    Effects of coal contamination on early life history processes of a reef-building coral, Acropora tenuis

    Successful reproduction and larval dispersal are important for the persistence ofmarine invertebrate populations, and these early life history processes can be sensitive to marine pollution. Coal is emerging as a contaminant of interest due to the proximity of ports and shipping lanes to coral reefs. To assess the potential hazard of this contaminant, gametes, newly developedembryos, larvae and juveniles of the coral Acropora tenuis were exposed to a range of coal leachate, suspended coal, and coal smothering treatments. Fertilisation was the most sensitive reproductive process tested. Embryo survivorship decreased with increasing suspended coal concentrations and exposure duration, effects on larval settlement varied between treatments, while effects on juvenile survivorship were minimal. Leachate exposures had negligible effects on fertilisation and larval settlement. These results indicate that coral recruitment could be affected by spills that produce plumes of suspended coal particles which interact with gametes and embryos soon after spawning

    Spatial distribution of beef cattle on a New Zealand hill country farm: monitoring the use of streams and wet areas

    Grazing livestock are an important source of contamination of freshwater, particularly when they have direct access to streams. Cattle in particular contribute to riparian habitat deterioration through stream bank destruction and direct defecation and urination in streams. Exclusion of stock or planting of riparian areas, are the most common catchment management methods used to protect waterways. Given the relatively low returns from beef and sheep farming, both of these strategies are very expensive and often logistically prohibitive in steep hill county landscapes. Despite this, policy trends indicate that fencing of streams in agricultural catchments may become mandatory in the future. It is important that we understand how much time cattle spend in and around hill country streams and wet areas (wetlands and hill side seeps), in order to quantify the likely environmental benefits from such policies. The current study examined cattle movement data obtained using Global Positioning System (GPS) collars from experiments undertaken at Massey University’s hill country research farm, Tuapaka, near Palmerston North, to investigate the amount of time cattle spent in and around streams and wet areas. Animal movement data were collected over seven grazing events, in three winter periods (2012, 2013 and 2015). Permanent streams and wet areas were identified using a digital elevation model derived from 1m LiDAR data, aerial RGB images and RTK measurements. Cattle spent 3.3 – 6% (48 – 86 min/day) of their day in streams and wet areas consistently across the 7 data collections. Cattle spent more time in streams and wet areas during the afternoon. There are differences in the median amount of time individual animals spend in non-risk areas. Further research is necessary to evaluate how we can influence the amount of time cattle spend in riparian areas on hill country and how stream bank behaviour varies at different times of the year

    The Need and Potential of Biosensors to Detect Dioxins and Dioxin-Like Polychlorinated Biphenyls along the Milk, Eggs and Meat Food Chain

    Dioxins and dioxin-like polychlorinated biphenyls (DL-PCBs) are hazardous toxic, ubiquitous and persistent chemical compounds, which can enter the food chain and accumulate up to higher trophic levels. Their determination requires sophisticated methods, expensive facilities and instruments, well-trained personnel and expensive chemical reagents. Ideally, real-time monitoring using rapid detection methods should be applied to detect possible contamination along the food chain in order to prevent human exposure. Sensor technology may be promising in this respect. This review gives the state of the art for detecting possible contamination with dioxins and DL-PCBs along the food chain of animal-source foods. The main detection methods applied (i.e., high resolution gas-chromatography combined with high resolution mass-spectrometry (HRGC/HRMS) and the chemical activated luciferase gene expression method (CALUX bioassay)), each have their limitations. Biosensors for detecting dioxins and related compounds, although still under development, show potential to overcome these limitations. Immunosensors and biomimetic-based biosensors potentially offer increased selectivity and sensitivity for dioxin and DL-PCB detection, while whole cell-based biosensors present interpretable biological results. The main shortcoming of current biosensors, however, is their detection level: this may be insufficient as limits for dioxins and DL-PCBs for food and feedstuffs are in pg per gram level. In addition, these contaminants are normally present in fat, a difficult matrix for biosensor detection. Therefore, simple and efficient extraction and clean-up procedures are required which may enable biosensors to detect dioxins and DL-PCBs contamination along the food chain

    High-affinity recombinant phage antibodies to the pan-carcinoma marker epithelial glycoprotein-2 for tumour targeting.

    The tumour-associated antigen epithelial glycoprotein-2 (EGP-2) is a promising target for detection and treatment of a variety of human carcinomas. Antibodies to this antigen have been successfully used in patients for imaging of small-cell lung cancer and for adjuvant treatment of minimal residual disease of colon cancer. We describe here the isolation and complete characterization of high-affinity single-chain variable fragments (scFv) to the EGP-2 antigen. First, the binding kinetics of four murine whole antibodies directed to EGP-2 (17-1A, 323/A3, MOC-31 and MOC-161) were determined using surface plasmon resonance (SPR). The MOC-31 antibody has the lowest apparent off-rate, followed by MOC-161 and 323/A3. The V-genes of the two MOC hybridomas were cloned as scFv in a phage display vector and antigen-binding phage were selected by panning on recombinant antigen. The scFvs compete with the original hybridoma antibodies for binding to antigen and specifically bind to human carcinomas in immunohistochemistry. MOC-31 scFv has an off-rate which is better than those of the bivalent 17-1A and 323/A3 whole antibodies, providing it with an essential characteristic for tumour retention in vivo. The availability of these high-affinity anti-EGP-2 antibody fragments and of their encoding V-genes creates a variety of possibilities for their future use as tumour-targeting vehicles

