    Creating a classroom culture that promotes inquiry-oriented discussions: Reading and talking about multiple texts

    In this report, we provide a rationale for making inquiry-oriented discussions a more integral part of classroom practice. We begin by identifying five features that make this type distinct from other types of discussion. Then, in the heart of the report we explain the concepts and practices that are central to creating inquiry-oriented discussions. We discuss these concepts and practices by depicting a framework of sorts for designing, implementing, and evaluating inquiry-oriented discussions. The framework focuses on nine elements key to making inquiry-oriented discussions more integral to classroom practice. Finally, we provide suggestions for getting started with inquiry-oriented discussions, and address questions often raised by those who start these discussions in their classroom

    Exploring the psychometric properties of the Working Alliance Inventory in general practice : a cross-sectional study

    Contains fulltext : 232432.pdf (Publisher’s version ) (Open Access)BACKGROUND: The therapeutic alliance is a framework from psychology that describes three components: goals, tasks, and bond. The Working Alliance Inventory adapted for general practice (WAI-GP) measures the strength of the therapeutic alliance between the patient and the clinician, and it could be useful in both research and clinical settings. AIM: To determine if the patient score on WAI-GP can delineate the three components (goals, tasks, and bond), and to test concurrent validity with the Consultation and Relational Empathy (CARE) measure and the Patient Perception of Patient-Centredness (PPPC) measure. DESIGN & SETTING: A cross-sectional study took place in 12 general practice waiting rooms in Australia. METHOD: The research instruments included the 12-item WAI-GP (the patient version), the CARE and PPPC measures, plus a survey of demographics and reason for consultation. To perform a principal components factor analysis of the WAI-GP, this dataset was combined with an existing dataset. The Spearman rank correlation was used to determine concurrent validity between the WAI-GP and the CARE and PPPC measures. RESULTS: Participants (97-99%) reported a strong positive alliance after the consultation (average WAI-GP mean 4.27 ± 0.67 out of 5, n = 146). Factor analysis could not separate the three components (one factor, eigenvalue >1; Cronbach's α = 0.957; n = 281). Concurrent validity was supported by moderate correlations with the other measures (PPPC ρ = -0.51, P<0.005, CARE ρ = 0.56, P<0.005). CONCLUSION: Three components could not be identified, but the WAI-GP has a high internal consistency and concurrent validity with moderate correlations with the CARE and PPPC. A more diverse sample may better distinguish the three components leading to more specific feedback to clinicians on their consultation practices

    Crohn\u27s disease-associated ATG16L1 T300A genotype is associated with improved survival in gastric cancer

    BACKGROUND: A non-synonymous single nucleotide polymorphism of the ATG16L1 gene, T300A, is a major Crohn\u27s disease (CD) susceptibility allele, and is known to be associated with increased apoptosis induction in the small intestinal crypt base in CD subjects and mouse models. We hypothesized that ATG16L1 T300A genotype also correlates with increased tumor apoptosis and therefore could lead to superior clinical outcome in cancer subjects. METHODS: T300A genotyping by Taqman assay was performed for gastric carcinoma subjects who underwent resection from two academic medical centers. Transcriptomic analysis was performed by RNA-seq on formalin-fixed paraffin-embedded cancerous tissue. Tumor apoptosis and autophagy were determined by cleaved caspase-3 and p62 immunohistochemistry, respectively. The subjects\u27 genotypes were correlated with demographics, various histopathologic features, transcriptome, and clinical outcome. FINDINGS: Of the 220 genotyped subjects, 163 (74%) subjects carried the T300A allele(s), including 55 (25%) homozygous and 108 (49%) heterozygous subjects. The T300A/T300A subjects had superior overall survival than the other groups. Their tumors were associated with increased CD-like lymphoid aggregates and increased tumor apoptosis without concurrent increase in tumor mitosis or defective autophagy. Transcriptomic analysis showed upregulation of WNT/β-catenin signaling and downregulation of PPAR, EGFR, and inflammatory chemokine pathways in tumors of T300A/T300A subjects. INTERPRETATION: Gastric carcinoma of subjects with the T300A/T300A genotype is associated with repressed EGFR and PPAR pathways, increased tumor apoptosis, and improved overall survival. Genotyping gastric cancer subjects may provide additional insight for clinical stratification

    Epitope Addition and Ablation via Manipulation of a Dengue Virus Serotype 1 Infectious Clone

