    In the recent years, girder bridge cranes are replaced by double-beam overhead cranes with beams of rectangular cross-section. In addition, new materials are used for their fabrication, characterized by different values of allowable loads and deformations. In the paper, two overhead cranes from JSC "Vilniaus kranai" are considered. A mathematical model is proposed that enables assessment of the impact of the hydraulic damper built in the cargo suspension system upon the dynamic features of the crane in the beginning of the lifting process. It was determined that in such a way the period of vibration damping is reduced. However, the impact of the damper upon the dynamics of the metal structure of the crane is limited because of high mobility of the rope as compared to the mobility of the total structure

    The article substantiates the necessity of development of municipal formations in the Russian Federation socio-economic development, in accordance with Federal law No. 172-FZ of 28.06.2014 «On strategic planning in the Russian Federation». Under this law, local self-government bodies independently decide on development Strategies for socio-economic development, taking into account the peculiarities of natural-climatic conditions, selection of priority areas, growth points, ensuring the strategic development of the municipality. The object of the study adopted Promyshlennovskiy municipal district of the Kemerovo region with the structure of the economy in agricultural sector, with a population of under fifty thousand people, there are promising natural reserves of coal. The authors consider the status and level of development of the district. The study was conducted in order to develop a «Strategy for socio-economic development of Promyshlennovskiy municipal area until 2035». This paper outlines the strategic directions of development of the district, a forecast of the main results obtained through the implementation of 16 programs, 34 projects, adopted a Strategy that is formed based on the presidential Decree 204 of7.05.2018 «On the national goals and strategic objectives development of the Russian Federation for the period up to 2024»and Ordinance No. 1288 «About the organization o fproject activities the government of the Russian Federation»dated 31.10.2018 G.В статье обоснована потребность разработки муниципальными образованиями в Российской Федерации Стратегий социально-экономического развития, в соответствии с ФЗ РФ № 172-ФЗ от 28.06.2014 г. «О стратегическом планировании в Российской Федерации». По этому закону органы местного самоуправления самостоятельно принимают решение о разработке Стратегий социально-экономического развития, с учетом особенностей природно-климатических условий, выбора приоритетных направлений, точек роста, обеспечивающих стратегическое развитие муниципального образования. Объектом исследования принят Промышленновский муниципальный район Кемеровской области со структурой экономики сельскохозяйственной направленности, с численностью населения до пятидесяти тысяч человек, имеются перспективные природные запасы угля. Авторами рассмотрено состояние и уровень развития района. Исследование проводилось с целью сформировать «Стратегию социально-экономического развития Промышленновского муниципального района до 2035 года». В этом документе рассмотрены стратегические направления развития района, прогноз основных результатов, получаемых за счет реализации 16 программ, 34 проектов, принятых к Стратегии, которые сформированы с учетом Указа Президента 204 от 7.05.2018 г. «О национальных целях и стратегических задачах развития Российской Федерации на период до 2024 года» и Постановления №1288 «Об организации проектной деятельности в Правительстве Российской Федерации» от 31.10.2018 г

    The algorithm of classification of municipal unions (MU is based on definition of a confidential interval of average long-term tick-borne encephalitis virus (TBE) sickness rate, as a quantitative integrated indicator of an epidemiological situation at the territory. It is proved that a decade is a sufficient period to reveal the patters that can be transferred to general totality and facilitate realization of MU classification. Distribution by MU groups is a steady characteristic of TBE morbidity at MU territories by decades in 95 % a confidential interval

    Our knowledge on tissue- and disease-specific functions of human genes is rather limited and highly context-specific. Here, we have developed a method for the comparison of mRNA expression levels of most human genes across 9,783 Affymetrix gene expression array experiments representing 43 normal human tissue types, 68 cancer types, and 64 other diseases. This database of gene expression patterns in normal human tissues and pathological conditions covers 113 million datapoints and is available from the GeneSapiens website

    Layered material platform for surface plasmon resonance biosensing

    Plasmonic biosensing has emerged as the most sensitive label-free technique to detect various molecular species in solutions and has already proved crucial in drug discovery, food safety and studies of bio-reactions. This technique relies on surface plasmon resonances in ~50 nm metallic films and the possibility to functionalize the surface of the metal in order to achieve selectivity. At the same time, most metals corrode in bio-solutions, which reduces the quality factor and darkness of plasmonic resonances and thus the sensitivity. Furthermore, functionalization itself might have a detrimental effect on the quality of the surface, also reducing sensitivity. Here we demonstrate that the use of graphene and other layered materials for passivation and functionalization broadens the range of metals which can be used for plasmonic biosensing and increases the sensitivity by 3-4 orders of magnitude, as it guarantees stability of a metal in liquid and preserves the plasmonic resonances under biofunctionalization. We use this approach to detect low molecular weight HT-2 toxins (crucial for food safety), achieving phase sensitivity~0.5 fg/mL, three orders of magnitude higher than previously reported. This proves that layered materials provide a new platform for surface plasmon resonance biosensing, paving the way for compact biosensors for point of care testing

