    Conical holographic heat engines

    We demonstrate that adding a conical deficit to a black hole holographic heat engine increases its efficiency; in contrast, allowing a black hole to accelerate decreases efficiency if the same average conical deficit is maintained. Adding other charges to the black hole does not change this qualitative effect. We also present a simple formula to calculate the efficiency of elliptical cycles for any black hole, which allows a more efficient numerical algorithm for computation

    Approximation of excitonic absorption in disordered systems using a compositional component weighted CPA

    Employing a recently developed technique of component weighted two particle Green's functions in the CPA of a binary substitutional alloy AcB1cA_cB_{1-c} we extend the existing theory of excitons in such media using a contact potential model for the interaction between electrons and holes to an approximation which interpolates correctly between the limits of weak and strong disorder. With our approach we are also able to treat the case where the contact interaction between carriers varies between sites of different types, thus introducing further disorder into the system. Based on this approach we study numerically how the formation of exciton bound states changes as the strengths of the contact potentials associated with either of the two site types are varied through a large range of parameter values.Comment: 27 pages RevTeX (preprint format), 13 Postscript figure file

    Trehalose Uptake through P2X7 Purinergic Channels Provides Dehydration Protection

    The tetra-anionic form of ATP (ATP4-) is known to induce monovalent and divalent ion fluxes in cells that express purinergic P2X7 receptors (Steinberg et al., 1987; Sung et al., 1985), and with sustained application of ATP it has been shown that dyes as large as 831 daltons can permeate the cell membrane (Steinberg et al, 1987). The current study explores the kinetics of loading α,α-trehalose (342 daltons) into ATP stimulated J774.A1 cells, which are known to express the purinergic P2X7 receptor (Steinberg et al., 1987). Cells that were incubated at 37 ̊C in a 50 mM phosphate buffer (pH 7.0) contailing 225 mM trehalose and 5 mM ATP, were shown to load trehalose linearly over time. Concentrations of ~50 mM were reached within 90 min of incubation. Cells incubated in the same solution at 4 ̊C loaded minimally, consistent with the inactivity of the receptor at low temperatures. However, extended incubation at 37 oC (\u3e60 min) resulted in zero next-day survival, with adverse effects appearing even with incubation periods as short as 30 min. By using a two-step protocol with a short time period at 37 oC to allow pore formation, followed by an extended loading period on ice, cells could be loaded with up to 50 mM trehalose while maintaining good next day recovery (49% ± 12 % by Trypan Blue exclusion, 56 ± 20% by Alamar BlueTM assay). Cells porated by this method and allowed an overnight recovery period exhibited improved dehydration tolerance suggesting a role for ATP poration in the anhydrous preservation of cells

    Human Tra2 proteins jointly control a CHEK1 splicing switch among alternative and constitutive target exons

    Alternative splicing—the production of multiple messenger RNA isoforms from a single gene—is regulated in part by RNA binding proteins. While the RBPs transformer2 alpha (Tra2α) and Tra2β have both been implicated in the regulation of alternative splicing, their relative contributions to this process are not well understood. Here we find simultaneous—but not individual—depletion of Tra2α and Tra2β induces substantial shifts in splicing of endogenous Tra2β target exons, and that both constitutive and alternative target exons are under dual Tra2α–Tra2β control. Target exons are enriched in genes associated with chromosome biology including CHEK1, which encodes a key DNA damage response protein. Dual Tra2 protein depletion reduces expression of full-length CHK1 protein, results in the accumulation of the DNA damage marker γH2AX and decreased cell viability. We conclude Tra2 proteins jointly control constitutive and alternative splicing patterns via paralog compensation to control pathways essential to the maintenance of cell viability

    Modelling sea breeze climatologies and interactions on coasts in the southern North Sea: Implications for offshore wind energy

    Current understanding of the behaviour of sea breezes in the offshore environment is limited but rapidly requires improvement due, not least, to the expansion of the offshore wind energy industry. Here we report on contrasting characteristics of three sea-breeze types on five coastlines around the southern North Sea from an 11 year model-simulated climatology. We present and test an identification method which distinguishes sea-breeze types which can, in principle, be adapted for other coastlines around the world. The coherence of the composite results for each type demonstrates that the method is very effective in resolving and distinguishing characteristics and features. Some features, such as jets and calm zones, are shown to influence offshore wind farm development areas, including the sites of the proposed wind farms up to 200 km offshore. A large variability in sea-breeze frequency between neighbouring coastlines of up to a factor of 3 is revealed. Additionally, there is a strong association between sea-breeze type on one coastline and that which may form coincidentally on another nearby. This association can be as high as 86% between, for example, the North Norfolk and East Norfolk coasts. We show, through associations between sea-breeze events on coastlines with contrasting orientations, that each coastline can be important for influencing the wind climate of another. Furthermore, we highlight that each sea-breeze type needs separate consideration in wind power resource assessment and that future larger turbines will be more sensitive to sea-breeze impacts
