    Contributions and perspectives of Indigenous Peoples to the study of mercury in the Arctic

    Arctic Indigenous Peoples are among the most exposed humans when it comes to foodborne mercury (Hg). In response, Hg monitoring and research have been on-going in the circumpolar Arctic since about 1991; this work has been mainly possible through the involvement of Arctic Indigenous Peoples. The present overview was initially conducted in the context of a broader assessment of Hg research organized by the Arctic Monitoring and Assessment Programme. This article provides examples of Indigenous Peoples' contributions to Hg monitoring and research in the Arctic, and discusses approaches that could be used, and improved upon, when carrying out future activities. Over 40 mercury projects conducted with/by Indigenous Peoples are identified for different circumpolar regions including the U.S., Canada, Greenland, Sweden, Finland, and Russia as well as instances where Indigenous Knowledge contributed to the understanding of Hg contamination in the Arctic. Perspectives and visions of future Hg research as well as recommendations are presented. The establishment of collaborative processes and partnership/co-production approaches with scientists and Indigenous Peoples, using good communication practices and transparency in research activities, are key to the success of research and monitoring activities in the Arctic. Sustainable funding for community-driven monitoring and research programs in Arctic countries would be beneficial and assist in developing more research/ monitoring capacity and would promote a more holistic approach to understanding Hg in the Arctic. These activities should be well connected to circumpolar/international initiatives to ensure broader availability of the information and uptake in policy development

    Brominated flame retardants (BFRs) represent a large group of chemicals used in a variety of household and commercial products to prevent fire propagation. The environmental persistence and toxicity of some of the most widely used BFRs has resulted in a progressive ban worldwide and the development of novel BFRs for which the knowledge on environmental health impacts remains limited. The objectives of this study were to evaluate the effects of two emerging BFRs, 1,2-bis(2,4,6-tribromophenoxy)ethane (BTBPE) and 2-ethylhexyl-2,3,4,5-tetrabromobenzoate (EH-TBB), in diet exposed juvenile rainbow trout (Oncorhynchus mykiss). Both compounds were detected in fish carcasses at 76% and 2% of the daily dosage of BTBPE and EH-TBB, respectively, indicating accumulation of BTBPE and by contrast extensive depuration/metabolism of EH-TBB. Liver gene transcription analysis using RNA-sequencing indicated that the chronic 28-d dietary exposure of trout to EH-TBB down-regulated one single gene related to endocrine-mediated processes, whereas BTBPE impacted the transcription of 33 genes, including genes involved in the immune response, reproduction, and oxidative stress. Additional analysis using qRT-PCR after 48-h and 28-d of exposure confirmed the impact of BTBPE on immune related genes in the liver (apolipoprotein A-I, lysozyme) and the head-kidney (complement c3-4).     A Multi-Matrix Metabolomic Approach in Ringed Seals and Beluga Whales to Evaluate Contaminant and Climate-Related Stressors

    As a high trophic-level species, ringed seals (Pusa hispida) and beluga whales (Delphinapterus leucas) are particularly vulnerable to elevated concentrations of biomagnifying contaminants, such as polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs) and mercury (Hg). These species also face climate-change-related impacts which are leading to alterations in their diet and associated contaminant exposure. The metabolomic profile of marine mammal tissues and how it changes to environmental stressors is poorly understood. This study characterizes the profiles of 235 metabolites across plasma, liver, and inner and outer blubber in adult ringed seals and beluga whales and assesses how these profiles change as a consequence of contaminants and dietary changes. In both species, inner and outer blubber were characterized by a greater proportion of lipid classes, whereas the dominant metabolites in liver and plasma were amino acids, carbohydrates, biogenic amines and lysophosphatidylcholines. Several metabolite profiles in ringed seal plasma correlated with δ13C, while metabolite profiles in blubber were affected by hexabromobenzene in ringed seals and PBDEs and Hg in belugas. This study provides insight into inter-matrix similarities and differences across tissues and suggests that plasma and liver are more suitable for studying changes in diet, whereas liver and blubber are more suitable for studying the impacts of contaminants

    Variation in body size of ringed seals (Pusa hispida hispida) across the circumpolar Arctic: evidence of morphs, ecotypes or simply extreme plasticity?

    The ringed seal is a small phocid seal that has a northern circumpolar distribution. It has long been recognized that body size is variable in ringed seals, and it has been suggested that ecotypes that differ in size exist. This study explores patterns of body size (length and girth) and age-at-maturity across most of the Arctic subspecies’ range using morphometric data from 35 sites. Asymptotic lengths varied from 113 to 151 cm, with sites falling into five distinct size clusters (for each sex). Age-at-maturity ranged from 3.1 to 7.4 years, with sites that had early ages of sexual maturity generally having small length-at-maturity and small final body length. The sexes differed in length at some sites, but not in a consistent pattern of dimorphism. The largest ringed seals occurred in western Greenland and eastern Canada, and the smallest occurred in Alaska and the White Sea. Latitudinal trends occurred only within sites in the eastern Canadian Arctic. Girth (with length and season accounted for) was also highly variable but showed no notable spatial pattern; males tended to be more rotund than females. Genetic studies are needed, starting with the “giants” at Kangia (Greenland) and in northern Canada to determine whether they are genetically distinct ecotypes. Additional research is also needed to understand the ecological linkages that drive the significant regional size differences in ringed seals that were confirmed in this study, and also to understand their implications with respect to potential adaptation to climate change

    Occurrence of substituted diphenylamine antioxidants and benzotriazole UV stabilizers in Arctic seabirds and seals

    Substituted diphenylamine antioxidants (SDPAs) and benzotriazole UV stabilizers (BZT-UVs) are contaminants of emerging environmental concern. However, little is known about the occurrence of these contaminants in the Arctic. In this study, we investigated the levels of 11 SDPAs and 6 BZT-UVs in livers and eggs of two seabird species, the black-legged kittiwake (Rissa tridactyla) and northern fulmar (Fulmarus glacialis), as well as the liver of ringed seals (Pusa hispida) from Canadian high- and sub-Arctic sites. The concentrations of ΣSDPAs in seabird livers (median 336 pg g−1, wet weight (ww)) were significantly higher than the eggs (median 24 pg g−1, ww) and the seal livers (median 38 pg g−1, ww), suggesting liver was a primary tissue of SDPA accumulation in seabirds and that seabirds were at greater risk of exposure to SDPAs than marine mammals in the Arctic. The predominant SDPA was monostyryl octyl-diphenylamine and this compound was detected in every seabird and seal sample, indicating the widespread distribution of this contaminant in Arctic food webs. Unlike SDPAs, the detection rate and concentrations of BZT-UVs in seals were higher than in seabirds. The compound 2-(2H-benzotriazol-2-yl)-4-(1,1,3,3-tetramethylbutyl)phenol (UV329) or its isomer 2-(2H-benzotriazol-2-yl)-4-(tert-butyl)-6-(sec-butyl) phenol (UV350) was the predominant BZT-UVs in seals, with the concentrations of ΣBZT-UVs between <method quantification limits and 1.66 × 104 pg g−1 (ww) (median: 2.36 × 103 pg g−1, ww). This is the first report of the different distribution patterns of SDPAs and BZT-UVs in wildlife from Canadian Arctic sites
