    Performance as a classification criterion of tourist origins and destinations

    The definition of performance variables, which can be used as indicators of the results obtained/to be obtained, is a fundamental prerequisite in the carrying out of any evaluation process. Portfolio analysis has been used as a tool for the study of market segments, namely of tourist destinations. However, a review of the literature did not identify a single case where the performance variable is adjusted to the nature of the object. In fact, contrary to many other situations, the use of performance variables in absolute terms does not appear to be the most recommendable solution given that the total volume is conditioned by the size of the territory and, above all, by the number of inhabitants. In other words, for a certain level of socio-economic development, larger countries (in terms of size and number of inhabitants) will tend to have higher absolute values. Besides this, the empirical studies reviewed contain little, or no, information about competition, being temporally static. Although in some cases growth rate is used as a variable, thus presupposing the consideration of two magnitudes in two different moments in time, more than one register per object is never considered, meaning that evolutive analyses are not viable. Consequently, a performance proxy was developed which enables a direct comparison between generating countries (origins) and between destinations, regardless of the number of inhabitants. The selection of the performance proxy can depend on the object of study. In any case, it should always permit the evaluation, in general terms, of the results obtained by the destination for the set of origins under analysis and the results generated by the origin for the set of destinations under study. The proposed tool is a component of a model for the identification of priority market segments, and is constituted by a system of orthogonal axes which produces four quadrants that are distinct in terms of intensity (axis OY) and growth (axis OX) of tourist flows. The tool, like the model, can be applied to any type of destination (city, region or country) and to any market segmentation criterion (geographic, demographic, behavioural, psychographic or other). In the study reported here, the instrument is applied to the 15 Member States of the European Union (before its expansion on May 1st 2004), for the period from 1996 to 2001. Eurostat was the sole source of data for the study. The UK and Ireland are the countries classified as ‘Progressing Origins’. With 16.8% of the population, they increased their contribution from 17.7% to 21.9% and are responsible for 38.7% of the growth that occurred between 1996 and 2001. Spain, Greece and Ireland are the countries classified as ‘Progressing Destinations’. With 14.4% of the population, they increased their share of the market from 21.5% to 24.2%, capturing 35.1% of the growth that occurred between 1996 and 2001

    Resistance to quambalaria shoot blight and myrtle rust in Corymbia calophylla seedlings

    Abstract Corymbia calophylla (marri), an endemic keystone tree species in southwest Western Australia, is increasingly impacted by the introduced basidiomycete smut Quambalaria pitereka. The basidiomycete rust Austropuccinia psidii (myrtle rust), an invasive pathogen recently introduced to Eastern Australia, is expected to spread to the southwest of Western Australia eventually. Austropuccinia psidii has similar epidemiology to Q. pitereka, and there is concern that C. calophylla may be susceptible. Preliminary pathogenicity tests showed significant differences in aggressiveness between twelve Q. pitereka isolates, and there was evidence of interactions between isolates and C. calophylla provenances. Seedlings from 59 open-pollinated families from 11 provenances covering the natural range of marri were screened for resistance to Q. pitereka and A. psidii under controlled glasshouse conditions. Resistance of seedlings within provenances to Q. pitereka and A. psidii differed significantly. There was no significant correlation between resistance to Q. pitereka and resistance to A. psidii. Seedlings of provenances from wetter regions were more resistant to both pathogens, but the correlation coefficients were insignificant. Seedlings of four families in three provenances (Serpentine, Chidlow, and Kingston) showed 100% resistance to Q. pitereka. Narrow-sense heritability estimates were 0.07 for quambalaria shoot blight resistance and 0.34 for myrtle rust resistance. The results indicate the potential to use selected families/individuals resistant to Q. pitereka and A. psidii for tree improvement programs and adaptive management strategies

    Quambalaria shoot blight resistance in marri (Corymbia calophylla) : genetic parameters and correlations between growth rate and blight resistance

