    Alfalfa genomic selection for different stress-prone growing regions

    Alfalfa (Medicago sativa L.) selection for stress-prone regions has high priority for sustainable crop–livestock systems. This study assessed the genomic selection (GS) ability to predict alfalfa breeding values for drought-prone agricultural sites of Algeria, Morocco, and Argentina; managed-stress (MS) environments of Italy featuring moderate or intense drought; and one Tunisian site irrigated with moderately saline water. Additional aims were to investigate genotype × environment interaction (GEI) patterns and the effect on GS predictions of three single-nucleotide polymorphism (SNP) calling procedures, 12 statistical models that exclude or incorporate GEI, and allele dosage information. Our study included 127 genotypes from a Mediterranean reference population originated from three geographically contrasting populations, genotyped via genotyping-by-sequencing and phenotyped based on multi-year biomass dry matter yield of their dense-planted half-sib progenies. The GEI was very large, as shown by 27-fold greater additive genetic variance × environment interaction relative to the additive genetic variance and low genetic correlation for progeny yield responses across environments. The predictive ability of GS (using at least 37,969 SNP markers) exceeded 0.20 for moderate MS (representing Italian stress-prone sites) and the sites of Algeria and Argentina while being quite low for the Tunisian site and intense MS. Predictions of GS were complicated by rapid linkage disequilibrium decay. The weighted GBLUP model, GEI incorporation into GS models, and SNP calling based on a mock reference genome exhibited a predictive ability advantage for some environments. Our results support the specific breeding for each target region and suggest a positive role for GS in most regions when considering the challenges associated with phenotypic selection.EEA Santiago del EsteroFil: Annicchiarico, Paolo.     SILS MRAT: A Multi-Agent Decision-Support System for Shipboard Integration of Logistics Systems

    This report describes work performed by CDM Technologies Inc. on subcontract to ManTech Advanced Systems International, Inc. (Fairmont, West Virginia), and under sponsorship of the Office of Naval Research (ONR). The principal aim of the SILS (Shipboard Integration of Logistics Systems) project is to provide a decision-support capability for Navy ships that integrates shipboard logistical and tactical systems within a near real-time, automated, computer-based shipboard readiness and situation awareness facility. Specifically, SILS is intended to provide the captain of a ship and his staff with an accurate evaluation of the current condition of the ship, based on the ability of all of its equipment, services and personnel to perform their intended functions. The SILS software system consists of two main subsystems, namely: the SILS IE (Interface Engine) subsystem for information interchange with heterogeneous external applications, developed by ManTech Advanced Systems International; and, the SILS MRAT (Mission Readiness Analysis Toolkit) subsystem for intelligent decision-support with collaborative software agents, developed by CDM Technologies. This report is focused specifically on the technical aspects of the SILS MRAT subsystem. The automated reasoning capabilities of SILS MRAT are supported by a knowledge management architecture that is based on information-centric principles. Such an architecture utilizes a virtual model of the real world problem situation, consisting of data objects with characteristics and a rich set of relationships. Commonly referred to as an ontology, this internal information model provides a common vocabulary and context for software agents with reasoning capabilities. The concurrent need for incremental capability increases implies a steadily increasing data load from diverse operational (dynamic) and historical (static) data sources, ranging from free text messages and Web content to highly structured data contained in consolidated operational data stores, Data Warehouses, and Data Marts. In order to provide useful high-level capabilities the architecture is required to support the transformation of these data flows into information and knowledge relevant to the concerns and operational context of individual shipboard users. Accordingly, the system must be capable of not only storing data but also the relationships and higher level concepts that place the data into context. For this reason, to manage an increasing number of relationships and concepts over time, the SILS MRAT subsystem was designed to employ a formalized ontological framework. There were four additional considerations in the selection of the overall SILS architecture. First, utility to support a useful level of automated information management (i.e., the ability to collaboratively analyze data, monitor dynamic operational context, formulate warnings and alerts, and generate recommendations). Second, flexibility to accommodate contributions from multiple team members that may employ differing technologies and implementation paradigms. Third, scalability to allow a progressive increase in the breadth and diversity of the data sources, the volume of data processed, the number of validated components, and the intelligence of the tools (i.e., agents). Fourth, adaptability to facilitate the tailoring of the information management capabilities to different data sources and existing data environments. The current SILS architecture addresses these desirable characteristics by partitioning the system into a lower-level data collection and integration layer, a higher-level information management layer (SILS MRAT), and a translation facility that is capable of mapping the data schema of the lower layer to the information representation (i.e., ontology) of the upper layer (SILS IE). The higher-level information management layer provides a collaborative, distributed communication facility that supports the development of semi-autonomous modules of capability referred to as agents. The agents employ the formalized ontology supported by the communication facility to collaborate with each other and the human users in a meaningful manner

    Governance, Coordination and Evaluation: the case for an epistemological focus and a return to C.E. Lindblom