    A data da semeadura é um dos fatores que mais afetam produtividade de milho de sequeiro. Utilizou-se o modelo Ceres-Maize para simular a produtividade de milho semedo em diferentes datas em Sete Lagoas, MG. Avaliou-se uma metodologia baseada em dados estatísticos, para auxiliar no estabelecimento do período adequado de semeadura de milho. Cabe ao tomador de decisão definir o nível de quebra de produtividade que ele estaria disposto a aceitar. Tolerando-se uma redução de 10% na produtividade em relação ao valor médio máximo, o período de semeadura na região deve ser de 26 de setembro a sete de novembro, embora o risco para a semeadura de setembro seja alto devido às instabilidades climáticas. Mesmo com semeaduras realizadas neste período, há uma probabilidade de cerca de 33% de que ocorram quebras de safra de até 10% em decorrência de déficit hídrico e de outros fatores como temperaturas noturnas altas

    O uso da irrigação se constitui numa alternativa para alavancar a produção de milho na região central de Minas Gerais, que é carente de grãos. Modelos de simulação de culturas acoplados a ferramentas de apoio à decisão podem ser empregados para auxiliar na avaliação de estratégias de semeadura que possibilitem a obtenção de produtividades elevadas com o uso racional dos recurso hídricos. Utilizaram-se dados de produtividade de grãos de milho simulados e uma metodologia estatística para desenvolver um critério de decisão para datas de semeadura em um sistema de produção de milho irrigado. Tolerando-se uma redução de 10% na produtividade, em relação ao valor médio máximo, o período de semeadura na região deve ser de 23 de janeiro a 27 de março

    The Density of States in High-Tc Superconductors Vortices

    We calculated the electronic structure of a vortex in a pseudogapped superconductor within a model featuring strong correlations. With increasing strength of the correlations, the BCS core states are suppressed and the spectra in and outside the core become similar. If the correlations are short-range, we find new core states in agreement with the observations in YBaCuO and BiSrCaCuO. Our results point to a common phenomenology for these two systems and indicate that normal-state correlations survive below Tc without taking part in the overall phase coherence.Comment: REVTeX 4, 5 pages, 2 EPS figures.     Responsive glyco-poly(2-oxazoline)s: synthesis, cloud point tuning, and lectin binding

    A new sugar-substituted 2-oxazoline monomer was prepared using the copper-catalyzed alkyne-azide cycloaddition (CuAAC) reaction. Its copolymerization with 2-ethyl-2-oxazoline as well as 2-(dec-9-enyl)-2-oxazoline, yielding well-defined copolymers with the possibility to tune the properties by thiol-ene "click" reactions, is described. Extensive solubility studies on the corresponding glycocopolymers demonstrated that the lower critical solution temperature behavior and pH-responsiveness of these copolymers can be adjusted in water and phosphate-buffered saline (PBS) depending on the choice of the thiol. By conjugation of 2,3,4,6-tetra-O-acetyl-1-thio-beta-D-glucopyranose and subsequent deprotection of the sugar moieties, the hydrophilicity of the copolymer could be increased significantly, allowing a cloud-point tuning in the physiological range.     Environmental drivers of distribution and reef development of the Mediterranean coral Cladocora caespitosa

    Cladocora caespitosa is the only Mediterranean scleractinian similar to tropical reef-building corals. While this species is part of the recent fossil history of the Mediterranean Sea, it is currently considered endangered due to its decline during the last decades. Environmental factors affecting the distribution and persistence of extensive bank reefs of this endemic species across its whole geographic range are poorly understood. In this study, we examined the environmental response of C. caespitosa and its main types of assemblages using ecological niche modeling and ordination analysis. We also predicted other suitable areas for the occurrence of the species and assessed the conservation effectiveness of Mediterranean marine protected areas (MPAs) for this coral. We found that phosphate concentration and wave height were factors affecting both the occurrence of this versatile species and the distribution of its extensive bioconstructions in the Mediterranean Sea. A set of factors (diffuse attenuation coefficient, calcite and nitrate concentrations, mean wave height, sea surface temperature, and shape of the coast) likely act as environmental barriers preventing the species from expansion to the Atlantic Ocean and the Black Sea. Uncertainties in our large-scale statistical results and departures from previous physiological and ecological studies are also discussed under an integrative perspective. This study reveals that Mediterranean MPAs encompass eight of the ten banks and 16 of the 21 beds of C. caespitosa.     Magnetic field induced charge and spin instabilities in cuprate superconductors

    A d-wave superconductor, subject to strong phase fluctuations, is known to suffer an antiferromagnetic instability closely related to the chiral symmetry breaking in (2+1)-dimensional quantum electrodynamics (QED3). On the basis of this idea we formulate a "QED3 in a box" theory of local instabilities of a d-wave superconductor in the vicinity of a single pinned vortex undergoing quantum fluctuations around its equilibrium position. As a generic outcome we find an incommensurate 2D spin density wave forming in the neighborhood of a vortex with a concomitant "checkerboard" pattern in the local electronic density of states, in agreement with recent neutron scattering and tunneling spectroscopy measurements.Comment: 4 pages REVTeX + 2 PostScript figures included in text.