    ABSTRACT Despite the clinical relevance, dengue virus (DENV) research has been hampered by the absence of robust reverse genetic systems to manipulate the viral serotypes for propagation and generation of mutant viruses. In this article, we describe application of an infectious clone system for DENV serotype 1 (DENV1). Similar to previous clones in both flaviviruses and coronaviruses, the approach constructs a panel of contiguous cDNAs that span the DENV genome and can be systematically and directionally assembled to produce viable, full-length viruses. Comparison of the virus derived from the infectious clone with the original viral isolate reveals identical sequence, comparable endpoint titers, and similar focus staining. Both focus-forming assays and percent infection by flow cytometry revealed overlapping replication levels in two different cell types. Moreover, serotype-specific monoclonal antibodies (MAbs) bound similarly to infectious clone and the natural isolate. Using the clone, we were able to insert a DENV4 type-specific epitope recognized by primate MAb 5H2 into envelope (E) protein domain I (EDI) of DENV1 and recover a viable chimeric recombinant virus. The recombinant DENV1 virus was recognized and neutralized by the DENV4 type-specific 5H2 MAb. The introduction of the 5H2 epitope ablated two epitopes on DENV1 EDI recognized by human MAbs (1F4 and 14C10) that strongly neutralize DENV1. Together, the work demonstrates the utility of the infectious clone and provides a resource to rapidly manipulate the DENV1 serotype for generation of recombinant and mutant viruses. IMPORTANCE Dengue viruses (DENVs) are significant mosquito-transmitted pathogens that cause widespread infection and can lead to severe infection and complications. Here we further characterize a novel and robust DENV serotype 1 (DENV1) infectious clone system that can be used to support basic and applied research. We demonstrate how the system can be used to probe the antigenic relationships between strains by creating viable recombinant viruses that display or lack major antibody epitopes. The DENV1 clone system and recombinant viruses can be used to analyze existing vaccine immune responses and inform second-generation bivalent vaccine designs

    A synchrotron self-Compton model with low energy electron cut-off for the blazar S5 0716+714

    Rapid inverse Compton cooling sets in when the brightness temperature (T_B) of a self-absorbed synchrotron source with power-law electrons reaches ~10^{12} K. However, T_B inferred from observations of intra-day variable sources (IDV) are well above the "Compton catastrophe" limit. This can be understood if the underlying electron distribution cuts off at low energy. We approximate a low-energy cut-off with monoenergetic electrons. We compute the synchrotron self-Compton (SSC) spectrum of such distribution, and using the IDV source S5~0716+714 as an example, we compare it to the observed SED of S5~0716+714. The hard radio spectrum is well-fitted by this model, and the optical data can be accommodated by a power-law extension to the electron spectrum. We therefore examine the scenario of an injection of electrons that is a double power law in energy with a hard low-energy component that does not contribute to the synchrotron opacity. We show that the double power-law injection model is in good agreement with the observed SED of S5~0716+714. For intrinsic variability, we find that a Doppler factor of D\geq30 can explain the observed SED provided that low-frequency (<32 GHz) emission originates from a larger region than the higher-frequency emission. To fit the entire spectrum, D\geq65 is needed. We find the constraint imposed by induced Compton scattering at high T_B is insignificant in our model. We confirm that electron distribution with a low-energy cut-off can explain the high T_B in compact radio sources. We show that synchrotron spectrum from such distributions naturally accounts for the observed hard radio continuum with a softer optical component, without the need for an inhomogeneous source.Comment: 13 pages, 5 figures, 1 table, to appear in A&A; refereces removed from caption of Fig.3, added acknowledgemen

    Limited T Cell Receptor Diversity of HCV-specific T Cell Responses Is Associated with CTL Escape

    Escape mutations are believed to be important contributors to immune evasion by rapidly evolving viruses such as hepatitis C virus (HCV). We show that the majority of HCV-specific cytotoxic T lymphocyte (CTL) responses directed against viral epitopes that escaped immune recognition in HCV-infected chimpanzees displayed a reduced CDR3 amino acid diversity when compared with responses in which no CTL epitope variation was detected during chronic infection or with those associated with protective immunity. Decreased T cell receptor (TCR) CDR3 amino acid diversity in chronic infection could be detected long before the appearance of viral escape mutations in the plasma. In both chronic and resolved infection, identical T cell receptor clonotypes were present in liver and peripheral blood. These findings provide a deeper understanding of the evolution of CTL epitope variations in chronic viral infections and highlight the importance of the generation and maintenance of a diverse TCR repertoire directed against individual epitopes

    We study Vasiliev's system of higher spin gauge fields coupled to massive scalars in AdS_3, and compute the tree level two and three point functions. These are compared to the large N limit of the W_N minimal model, and nontrivial agreements are found.
