Wound degradomics - current status and future perspectives
Proteases are pivotal modulators of extracellular matrix components and bioactive proteins at all phases of cutaneous wound healing and thereby essentially contribute to the successful reestablishment of skin integrity upon injury. As a consequence, disturbance of proteolytic activity at the wound site is a major factor in the pathology of chronic wounds. A large body of data acquired in many years of research provide a good understanding of how individual proteases may influence the repair process. The next challenge will be to integrate these findings and to elucidate the complex interactions of proteolytic enzymes, their inhibitors and substrates on a system-wide level. Here, we present novel approaches that might help to achieve this ambitious goal in cutaneous wound healing researc
Measured and simulated heavy-ion beam loss patterns at the CERN Large Hadron Collider
The Large Hadron Collider (LHC) at CERN pushes forward to new regimes in terms of beam energy and intensity. In view of the combination of very energetic and intense beams together with sensitive machine components, in particular the superconducting magnets, the LHC is equipped with a collimation system to provide protection and intercept uncontrolled beam losses. Beam losses could cause a superconducting magnet to quench, or in the worst case, damage the hardware. The collimation system, which is optimized to provide a good protection with proton beams, has shown a cleaning efficiency with heavy-ion beams which is worse by up to two orders of magnitude. The reason for this reduced cleaning efficiency is the fragmentation of heavy-ion beams into isotopes with a different mass to charge ratios because of the interaction with the collimator material. In order to ensure sufficient collimation performance in future ion runs, a detailed theoretical understanding of ion collimation is needed. The simulation of heavy-ion collimation must include processes in which Pb82+208 ions fragment into dozens of new isotopes. The ions and their fragments must be tracked inside the magnetic lattice of the LHC to determine their loss positions. This paper gives an overview of physical processes important for the description of heavy-ion loss patterns. Loss maps simulated by means of the two tools ICOSIM [1,2] and the newly developed STIER (SixTrack with Ion-Equivalent Rigidities) are compared with experimental data measured during LHC operation. The comparison shows that the tool STIER is in better agreement.peer-reviewe
Exponential Stabilization of Nonlinear Driftless Systems with Robustness to Unmodeled Dynamics
Exponential stabilization of nonlinear driftless affine control systems is addressed with the concern of achieving robustness with respect to imperfect knowledge of the system's control vector fields. In order to satisfy this robustness requirement, and inspired by \cite{bennani95} where the same issue was first addressed, we consider a control strategy which consists of applying periodically updated open-loop controls that are continuous with respect to state initial conditions. These controllers are more precisely described as continuous time-periodic feedbacks associated with a specific dynamic extension of the original system. Sufficient conditions which, if they are satisfied by the control law, ensure that the control is a robust exponential stabilizer for the extended system are given. Explicit and simple control expressions which satisfy these conditions in the case of -dimensional chained systems are proposed. A constructive algorithm for the design of such control laws, which applies to any (sufficiently regular) driftless control system, is described
Improved aperture measurements at the LHC and results from their application in 2015
A good knowledge of the available aperture in the LHC
is essential for a safe operation due to the risk of magnet
quenches or even damage in case of uncontrolled beam
losses. Experimental validations of the available aperture
are therefore crucial and were in the past carried out by either a collimator scan combined with beam excitations or
through the use of local orbit bumps. In this paper, we show
a first comparison of these methods in the same machine
configuration, as well as a new very fast method based on a
beam-based collimator alignment and a new faster variant
of the collimator scan method. The methods are applied
to the LHC operational configuration for 2015 at injection
and with squeezed beams and the measured apertures are
presented.peer-reviewe
Studies on heavy ion losses from collimation cleaning at the LHC
The LHC collimation system protects superconducting
magnets from beam losses. By design, it was optimized
for the high-intensity proton challenges but so far provided
adequate protection also during the LHC heavy-ion runs
with 208Pb82+ ions up to a beam energy of 4 Z TeV. Ion beam
cleaning brings specific challenges due to different physical
interactions with the collimator materials and might require
further improvements for operation at 7 Z TeV. In this article,
we study heavy-ion beam losses leaking out of the LHC
collimation system, both in measurement and simulations.
The simulations are carried out using both ICOSIM, with a
simplified ion physics model implemented, and SixTrack, including more detailed starting conditions from FLUKA but
without including online scattering in subsequent collimator hits. The results agree well with measurements overall,
although some discrepancies are present. The reasons for
the discrepancies are investigated and, on this basis, the
requirements for an improved simulation tool are outlined.peer-reviewe
Performance of the LHC collimation system during 2015
During the long shutdown, a total of 32 collimators were
replaced or installed to improve the Run 1 system, including 18 new collimators with embedded beam position monitors, additional physics debris collimators, additional passive absorbers and the re-installation or displacement of existing collimators. The commissioning experience as well
as the performance of the collimation system in 2015 is
reviewed following these upgrades. In particular, the operational experience with the embedded BPM collimators
is discussed, emphasizing the improvements on alignment
efficiency and the strategy for better orbit monitoring. Improvements of collimation setting validation through an optimized loss map strategy are also presented. Further improvements planned for 2016 are discussed.peer-reviewe
Cardiovascular disease, chronic kidney disease, and diabetes mortality burden of cardiometabolic risk factors from 1980 to 2010: A comparative risk assessment
Background: High blood pressure, blood glucose, serum cholesterol, and BMI are risk factors for cardiovascular diseases and some of these factors also increase the risk of chronic kidney disease and diabetes. We estimated mortality from cardiovascular diseases, chronic kidney disease, and diabetes that was attributable to these four cardiometabolic risk factors for all countries and regions from 1980 to 2010. Methods: We used data for exposure to risk factors by country, age group, and sex from pooled analyses of population-based health surveys. We obtained relative risks for the effects of risk factors on cause-specific mortality from meta-analyses of large prospective studies. We calculated the population attributable fractions for each risk factor alone, and for the combination of all risk factors, accounting for multicausality and for mediation of the effects of BMI by the other three risks. We calculated attributable deaths by multiplying the cause-specific population attributable fractions by the number of disease-specific deaths. We obtained cause-specific mortality from the Global Burden of Diseases, Injuries, and Risk Factors 2010 Study. We propagated the uncertainties of all the inputs to the final estimates. Findings: In 2010, high blood pressure was the leading risk factor for deaths due to cardiovascular diseases, chronic kidney disease, and diabetes in every region, causing more than 40% of worldwide deaths from these diseases; high BMI and glucose were each responsible for about 15% of deaths, and high cholesterol for more than 10%. After accounting for multicausality, 63% (10·8 million deaths, 95% CI 10·1-11·5) of deaths from these diseases in 2010 were attributable to the combined effect of these four metabolic risk factors, compared with 67% (7·1 million deaths, 6·6-7·6) in 1980. The mortality burden of high BMI and glucose nearly doubled from 1980 to 2010. At the country level, age-standardised death rates from these diseases attributable to the combined effects of these four risk factors surpassed 925 deaths per 100 000 for men in Belarus, Kazakhstan, and Mongolia, but were less than 130 deaths per 100 000 for women and less than 200 for men in some high-income countries including Australia, Canada, France, Japan, the Netherlands, Singapore, South Korea, and Spain. Interpretation: The salient features of the cardiometabolic disease and risk factor epidemic at the beginning of the 21st century are high blood pressure and an increasing effect of obesity and diabetes. The mortality burden of cardiometabolic risk factors has shifted from high-income to low-income and middle-income countries. Lowering cardiometabolic risks through dietary, behavioural, and pharmacological interventions should be a part of the global response to non-communicable diseases.
