    Transcranial Direct Current Stimulation Studies Investigating the Role of the DLPFC in Memory and Metamemory

    Metamemory, broadly defined as knowledge about one’s own memory, is thought to consist of two separate processes, monitoring, the ability to assess the contents of your own memory, and control, altering behavior in service of memory, that work together to aide in encoding and retrieval processes. Prior work has shown that metamemory monitoring accuracy can be improved in a semantic memory task via high-definition transcranial direct current stimulation (HD-tDCS), a form of non-invasive brain stimulation, over the left dorsolateral prefrontal cortex (DLPFC). Over three experiments, the current dissertation sought to replicate and extend this work by: 1) testing the effects of HD-tDCS over the DLPFC in memory and metamemory monitoring for an episodic memory task, 2) testing of the effects of conventional tDCS over the DLPFC in memory and metamemory monitoring for both a semantic and episodic task using remotely supervised (rs) tDCS, 3) using HD-tDCS to examine the role of the DLPFC in memory and metamemory monitoring and additionally metamemory control. There were some effects of brain stimulation on memory performance, with greater cued recall when HD-tDCS was applied over the DLPFC right before cued recall compared to sham. Additionally, there were rs-tDCS induced improvements in recognition accuracy, but only in certain task orders. However, across three experiments there was no evidence that either HD or conventional rs-tDCS over the left DLPFC results in improved metamemory monitoring accuracy. A follow-up behavioral study suggested that, in one experiment, the lack of improvement in metamemory monitoring accuracy was related to changes made in the behavioral task. Turning to metamemory control, there were no effects of brain stimulation, but there was evidence that individuals use their metamemory monitoring judgments to control their memory. Although there were some interesting findings, there were mostly null effects of tDCS, suggesting a need for more systematic testing of tDCS as an experimental tool, specifically regarding the parameters in which tDCS does and does not produce changes in cognition

    Differential effects of remotely supervised transcranial direct current stimulation on recognition memory depending on task order

    BackgroundPrior work has shown positive effects of High Definition transcranial direct current stimulation (HD-tDCS) over the dorsolateral prefrontal cortex (DLPFC) on semantic memory performance and metamemory monitoring accuracy. However, HD-tDCS requires setup by a trained researcher, which is not always feasible. Few studies have used remotely supervised (rs) tDCS in healthy populations, and remote supervision has strong practical benefits.Objective/hypothesisThe goal of the current study was to test if previously shown effects of HD-tDCS over the left DLPFC on semantic memory performance and metamemory monitoring accuracy extended to conventional rs-tDCS, which is less focal than HD-tDCS, and to episodic memory and metamemory tasks.Materials and methodsA total of 36 healthy participants completed 6 weeks of rs-tDCS sessions, with either active left or right anodal DLPFC stimulation, or sham. Participants completed semantic and episodic memory and metamemory tasks, which each lasted for three consecutive sessions, and session order was counterbalanced across participants.ResultsOverall, there were no main effects of rs-tDCS on metamemory monitoring accuracy or memory performance for either the semantic or the episodic tasks. However, there were effects of rs-tDCS that depended on the order of completing the episodic and semantic task sessions. When participants completed the semantic task sessions after the episodic task sessions, semantic recognition was greater in the left anodal DLPFC condition. In a parallel effect, when participants completed the episodic task sessions after the semantic task sessions, episodic recognition was greater in the right anodal DLPFC condition.ConclusionPrior experience with tDCS is a factor for effects of rs-tDCS on cognition. Additionally, the current experiment provides evidence for the feasibility of fully remotely supervised tDCS in healthy participants

    Validity and everyday clinical applicability of lumbar muscle fatigue assessment methods in patients with chronic non-specific low back pain: a systematic review

    Purpose: This systematic literature review aimed at examining the validity and applicability in everyday clinical rehabilitation practise of methods for the assessment of back muscle fatiguability in patients with chronic non-specific low back pain (CNSLBP). Methods: Extensive research was performed in MEDLINE, Cumulative Index of Nursing and Allied Health Literature (CINAHL), Embase, Physiotherapy Evidence Database (PEDro) and Cochrane Central Register of Controlled Trials (CENTRAL) databases from their inception to September 2014. Potentially relevant articles were also manually looked for in the reference lists of the identified publications. Studies examining lumbar muscle fatigue in people with CNSLBP were selected. Two reviewers independently selected the articles, carried out the study quality assessment and extracted the results. A modified Strengthening the Reporting of Observational Studies in Epidemiology (STROBE) scale was used to evaluate the scientific rigour of the selected works. Results: Twenty-four studies fulfilled the selection criteria and were included in the systematic review. We found conflicting data regarding the validity of methods used to examine back muscle fatigue. The Biering-Sorensen test, performed in conjunction with surface electromyography spectral analysis, turned out to be the most widely used and comparatively, the most optimal modality currently available to assess objective back muscle fatigue in daily clinical practise, even though critical limitations are discussed. Conclusions: Future research should address the identification of an advanced method for lower back fatigue assessment in patients with CNSLBP which, eventually, might provide physical therapists with an objective and reliable test usable in everyday clinical practise