    While much political science research focuses on conceptualizing and analyzing various forms of governance, there remains a need to develop frameworks and criteria for governance evaluation (Torfing et al 2012). The post-positivist turn, influential in recent governance theory, emphasizes the complexity, uncertainty and the contested normative dimensions of policy analysis. Yet a central evaluative question still arises concerning the capacity of governance networks to facilitate ‘coordination’. The classic contributions of Charles Lindblom, although pre-dating the contemporary governance literature, can enable further elaboration of and engagement with this question. Lindblom’s conceptualisation of coordination challenges in the face of complexity shares with post-positivism a recognition of the inevitably contested nature of policy goals. Yet Lindblom suggests a closer focus on the complex, dynamically evolving, broadly ‘economic’ choices and trade-offs involved in defining and delivery policy for enabling these goals to be achieved and the significant epistemological challenges that they raise for policy-makers. This focus can complement and enrich both post-positivist scholarship and the process and incentives-orientated approaches which predominate in contemporary political science research on coordination in governance. This is briefly illustrated through a short case study evaluating governance for steering markets towards delivering low and zero carbon homes in England

    John Stuart Mill and Fourierism: ‘association’, ‘friendly rivalry’ and distributive justice

    John Stuart Mill’s self-description as ‘under the general designation of Socialist’ has been under-explored. It is an important feature of something else often overlooked: the importance of the French context of Mill’s thought. This article focuses on the role of Fourierism in the development of Mill’s ideas, exploring the links to Fourierism in Mill’s writing on profit-sharing; his use of the words ‘association’ and ‘friendly rivalry’; and his views concerning distributive justice. It then reconsiders his assessment of Fourierism as a desirable, workable and immediately implementable form of social reform, ultimately arguing it was Mill’s most-preferred form of ‘utopian’ socialism

    Liberalization, globalization and the dynamics of democracy in India

    In the closing decades of the twentieth century there has been an almost complete intellectual triumph of the twin principles of marketization (understood here as referring to the liberalization of domestic markets and freer international mobility of goods, services, financial capital and perhaps, more arguably, labour) and democratization . A paradigm shift of this extent and magnitude would not have occurred in the absence of some broad consensus among policymakers and (sections of) intellectuals around the globe on the desirability of such a change. There seems to be a two-fold causal nexus between marketization and democracy. The first is more direct, stemming from the fact of both systems sharing certain values and attitudes in common. But there is also a second more indirect chain from marketization to democracy, which is predicated via three sub-chains (i) from marketization to growth, (ii) from growth to overall material development welfare and (iii) from material development to social welfare and democracy. We examine each of these sub-links in detail with a view to obtaining a greater understanding of the hypothesized role of free markets in promoting democracies. In the later part of the paper we examine the socio-economic outcomes governing the quality of democracy in a specifically Indian context

    The SDF-1α/CXCR4 Axis is Required for Proliferation and Maturation of Human Fetal Pancreatic Endocrine Progenitor Cells

    The chemokine receptor CXCR4 and ligand SDF-1α are expressed in fetal and adult mouse islets. Neutralization of CXCR4 has previously been shown to diminish ductal cell proliferation and increase apoptosis in the IFNÎł transgenic mouse model in which the adult mouse pancreas displays islet regeneration. Here, we demonstrate that CXCR4 and SDF-1α are expressed in the human fetal pancreas and that during early gestation, CXCR4 colocalizes with neurogenin 3 (ngn3), a key transcription factor for endocrine specification in the pancreas. Treatment of islet like clusters (ICCs) derived from human fetal pancreas with SDF-1α resulted in increased proliferation of epithelial cells in ICCs without a concomitant increase in total insulin expression. Exposure of ICCs in vitro to AMD3100, a pharmacological inhibitor of CXCR4, did not alter expression of endocrine hormones insulin and glucagon, or the pancreatic endocrine transcription factors PDX1, Nkx6.1, Ngn3 and PAX4. However, a strong inhibition of ÎČ cell genesis was observed when in vitro AMD3100 treatment of ICCs was followed by two weeks of in vivo treatment with AMD3100 after ICC transplantation into mice. Analysis of the grafts for human C-peptide found that inhibition of CXCR4 activity profoundly inhibits islet development. Subsequently, a model pancreatic epithelial cell system (CFPAC-1) was employed to study the signals that regulate proliferation and apoptosis by the SDF-1α/CXCR4 axis. From a selected panel of inhibitors tested, both the PI 3-kinase and MAPK pathways were identified as critical regulators of CFPAC-1 proliferation. SDF-1α stimulated Akt phosphorylation, but failed to increase phosphorylation of Erk above the high basal levels observed. Taken together, these results indicate that SDF-1α/CXCR4 axis plays a critical regulatory role in the genesis of human islets

    Institutional Solutions to Precariousness and Inequality in Labour Markets

    It has become widely assumed that the standard employment relationship (SER) is in irreversible decline in industrialized societies. However, non-standard and precarious work relationships often complement the SER via labour market transitions, and are not displacing it as the focal point of labour market regulation. The co-ordination and risk management functions of the SER continue to be relevant in market economies, and the SER is adjusting to new conditions. The SER has a complex and evolving relationship to gender and to social stratification. In the European context where the SER originated and achieved its clearest legal expression, institutional solutions to precariousness and inequality are being developed, the most innovative of which avoid simple deregulation in favour of integrated policy responses involving a range of complementary regulatory mechanisms.We are grateful for funding from the Cambridge Political Economy Society TrustThis is the author accepted manuscript.
