    Technological appropriateness of biomass production in rural settings:Addressing water hyacinths (E. crassipes) problem in Lake Tondano, Indonesia

    Climate change has induced an excessive growth of water hyacinths, which produces unintended consequences for the surrounding ecosystem. Particularly, water hyacinth is a major problem throughout the world's tropical zone, which largely consists of rural regions. One way to address the water hyacinths problem is to convert them into biomass. However, typical biomass production technologies have not considered local settings when they are installed in rural areas lacking knowledge and resources. This study aims at assessing the technological appropriateness of biomass production from water hyacinths in rural settings under limited resources and knowledge. This research proposes two scenarios (i.e., high-tech and low-tech) to utilise water hyacinths from Lake Tondano, Indonesia, as the case study. The scenarios consider local settings of communities living around the lake by applying scenario-based design science according to Weiringa's adaptation of the five-stage regulative cycle of Van Strien. The assessment stage employs three levels of technological appropriateness (techno-economic, environmental, social) to assess each scenario for the rural context. Results show that the low-tech design is more appropriate for rural settings around Lake Tondano. Both designs are technically able to resolve the water hyacinths problem; however, the low-tech design is more practical for local communities, addressing the environmental problem while simultaneously boosting socioeconomic developments. In general, the small-scale nature of the more appropriate design applies to other rural areas, with which those areas can utilise various biomass sources while benefitting their socioeconomic situations. Further studies need to assess the technological appropriateness of the appropriate design again based on rural contexts in their location(s)

    A toolkit for algorithmic equity and community empowerment

    A wave of recent scholarship documenting the discriminatory harms of algorithmic systems has spurred widespread interest in algorithmic accountability and regulation. Yet effective accountability and regulation is stymied by a persistent lack of resources supporting public understanding of algorithms and artificial intelligence. We present a toolkit for algorithmic legibility developed using participatory design methodologies. Through interactions with a US-based civil rights organization and their coalition of community organizations, we iden- tify a need for (i) 'street level' heuristics that aid stakeholders in distinguishing between types of analytic and information systems in lay language, and (ii) risk assessment tools for such systems that begin by making algorithms more legible. The present work delivers a toolkit to achieve these aims

    Hepatitis in a patient with SLE: Is it autoimmune hepatitis?

    In a patient with systemic lupus erythematosus (SLE), we considered the diagnosis of autoimmune hepatitis (AIH) in view of raised serum aminotransferases, hypergammaglobulinaemia, antinuclear antibodies (titre 1:10240), seronegative of markers for viral hepatitis and absence of recent hepatotoxic drug usage. The diagnosis of AIH was supported by using the scoring system, recently developed by the International Autoimmune Hepatitis Group and the excellent response to treatment with prednisone. Liver histology, however, showed no characteristic features of AIH. The relevance of liver histology and scoring for AIH in SLE with hepatic involvement is discussed

    Restoration of estuarine tidal mudflat sediments after hypoxia

    Ecosystem function recovery and benthic community recovery was investigated after experimentally induced depleted oxygen bottom water concentrations in a tidal mudflat (Paulinapolder, Westerschelde estuary). Macrofauna recovery developed through different succession stages and was structured by facilitative and inhibitive interactions: early colonizers had a positive effect on subsequent colonizers, while later succession species negatively affected the stable conditions created by the early colonizing tube-builders. Transitions between different stages were related to changes in environmental characteristics and biotic-environmental interactions (e.g. exploitation competition for food). Nematode community -and biogeochemical recovery were related to macrobenthic succession. Dense polychaete tube aggregations and the development of a fresh diatom bloom, as a result of the low grazing pressure by surface deposit feeding macrofauna during the first stage, stabilized the sediment and thereby enhanced macrobenthic and nematode recruitment success. Bioturbation impact of later succession species increased oxygen input in the sediment, resulting in an enhanced nitrification, denitrification and energy use

    The CXCL12/CXCR4 signaling axis retains neutrophils at inflammatory sites in zebrafish

    The inappropriate retention of neutrophils at inflammatory sites is a major driver of the excessive tissue damage characteristic of respiratory inflammatory diseases including COPD, ARDS, and cystic fibrosis. The molecular programmes which orchestrate neutrophil recruitment to inflammatory sites through chemotactic guidance have been well-studied. However, how neutrophil sensitivity to these cues is modulated during inflammation resolution is not understood. The identification of neutrophil reverse migration as a mechanism of inflammation resolution and the ability to modulate this therapeutically has identified a new target to treat inflammatory disease. Here we investigate the role of the CXCL12/CXCR4 signaling axis in modulating neutrophil retention at inflammatory sites. We used an in vivo tissue injury model to study neutrophilic inflammation using transgenic zebrafish larvae. Expression of cxcl12a and cxcr4b during the tissue damage response was assessed using in situ hybridization and analysis of RNA sequencing data. CRISPR/Cas9 was used to knockdown cxcl12a and cxcr4b in zebrafish larvae. The CXCR4 antagonist AMD3100 was used to block the Cxcl12/Cxcr4 signaling axis pharmacologically. We identified that cxcr4b and cxcl12a are expressed at the wound site in zebrafish larvae during the inflammatory response. Following tail-fin transection, removal of neutrophils from inflammatory sites is significantly increased in cxcr4b and cxcl12a CRISPR knockdown larvae. Pharmacological inhibition of the Cxcl12/Cxcr4 signaling axis accelerated resolution of the neutrophil component of inflammation, an effect caused by an increase in neutrophil reverse migration. The findings of this study suggest that CXCR4/CXCL12 signaling may play an important role in neutrophil retention at inflammatory sites, identifying a potential new target for the therapeutic removal of neutrophils from the lung in chronic inflammatory disease

