    Fabrication of mirror templates in silica with micron-sized radii of curvature

    We present the fabrication of exceptionally small-radius concave microoptics on fused silica substrates using CO2 laser ablation and subsequent reactive ion etching. The protocol yields on-axis near-Gaussian depressions with radius of curvature ā‰²5\lesssim5 microns at shallow depth and low surface roughness of 2 angstroms. This geometry is appealing for cavity quantum electrodynamics where small mode volumes and low scattering losses are desired. We study the optical performance of the structure within a tunable Fabry-Perot type microcavity, demonstrate near-coating-limited loss rates (F = 25,000) and small focal lengths consistent with their geometrical dimensions.Comment: 5 pages, 4 figure

    An artificial Rb atom in a semiconductor with lifetime-limited linewidth

    We report results important for the creation of a best-of-both-worlds quantum hybrid system consisting of a solid-state source of single photons and an atomic ensemble as quantum memory. We generate single photons from a GaAs quantum dot (QD) frequency-matched to the Rb D2-transitions and then use the Rb transitions to analyze spectrally the quantum dot photons. We demonstrate lifetime-limited QD linewidths (1.48 GHz) with both resonant and non-resonant excitation. The QD resonance fluorescence in the low power regime is dominated by Rayleigh scattering, a route to match quantum dot and Rb atom linewidths and to shape the temporal wave packet of the QD photons. Noise in the solid-state environment is relatively benign: there is a blinking of the resonance fluorescence at MHz rates but negligible upper state dephasing of the QD transition. We therefore establish a close-to-ideal solid-state source of single photons at a key wavelength for quantum technologies

    Design, Implementation and Evaluation of a National Campaign to Deliver 18 Million Free Long-Lasting Insecticidal Nets to Uncovered Sleeping Spaces in Tanzania.

    Since 2004, the Tanzanian National Voucher Scheme has increased availability and accessibility of insecticide-treated nets (ITNs) to pregnant women and infants by subsidizing the cost of nets purchased. From 2008 to 2010, a mass distribution campaign delivered nine million long-lasting insecticidal nets (LLINs) free-of-charge to children under-five years of age in Tanzania mainland. In 2010 and 2011, a Universal Coverage Campaign (UCC) led by the Ministry of Health and Social Welfare (MoHSW) was implemented to cover all sleeping spaces not yet reached through previous initiatives. The UCC was coordinated through a unit within the National Malaria Control Programme. Partners were contracted by the MoHSW to implement different activities in collaboration with local government authorities. Volunteers registered the number of uncovered sleeping spaces in every household in the country. On this basis, LLINs were ordered and delivered to village level, where they were issued over a three-day period in each zone (three regions). Household surveys were conducted in seven districts immediately after the campaign to assess net ownership and use. The UCC was chiefly financed by the Global Fund to Fight AIDS, Tuberculosis and Malaria with important contributions from the US President's Malaria Initiative. A total of 18.2 million LLINs were delivered at an average cost of USD 5.30 per LLIN. Overall, 83% of the expenses were used for LLIN procurement and delivery and 17% for campaign associated activities. Preliminary results of the latest Tanzania HIV Malaria Indicator Survey (2011-12) show that household ownership of at least one ITN increased to 91.5%. ITN use, among children under-five years of age, improved to 72.7% after the campaign. ITN ownership and use data post-campaign indicated high equity across wealth quintiles. Close collaboration among the MoHSW, donors, contracted partners, local government authorities and volunteers made it possible to carry out one of the largest LLIN distribution campaigns conducted in Africa to date. Through the strong increase of ITN use, the recent activities of the national ITN programme will likely result in further decline in child mortality rates in Tanzania, helping to achieve Millennium Development Goals 4 and 6

    Expression of Procyclin mRNAs during Cyclical Transmission of Trypanosoma brucei

    Trypanosoma brucei, the parasite causing human sleeping sickness, relies on the tsetse fly for its transmission. In the insect, EP and GPEET procyclins are the major surface glycoproteins of procyclic (midgut) forms of the parasite, with GPEET predominating in the early procyclic form and two isoforms of EP in the late procyclic form. EP procyclins were previously detected on salivary gland trypanosomes, presumably epimastigotes, by immunoelectron microscopy. However, no procyclins could be detected by mass spectrometry when parasites were isolated from infected glands. We have used qualitative and quantitative RT-PCR to analyse the procyclin mRNAs expressed by trypanosomes in the tsetse midgut and salivary glands at different time points after infection. The coding regions of the three EP isoforms (EP1, EP2 and EP3) are extremely similar, but their 3ā€² untranslated regions contain unique sequences that make it possible to assign the cDNAs amplified by this technique. With the exception of EP2, we found that the spectrum of procyclin mRNAs expressed in the midgut mirrors the protein repertoire of early and established procyclic forms. Surprisingly, procyclin mRNAs, including that of GPEET, are present at relatively high levels in salivary gland trypanosomes, although the proteins are rarely detected by immunofluorescence. Additional experiments using transgenic trypanosomes expressing reporter genes or mutant forms of procyclin point to a mechanism of translational or post-translational control, involving the procyclin coding regions, in salivary gland trypanosomes. It is widely accepted that T. brucei always has a coat of either variant surface glycoprotein or procyclin. It has been known for many years that the epimastigote form does not have a variant surface glycoprotein coat. The finding that this life cycle stage is usually negative for procyclin as well is new, and means that the paradigm will need to be revised

