Análise do fosfolipidoma plasmático revela diferenças entre o perfil de crianças com fenilcetonúria e de crianças saudáveis
Phenylketonuria (PKU) is the most prevalent innate error in amino acid
metabolism. PKU is characterized by the deficiency of a phenylalanine (Phe)
metabolism enzyme, phenylalanine hydroxylase, which is responsible for the
conversion of Phe into tyrosine (Tyr). Deficiency of this enzyme causes the
accumulation of Phe, which may cause severe cognitive impairment. Early
diagnosis through neonatal screening and a rapid therapeutic implementation
are essential to prevent irreversible sequelae. Therapeutic approach for PKU is
based on strict control of Phe intake through a lifelong diet restricted in proteinrich food. High dietary restrictions can lead to imbalances in other nutrients,
notably lipids.
Previous plasma/serum and red blood cells studies of PKU patients
with Phe-restricted diet revealed lipid changes, namely in lipoprotein’s
components and fatty acid profile. Despite the changes already reported,
further studies are needed to understand in detail the changes in the lipid
profile of PKU patients, particularly at the level of phospholipids, which also
have important signalling and regulatory functions. Thus, the main aim of this
work was to study the phenylketonuric phospholipidome in order to identify the
occurrence of changes in lipid profile. To this end, the plasma phospholipidenriched extracts obtained from PKU and healthy children (control group, CT)
were analysed by HILIC-MS/MS and GC-MS.
Using this approach, 187 lipid species belonging to 9 different
phospholipid classes and 3 ceramides were identified. Principal component
analysis of lipid species dataset showed a distinction between PKU and CT
groups. Univariate analysis revealed that 146 phospholipid species were
significantly different between groups. The species with major variation
included phosphatidylcholines (PC), bearing polyunsaturated fatty acids
(PUFA), which were more abundant in the PKU group. For patients with diet
supplemented with PUFA, the higher level of PUFA-containing lipid species
may be related with such supplementation. This study was the first report
comparing the plasma polar lipidome of PKU and healthy children, highlighting
that the phospholipidome of PKU children is significantly altered when
compared with healthy children.
In conclusion, the lipidomic approach used in this work allowed the
analysis and identification of the phospholipid profile of PKU and healthy
children. However, further studies with larger cohorts are needed to clarify if the
changes identified are specific to the phenylketonuric children, namely those
with PUFA supplementation.A fenilcetonúria (PKU) é o erro inato mais prevalente no metabolismo
de aminoácidos. A PKU é caracterizada pela deficiência de uma enzima do
metabolismo da fenilalanina (Phe), a fenilalanina hidroxilase, a qual é
responsável pela conversão de Phe em tirosina (Tyr). A deficiência desta
enzima causa a acumulação de Phe, podendo causar um comprometimento
cognitivo grave. Assim, o diagnóstico precoce através do rastreio neonatal e
uma rápida implementação do tratamento são essenciais para evitar sequelas
irreversíveis. A abordagem terapêutica usada na PKU baseia-se no controlo
rigoroso do aporte de Phe através uma dieta restrita em alimentos ricos em
proteínas que deve ser mantida durante toda a vida. As elevadas restrições na
dieta podem levar a desequilíbrios noutros nutrientes, nomeadamente em
lípidos.
Estudos anteriores com plasma/soro e glóbulos vermelhos de
pacientes com PKU com dieta restrita em Phe revelaram alterações lipídicas,
nomeadamente nos componentes de lipoproteínas e no perfil de ácidos
gordos. Apesar das alterações já reportadas, são necessários mais estudos
para compreender em detalhe as alterações no perfil lipídico de pacientes com
PKU, nomeadamente a nível dos fosfolípidos, os quais também têm
importantes funções de sinalização e regulação. Assim, o principal objetivo
deste trabalho foi estudar o fosfolipidoma de fenilcetonúricos, a fim de
identificar a ocorrência de alterações no perfil lipídico. Para tal, extratos
plasmáticos enriquecidos em fosfolípidos obtidos a partir de crianças com PKU
e de crianças saudáveis (grupo controlo, CT) foram analisados por HILICMS/MS e GC-MS.
