    Development of specific PCR assays for the detection of Cryptocaryon irritans

    Cryptocaryon irritans is one of the most important protozoan pathogens of marine fish, causing the “white spot” disease and posing a significant problem to marine aquaculture. In the present study, a C. irritans-specific reverse primer (S15) was designed based on the published sequence of the second internal transcribed spacer (ITS-2) of ribosomal DNA (rDNA) of C. irritans and used together with the conserved forward primer P1 to develop a specific polymerase chain reaction (PCR) assay for direct, rapid, and specific detection of C. irritans. The specificity of these primers was tested with both closely and distantly related ciliates (Pseudokeroronpsis rubra, Pseudokeroronpsis carnae, Euplotes sp. 1, Ichthyophthirius multifiliis, Pseudourostyla cristata, and Paramecium caudaium), and only C. irritans was detected and no product was amplified from any other ciliates examined in this study using the specific primer set P1-S15. The specific PCR assay was able to detect as low as 45 pg of C. irritans DNA and a nested PCR assay using two primer sets (P1/NC2, P1/S15) increased the sensitivity, allowing the detection of a single C. irritans. The species-specific PCR assays should provide useful tools for the diagnosis, prevention, and molecular epidemiological investigations of C. irritans infection in marine fish

    Enfermagem médico-cirúrgica: uma nova abordagem de ensino e sua avaliação pelo aluno

    This issue is about a teaching methodology developed by the Medical-Surgical subject of Nursing on Graduate Course at Universidade Estadual de Londrina. The teachers used the student-centered teaching process based on interpersonal relationship and problem solving method. The aim was to evaluate the last period of nursing students perception about developing of cognitive, affective and psychomotor abilities when compared with others subjects of nursing course since 1985 until 1990. This method of teaching supplied more integration among theory and practice, more ability in developing library search and oral exposition, besides more active student participation on his apprenticeship process.Este trabajo trata de la metodologia de enseñanza utilizada por la disciplina de Enfermería Médico-Quirúrgica de la Universidade Estadual de Londrina, modificada con el propósito de hacerla centrada en el alumno, con énfasis mayor en las relaciones interpersonales y estruturada en la resolución de los problemas. Tuvo como objetivo verificar la percepción de los alumnos del último semestre del curso con relación al desarrollo de habilidades cognoscitivas, afectivas y psicomotoras por la disciplina sobredicha, cuando comparada a las otras disciplinas del curso. Los datos fueron obtenidos entre 1985 y 1990 bajo la aplicación de un cuestionario, aplicativo, a los alumnos que recibirían el Grado (de bachiller) a fin de año, en sesiones específicas. Se ha verificado que este método de enseñanza ha proporcionado al alumno una integración mayor entre teoría y práctica, mayor desarrollo en competencia de investigación en biblioteca, participación más activa en su proceso de aprendizaje y mayor capacidad como expositor verbal.Este trabalho versa sobre a metodologia de ensino adotada pela disciplina Enfermagem Médico-Cirúgica da Universidade Estadual de Londrina, modificada no sentido de torná-la centrada no aluno, com maior ênfase nas relações interpessoais e estruturada na resolução de problemas. Teve como objetivo verificar a percepção dos alunos do último período do curso em relação ao desenvolvimento de habilidades cognitivas, afetivas e psicomotoras pela disciplina em questão, quando comparada as demais disciplinas do curso. Os dados foram coletados através de um questionário aplicado aos formandos dos anos de 1985 a 1990, em sessões específicas. Verificou-se que este método de ensino proporcionou ao aluno uma maior integração entre teoria e prática, maior desenvolvimento de habilidade de pesquisa em biblioteca, participação mais ativa no seu processo de aprendizagem e maior habilidade para exposição oral

    障害者の情報環境改善の一試案として 「障害者支援ソフトウェア導入 one CD linux」 の自動生成システム (Assistive Tools Using Linux On Demand System) を提案する.本研究では, まずネットワーク上にある障害者支援ソフトウェアを one CD linux 上に実装を行った. 実装には, 視覚障害者用音声読み上げエディター emacspeak を試用した. その上で, ソフトウェアの選択/実装/iso イメージ生成を半自動で行えるインストーラを製作し、 そのインストーラを含む one CD linux の試作を行った. 本システムの試作により, 障害者に必要とされる機能を取捨選択し個別の障害に対応できる OS 製作の可能性を検討した

    Bactericidal activity of caprylic acid entrapped in mesoporous silica nanoparticles

    [EN] Development of nanotechnologies to improve the functionality of natural antimicrobials for food applications has received much attention in recent years. Mesoporous silica particles, such as MCM-41, have been recently proposed as smart delivery devices capable of loading and releasing large amounts of cargo. In this study, the antimicrobial activity of caprylic acid entrapped in MCM-41 nanoparticles against Escherichia coli, Salmonella enterica, Staphylococcus aureus and Listeria monocytogenes was tested and compared with the bactericidal effect of free caprylic acid using the macrodilution method. The minimum bactericidal concentration for free caprylic acid was established to be below 18.5 mM for S. aureus and L. monocytogenes and within the 18.5-20 mM range for E. coli and S. enterica. Moreover, caprylic acid loaded nanoparticles showed a total inhibition of the growth within the 18.5-20 mM range for the tested bacteria, and therefore the antimicrobial activity was preserved. Transmission electron microscopy images revealed that bacteria treatment with the caprylic acid-loaded nanoparticles generated disruption of cell envelope and leakage of cytoplasmic content, which resulted in cell death. We believe that caprylic acid encapsulation in nanoparticles MCM-41 can provide an effective system for potential applications in food safety in the food industry due to the possible controlled release of fatty acid and the masking of its unpleasant organoleptic properties.