    Quambalaria shoot blight (QSB) has emerged recently as a severe disease of Corymbia calophylla (marri). In this study, QSB damage and growth were assessed in Corymbia calophylla trees at 4 and 6 years of age in two common gardens consisting of 165 and 170 open-pollinated families representing 18 provenances across the species’ natural distribution. There were significant differences between provenances for all traits. The narrow-sense heritability for growth traits and QSB damage at both sites were low to moderate. The genetic correlation between QSB damage and growth traits was negative; fast-growing families were less damaged by QSB disease. Age-age genetic correlations for individual traits at four and six years were very strong, and the type-B (site–site) correlations were strongly positive for all traits. Provenances from cooler wetter regions showed higher resistance to QSB. The QSB incidence at 6 years was significantly correlated with environmental factors of the provenance’s origin. The QSB incidence at years four and six was not correlated with the QSB expression in 3-month-old seedlings. Based on these results, selection for resistance could be undertaken using 4-year-old trees.     Stalking influenza by vaccination with pre-fusion headless HA mini-stem.

    Inaccuracies in prediction of circulating viral strain genotypes and the possibility of novel reassortants causing a pandemic outbreak necessitate the development of an anti-influenza vaccine with increased breadth of protection and potential for rapid production and deployment. The hemagglutinin (HA) stem is a promising target for universal influenza vaccine as stem-specific antibodies have the potential to be broadly cross-reactive towards different HA subtypes. Here, we report the design of a bacterially expressed polypeptide that mimics a H5 HA stem by protein minimization to focus the antibody response towards the HA stem. The HA mini-stem folds as a trimer mimicking the HA prefusion conformation. It is resistant to thermal/chemical stress, and it binds to conformation-specific, HA stem-directed broadly neutralizing antibodies with high affinity. Mice vaccinated with the group 1 HA mini-stems are protected from morbidity and mortality against lethal challenge by both group 1 (H5 and H1) and group 2 (H3) influenza viruses, the first report of cross-group protection. Passive transfer of immune serum demonstrates the protection is mediated by stem-specific antibodies. Furthermore, antibodies indudced by these HA stems have broad HA reactivity, yet they do not have antibody-dependent enhancement activity

    Consumer behaviour in tourism: Concepts, influences and opportunities

    Although consumer behaviour (CB) is one of the most researched areas in the field of tourism, few extensive reviews of the body of knowledge in this area exist. This review article examines what we argue are the key concepts, external influences and opportune research contexts in contemporary tourism CB research. Using a narrative review, we examine the CB literature published in three major tourism journals from 2000 to 2012. Of 519 articles identified and reviewed, 191 are included in this article. We examine the development of and scope for future research on nine key concepts, including decision-making, values, motivations, self-concept and personality, expectations, attitudes, perceptions, satisfaction, trust and loyalty. We then examine three important external influences on tourism behaviour, technology, Generation Y and the rise in concern over ethical consumption. Finally, we identify and discuss five research contexts that represent major areas for future scholarship: group and joint decision-making, under-researched segments, cross-cultural issues in emerging markets, emotions and consumer misbehaviour. Our examination of key research gaps is concluded by arguing that the hedonic and affective aspects of CB research in tourism must be brought to bear on the wider CB and marketing literature

    Relative contribution of mutations in genes for autosomal dominant distal hereditary motor neuropathies: a genotype-phenotype correlation study

    Distal hereditary motor neuropathy (HMN) is a clinically and genetically heterogeneous group of disorders affecting spinal α-motor neurons. Since 2001, mutations in six different genes have been identified for autosomal dominant distal HMN; glycyl-tRNA synthetase (GARS), dynactin 1 (DCTN1), small heat shock 27 kDa protein 1 (HSPB1), small heat shock 22 kDa protein 8 (HSPB8), Berardinelli-Seip congenital lipodystrophy (BSCL2) and senataxin (SETX). In addition a mutation in the (VAMP)-associated protein B and C (VAPB) was found in several Brazilian families with complex and atypical forms of autosomal dominantly inherited motor neuron disease. We have investigated the distribution of mutations in these seven genes in a cohort of 112 familial and isolated patients with a diagnosis of distal motor neuropathy and found nine different disease-causing mutations in HSPB8, HSPB1, BSCL2 and SETX in 17 patients of whom 10 have been previously reported. No mutations were found in GARS, DCTN1 and VAPB. The phenotypic features of patients with mutations in HSPB8, HSPB1, BSCL2 and SETX fit within the distal HMN classification, with only one exception; a C-terminal HSPB1-mutation was associated with upper motor neuron signs. Furthermore, we provide evidence for a genetic mosaicism in transmitting an HSPB1 mutation. This study, performed in a large cohort of familial and isolated distal HMN patients, clearly confirms the genetic and phenotypic heterogeneity of distal HMN and provides a basis for the development of algorithms for diagnostic mutation screening in this group of disorder