Deste modo, foram identificadas 187 espécies lipídicas pertencentes a
9 classes diferentes de fosfolípidos e 3 ceramidas. A análise de componentes
principais do conjunto de dados das espécies lipídicas mostrou uma distinção
entre os grupos PKU e CT. A análise univariada revelou que 146 espécies de
fosfolípidos foram significativamente diferentes entre os dois grupos. As
espécies com maior variação incluíram principalmente fosfatidilcolinas,
contendo ácidos gordos polinsaturados (PUFA), estando estas mais
abundantes no grupo PKU. Para doentes com uma dieta suplementada com
PUFA, os níveis elevados de espécies contendo PUFA podem estar
relacionados com a suplementação. Este estudo foi o primeiro a comparar o
lipidoma polar plasmático de crianças com PKU e de crianças saudáveis,
realçando que o fosfolipidoma de crianças com PKU está significativamente
alterado quando comparado com crianças saudáveis.
Em suma, a abordagem lipidómica usada neste estudo permitiu a
análise e identificação do perfil fosfolipídico de crianças com PKU e de
crianças saudáveis. Contudo, mais estudos são necessários para esclarecer
se alterações identificadas são específicas de crianças fenilcetonúricas,
nomeadamente daquelas que fazem suplementação com PUFA.Mestrado em Bioquímic

A consequência é normalmente a perda dos contornos do rebordo alveolar, levando a compromissos estéticos e a invibilização da colocação ideal de um implante.
É cada vez mais notório que a regeneração óssea se mostra fundamental no que respeita à reabilitação implantosuportada, devido a muitos pacientes apresentarem estas reabsorções do rebordo alveolar.
Neste sentido, várias técnicas estão descritas na literatura, visando à preservação do alvéolo pós-extracção, como o caso da extracção atraumática, regeneração óssea guiada utilizando membranas e enxertos, colocação imediata de implantes, entre outras. Deste modo, é possível proporcionar aos pacientes uma boa reabilitação, tanto funcionalmente como esteticamente.
Com base numa revisão bibliográfica, a presente monografia tem como objectivo descrever as técnicas utilizadas actualmente na preservação do alvéolo pós-extracção, os biomateriais que se destacam nestes procedimentos e discussão dos seus resultados clínicos
As intervenções do enfermeiro especialista em enfermagem de reabilitação (EEER) na prevenção de complicações no utente submetido a ventilação mecânica invasiva
O progresso técnico e científico possibilitou o desenvolvimento de técnicas e procedimentos revolucionários na área da saúde, que permitiram não só um incremento na sobrevida, mas também na qualidade de vida dos indivíduos com doença crítica. As técnicas de substituição de funções vitais, entre as quais se destaca a Ventilação Mecânica Invasiva (VMI), constituem um impressionante exemplo do progresso médico.
A VMI assume-se como uma terapêutica de suporte respiratório eficaz, necessária à manutenção da vida em determinadas circunstâncias e patologias. No entanto, à semelhança de qualquer outra terapêutica, não se assume como inócua podendo induzir na pessoa alterações da sua função. Das potenciais complicações destacam-se como mais comuns as alterações neurológicas, respiratórias e músculo-esqueléticas, comprometendo o autocuidado da pessoa e a sua qualidade de vida.
O cuidado ao utente submetido a VMI assume-se como uma área prioritária para o Enfermeiro Especialista em Enfermagem de Reabilitação, sobretudo pelo seu cariz urgente/emergente na prevenção de possíveis complicações decorrentes não só do processo de doença, mas também da própria técnica de suporte vital.
A elaboração deste relatório de aprendizagem visa dar resposta à necessidade de investigação identificada no projeto de aprendizagem: “As intervenções do Enfermeiro Especialista em Enfermagem de Reabilitação (EEER) na prevenção de complicações no utente submetido a ventilação mecânica invasiva”.
