    Diplomová práce je zaměřená na novostavbu umělecké školy ve Valašském Meziříčí. Objekt je třípodlažní, s jedním podzemním podlažím a dvěma nadzemními podlažími s plochou střechou. Nosná konstrukce je tvořena skeletem - železobetonové monolitické sloupy a železobetonová monolitická stropní deska. Výplňové zdivo je z keramických tvarovek. Objekt je zateplen minerální vatou. Podrobnější řešení diplomové práce se zabývá řešením hlavní části objektu umělecké školy, kde se nachází učebny a technické zázemí objektu. Ve vedlejších částech se nachází zázemí zaměstnanců, víceúčelový sál, kavárna a podzemní garáže.The final thesis is focused on a new building of School of arts in Valašské Meziříčí. The object has three floors, it has a basement and two above-ground floors witch a flat roof. A loadbearing structure is made of skeleton - reinforced concrete monolithic columns and reinforced concrete monolithic floor slabs. Filling masonry is made of ceramic tiles. Object has thermal insulation by mineral wool. Detailed solution of final thesis deals with the solution of main building School of arts, where are the schoolrooms and technical facilities. In the other parts of the object are a staff facilities, multipurpose hall, café and underground garage.

    Předmětem této bakalářské práce je návrh a projektová dokumentace dvougeneračního rodinného domu se dvěma bytovými jednotkami a s prodejnou uměleckých předmětů ve Vyškově. Rodinný dům je částečně podsklepený a má tři nadzemní podlaží. Součástí domu je i garáž pro oba byty. Objekt je zčásti zastřešen jednoplášťovou plochou střechou a z části pultovou střechou. Kromě vlastního projektu je součástí i architektonická studie, požární zpráva a tepelně technické posouzení domu.This Bachelor’s work is focused on the study and contract documents of one duplex family house with two dwelling units with artworks shop in Vyškov. The house is partly with a cellar and has three floors. A garage for both the flats belongs to the house too. The building is partly roofed with a single coat flat roof and partly with an aisle roof. The work also includes architectonical drawings, fire approval and technical appraisal of heating the house.

    Cílem této bakalářské práce je analýza recyklace plastů a analýza recyklačního procesu PET lahví ve společnosti PETKA CZ, a.s., kde jsem po dobu studia vykonával svou praxi. V neposlední řadě se práce zabývá způsoby, jak se získává vstupní materiál pro výrobu recyklátu z PET lahví, hospodařením s plastovým odpadem, návrhy na zlepšení ve výrobě tohoto podniku a zároveň možnostmi, jak snížit odpadovost při výrobě recyklátu. Dále tato práce popisuje legislativní úpravu ochrany životního prostředí, všeobecné principy recyklace, jak se recyklát společnosti PETKA CZ, a.s. v současné době využívá a zároveň, jak bychom mohli tento recyklát využít v budoucnosti.The aim of this bachelor's thesis is to analyze the recycling of plastics and the analysis of the recycling process of PET bottles in the company PETKA CZ, a.s. In the last analysis of the work influencing the ways to obtain input material for the production of recycling from PET bottles, management of plastic waste, proposals for improving the production of this company and at the same time the possibilities of reducing waste in the production of recycling. Furthermore, this work describes the legislative regulation of environmental protection, general principles of recycling, how to recycle the company PETKA CZ, a.s. It currently determines and at the same time how we could recycle this to use in the future.

    Constraining long-term denudation and faulting history in intraplate regions by multisystem thermochronology: An example of the Sudetic Marginal Fault (Bohemian Massif, central Europe)