    Special considerations for studies of extracellular vesicles from parasitic helminths: a community-led roadmap to increase rigour and reproducibility

    Over the last decade, research interest in defining how extracellular vesicles (EVs) shape cross-species communication has grown rapidly. Parasitic helminths, worm species found in the phyla Nematoda and Platyhelminthes, are well-recognised manipulators of host immune function and physiology. Emerging evidence supports a role for helminth-derived EVs in these processes and highlights EVs as an important participant in cross-phylum communication. While the mammalian EV field is guided by a community-agreed framework for studying EVs derived from model organisms or cell systems [e.g., Minimal Information for Studies of Extracellular Vesicles (MISEV)], the helminth community requires a supplementary set of principles due to the additional challenges that accompany working with such divergent organisms. These challenges include, but are not limited to, generating sufficient quantities of EVs for descriptive or functional studies, defining pan-helminth EV markers, genetically modifying these organisms, and identifying rigorous methodologies for in vitro and in vivo studies. Here, we outline best practices for those investigating the biology of helminth-derived EVs to complement the MISEV guidelines. We summarise community-agreed standards for studying EVs derived from this broad set of non-model organisms, raise awareness of issues associated with helminth EVs and provide future perspectives for how progress in the field will be achieved

    Efficacy and Safety of Lacosamide in Painful Diabetic Neuropathy

    OBJECTIVE: To evaluate efficacy and safety of lacosamide compared with placebo in painful diabetic polyneuropathy. RESEARCH DESIGN AND METHODS: Diabetic patients with at least moderate neuropathic pain were randomized to placebo or lacosamide 400 (in a slow or standard titration) or 600 mg/day over 6-week titration and 12-week maintenance periods. Primary efficacy criterion was intra-individual change in average daily Numeric Pain Rating Scale score from baseline to the last 4 weeks. RESULTS: For the primary end point, pain reduction was numerically but not statistically greater with lacosamide compared with placebo (400 mg/day, P = 0.12; 600 mg/day, P = 0.18). Both doses were significantly more effective compared with placebo over the titration (P = 0.03, P = 0.006), maintenance (P = 0.01, P = 0.005), and entire treatment periods (P = 0.03, P = 0.02). Safety profiles between titration schemes were similar. CONCLUSIONS: Lacosamide reduced neuropathic pain and was well tolerated in diabetic patients, but the primary efficacy criterion was not met, possibly due to an increased placebo response over the last 4 weeks.status: publishe

    Loss of function mutations in HARS cause a spectrum of inherited peripheral neuropathies.

    Inherited peripheral neuropathies are a genetically heterogeneous group of disorders characterized by distal muscle weakness and sensory loss. Mutations in genes encoding aminoacyl-tRNA synthetases have been implicated in peripheral neuropathies, suggesting that these tRNA charging enzymes are uniquely important for the peripheral nerve. Recently, a mutation in histidyl-tRNA synthetase (HARS) was identified in a single patient with a late-onset, sensory-predominant peripheral neuropathy; however, the genetic evidence was lacking, making the significance of the finding unclear. Here, we present clinical, genetic, and functional data that implicate HARS mutations in inherited peripheral neuropathies. The associated phenotypic spectrum is broad and encompasses axonal and demyelinating motor and sensory neuropathies, including four young patients presenting with pure motor axonal neuropathy. Genome-wide linkage studies in combination with whole-exome and conventional sequencing revealed four distinct and previously unreported heterozygous HARS mutations segregating with autosomal dominant peripheral neuropathy in four unrelated families (p.Thr132Ile, p.Pro134His, p.Asp175Glu and p.Asp364Tyr). All mutations cause a loss of function in yeast complementation assays, and p.Asp364Tyr is dominantly neurotoxic in a Caenorhabditis elegans model. This study demonstrates the role of HARS mutations in peripheral neuropathy and expands the genetic and clinical spectrum of aminoacyl-tRNA synthetase-related human disease