    Ethnicity, schooling, and merit in the Netherlands

    We examine to what extent ethnicity affects academic ability measured in the first year of secondary school and secondary school type in the Netherlands. We focus on second-generation immigrants. The empirical results indicate that academic ability (both in mathematics and language) is not affected by ethnicity, independent of parents’ occupation, education, and resources. On a bivariate level, children of Turkish and Moroccan immigrants in the Netherlands are found relatively often in lower tracks in secondary school. This relationship is fully driven by social class and merit, operationalized as including ability and effort. Moreover, children of Turkish, Surinamese and Antillean migrants are, relative to Dutch children from similar backgrounds and merit, more often found in higher tracks in secondary school. However, given the very skewed distribution of educational attainment of immigrants, it is questionable whether ‘class versus ethnicity models’ can accurately compare achievements of native and immigrant children in the Netherlands

    Computer modeling of diabetes and Its transparency: a report on the Eighth Mount Hood Challenge

    Objectives The Eighth Mount Hood Challenge (held in St. Gallen, Switzerland, in September 2016) evaluated the transparency of model input documentation from two published health economics studies and developed guidelines for improving transparency in the reporting of input data underlying model-based economic analyses in diabetes. Methods Participating modeling groups were asked to reproduce the results of two published studies using the input data described in those articles. Gaps in input data were filled with assumptions reported by the modeling groups. Goodness of fit between the results reported in the target studies and the groups’ replicated outputs was evaluated using the slope of linear regression line and the coefficient of determination (R2). After a general discussion of the results, a diabetes-specific checklist for the transparency of model input was developed. Results Seven groups participated in the transparency challenge. The reporting of key model input parameters in the two studies, including the baseline characteristics of simulated patients, treatment effect and treatment intensification threshold assumptions, treatment effect evolution, prediction of complications and costs data, was inadequately transparent (and often missing altogether). Not surprisingly, goodness of fit was better for the study that reported its input data with more transparency. To improve the transparency in diabetes modeling, the Diabetes Modeling Input Checklist listing the minimal input data required for reproducibility in most diabetes modeling applications was developed. Conclusions Transparency of diabetes model inputs is important to the reproducibility and credibility of simulation results. In the Eighth Mount Hood Challenge, the Diabetes Modeling Input Checklist was developed with the goal of improving the transparency of input data reporting and reproducibility of diabetes simulation model results

    Comparison of soft and hard tissue ablation with sub-ps and ns pulse lasers

    Tissue ablation with ultrashort laser pulses offers several unique advantages. The nonlinear energy deposition is insensitive to tissue type, allowing this tool to be used for soft and hard tissue ablation. The localized energy deposition lead to precise ablation depth and minimal collateral damage. This paper reports on efforts to study and demonstrate tissue ablation using an ultrashort pulse laser. Ablation efficiency and extent of collateral damage for 0.3 ps and 1000 ps duration laser pulses are compared. Temperature measurements of the rear surface of a tooth section is also presented

    Maternal plasma folate impacts differential DNA methylation in an epigenome-wide meta-analysis of newborns

    Folate is vital for fetal development. Periconceptional folic acid supplementation and food fortification are recommended to prevent neural tube defects. Mechanisms whereby periconceptional folate influences normal development and disease are poorly understood: epigenetics may be involved. We examine the association between maternal plasma folate during pregnancy and epigenome-wide DNA methylation using Illumina" s HumanMethyl450 Beadchip in 1,988 newborns from two European cohorts. Here we report the combined covariate-adjusted results using meta-analysis and employ pathway and gene expression analyses. Four-hundred forty-three CpGs (320 genes) are significantly associated with maternal plasma folate levels during pregnancy (false discovery rate 5%); 48 are significant after Bonferroni correction. Most genes are not known for folate biology, including APC2, GRM8, SLC16A12, OPCML, PRPH, LHX1, KLK4 and PRSS21. Some relate to birth defects other than neural tube defects, neurological functions or varied aspects of embryonic development. These findings may inform how maternal folate impacts the developing epigenome and health outcomes in offspring
