Multifunctional enzymatically generated hydrogels for chronic wound application

The healing of chronic wounds requires intensive medical intervention at huge healthcare costs. Dressing materials should consider the multifactorial nature of these wounds comprising deleterious proteolytic and oxidative enzymes and high bacterial load. In this work, multifunctional hydrogels for chronic wound application were produced by enzymatic cross- linking of thiolated chitosan and gallic acid. The hydrogels combine several beneficial to wound healing properties, controlling the matrix metalloproteinases (MMPs) and myeloperoxidase (MPO) activities, oxidative stress, and bacterial contamination. In vitro studies revealed above 90% antioxidant activity, and MPO and collagenase inhibition by up to 98 and 23%, respectively. Ex vivo studies with venous leg ulcer exudates confirmed the inhibitory capacity of the dressings against MPO and MMPs. Additionally, the hydrogels reduced the population of the most frequently encountered in nonhealing wounds bacterial strains. New myeloperoxidase detection system based on enzyme-catalysed oxidative synthesis of a dye for paper-based diagnostic devices

The severity and cost of wound infections strongly demands for simple and fast methods for wound infection determination. Point-of-care testing devices play a crucial role in order to achieve a fast diagnosis and early treatment. Myeloperoxidase (MPO) enzyme, detected in fluids of infected wounds has been postulated as a suitable biomarker for wound diagnostics. Here we present a new system for MPO detection, based on enzyme-catalysed oxidative synthesis of a dye that can be incorporated into paper-based point of care devices. Visual MPO detection has been achieved through the use of phenylenediamine, a common colourless hair dye precursor. MPO oxidation of these compounds yielded bright coloured products distinguishable from the colour of the wound environment. Immobilisation of the MPO substrates on paper strips was achieved through in situ interaction of the oxidised coloured product with branched polyethyleneimine. The colour reaction of the immobilized substrates, detectable by naked eye, responds to the MPO levels present in infected wound fluids revealing an easy system for incorporation of MPO detection in paper based diagnostic devices.Peer ReviewedPostprint (author's final draft

Introducing entrepreneurship in engineering project management engineering

Según el diario el Mundo, por primera vez los estudiantes universitarios españoles que tienen pensado crear un negocio (26,8%) supera a los que se ven trabajando en la Administración Pública el (25%), ese fue el resultados de las 8600 encuestas realizadas por GAD3 para el estudio encargado por Educa 2020, donde Ingeniería y Arquitectura son las ramas con mayores porcentajes de alumnado que quieren crear una empresa. Con estos datos, se pretende trabajar en esta línea para estimular al alumnado en la generación de ideas de negocio y dar soporte para llevarlas a cabo. Además, a través del trabajo coordinado con el Vicerrectorado de Coordinación Institucional e Infraestructuras se intentará acercar al alumnado cómo es el proceso de licitación de obras, servicios o equipamiento en una Administración Pública como es la UCO, mostrando la transparencia del proceso. En la experiencia piloto del anterior curso académico, el alumnado valoró muy positivamente la asistencia a un acto público de apertura de ofertas económicas de un proceso de licitación real desarrollado por la UCO, donde se mostraron participativos y vieron de primera mano cómo todo el proceso es transparente y sigue rigurosamente la Ley de Contratos del Sector Público.According to the El Mundo newspaper, for the first time, Spanish university students who plan to create a business (26.8%) exceed those who are working in the Public Administration (25%), that was the result of the 8600 surveys conducted by GAD3 for the study commissioned by Educa 2020, where Engineering and Architecture are the branches with the highest percentages of students that want to start a business. With these data, it is recommended to work in this line to stimulate the student in the generation of business ideas and give support to carry them out. In addition, through coordinated work with the Office of the Vice Chancellor for Institutional Coordination and Infrastructures, students are approached about the process of bidding for works, services or equipment in a Public Administration such as the UCO, showing the transparency of the process. In the pilot experience of the previous academic course, the students positively valued attendance at a public act of opening economic offers of a real bidding process developed by the UCO, where they participated and showed how the whole transparent process rigorously follows the Law of Public Sector Contracts