O principal objetivo deste trabalho prende-se com demonstrar as competências adquiridas de acordo com o preconizado pela Teoria do Autocuidado de Dorothea Orem, bem como proceder à análise reflexiva do desempenho ao longo do processo de aprendizagem
Una enfermedad lejana: la información sobre poliomielitis y síndrome post-polio en la prensa hispanolusa, 1995-2009
[ES] Se explora el cambio en la percepción social de la polio en la Península Ibérica a través del análisis de contenidos, entre 1995 y 2009, de dos periódicos de gran tirada. La desaparición en la agenda periodística de la polio y de las personas que viven con sus secuelas influyó en el olvido de la misma en la agenda pública. La poliomielitis se vinculó a la pobreza y la ignorancia en países lejanos, susceptibles de acciones de cooperación, siendo objeto de atención solo cuando es percibida como amenaza para Occidente, vinculada a crisis sanitarias o en un sentido metafórico. Así, el síndrome post-polio fue invisibilizado en el caso portugués y débilmente representado en España por el movimiento asociativo.[EN] The change in the social perception of poliomyelitis in the Iberian Peninsula through content analysis of two largecirculation newspapers between 1995 and 2009 is examined. The disappearance from the journalistic agenda of poliomyelitis and people living with the after-effects of the disease led it to be excluded from the public agenda. Poliomyelitis was associated with poverty and ignorance in distant countries that were susceptible to cooperation activities and only came to public attention when it was perceived as a threat to the West, linked to health crises or in a metaphorical sense. Operation regimes: A comparison based on Nannochloropsis oceanica biomass and lipid productivity
Microalgae are currently considered to be a promising feedstock for biodiesel production. However, significant research efforts are crucial to improve the current biomass and lipid productivities under real outdoor production conditions. In this context, batch, continuous and semi-continuous operation regimes were compared during the Spring/Summer seasons in 2.6 m(3) tubular photobioreactors to select the most suitable one for the production of the oleaginous microalga Nannochloropsis oceanica. Results obtained revealed that N. oceanica grown using the semi-continuous and continuous operation regimes enabled a 1.5-fold increase in biomass volumetric productivity compared to that cultivated in batch. The lipid productivity was 1.7-fold higher under semi-continuous cultivation than that under a batch operation regime. On the other hand, the semi-continuous and continuous operation regimes spent nearly the double amount of water compared to that of the batch regime. Interestingly, the biochemical profile of produced biomass using the different operation regimes was not affected regarding the contents of proteins, lipids and fatty acids. Este estudo inscreve-se no âmbito do projecto intitulado “Imigração: implicações económicas, profissionais, socioculturais e linguísticas”. Uma vasta literatura empírica revela o fenómeno do despedimento no
mercado de trabalho, sendo que os custos em termos de desemprego (incidência e duração) e de perdas salariais (magnitude e persistência) têm sido os aspectos mais estudados. Neste estudo procura-se
compreender as consequências deste fenómeno na deslocalização dos recursos humanos de outros países para Portugal e avaliar a incidência dos imigrantes na região norte de Portugal, nomeadamente ajudar a
clarificar os mecanismos de determinação das medidas de política económica. (sectores mais procurados, integração, interacções nas organizações). Com a descolonização e, mais tarde, com a integração na União
Europeia, o tecido social e cultural sofreu profundas alterações. Portugal foi-se tornando país de acolhimento.
Foi criada uma base de dados com informação sobre um conjunto significativo de empresas (distrito, concelho, actividade económica, natureza jurídica e forma de gestão), bem como informação individualizada sobre características do pessoal imigrante em serviço (sexo, idade, nível de escolaridade, nível de qualificação, profissão, remunerações, horas de trabalho). Optou-se por métodos quantitativos (foram desenhados e aplicados inquéritos) e qualitativos, por concordarmos com Stiles (1993, in Gonçalves, 2000), ao defender que a condensação da experiência humana em palavras é menos redutora que a sua simbolização em

Assessment of carbon metabolism of Coffee Kawisari hybrid challenged by Hemileia vastatrix, the causal agent of Coffee Leaf Rust.
