Spectra of ergot alkaloids produced by Claviceps purpurea 1029c in solid-state fermentation system : influence of the composition of liquid medium used for impregnating sugar-cane pith bagasse
A total of 16 different combinations of the liquid nutrient medium used for impregnating sugar-cane pith bagasse have been evaluated for the production of total alkaloids by #Claviceps purpurea$ 1029c in a solid-state fermentation system, and for their effects on the spectra of the alkaloids produced. The data indicated large differences in the alkaloids spectra. There is therefore the possibility of achieving tailor-made spectra of ergot alkaloids by changing the liquid nutrient media composition. (Résumé d'auteur
Potential of solid state fermentation for production of ergot alkaloids
Production of total ergot alkaloids by #Claviceps fusiformis in solid state fermentation was 3.9 times higher compared to that in submerged fermentation. Production was equal in the case of #Claviceps purpurea but the spectra of alkaloids were advantageous with the use of solid state fermentation. The data establish potential of solid state fermentation which was not explored earlier for production of ergot alkaloids. (Résumé d'auteur
Solid substrate mediated changes in ergot alkaloid spectra in solid state fermentation system
Use of different solid substrates resulted in minor alterations in total alkaloid production by #Claviceps purpurea$ 1029c in solid state fermentation system but the changes in the spectra of ergot alkaloids were of significantly higher magnitudes. Ergonovine accounted for 93% of the total alkaloid production in wheat grain medium while lysergic acid derivatives and ergonovine comprised of 66% and 32% of total alkaloids in rye grain medium. In contrast, ergonovine, ergotamine, and lysergic acid derivatives were 35, 35, and 25% respectively of the total alkaloids with the use of sugar cane pith bagasse impregnated with sucrose. Influence of antioxidant location on the protection of oil encapsulated in powder
Encapsulation of Poly Unsaturated Fatty Acids (PUFAs) in solid matrix, by providing a physical barrier, is used to prevent or delay their degradation; and the use of antioxidant is expected to enhance PUFAs oxidative stability. In liquid emulsions, the effectiveness of antioxidants is known to depend on their distribution between the oil and aqueous phase. In this study, the impact of the lipophilic or hydrophilic character of phenolic antioxidants on the oxidative stability of encapsulated PUFAs was investigated following the evolution of conjugated dienes and antioxidant residual content during accelerated ageing test (50\ub0C, 60% RH). Dry emulsions containing 10%wt stripped sunflower oil (60% PUFAs), 89.7%wt maltodextrin DE12 (wall material) and 0.3%wt Tween\uae20 (surfactant) were produced by spray drying. Some were enriched with alpha-tocopherol (lipophilic - 500 ppm in oil) or chlorogenic acid (hydrophilic, 1000 ppm in maltodextrin), two scavengers of lipid radicals implied in oxidation.Antioxidants improved the oxidative stability of encapsulated oil. With chlorogenic acid, oil oxidation occurred after a two days lag phase whilst for alpha-tocopherol, no lag phase was observed but the oxidation rate was smaller than in control and chlorogenic acid powders during the ten first days of ageing (Fig, 1a). The residual concentration of chlorogenic acid deceased rapidly during the first two days and then remained constant whilst the concentration of alphatocopherol decreased regularly ensuring oil protection until it has been totally consumed after ten days (Fig.1b). The better oil protection provided by alpha-tocopherol during the first ten days of storage was attributed to the different location of both antioxidants within the macro-domains of the dry emulsion. Alpha-tocopherol, in oil droplets, was directly in contact with the oil to protect whilst for chlorogenic acid, entrapped in the solid matrix, only the fraction in contact with the oil droplets brought protection and 60% of initial chlorogenic acid remained preserved in the matrix
Modulation of extracellular matrix by nutritional hepatotrophic factors in thioacetamide-induced liver cirrhosis in the rat
