    Genome sequencing reveals fine scale diversification and reticulation history during speciation in Sus

    Background Elucidating the process of speciation requires an in-depth understanding of the evolutionary history of the species in question. Studies that rely upon a limited number of genetic loci do not always reveal actual evolutionary history, and often confuse inferences related to phylogeny and speciation. Whole-genome data, however, can overcome this issue by providing a nearly unbiased window into the patterns and processes of speciation. In order to reveal the complexity of the speciation process, we sequenced and analyzed the genomes of 10 wild pigs, representing morphologically or geographically well-defined species and subspecies of the genus Sus from insular and mainland Southeast Asia, and one African common warthog. Results Our data highlight the importance of past cyclical climatic fluctuations in facilitating the dispersal and isolation of populations, thus leading to the diversification of suids in one of the most species-rich regions of the world. Moreover, admixture analyses revealed extensive, intra- and inter-specific gene-flow that explains previous conflicting results obtained from a limited number of loci. We show that these multiple episodes of gene-flow resulted from both natural and human-mediated dispersal. Conclusions Our results demonstrate the importance of past climatic fluctuations and human mediated translocations in driving and complicating the process of speciation in island Southeast Asia. This case study demonstrates that genomics is a powerful tool to decipher the evolutionary history of a genus, and reveals the complexity of the process of speciation

    Evolution of a Membrane Protein Regulon in Saccharomyces

    Expression variation is widespread between species. The ability to distinguish regulatory change driven by natural selection from the consequences of neutral drift remains a major challenge in comparative genomics. In this work, we used observations of mRNA expression and promoter sequence to analyze signatures of selection on groups of functionally related genes in Saccharomycete yeasts. In a survey of gene regulons with expression divergence between Saccharomyces cerevisiae and S. paradoxus, we found that most were subject to variation in trans-regulatory factors that provided no evidence against a neutral model. However, we identified one regulon of membrane protein genes controlled by unlinked cis- and trans-acting determinants with coherent effects on gene expression, consistent with a history of directional, nonneutral evolution. For this membrane protein group, S. paradoxus alleles at regulatory loci were associated with elevated expression and altered stress responsiveness relative to other yeasts. In a phylogenetic comparison of promoter sequences of the membrane protein genes between species, the S. paradoxus lineage was distinguished by a short branch length, indicative of strong selective constraint. Likewise, sequence variants within the S. paradoxus population, but not across strains of other yeasts, were skewed toward low frequencies in promoters of genes in the membrane protein regulon, again reflecting strong purifying selection. Our results support a model in which a distinct expression program for the membrane protein genes in S. paradoxus has been preferentially maintained by negative selection as the result of an increased importance to organismal fitness. These findings illustrate the power of integrating expression- and sequence-based tests of natural selection in the study of evolutionary forces that underlie regulatory change

    Primary healthcare and the construction of thematic health networks: what role can they play?

    The enhancement of primary healthcare has been a core strategy for the empowerment of the Brazilian Unified Health System (SUS). Recent guidelines issued by OPAS and the Ministry of Health highlight the role it has played as a thematic communication network center, a regulating agent for the access and use of services required for comprehensive healthcare. Sponsored by PPSUS/Fapesp, this study examines the possibilities of the primary healthcare network exercising such a strategic function. Life narratives involving 15 regular users were produced in two cities of ABC Paulista, which have adopted the Family Health Strategy for the organization of their primary healthcare networks. The study presents three main findings: the primary healthcare network serves as an outpost of SUS by producing user values even for high complexity service users; the primary network is perceived is a place for simple care needs; there is shared impotence between users and teams when it comes to the network functioning as the coordinator of care, indicating that it does not possess the technological, operational and organizational material conditions or symbolic conditions (values, meanings, and representations) to be in a central position in the coordination of thematic healthcare networks.O fortalecimento da atenção bĂĄsica tem sido valorizado como estratĂ©gia central para a construção do SUS. Diretrizes recentes emanadas pela OPAS e pelo MS destacam seu papel como centro de comunicação de redes temĂĄticas, como reguladora do acesso e utilização dos serviços necessĂĄrios para a integralidade do cuidado. O presente estudo, financiado com recursos PPSUS/Fapesp, problematiza as possibilidades da rede bĂĄsica exercer tal função estratĂ©gica. Foram produzidas narrativas de vida de 15 usuĂĄrios altamente utilizadores de serviços de saĂșde em dois municĂ­pios do ABC paulista, que adotaram a EstratĂ©gia de SaĂșde da FamĂ­lia para organização de suas redes bĂĄsicas.     Intimate partner violence against women in an economically vulnerable urban area, Central-West Brazil