Plants have evolved sophisticated mechanisms to coordinate carbon metabolism during growth and development both under optimal and stress conditions. In coffee-rust biotrophic interactions, plants try to limit pathogen access to nutrients (e.g., sugars and sugar derivatives) and trigger immune responses, while Hemileia vastatrix (Hv) attempts to circumvent plant defences and control the host's primary metabolism for its own benefit. Previous proteomics data highlighted the up-regulation of proteins from photosynthesis, primary metabolism, and redox-related enzymes along the coffee resistance response. Coffee Kawisari hybrid - Hv interactions (resistant and susceptible reactions) were evaluated using a single sample fractionation method for metabolite and protein extraction. The microscopic evaluation of the Hv infection process revealed that coffee resistance was associated with early hypersensitive response and accumulation of phenolic-like compounds in host cell walls. GC-TOF-MS untargeted metabolomics allowed the identification of metabolic components, such as sugars, sugar derivatives, amino acids, phenylpropanoids, chlorogenic acids, alkaloids, and fatty acids (using the Golm Metabolome database). The overrepresentation of several caffeoylquinic acids in the resistance reaction may be linked to the accumulation of the phenolic-like compounds that were cytologically observed at the infection sites. Furthermore, sugar-related features also played a role in distinguishing between resistant and susceptible reactions, such as glucose and galactose. The cellular availability of mono and disaccharides is the result of the activity of several enzymes, e.g., sucrose synthase and invertases, that can be targeted by Hv in its strategy to manipulate plant carbon metabolism. The activity profile of these enzymes along the infection will be discussed. Proteomic analysis of the same samples (using the single sample fractionation method previously mentioned) is foreseen. Heterotrophic and photoautotrophic media optimization using response surface methodology for the Novel Microalga Chlorococcum amblystomatis
The nutritional requirements of novel microalgal strains are key for their effective cultivation and metabolite content. Therefore, the optimization of heterotrophic and photoautotrophic culture media is crucial for novel Chlorococcum amblystomatis growth. Heterotrophic and photoautotrophic biomass samples were characterized to identify the differences between their heterotrophic and photoautotrophic biomass composition and their biotechnological potential. Media optimization through surface response methodology led to 44.9 and 51.2% increments in C. amblystomatis-specific growth rates under heterotrophic and photoautotrophic growth, respectively. This microalga registered high protein content (61.49–73.45% dry weight), with the highest value being observed in the optimized photoautotrophic growth medium. The lipid fraction mainly constituted polyunsaturated fatty acids, ranging from 44.47 to 51.41% for total fatty acids (TFA) in cells under heterotrophy. However, these contents became significantly higher (70.46–72.82% TFA) in cultures cultivated under photoautotrophy. An interesting carotenoids content was achieved in the cultures grown in optimized photoautotrophic medium: 5.84 mg·g−1 β-carotene, 5.27 mg·g−1 lutein, 3.66 mg·g−1 neoxanthin, and 0.75 mg·g−1 violaxanthin. Therefore, C. amblystomatis demonstrated an interesting growth performance and nutritional profile for food supplements and feed products that might contribute to meeting the world’s nutritional demand.info:eu-repo/semantics/publishedVersio
Effects of LED lighting on Nannochloropsis oceanica grown in outdoor raceway ponds
Growth in most microalgal mass cultivation systems is light-limited, particularly in raceway ponds (RWP) where the light path is higher. Artificial lighting can be a promising solution to diminishing dark zones and enhance microalgal productivity. Therefore, our goal was to prevent the cell shift from photosynthesis to a respiration-only stage by resorting to LED illumination. Nannochloropsis oceanica cultures were accordingly grown out-doors in a preliminary small-scaleexperiment, followed by pilot-scale trials. In the former, three 3.0-m(2) RWP were set up under three distinct conditions: 1) without LEDs (control); 2) LEDs turned on during the night; and 3) LEDs turned on for 24 h. In the pilot-scale trial, one of two 28.9-m(2) pilot-scale RWPs was coupled to the best LED setup - determined in the small-scale preliminary experiment - using the same light intensity (normal mode) and half of the intensity (economy mode), with the second RWP serving as a control. In the preliminary experiment, the use of LEDs for 24 h was deemed as not helpful during daytime, before the culture reached asymptotic to 0.5 g DW L-1 - when dark zones appeared during the day due to sunlight attenuation in the 0.1 m-deep cultures. Overall, use of LEDs increased biomass growth chiefly by increasing nighttime productivities - materialized in higher chlorophyll, protein, and carbohydrate productivities in LED-lit cultures. A higher impact of LED lighting was observed under lower sunlight irradiances. A preliminary economic analysis indicates that use of LEDs in RWPs outdoors should be considered for high-value metabolites only.info:eu-repo/semantics/publishedVersio