Nutritional substances associated to some hormones enhance liver regeneration when injected intraperitoneally, being denominated hepatotrophic factors (HF). Here we verified if a solution of HF (glucose, vitamins, salts, amino acids, glucagon, insulin, and triiodothyronine) can revert liver cirrhosis and how some extracellular matrices are affected. Cirrhosis was induced for 14 weeks in 45 female Wistar rats (200 mg) by intraperitoneal injections of thioacetamide (200 mg/kg). Twenty-five rats received intraperitoneal HF twice a day for 10 days (40 mL·kg-1·day-1) and 20 rats received physiological saline. Fifteen rats were used as control. The HF applied to cirrhotic rats significantly: a) reduced the relative mRNA expression of the genes: Col-α1 (-53%), TIMP-1 (-31.7%), TGF-β1 (-57.7%), and MMP-2 (-41.6%), whereas Plau mRNA remained unchanged; b) reduced GGT (-43.1%), ALT (-17.6%), and AST (-12.2%) serum levels; c) increased liver weight (11.3%), and reduced liver collagen (-37.1%), regenerative nodules size (-22.1%), and fibrous septum thickness. Progranulin protein (immunohistochemistry) and mRNA (in situ hybridization) were found in fibrous septa and areas of bile duct proliferation in cirrhotic livers. Concluding, HF improved the histology and serum biochemistry of liver cirrhosis, with an important reduction of interstitial collagen and increased extracelullar matrix degradation by reducing profibrotic gene expression
Explorar e Investigar em Matemática: Uma Actividade Fundamental no Ensino e na Aprendizagem
Investigar, ensinar e aprender são actividades que podem estar presentes, de forma
articulada, no ensino-aprendizagem da Matemática e na actividade profissional do
professor. Para isso, é necessário conceber tarefas que possam ser o ponto de partida para
investigações e explorações matemáticas dos alunos e discutir o modo como podem ser
trabalhadas na sala de aula. Recorrendo a exemplos de actividades realizadas por
professores de Matemática portugueses, analiso a actividade de aprendizagem suscitada
por tarefas deste tipo e discuto as respectivas potencialidades. Finalmente, refiro as
condições respeitantes à cultura profissional dos professores que podem favorecer uma
actividade de investigação sobre a sua própria profissional, com relevo para a colaboração
e a dimensão associativa.Researching, teaching and learning are activities that may be present, in coordination,
teaching and learning of mathematics in the work of the teacher. Therefore, it is necessary
to design tasks that may be the starting point for mathematical investigations and
explorations of the students and discuss how they can be worked in the classroom. Drawing
on examples of activities carried out by Portuguese teachers of mathematics by examining
the activity of learning raised by such tasks and discuss their potencial. Finally, I refer to
conditions relating to the professional culture of teachers who can promote the research
activity on their own training, with an emphasis on collaboration and associative dimension
Emergent geometry from q-deformations of N=4 super Yang-Mills
We study BPS states in a marginal deformation of super Yang-Mills on R x S^3
using a quantum mechanical system of q-commuting matrices. We focus mainly on
the case where the parameter q is a root of unity, so that the AdS dual of the
field theory can be associated to an orbifold of AdS_5x S^5. We show that in
the large N limit, BPS states are described by density distributions of
eigenvalues and we assign to these distributions a geometrical spacetime
interpretation. We go beyond BPS configurations by turning on perturbative
non-q-commuting excitations. Considering states in an appropriate BMN limit, we
use a saddle point approximation to compute the BMN energy to all perturbative
orders in the 't Hooft coupling. We also examine some BMN like states that
correspond to twisted sector string states in the orbifold and we show that our
geometrical interpretation of the system is consistent with the quantum numbers

Analytical study of non-linear transport across a semiconductor-metal junction
In this paper we study analytically a one-dimensional model for a
semiconductor-metal junction. We study the formation of Tamm states and how
they evolve when the semi-infinite semiconductor and metal are coupled
together. The non-linear current, as a function of the bias voltage, is studied
using the non-equilibrium Green's function method and the density matrix of the
interface is given. The electronic occupation of the sites defining the
interface has strong non-linearities as function of the bias voltage due to
strong resonances present in the Green's functions of the junction sites. The
surface Green's function is computed analytically by solving a quadratic matrix
equation, which does not require adding a small imaginary constant to the