En este trabajo se presentan las interrogantes surgidas después de que una de las autoras ganara un premio internacional en una startup. Las consideraciones versan sobre el sistema capitalista y las startup como organizaciones que fomentan el emprendimiento; lo que obliga a las autoras a explicar qué significa emprendimiento y específicamente emprendimiento femenino, para poder exponer el caso LIA, que es el emprendimiento femenino ganador del premio internacional

Characteristics and evolution of patients with out of hospital cardiac arrest treated with therapeutic hypothermia

[ES] Introducción: La parada cardiorrespiratoria (PCR) presenta múltiples morbilidades secundarias, especialmente las neurológicas, por tanto es necesario un correcto manejo posparada. El objetivo de este trabajo consiste en analizar las características y los resultados de pacientes sometidos a hipotermia tras una PCR extrahospitalaria resucitada. Material y métodos: Realizamos un estudio prospectivo observacional de dos años, donde se incluyeron pacientes que ingresaron en nuestra unidad mayores de 18 años y con PCR prolongada extrahospitalaria resucitada, en los cuales se realizó un protocolo consensuado de cuidados posresucitación e hipotermia terapéutica. Resultados: Se incluyeron 33 pacientes. Se observó una media de 31,6 min de tiempo de PCR. Un 32,3% presentaron encefalopatía hipóxico-anóxica, siendo más frecuente en fumadores (p = 0,04), y con mayores tiempos de PCR (p= 0,049). También el 66,7% de los pacientes presentaron mayores datos de hipoperfusión (p = 0,049). Conclusión: Observamos que la hipotermia podría ser un instrumento útil en el manejo de las PCR extrahospitalarias. Además, encontramos que el pronóstico neurológico se asocia más a factores no modificables.[EN] Introduction: Cardiac arrest have secondary co-morbidities, especially neurological outcome. Due to this cirumstances is mandatory to optimized postcardiac arrest management. The objective of this research is to analyze patient's characteristics and outcome after out-hospital cardiac arrest in patients that have undergone therapeutic hypothermia. Matherial and methods: We conducted a prospective observational study during a two-year period. We included resuscitated patients over eighteen years old who were admitted in to our intensive care unit. These patients had undergone prolonged resuscitation measures and were included in an implemented post-resuscitation protocol which included therapeutic hypothermia. Results: Thirty-three patients were analyzed. The median time of cardiopulmonary resuscitation was 31.6 minutes. We observed hypoxic/anoxic encephalopathy in 32.3% of cases. This prognosis was associated in those who smoked (p = .04) and with longer time of cardiac arrest (p = .049). Also, the 66.7% of the patients showed a higher incidence of hypoperfusion (p = .049). Conclusion: In our series hypothermia could be a useful tool in post-resucitation cardiac arrest managment. We also found that the neurological prognosis was associated to non-modifiable risk factors

Nanoparticle-driven self-assembling injectable hydrogels provide a multi-factorial approach for chronic wound treatment

Chronic wounds represent a major health burden and drain on medical system. Efficient wound repair is only possible if the dressing materials target simultaneously multiple factors involved in wound chronicity, such as deleterious proteolytic and oxidative enzymes and high bacterial load. Here we develop multifunctional hydrogels for chronic wound management through self-assembling of thiolated hyaluronic acid (HA-SH) and bioactive silver-lignin nanoparticles (Ag@Lig NPs). Dynamic and reversible interactions between the polymer and Ag@Lig NPs yield hybrid nanocomposite hydrogels with shear thinning and self-healing properties, coupled to zero-order kinetics release of antimicrobial silver in response to infection-related hyaluronidase (HNase). The hydrogels inhibit the major enzymes myeloperoxidase (MPO) and matrix metalloproteinases (MMPs) responsible for wound chronicity in a patient’s wound exudate. Furthermore, the lignin-capped AgNPs provide the hydrogel with antioxidant properties and strong antibacterial activity against Staphylococcus aureus and Pseudomonas aeruginosa. The nanocomposite hydrogels are not toxic to human keratinocytes after 7 days of direct contact. Complete tissue remodeling and restoration of skin integrity is demonstrated in vivo in a diabetic mouse model. Hematological analysis reveals lack of wound inflammation due to bacterial infection or toxicity, confirming the potential of HA-SH/Ag@Lig NPs hydrogels for chronic wound management.Peer ReviewedPostprint (author's final draft
