An Introduction to Harmonic Manifolds and the Lichnerowicz Conjecture
The title is self-explanatory. We aim to give an easy to read and
self-contained introduction to the field of harmonic manifolds. Only basic
knowledge of Riemannian geometry is required. After we gave the definition of
harmonicity and derived some properties, we concentrate on Z. I. Szab\'o's
proof of Lichnerowicz's conjecture in the class of compact simply connected
manifolds
Serotonin-mediated tuning of human helper T cell responsiveness to the chemokine CXCL12
In addition to its role as neurotransmitter, serotonin (5-HT) is an important modulator of inflammation and immunity. Here, we report novel findings suggesting a 5-HT involvement in T cell migration. In particular, we show that 5-HT tunes the responsiveness of human T lymphocytes to the broadly expressed chemokine CXCL12 in transwell migration assays. By real-time PCR, western blot analysis and electrophysiological patch clamp experiments, we demonstrate that the type 3 5-HT receptor (5-HT 3) is functionally expressed in human primary T cells. In addition, specific 5-HT 3 receptor agonists selectively decrease T cell migration towards gradients of CXCL12 but not of inflammatory chemokines, such as CCL2 and CCL5. In transmigration experiments, 5-HT 3 receptor stimulation reverts the inhibitory effect of endothelial-bound CXCL12 on T cell migration. Our data suggest that the reduced T cell responsiveness to CXCL12 induced by 5-HT may occur to facilitate T cell extravasation and migration into inflamed tissues
QUASI-CONFORMAL CURVATURE TENSOR OF GENERALIZED SASAKIAN-SPACE-FORMS
The present paper deals the study of generalised Sasakian-space-forms with the conditions Cq(Ο,X).S = 0, Cq(Ο,X).R = 0 and Cq(Ο,X).Cq = 0, where R, S and Cq denote Riemannian curvature tensor, Ricci tensor and quasi-conformal curvature tensor of the space-form, respectively and at last, we have given some examples to improve our results
Inclusion complexation of the anticancer drug pomalidomide with cyclodextrins: fast dissolution and improved solubility
Pomalidomide (POM), a potent anticancer thalidomide analogue was characterized in terms of cyclodextrin complexation to improve its aqueous solubility and maintain its anti-angiogenic activity. The most promising cyclodextrin derivatives were selected by phase-solubility studies. From the investigated nine cyclodextrins â differing in cavity size, nature of substituents, degree of substitution and charge â the highest solubility increase was observed with sulfobutylether-ÎČ-cyclodextrin (SBE-ÎČ-CD). The inclusion complexation between POM and SBE-ÎČ-CD was further characterized with a wide variety of state-of-the-art analytical techniques, such as nuclear magnetic resonance spectroscopy (NMR), infrared spectroscopy (IR), circular dichroism spectroscopy, fluorescence spectroscopy as well as X-ray powder diffraction method (XRD). Job plot titration by NMR and the AL-type phase-solubility diagram indicated 1:1 stoichiometry in a liquid state. Complementary analytical methods were employed for the determination of the stability constant of the complex; the advantages and disadvantages of the different approaches are also discussed. Inclusion complex formation was also assessed by molecular modelling study. Solid state complexation in a 1:1 M ratio was carried out by lyophilization and investigated by IR and XRD. The complex exhibited fast-dissolution with immediate release of POM, when compared to the pure drug at acidic and neutral pH. Kinetic analysis of POM release from lyophilized complex shows that Korsmeyer-Peppas and Weibull model described the best the dissolution kinetics. The cytotoxicity of the complex was tested against the LP-1 human myeloma cell line which revealed that supramolecular interactions did not significantly affect the anti-cancer activity of the drug. Overall, our results suggest that the inclusion complexation of POM with SBE-ÎČ-CD could be a promising approach for developing more effective POM formulations with increased solubility
Prevalence of Coxiella burnetii in cows' and ewes' bulk tank milk samples from selected dairy farms of Central Italy
The prevalence of Coxiella burnetii, the causative agent of Q fever, in cattle and sheep raw milk farms was determined in Central Italy, an area in which dairy production plays an important economic role. Milk samples (n. 189), collected from 66 dairy farms in 2012â2013, were tested by a commercial real-time PCR assay. Seventeen dairy farms had at least one positive milk sample; percent positive was higher for cattle (50%) than sheep (21%) farms. Concerning milk, 15% of samples tested overall gave a positive result, with the highest percentage of positivity observed for bovine milk compared with sheep milk (41% and 12%, respectively). In the only bovine farm repeatedly sampled during the study, C. burnetii contamination was persistently found for almost a year. The prevalence calculated for the sheep farms showed a discontinuous trend with a maximum peak in February. The results obtained underline the widespread presence of the pathogen in the considered geographical area, giving new epidemiological information. Since the milk route of elimination is a potential vehicle of infection for farmers, veterinarians and for dairy stakeholders in general, BTM screening by real-time PCR can be applied as a useful surveillance tool both for the identification of infected flocks and implementation of control programmes
