evaluated in the @Risk regarding their probability distribution. Thereafter, probability calculations of biomass production were performed, based on different levels of warranty in SigmaPlot software (11.0). The model underestimated the BFT collected in two locations and overestimated BFT in the others. The Weibull function was the best one to describe the data. Regarding biomass production under a 95% natural warranty, it was observed that leucaena showed low variation among locations (2240 ± 752 kg of DM ha-1 year-1 ). The PHYGROW model accurately predicted the leucaena BFT which, in turn, demonstrated significant adaptation potential to the various soil and climate conditions of Brazilian Semiarid. The use of probability analysis can contribute to forage planning, thus reducing the uncertainties related to climate variability, especially in rainfed production systems of dry areas

    Archaeometric analysis of ceramic production in Tiwanaku state (c.500-1000ce) : An exploratory study

    The chemical and mineralogical characterization of seven ceramic fragments produced within Tiwanaku state (c.500-1000 ce) is reported. The instrumental techniques used included X-ray elemental and mineralogical chemical analysis, Raman spectroscopy, and scanning and light microscopy. The results indicate there are several clay types, although they show similarities, such as the use of a plant-based temper. The red colour of the decoration is hematite, and manganese oxides such as jacobsite are present in the black. The white colour is a mixture of gypsum and clay, and the orange is a mixture of hematite and clay. The use of colours, the quality of the clays and the temperatures reached during pottery firing point to expertise in ceramic production and to complex decision-making processes. The multi-elemental archaeometric approach documented here could become an important tool to shed a light on ancient ceramic technology and the internal variance of Tiwanaku pottery.Peer reviewe

    Molecular basis for the folding of β-helical autotransporter passenger domains

    Bacterial autotransporters comprise a C-terminal β-barrel domain, which must be correctly folded and inserted into the outer membrane to facilitate translocation of the N-terminal passenger domain to the cell exterior. Once at the surface, the passenger domains of most autotransporters are folded into an elongated β-helix. In a cellular context, key molecules catalyze the assembly of the autotransporter β-barrel domain. However, how the passenger domain folds into its functional form is poorly understood. Here we use mutational analysis on the autotransporter Pet to show that the β-hairpin structure of the fifth extracellular loop of the β-barrel domain has a crucial role for passenger domain folding into a β-helix. Bioinformatics and structural analyses, and mutagenesis of a homologous autotransporter, suggest that this function is conserved among autotransporter proteins with β-helical passenger domains. We propose that the autotransporter β-barrel domain is a folding vector that nucleates folding of the passenger domain

    Quantum algorithm and circuit design solving the Poisson equation

    The Poisson equation occurs in many areas of science and engineering. Here we focus on its numerical solution for an equation in d dimensions. In particular we present a quantum algorithm and a scalable quantum circuit design which approximates the solution of the Poisson equation on a grid with error \varepsilon. We assume we are given a supersposition of function evaluations of the right hand side of the Poisson equation. The algorithm produces a quantum state encoding the solution. The number of quantum operations and the number of qubits used by the circuit is almost linear in d and polylog in \varepsilon^{-1}. We present quantum circuit modules together with performance guarantees which can be also used for other problems.Comment: 30 pages, 9 figures.
