    Molecular determinants of improved cathepsin B inhibition by new cystatins obtained by DNA shuffling

    Background: Cystatins are inhibitors of cysteine proteases. The majority are only weak inhibitors of human cathepsin B, which has been associated with cancer, Alzheimer's disease and arthritis. Results: Starting from the sequences of oryzacystatin-1 and canecystatin-1, a shuffling library was designed and a hybrid clone obtained, which presented higher inhibitory activity towards cathepsin B. This clone presented two unanticipated point mutations as well as an N-terminal deletion. Reversing each point mutation independently or both simultaneously abolishes the inhibitory activity towards cathepsin B. Homology modeling together with experimental studies of the reverse mutants revealed the likely molecular determinants of the improved inhibitory activity to be related to decreased protein stability. Conclusion: A combination of experimental approaches including gene shuffling, enzyme assays and reverse mutation allied to molecular modeling has shed light upon the unexpected inhibitory properties of certain cystatin mutants against Cathepsin B. We conclude that mutations disrupting the hydrophobic core of phytocystatins increase the flexibility of the N-terminus, leading to an increase in inhibitory activity.     OCCURRENCE OF CRYPTOSPORIDIUM SP. IN DOGS AND CATS FROM CURITIBA AND ITS METROPOLITAN AREA

    The present study was carried out with the aim of assess the occurrence of Cryptosporidium sp. infection in dogs and cats in Curitiba and its metropolitan area, state of Paraná, Brazil. Techniques employed to detect the protozoan in fecal samples were: staining by Ziehl-Neelsen for oocysts search and nested polymerase chain reaction (nPCR) targeting the 18SSU rDNA gene. To attempt the proposed aim, 91 feces samples of dogs and 25 of cats were collected and analyzed. Ziehl-Neelsen technique was unable to detect any oocyst in both groups analyzed, showing a very low sensitivity. Results of nPCR showed an infection rate of 13.2% (12/91) and 4% (1/25) in dogs and cats respectively.  The implications of these epidemiological data are discussed in this work

    The use of Maytenus ilicifolia to prevent cisplatin-induced ototoxicity

    Maytenus ilicifolia é uma planta sul americana apresenta várias propriedades medicinais, entre elas, a ação antioxidante. OBJETIVO: Por meio de um modelo original de ototoxicidade induzida pela cisplatina, verificar uma possível ação otoprotetora do extrato aquoso desta planta. MATERIAL E MÉTODO: Estudo clínico e experimental com cobaias fêmeas, albinas divididas em 5 grupos: 9 animais recebendo somente 3 doses de 7,5mg/kg/d do protocolo de cisplatina, 4 animais somente com o extrato, 10 animais com cisplatina e 1g/kg/d de extrato por 8 dias, 5 animais com cisplatina e 3g/kg/d do extrato por 8 dias e 5 animais recebendo extrato por 3 semanas e cisplatina na última semana. Os exames foram emissões otoacústica por produtos de distorção, potencial de tronco encefálico pré e após administração de cisplatina e, microscopia eletrônica de varredura. RESULTADOS: Os animais que receberam a cisplatina com o extrato, independente da dose, obtiveram alterações em todos os testes, com lesões na região basal na microscopia eletrônica. CONCLUSÃO: Apesar do efeito antioxidante da Maytenus ilicifolia, ela não foi suficiente para bloquear o efeito ototóxico da cisplatina.Maytenus ilicifolia is a native plant from South America, with several medicinal properties including antioxidant effects. AIM: using an original cisplatin induced ototoxicity model, we evaluated a possible otoprotection caused by Maytenus ilicifolia extract. MATERIALS AND METHODS: clinical and experimental study design with female albino guinea pigs divided in groups as follows: 9 animals receiving cisplatin only (three doses of 7.5mg/kg/day), 4 animals receiving the plant extract only, 10 animals receiving the cisplatin protocol and 1g/kg/day of extract for 8 days, 5 animals with cisplatin and 3g/kg/day of extract for 8 days, and 5 animals receiving extract for 3 weeks and cisplatin in the last week. The tests were distortion product otoacoustic emissions, brainstem auditory response, before and after medication and scanning electron microscopy. RESULTS: the animals receiving cisplatin plus plant extract, had alterations in all the tests, showing lesions on the basal cochlear region under electron microscopy. CONCLUSIONS: Despite of the plant extract's antioxidant effect, it was not sufficient to protect the cochlea against cisplatin ototoxicity

