    Low-noise, high-strength, spiral-bevel gears for helicopter transmissions

    Improvements in spiral-bevel gear design were investigated to support the Army/NASA Advanced Rotorcraft Transmission program. Program objectives were to reduce weight by 25 percent, reduce noise by 10 dB, and increase life to 5000 hr mean-time-between-removal. To help meet these goals, advanced-design spiral-bevel gears were tested in an OH-58D helicopter transmission using the NASA 500-hp Helicopter Transmission Test Stand. Three different gear designs tested included: (1) the current design of the OH-58D transmission except gear material X-53 instead of AISI 9310; (2) a higher-strength design the same as the current but with a full fillet radius to reduce gear tooth bending stress (and thus, weight); and (3) a lower-noise design the same as the high-strength but with modified tooth geometry to reduce transmission error and noise. Noise, vibration, and tooth strain tests were performed and significant gear stress and noise reductions were achieved

    How pharmacoepidemiology networks can manage distributed analyses to improve replicability and transparency and minimize bias

    Several pharmacoepidemiology networks have been developed over the past decade that use a distributed approach, implementing the same analysis at multiple data sites, to preserve privacy and minimize data sharing. Distributed networks are efficient, by interrogating data on very large populations. The structure of these networks can also be leveraged to improve replicability, increase transparency, and reduce bias. We describe some features of distributed networks using, as examples, the Canadian Network for Observational Drug Effect Studies, the Sentinel System in the USA, and the European Research Network of Pharmacovigilance and Pharmacoepidemiology. Common protocols, analysis plans, and data models, with policies on amendments and protocol violations, are key features. These tools ensure that studies can be audited and repeated as necessary. Blinding and strict conflict of interest policies reduce the potential for bias in analyses and interpretation. These developments should improve the timeliness and accuracy of information used to support both clinical and regulatory decisions

    Inactivation of cloned Na channels expressed in Xenopus oocytes

    This study investigates the inactivation properties of Na channels expressed in Xenopus oocytes from two rat IIA Na channel cDNA clones differing by a single amino acid residue. Although the two cDNAs encode Na channels with substantially different activation properties (Auld, V. J., A. L. Goldin, D. S. Krafte, J. Marshall, J. M. Dunn, W. A. Catterall, H. A. Lester, N. Davidson, and R. J. Dunn. 1988. Neuron. 1:449-461), their inactivation properties resemble each other strongly but differ markedly from channels induced by poly(A+) rat brain RNA. Rat IIA currents inactivate more slowly, recover from inactivation more slowly, and display a steady-state voltage dependence that is shifted to more positive potentials. The macroscopic inactivation process for poly(A+) Na channels is defined by a single exponential time course; that for rat IIA channels displays two exponential components. At the single-channel level these differences in inactivation occur because rat IIA channels reopen several times during a depolarizing pulse; poly(A+) channels do not. Repetitive stimulation (greater than 1 Hz) produces a marked decrement in the rat IIA peak current and changes the waveform of the currents. When low molecular weight RNA is coinjected with rat IIA RNA, these inactivation properties are restored to those that characterize poly(A+) channels. Slow inactivation is similar for rat IIA and poly(A+) channels, however. The data suggest that activation and inactivation involve at least partially distinct regions of the channel protein