    OBJETIVO: Estimar a prevalĂȘncia de tipos de violĂȘncia e de comportamentos de controle praticados por parceiros Ă­ntimos contra mulheres residentes em ĂĄrea economicamente vulnerĂĄvel. MÉTODOS: Conduziu-se estudo transversal com 278 mulheres de 15 a 49 anos que tiveram parceiros Ă­ntimos alguma vez na vida, residentes em uma ĂĄrea metropolitana de BrasĂ­lia, DF, em 2007. Utilizou-se processo de amostragem aleatĂłria sistemĂĄtica. O instrumento de pesquisa constou de um questionĂĄrio com 58 perguntas desenvolvido pela Organização Mundial de SaĂșde. Foram analisadas as prevalĂȘncias de violĂȘncia fĂ­sica, psicolĂłgica e sexual. As variĂĄveis independentes consideradas foram caracterĂ­sticas sociodemogrĂĄficas da mulher, de contexto familiar e comunitĂĄrio bem como as sociodemogrĂĄfi cas do parceiro, de comportamento (freqĂŒĂȘncia do uso de bebidas ou drogas ilĂ­citas e relacionamento extraconjugal). RESULTADOS: A prevalĂȘncia de violĂȘncia psicolĂłgica foi a mais alta: 80,2% (n=223) das mulheres entrevistadas relataram pelo menos um ato no decorrer da vida e 50% (n=139) nos Ășltimos 12 meses. A prevalĂȘncia de violĂȘncia fĂ­sica ao longo da vida foi (58,6%) e nos Ășltimos 12 meses (32%), enquanto a prevalĂȘncia de mulheres que sofreram violĂȘncia sexual foi de 28,8% e 15,5%, respectivamente. CONCLUSÕES: As altas prevalĂȘncias das violĂȘncias mostram a magnitude da vulnerabilidade e das agressĂ”es praticadas contra mulheres nas relaçÔes com parceiros Ă­ntimos.OBJECTIVE: To estimate the prevalence of gender-based controlling behavior and types of violence committed by intimate partners against women living in an economically vulnerable area. METHODS: A cross-sectional study was performed with 278 women aged between 15 and 49 years, who had had at least one male intimate partner in their lives and lived in a metropolitan area of the city of BrasĂ­lia, Central-West Brazil, in 2007. Systematic random sampling process was used. The research instrument consisted of a questionnaire with 58 questions, developed by the World Health Organization. Prevalences of physical, psychological and sexual violence were analyzed. Independent variables considered were women’s sociodemographic, family and community context characteristics, in addition to their partners’ sociodemographic and behavior characteristics (frequency of alcohol or illicit drug use and extra-marital relationship). RESULTS: The highest prevalence was that of psychological violence: 80.2% (n=223) of the women interviewed reported at least one act throughout their lives and 50% (n=139) in the last 12 months. Prevalence of physical violence was 58.6% throughout life and 32% in the last 12 months, whereas those of sexual violence were 28.8% and 15.5%, respectively. CONCLUSIONS: High prevalences of violence show the magnitude of vulnerability and aggressions committed against women in relationships with intimate partners