    Inter-individual differences in foraging tactics of a colonial raptor : consistency, weather effects, and fitness correlates

    Background: Consistent inter-individual differences in behavioural phenotypes may entail differences in energy efficiency and expenditure, with different fitness payoffs. In colonial-breeding species, inter-individual differences in foraging behaviour may evolve to reduce resource use overlap among conspecifics exploiting shared foraging areas. Furthermore, individual differences in foraging behaviour may covary with individual characteristics, such as sex or physiological conditions. Methods: We investigated individual differences in foraging tactics of a colonial raptor, the lesser kestrel (Falco naumanni). We tracked foraging trips of breeding individuals using miniaturized biologgers. We classified behaviours from GPS data and identified tactics at the foraging trip level by cluster analysis. We then estimated energy expenditure associated to each tactic from tri-axial accelerometer data. Results: We obtained 489 foraging trips by 36 individuals. Two clusters of trips were identified, one (SF) characterized by more static foraging behaviour and the other (DF) by more dynamic foraging behaviour, with a higher proportion of flying activity and a higher energy expenditure compared to SF. Lesser kestrels showed consistent inter-individual differences in foraging tactics across weather condition gradients, favouring DF trips as solar radiation and crosswind intensity increased. DF trips were more frequent during the nestling-rearing than during the egg incubation stage. Nestlings whose tracked parent was more prone to perform DF trips experienced higher daily mass increase, irrespective of nestling feeding rates. Conclusions: Our study provided evidence that breeding lesser kestrels flexibly adopted different foraging tactics according to contingent weather landscapes, with birds showing consistent inter-individual differences in the tendency to adopt a given tactic. The positive correlation between the tendency to perform more energy-demanding DF trips and nestling growth suggests that individual differences in foraging behaviour may play a role in maintaining key life-history trade-offs between reproduction and self-maintenance

    Mode-coupling theory predictions for a limited valency attractive square-well model

    Recently we have studied, using numerical simulations, a limited valency model, i.e. an attractive square well model with a constraint on the maximum number of bonded neighbors. Studying a large region of temperatures TT and packing fractions ϕ\phi, we have estimated the location of the liquid-gas phase separation spinodal and the loci of dynamic arrest, where the system is trapped in a disordered non-ergodic state. Two distinct arrest lines for the system are present in the system: a {\it (repulsive) glass} line at high packing fraction, and a {\it gel} line at low ϕ\phi and TT. The former is essentially vertical (ϕ\phi-controlled), while the latter is rather horizontal (TT-controlled) in the (ϕ−T)(\phi-T) plane. We here complement the molecular dynamics results with mode coupling theory calculations, using the numerical structure factors as input. We find that the theory predicts a repulsive glass line -- in satisfactory agreement with the simulation results -- and an attractive glass line which appears to be unrelated to the gel line.Comment: 12 pages, 6 figures.     Fluid-fluid demixing transitions in colloid--polyelectrolyte star mixtures

    We derive effective interaction potentials between hard, spherical colloidal particles and star-branched polyelectrolytes of various functionalities and smaller size than the colloids. The effective interactions are based on a Derjaguin-like approximation, which is based on previously derived potentials acting between polyelectrolyte stars and planar walls. On the basis of these interactions we subsequently calculate the demixing binodals of the binary colloid--polyelectrolyte star mixture, employing standard tools from liquid-state theory. We find that the mixture is indeed unstable at moderately high overall concentrations. The system becomes more unstable with respect to demixing as the star functionality and the size ratio grow.Comment: 24 pages, 9 figures, submitted to Journal of Physics: Condensed Matte