    Effect of Gender, Energetics, and Biomechanics on Swimming Masters Performance

    The purpose of this study was to analyze the effect of gender and energetics on biomechanics and performance of masters swimmers over 1 season. Twenty-five masters swimmers (14 male and 11 female) were assessed 3 times (TP1, TP2, and TP3) during a season (male personal record in 200-m freestyle event: 173.00 ± 31.41 seconds: female personal record in 200-m freestyle event: 200.73 ± 25.02 seconds). An incremental 5 × 200-m step test was selected to evaluate velocity at 4 mmol·l⁻¹ of blood lactate concentration (v4), maximal blood lactate concentration after exercise (La(peak)), maximal oxygen uptake (V̇O2max), stroke frequency, stroke length (SL), stroke index (SI), and propelling efficiency of the arm stroke (η(p)). The 200-m freestyle performance and average swimming velocity (v200) were also monitored. Significant differences were observed between males and females for the 200-m freestyle performance, SL, SI, and La(peak). Performance (205.18 ± 24.47 seconds; 197.45 ± 20.97 seconds; 193.45 ± 18.12 seconds), SL (1.69 ± 0.17 m; 1.79 ± 0.13 m; 1.78 ± 0.15 m), SI (1.68 ± 0.31 m²·c⁻¹·s⁻¹; 1.83 ± 0.27 m²·c⁻¹·s⁻¹; 1.85 ± 0.27 m²·c⁻¹·s⁻¹), η(p) (0.32 ± 0.04; 0.33 ± 0.03; 0.33 ± 0.04), and V̇O2max (38.71 ± 3.44 ml·kg⁻¹·min⁻¹; 43.43 ± 3.71 ml·kg⁻¹·min⁻¹; 43.95 ± 7.02 ml·kg⁻¹·min⁻¹) have changed significantly throughout the season (TP1, TP2, and TP3, respectively) in female swimmers. In male, significant changes were found in η(p) (0.33 ± 0.07; 0.36 ± 0.05; 0.36 ± 0.06) and V̇O2max (41.65 ± 7.30 ml·kg⁻¹·min⁻¹; 45.19 ± 6.55 ml·kg⁻¹·min⁻¹; 50.19 ± 9.65 ml·kg⁻¹·min⁻¹) over the season (TP1, TP2, and TP3, respectively). Gender presented a significant effect on SL (TP2: η(p)² = 0.29; TP3: η(p)² = 0.37), SI (TP2: η(p)² = 0.25), and La(peak) (TP3: η(p)² = 0.42). v4 (TP1: η(p)² = 0.23), SL (TP1: η(p)² = 0.46), SI (TP1: η(p)² = 0.78; TP2: η(p)² = 0.37; TP3: η(p)² = 0.32), and η(p) (TP1: η(p)² = 0.28) had a significant effect on performance. Male masters swimmers have better performance, SL, SI, and La(peak) than female counterparts. Female masters swimmers enhanced significantly the 200-m freestyle performance over the season due to the improvement in swimming technique (SL, SI, and η(p)) and energetic factors (v4 and V̇O2max). Nonsignificant improvements were observed for the males' performance. Gender has a significant effect on SL, SI, and La(peak). Com vista a uma avaliação correcta das fontes sismogénicas em futuros estudos de perigosidade sísmica, integraram-se dados de reflexão sísmica, com dados de métodos potenciais e dados de epicentros relocalizados. As informações recolhidas a partir dos parâmetros das falhas irão ajudar à localização de fontes de sismos históricos utilizando elementos finitos de simulação de movimentos fortes

    Optical gas sensing with liquid crystal droplets and convolutional neural networks

    UIDB/50009/2020 UIDB/ 04378/2020 SCENT-ERC-2014-STG-639123, 2015-2022Liquid crystal (LC)-based materials are promising platforms to develop rapid, miniaturised and low-cost gas sensor devices. In hybrid gel films containing LC droplets, characteristic optical texture variations are observed due to orientational transitions of LC molecules in the presence of distinct volatile organic compounds (VOC). Here, we investigate the use of deep convolutional neural networks (CNN) as pattern recognition systems to analyse optical textures dynamics in LC droplets exposed to a set of different VOCs. LC droplets responses to VOCs were video recorded under polarised optical microscopy (POM). CNNs were then used to extract features from the responses and, in separate tasks, to recognise and quantify the vapours exposed to the films. The impact of droplet diameter on the results was also analysed. With our classification models, we show that a single individual droplet can recognise 11 VOCs with small structural and functional differences (F1-score above 93%). The optical texture variation pattern of a droplet also reflects VOC concentration changes, as suggested by applying a regression model to acetone at 0.9–4.0% (v/v) (mean absolute errors below 0.25% (v/v)). The CNN-based methodology is thus a promising approach for VOC sensing using responses from individual LC-droplets.publishersversionpublishe