    Cytotoxic activity of cell free fraction (CFF) of the broncho-alveolar lavage (BAL) according to its ability to lyse the cells from the monocytoid human blast cell line J96 was assessed 8 asthmatics and 4 normals. Immunoregulatory properties of CFF were assessed according to its influence on spontaneous and konkanavaline A induced proliferation of peripheric human blood lymphocytes, cultivated in the presence of 3H-timidine. As a rule during «normal» stage CFF has no cytotoxic and mitogenic activity, and very slightly inhibits mitogenic response of lymphocytes stimulated by konkavaline in vitro. During the state of bronchial asthma CF of BAL presents high cytotoxic activity, which is not followed by appearance or rise in level of tumor necrotizing factor a. Immunoregulatory properties of BAL CFF are changing during asthma exacerbations: There appear some factors capable of inhibiting the spontaneous proliferation of peripheric human blood lymphocytes and strengthen their response to mitogens in comparison with the «normal» BAL CFF.У 8 больных бронхиальной астмой и 4 лиц без легочной патологии определяли цитотоксическую активность бесклеточной фракции (БФ) бронхоальвеолярных смывов (БАС) по её способности лизировать клетки моноцитоидной линии опухолевых клеток человека J 96. Иммунорегуляторные свойства БФ оценивали по её влиянию на спонтанную и индуцированную конканавалином А пролиферацию лимфоцитов периферической крови человека, культивируемых в присутствии 3Н-тимидина. В «норме» БФ смыва, как правило, не обладает цитотоксической и митогенной активностью, а также незначительно ингибирует индуцируемый конканавалином митогенной ответ лимфоцитов in vitro. При бронхиальной астме БФ БАС обладает высокой цитотоксической активностью, которая не сопровождается появлением или повышением концентрации фактора некроза опухоли а. Иммунорегуляторные свойства БФ БАС во время обострения бронхиальной астмы изменяются: в ней появляются факторы, способные подавлять спонтанную пролиферацию лимфоцитов периферической крови человека и усиливать их ответ на митогены по сравнению с БФ БАС в «норме»

    Heterogeneity and clinical significance of ETV1 translocations in human prostate cancer

    A fluorescence in situ hybridisation (FISH) assay has been used to screen for ETV1 gene rearrangements in a cohort of 429 prostate cancers from patients who had been diagnosed by trans-urethral resection of the prostate. The presence of ETV1 gene alterations (found in 23 cases, 5.4%) was correlated with higher Gleason Score (P=0.001), PSA level at diagnosis (P=<0.0001) and clinical stage (P=0.017) but was not linked to poorer survival. We found that the six previously characterised translocation partners of ETV1 only accounted for 34% of ETV1 re-arrangements (eight out of 23) in this series, with fusion to the androgen-repressed gene C15orf21 representing the commonest event (four out of 23). In 5′-RACE experiments on RNA extracted from formalin-fixed tissue we identified the androgen-upregulated gene ACSL3 as a new 5′-translocation partner of ETV1. These studies report a novel fusion partner for ETV1 and highlight the considerable heterogeneity of ETV1 gene rearrangements in human prostate cancer

    Investigation of the key chemical structures involved in the anticancer activity of disulfiram in A549 non-small cell lung cancer cell line

    © 2018 The Author(s). Background: Disulfiram (DS), an antialcoholism medicine, demonstrated strong anticancer activity in the laboratory but did not show promising results in clinical trials. The anticancer activity of DS is copper dependent. The reaction of DS and copper generates reactive oxygen species (ROS). After oral administration in the clinic, DS is enriched and quickly metabolised in the liver. The associated change of chemical structure may make the metabolites of DS lose its copper-chelating ability and disable their anticancer activity. The anticancer chemical structure of DS is still largely unknown. Elucidation of the relationship between the key chemical structure of DS and its anticancer activity will enable us to modify DS and speed its translation into cancer therapeutics. Methods: The cytotoxicity, extracellular ROS activity, apoptotic effect of DS, DDC and their analogues on cancer cells and cancer stem cells were examined in vitro by MTT assay, western blot, extracellular ROS assay and sphere-reforming assay. Results: Intact thiol groups are essential for the in vitro cytotoxicity of DS. S-methylated diethyldithiocarbamate (S-Me-DDC), one of the major metabolites of DS in liver, completely lost its in vitro anticancer activity. In vitro cytotoxicity of DS was also abolished when its thiuram structure was destroyed. In contrast, modification of the ethyl groups in DS had no significant influence on its anticancer activity. Conclusions: The thiol groups and thiuram structure are indispensable for the anticancer activity of DS. The liver enrichment and metabolism may be the major obstruction for application of DS in cancer treatment. A delivery system to protect the thiol groups and development of novel soluble copper-DDC compound may pave the path for translation of DS into cancer therapeutics.This work was supported by grant from British Lung Foundation (RG14–8) and Innovate UK (104022).Published versio
