    O6-methylguanine-DNA methyltransferase (MGMT) Promoter methylation is a rare event in soft tissue sarcoma

    BACKGROUND: Gene silencing of O6-methylguanine–DNA methyltransferase (MGMT) by promoter methylation improves the outcome of glioblastoma patients after combined therapy of alkylating chemotherapeutic agents and radiation. The purpose of this study was to assess the frequency of MGMT promoter methylation in soft tissue sarcoma to identify patients eligible for alkylating agent chemotherapy such as temozolomide. FINDINGS: Paraffin tumor blocks of 75 patients with representative STS subtypes were evaluated. The methylation status of the MGMT promoter was assessed by methylation-specific polymerase-chain-reaction analysis (PCR). Furthermore, immunohistochemistry was applied to verify expression of MGMT. MGMT gene silencing was assumed if MGMT promoter methylation was present and the fraction of tumor cells expressing MGMT was 20% or less. Methylation specific PCR detected methylated MGMT promoter in 10/75 cases. Immunohistochemical staining of nuclear MGMT was negative in 15/75 cases. 6/75 tumor samples showed MGMT promoter methylation and negative immunohistochemical nuclear staining of MGMT. In none of the tested STS subtypes we found a fraction of tumors with MGMT silencing exceeding 22%. CONCLUSION: MGMT gene silencing is a rare event in soft tissue sarcoma and cannot be recommended as a selection criterion for the therapy of STS patients with alkylating agents such as temozolomide

    Das Bremer Energie-Institut initiierte Ende 1998 eine Untersuchung zu den "Auswirkungen eines im GebĂ€udebereich sinkenden WĂ€rmebedarfs auf die Struktur der Energieversorgung im WĂ€rmemarkt". Die Bearbeitung dieses Vorhabens ist in drei Phasen mit unterschiedlichen Zielrichtungen, Arbeitsmethoden und Kooperationspartnern gegliedert. In der ersten Phase wurde eine Grundlagenerhebung durchgefĂŒhrt, die die aktuelle Situation sowohl auf der "Nachfrageseite" (Stand des WĂ€rmeschutzes in Deutschland sowie in beispielhaften europĂ€ischen NachbarlĂ€ndern, neue Forschungsergebnisse zur Energieversorgung von WohngebĂ€uden mit niedrigem WĂ€rmebedarf usw.) als auch auf der "Angebotsseite" (Strukturen der Energieversorgung, Heizungssysteme, EnergietrĂ€ger-MĂ€rkte usw.) darstellt. Ein Abschlussbericht steht auf der Homepage der Bremer Energie-Konsens GmbH, die dieses Vorhaben gefördert hat, zum downloaden bereit. (www.energiekonsens.de‟\underline{www.energiekonsens.de}) In einer nachfolgenden zweiten Projektphase werden in Kooperation mit der Programmgruppe STE des Forschungszentrums JĂŒlich Prognosen fĂŒr die Entwicklung des WĂ€rmebedarfs bezogen auf definierte Siedlungsstrukturen erstellt und die Auswirkungen auf die Energieversorgungswirtschaft analysiert. Dabei werden insbesondere die VerĂ€nderungen der heute bestehenden Marktstrukturen der Versorgungswirtschaft als Folge einer sowohl quantitativen als auch qualitativen VerĂ€nderung des WĂ€rmeabsatzes untersucht und bewertet. Der vorliegende Bericht "Die Entwicklung des Wašrmemarktes fušr den Gebašudesektor bis 2050\textbf{Die Entwicklung des WĂ€rmemarktes fĂŒr den GebĂ€udesektor bis 2050}" ist ein Ergebnis der Phase II. Zeitlich etwa parallel zur zweiten Phase sollen in einer Phase III in Zusammenarbeit mit verschiedenen Energieversorgungsunternehmen Fallstudien durchgefĂŒhrt werden, die eine Verifizierung der in den vorangegangenen Phasen erarbeiteten Grundlagen, Prognosen und Wirkungsanalysen an konkreten Objekten und Versorgungsgebieten ermöglichen. Innerhalb der einzelnen Phasen wird das Bremer Energie-Institut federfĂŒhrend die quantitativen Auswirkungen eines reduzierten WĂ€rmeabsatzes sowie die aufgrund innovativer Energieversorgungssysteme verĂ€nderten qualitativen Anforderungen an die Energieversorgungsunternehmen ermitteln und hinsichtlich ihrer Relevanz fĂŒr die zukĂŒnftige Entwicklung der Energieversorgungswirtschaft bewerten. Das Gesamt-Vorhaben soll im Jahre 2001 mit einem umfassenden Bericht zu den "Auswirkungen eines im GebĂ€udebereich sinkenden WĂ€rmebedarfs auf die Struktur der Energieversorgung im WĂ€rmemarkt" abgeschlossen werden

    Social networks of men who have sex with men engaging in chemsex in Germany: differences in social resources and sexual health

    Abstract Meyer’s minority stress model posits that sexual minority communities can act as protective factors for individuals within the sexual minority. Given that existing evidence on this proposition is inconclusive, a social network approach was employed to capture diversity in the social environment of individuals involved in chemsex that might account for variations in social resources and sexual health. This study examined the social networks of men who have sex with men (MSM) involved in sexualised drug use, using data from a cross-sectional online survey. Utilising cluster analysis, four distinct social network types were identified based on network composition: MSM-diverse, partner-focused, family-diverse, and chemsex-restricted. In terms of social resources, the four network types did not exhibit significant differences in social support. However, individuals with a chemsex-restricted social network reported stronger social influence related to chemsex and less social engagement outside of chemsex. Contrary to initial expectations, the four network types did not differ in chemsex-related consequences or sexual satisfaction. MSM engaged in chemsex for over 5 years reported more chemsex-related consequences and lower sexual satisfaction, particularly those with a family-diverse social network. Additionally, indicators of network quality, such as perceived emotional closeness, reciprocity with network members, and overall satisfaction with the network, were more influential in predicting sexual health outcomes than social resources. The findings of the study suggest that the social environment of MSM engaged in chemsex plays a role in shaping their experiences. Insufficient inclusion in a sexual minority community is potentially associated with an elevated risk of poor sexual health. These findings underscore the importance of tailoring interventions to address the diverse needs of individuals exposed to different social environments

    Environmental changes in the central Baltic Sea during the past 1000 years: inferences from sedimentary records, hydrography and climate

    Short sediment cores from the eastern Gotland Basin were investigated using a multi-proxy approach in order to reconstruct the environmental conditions of the area during the past 1000 years. Sediment data and facies were discussed in relation to hydrographic features (salinity, oxygen) and climate change. During the medieval warm period (MWP), from about 900 to 1250 AD, the hydrographic and environmental conditions were similar to those of the present time (modern warm period, since about 1850): a temporally stable halocline, caused by regular saline water inflows from the North Sea, prevents vertical mixing and leads to bottom water anoxia and the deposition of laminated, organic-rich sapropels. During the period from about 1250 to 1850, referred to as the cold phase (including the Little Ice Age), the environmental conditions of the central Baltic Sea were distinctly different: the lower salinity, resulting from reduced North Sea water inflows, allowed vertical convection of the water column and long-term stable ventilation of the sea bed (oxic stage). Both the productivity of the planktonic ecosystem as well as the preservation of organic matter in the sediments improved during the warm periods. The anthropogenic impact can be identified within the recent laminated sequence by a temporal reconstruction of pollutant deposition.