    Theranostics in Boron neutron capture therapy

    Boron neutron capture therapy (BNCT) has the potential to specifically destroy tumor cells without damaging the tissues infiltrated by the tumor. BNCT is a binary treatment method based on the combination of two agents that have no effect when applied individually:10B and thermal neutrons. Exclusively, the combination of both produces an effect, whose extent depends on the amount of10B in the tumor but also on the organs at risk. It is not yet possible to determine the10B concentration in a specific tissue using non-invasive methods. At present, it is only possible to measure the10B concentration in blood and to estimate the boron concentration in tissues based on the assumption that there is a fixed uptake of10B from the blood into tissues. On this imprecise assumption, BNCT can hardly be developed further. A therapeutic approach, combining the boron carrier for therapeutic purposes with an imaging tool, might allow us to determine the10B concentration in a specific tissue using a non-invasive method. This review provides an overview of the current clinical protocols and preclinical experiments and results on how innovative drug development for boron delivery systems can also incorporate concurrent imaging. The last section focuses on the importance of proteomics for further optimization of BNCT, a highly precise and personalized therapeutic approach

    Predictive Criteria to Study the Pathogenesis of Malaria-Associated ALI/ARDS in Mice

    Malaria-associated acute lung injury/acute respiratory distress syndrome (ALI/ARDS) often results in morbidity and mortality. Murine models to study malaria-associated ALI/ARDS have been described; we still lack a method of distinguishing which mice will develop ALI/ARDS before death. This work aimed to characterize malaria-associated ALI/ARDS in a murine model and to demonstrate the first method to predict whether mice are suffering from ALI/ARDS before death. DBA/2 mice infected with Plasmodium berghei ANKA developing ALI/ARDS or hyperparasitemia (HP) were compared using histopathology, PaO2 measurement, pulmonary X-ray, breathing capacity, lung permeability, and serum vascular endothelial growth factor (VEGF) levels according to either the day of death or the suggested predictive criteria. We proposed a model to predict malaria-associated ALI/ARDS using breathing patterns (enhanced pause and frequency respiration) and parasitemia as predictive criteria from mice whose cause of death was known to retrospectively diagnose the sacrificed mice as likely to die of ALI/ARDS as early as 7 days after infection. Using this method, we showed increased VEGF levels and increased lung permeability in mice predicted to die of ALI/ARDS.     Interactions between global climate change strategies and local air pollution: lessons learnt from the expansion of the power sector in Brazil

    This study examines the interactions between local air pollutants and greenhouse gas emissions to assess potential synergies and trade-offs between local environmental pollution and climate policies, using the power generation sector in Brazil under different carbon scenarios up to 2050 as a case study. To this end, an integrated approach was developed, combining energy scenarios under different carbon mitigation targets and a local air pollution assessment model, tailored to the context of the Brazilian power sector. Results reveal that there are deep interactions between climate change mitigation and local air pollution abatement strategies. Increasing the diffusion of low-carbon technologies results in both mitigation of climate change and lower terrestrial acidification potential impacts, due to the rapid phase-out of fossil fuel power technologies. However, local air pollution indicators for particulate matter formation and human toxicity may rise in response to greenhouse gas emission mitigation constraints, indicating the existence of potential trade-offs. Some of these trade-offs can be offset by introducing available end-of-pipe pollution control measures reinforced by dedicated air quality policies

    Altered umbilical cord blood nutrient levels, placental cell turnover and transporter expression in human term pregnancies conceived by intracytoplasmic sperm injection (Icsi)

    Assisted reproductive technologies (ART) may increase risk for abnormal placental development, preterm delivery and low birthweight. We investigated placental morphology, transporter expression and paired maternal/umbilical fasting blood nutrient levels in human term pregnancies conceived naturally (n = 10) or by intracytoplasmic sperm injection (ICSI; n = 11). Maternal and umbilical vein blood from singleton term (>37 weeks) C-section pregnancies were assessed for levels of free amino acids, glucose, free fatty acids (FFA), cholesterol, high density lipoprotein (HDL), low density lipoprotein (LDL), very low-density lipoprotein (VLDL) and triglycerides. We quantified placental expression of GLUT1 (glucose), SNAT2 (amino acids), P-glycoprotein (P-gp) and breast cancer resistance protein (BCRP) (drug) transporters, and placental morphology and pathology. Following ICSI, placental SNAT2 protein expression was downregulated and umbilical cord blood levels of citrulline were increased, while FFA levels were decreased at term (p < 0.05). Placental proliferation and apoptotic rates were increased in ICSI placentae (p < 0.05). No changes in maternal blood nutrient levels, placental GLUT1, P-gp and BCRP expression, or placental histopathology were observed. In term pregnancies, ICSI impairs placental SNAT2 transporter expression and cell turnover, and alters umbilical vein levels of specific nutrients without changing placental morphology. These may represent mechanisms through which ICSI impacts pregnancy outcomes and programs disease risk trajectories in offspring across the life course