    Magma mixing enhanced by bubble segregation

    In order to explore the materials' complexity induced by bubbles rising through mixing magmas, bubble-advection experiments have been performed, employing natural silicate melts at magmatic temperatures. A cylinder of basaltic glass was placed below a cylinder of rhyolitic glass. Upon melting, bubbles formed from interstitial air. During the course of the experimental runs, those bubbles rose via buoyancy forces into the rhyolitic melt, thereby entraining tails of basaltic liquid. In the experimental run products, these plume-like filaments of advected basalt within rhyolite were clearly visible and were characterised by microCT and high-resolution EMP analyses. The entrained filaments of mafic material have been hybridised. Their post-experimental compositions range from the originally basaltic composition through andesitic to rhyolitic composition. Rheological modelling of the compositions of these hybridised filaments yield viscosities up to 2 orders of magnitude lower than that of the host rhyolitic liquid. Importantly, such lowered viscosities inside the filaments implies that rising bubbles can ascend more efficiently through pre-existing filaments that have been generated by earlier ascending bubbles. MicroCT imaging of the run products provides textural confirmation of the phenomenon of bubbles trailing one another through filaments. This phenomenon enhances the relevance of bubble advection in magma mixing scenarios, implying as it does so, an acceleration of bubble ascent due to the decreased viscous resistance facing bubbles inside filaments and yielding enhanced mass flux of mafic melt into felsic melt via entrainment. In magma mixing events involving melts of high volatile content, bubbles may be an essential catalyst for magma mixing. Moreover, the reduced viscosity contrast within filaments implies repeated replenishment of filaments with fresh end-member melt. As a result, complex compositional gradients and therefore diffusion systematics can be expected at the filament-host melt interface, due to the repetitive nature of the process. However, previously magmatic filaments were tacitly assumed to be of single-pulse origin. Consequently, the potential for multi-pulse filaments has to be considered in outcrop analyses. As compositional profiles alone may remain ambiguous for constraining the origin of filaments, and as 3-D visual evidence demonstrates that filaments may have experienced multiple bubbles passages even when featuring standard diffusion gradients, therefore, the calculation of diffusive timescales may be inadequate for constraining timescales in cases where bubbles have played an essential role in magma mixing. Data analysis employing concentration variance relaxation in natural samples can distinguish conventional single-pulse filaments from advection via multiple bubble ascent advection in natural samples, raising the prospect of yet another powerful application of this novel petrological tool

    Malaria sporozoites induce swift activation of antigen-specific CD8(+) T cells that inhibit the intracellular development of liver-stage parasites. The length of time of functional in vivo antigen presentation, estimated by monitoring the activation of antigen-specific CD8(+) T cells, is of short duration, with maximum T cell activation occurring within the first 8 h after immunization and lasting approximately 48 h. Although the magnitude of the CD8(+) T cell response closely correlates with the number of parasites used for immunization, increasing the time of antigen presentation by daily immunizations does not enhance the magnitude of this response. Thus, once a primary clonal burst is established, the CD8(+) T cell response becomes refractory or unresponsive to further antigenic stimulation. These findings strongly suggest that the most efficient strategy for the induction of primary CD8(+) T cell responses is the delivery of a maximal amount of antigen in a single dose, thereby ensuring a clonal burst that involves the largest number of precursors to become memory cells

    A Homogeneous Actor-Based Monitor Language for Adaptive Behaviour

    This paper describes a structured approach to encoding monitors in an actor language. Within a configuration of actors, each of which publishes a history, a monitor is an independent actor that triggers an action based on patterns occurring in the histories. We define a monitor language based on linear temporal logic and show how it can be homogeneously embedded within an actor language. The approach is demonstrated through a number of examples and evaluated in terms of a real-world actor-based simulation

    Use of a Semi-field System to Evaluate the Efficacy of Topical Repellents under user Conditions Provides a Disease Exposure free Technique Comparable with Field Data.

    Before topical repellents can be employed as interventions against arthropod bites, their efficacy must be established. Currently, laboratory or field tests, using human volunteers, are the main methods used for assessing the efficacy of topical repellents. However, laboratory tests are not representative of real life conditions under which repellents are used and field-testing potentially exposes human volunteers to disease. There is, therefore, a need to develop methods to test efficacy of repellents under real life conditions while minimizing volunteer exposure to disease. A lotion-based, 15% N, N-Diethyl-3-methylbenzamide (DEET) repellent and 15% DEET in ethanol were compared to a placebo lotion in a 200 sq m (10 m x 20 m) semi-field system (SFS) against laboratory-reared Anopheles arabiensis mosquitoes and in full field settings against wild malaria vectors and nuisance-biting mosquitoes. The average percentage protection against biting mosquitoes over four hours in the SFS and field setting was determined. A Poisson regression model was then used to determine relative risk of being bitten when wearing either of these repellents compared to the placebo. Average percentage protection of the lotion-based 15% DEET repellent after four hours of mosquito collection was 82.13% (95% CI 75.94-88.82) in the semi-field experiments and 85.10% (95% CI 78.97-91.70) in the field experiments. Average percentage protection of 15% DEET in ethanol after four hours was 71.29% (CI 61.77-82.28) in the semi-field system and 88.24% (84.45-92.20) in the field. Semi-field evaluation results were comparable to full-field evaluations, indicating that such systems could be satisfactorily used in measuring efficacy of topically applied mosquito repellents, thereby avoiding risks of exposure to mosquito-borne pathogens, associated with field testing
