    Common activation of canonical Wnt signaling in pancreatic adenocarcinoma.

    Pancreatic ductal adenocarcinoma (PDA) is an extremely aggressive malignancy, which carries a dismal prognosis. Activating mutations of the Kras gene are common to the vast majority of human PDA. In addition, recent studies have demonstrated that embryonic signaling pathway such as Hedgehog and Notch are inappropriately upregulated in this disease. The role of another embryonic signaling pathway, namely the canonical Wnt cascade, is still controversial. Here, we use gene array analysis as a platform to demonstrate general activation of the canonical arm of the Wnt pathway in human PDA. Furthermore, we provide evidence for Wnt activation in mouse models of pancreatic cancer. Our results also indicate that Wnt signaling might be activated downstream of Hedgehog signaling, which is an early event in PDA evolution. Wnt inhibition blocked proliferation and induced apoptosis of cultured adenocarcinoma cells, thereby providing evidence to support the development of novel therapeutical strategies for Wnt inhibition in pancreatic adenocarcinoma

    What, if anything, are hybrids: enduring truths and challenges associated with population structure and gene flow

    Hybridization is a potent evolutionary process that can affect the origin, maintenance, and loss of biodiversity. Because of its ecological and evolutionary consequences, an understanding of hybridization is important for basic and applied sciences, including conservation biology and agriculture. Herein, we review and discuss ideas that are relevant to the recognition of hybrids and hybridization. We supplement this discussion with simulations. The ideas we present have a long history, particularly in botany, and clarifying them should have practical consequences for managing hybridization and gene flow in plants. One of our primary goals is to illustrate what we can and cannot infer about hybrids and hybridization from molecular data; in other words, we ask when genetic analyses commonly used to study hybridization might mislead us about the history or nature of gene flow and selection. We focus on patterns of variation when hybridization is recent and populations are polymorphic, which are particularly informative for applied issues, such as contemporary hybridization following recent ecological change. We show that hybridization is not a singular process, but instead a collection of related processes with variable outcomes and consequences. Thus, it will often be inappropriate to generalize about the threats or benefits of hybridization from individual studies, and at minimum, it will be important to avoid categorical thinking about what hybridization and hybrids are. We recommend potential sampling and analytical approaches that should help us confront these complexities of hybridization

    Three-way symbolic tree-maps and ultrametrics

    Three-way dissimilarities are a generalization of (two-way) dissimilarities which can be used to indicate the lack of homogeneity or resemblance between any three objects. Such maps have applications in cluster analysis and have been used in areas such as psychology and phylogenetics, where three-way data tables can arise. Special examples of such dissimilarities are three-way tree-metrics and ultrametrics, which arise from leaf-labelled trees with edges labelled by positive real numbers. Here we consider three-way maps which arise from leaf-labelled trees where instead the interior vertices are labelled by an arbitrary set of values. For unrooted trees, we call such maps three-way symbolic tree-maps; for rooted trees, we call them three-way symbolic ultrametrics since they can be considered as a generalization of the (two-way) symbolic ultrametrics of Bocker and Dress. We show that, as with two- and three-way tree-metrics and ultrametrics, three-way symbolic tree-maps and ultrametrics can be characterized via certain k-point conditions. In the unrooted case, our characterization is mathematically equivalent to one presented by Gurvich for a certain class of edge-labelled hypergraphs. We also show that it can be decided whether or not an arbitrary three-way symbolic map is a tree-map or a symbolic ultrametric using a triplet-based approach that relies on the so-called BUILD algorithm for deciding when a set of 3-leaved trees or triplets can be displayed by a single tree. We envisage that our results will be useful in developing new approaches and algorithms for understanding 3-way data, especially within the area of phylogenetics

    Trophic stability and change across a sea ice cover gradient on the western Antarctic Peninsula

    The western Antarctic Peninsula (AP) is experiencing significant changes to sea ice cover, altering the macroalgal cover and potentially affecting the foundation of benthic food webs. We used fatty acid signatures as dietary and physiological trophic biomarkers to test the hypothesis that a gradient of 36-88% mean annual ice cover would affect the trophic ecology of fleshy macroalgae and diverse benthic invertebrate consumers along the western AP. We used SCUBA to collect organisms from benthic rocky nearshore habitats, 5-35 m depth, at 15 study sites during April-May of 2019. There were no consistent ecosystem-scale differences in the nutritionally important polyunsaturated fatty acids or other univariate fatty acid summary categories in either the seaweeds or invertebrates across the ice gradient, but we did find site-level differences in the multivariate fatty acid signatures of all seaweeds and invertebrates. Ice cover was a significant driver of the fatty acid signatures of 5 invertebrates, including 3 sessile (an anemone, a sponge, and a tunicate) and 2 mobile consumers (a sea star and a sea urchin). The multivariate fatty acid signatures of 2 other sea stars and a limpet were not affected by the ice gradient. These results indicate that the trophic ecology and resource assimilation of sessile consumers that are more connected to the macroalgal-derived food web will be more sensitive than mobile consumers to impending changes to annual ice and macroalgal cover along the western AP.National Science Foundation.Abstract -- Key words -- 1.     Bcl-2 protein family: Implications in vascular apoptosis and atherosclerosis

    Apoptosis has been recognized as a central component in the pathogenesis of atherosclerosis, in addition to the other human pathologies such as cancer and diabetes. The pathophysiology of atherosclerosis is complex, involving both apoptosis and proliferation at different phases of its progression. Oxidative modification of lipids and inflammation differentially regulate the apoptotic and proliferative responses of vascular cells during progression of the atherosclerotic lesion. Bcl-2 proteins act as the major regulators of extrinsic and intrinsic apoptosis signalling pathways and more recently it has become evident that they mediate the apoptotic response of vascular cells in response to oxidation and inflammation either in a provocative or an inhibitory mode of action. Here we address Bcl-2 proteins as major therapeutic targets for the treatment of atherosclerosis and underscore the need for the novel preventive and therapeutic interventions against atherosclerosis, which should be designed in the light of molecular mechanisms regulating apoptosis of vascular cells in atherosclerotic lesions

    Applying Contextual integrity to Open Data Publishing

    Open data publishing by both corporate and public bodies has increased significantly in recent years and this type of data could soon be developing into a real commodity. However, not all organisations pay sufficient heed to privacy as part of the decision-making process around open data publication, leaving both the organisation and the users whose data they handle vulnerable to privacy breaches. We present a case study in which we applied contextual integrity in practice, working with a UK local authority using real data. This illustrated how privacy can be incorporated into the decision-making process prior to publication taking place. Our results illustrate the application of Nissenbaum's Contextual Integrity Framework (CI) to the open data domain, and shows that CI is usable in practice