    The Rychlebské hory Mountain region in the Sudetes (NE Bohemian Massif) provides a natural laboratory for studies of postorogenic landscape evolution. This work reveals both the exhumation history of the region and the paleoactivity along the Sudetic Marginal Fault (SMF) using zircon (U-Th)/He (ZHe), apatite fission track (AFT), and apatite (U-Th)/He (AHe) dating of crystalline basement and postorogenic sedimentary samples. Most significantly, and in direct contradiction of traditional paleogeographic reconstructions, this work has found evidence of a large Cretaceous sea and regional burial (to >6.5 km) of the Carboniferous-Permian basement in the Late Cretaceous (~95–80 Ma). During the burial by sediments of the Bohemian Cretaceous Basin System, the SMF acted as a normal fault as documented by offset ZHe ages across the fault. At 85–70 Ma, the basin was inverted, Cretaceous strata eroded, and basement blocks were exhumed to the near surface at a rate of ~300 m/Ma as evidenced by Late Cretaceous–Paleocene AFT ages and thermal modeling results. There is no appreciable difference in AFT and AHe ages across the fault, suggesting that the SMF acted as a reverse fault during exhumation. In the late Eocene–Oligocene, the basement was locally heated to <70°C by magmatic activity related to opening of the Eger rift system. Neogene or younger thermal activity was not recorded in the thermochronological data, confirming that late Cenozoic uplift and erosion of the basement blocks was limited to less than ∼1.5 km in the study area

    P2RX7 inhibitor suppresses exosome secretion and disease phenotype in P301S tau transgenic mice

    Background: Neuronal accumulation of misfolded microtubule-associated protein tau is a hallmark of neuropathology in Alzheimer’s disease, frontotemporal dementia, and other tauopathies, and has been a therapeutic target. Microglia can spread tau pathology by secreting tau-containing exosomes, although the specific molecular target is yet to be identified for the therapeutic intervention. P2X purinoceptor 7 (P2RX7) is an ATP-gated cation channel, enriched in microglia and triggers exosome secretion. The purpose of the study is to examine the therapeutic effect of an orally applicable, CNS-penetrant P2RX7 specific inhibitor on the early disease stage of a tauopathy mouse model. Methods: Three-months-old P301S tau mice were treated with P2RX7-specific inhibitor GSK1482160 or vehicle for 30 days, followed by behavioral, biochemical and immunohistochemical assessment. GSK1482160 was also tested for exosome secretion from primary cultured murine astrocytes, neurons and microglia in vitro. Results: Oral administration of GSK1482160 significantly reduced accumulation of MC1+ and Alz50+ misfolded tau in hippocampal regions, which was accompanied with reduced accumulation of Tsg101, an exosome marker, in hippocampal neurons. Proximity ligation assay demonstrated complex formation of Alz50+ tau and Tsg101 in hippocampal neurons, which was reduced by GSK1482160. On the other hand, GSK1482160 had no effect on microglial ramification or CD68 expression, which was significantly enhanced in P301S mice, or pro/anti-inflammatory cytokine gene expression. Strikingly, GSK1482160-treated P301S mice show significantly improved working and contextual memory as determined by Y-maze and fear conditioning tests. GSK1482160 also significantly increased accumulation of Tsg101 and CD81 in microglia in vivo, suggesting its suppression of P2RX7-induced exosome secretion from microglia. This effect was confirmed in vitro, as ATP-induced secretion of tau-containing exosome was significantly suppressed by GSK1482160 treatment from primary murine microglia, but not from neurons or astrocytes. Discussion: The oral administration of P2RX7 inhibition mitigates disease phenotypes in P301S mice, likely by suppressing release of microglial exosomes. P2RX7 could be a novel therapeutic target for the early stage tauopathy development

    Design of a new production line

    This diploma thesis deals with the project of implementing a production line in a selected company dealing with the recycling of plastic waste and the production of rPET flakes as a secondary raw material for the production of plastic products. First, it deals with the description of the current state and the production process in the company, including the layout of the equipment, and then defines the requirements for the project by the management of the company and individual shareholders of the company. Defining the requirements for the project and at the same time the method of obtaining key information took place on the basis of in-depth interviews with the interested parties of this project, while a pilot design of the layout of the machinery was created, including a description of the technology and a flowchart of the required production process. The main content of the design part is the creation of a proposal for a possible implementation of the production line using a network diagram using the PERT method, which indicates that the duration of the project was calculated to be 106.6 days. Together with the network diagram of the project, an analysis and proposals for managing the risks of this project were prepared. At the end of the draft chapter, a financial analysis of the project's payback period was carried out, as the condition of a payback period of 3-5 years must be met in order for the project to be approved by the shareholders, which this project fulfilled in all three expected progress scenarios
