    Optimal Investment in the Development of Oil and Gas Field

    Let an oil and gas field consists of clusters in each of which an investor can launch at most one project. During the implementation of a particular project, all characteristics are known, including annual production volumes, necessary investment volumes, and profit. The total amount of investments that the investor spends on developing the field during the entire planning period we know. It is required to determine which projects to implement in each cluster so that, within the total amount of investments, the profit for the entire planning period is maximum. The problem under consideration is NP-hard. However, it is solved by dynamic programming with pseudopolynomial time complexity. Nevertheless, in practice, there are additional constraints that do not allow solving the problem with acceptable accuracy at a reasonable time. Such restrictions, in particular, are annual production volumes. In this paper, we considered only the upper constraints that are dictated by the pipeline capacity. For the investment optimization problem with such additional restrictions, we obtain qualitative results, propose an approximate algorithm, and investigate its properties. Based on the results of a numerical experiment, we conclude that the developed algorithm builds a solution close (in terms of the objective function) to the optimal one

    Demethylation and cleavage of dimethylsulfoniopropionate and reduction of dimethyl sulfoxide by sulfate-reducing bacteria

    Many marine algae contain high concentrations of dimethylsulfoniopropionate (DMSP); most likely this compound functions mainly as an osmolyte. In anoxic marine sediments DMSP can be degraded in two ways: via an initial demethylation, or via a cleavage to dimethyl sulfide (DMS) and acrylate. Although the occurrence of these processes in sediments was known, the types of organisms responsible for them were not. Recent data from out laboratory, however, have shown that certain types of sulfate-reducing bacteria can carry out a demethylation of DMSP, whereas another sulfate reducer was found to cleave DMSP to DMS and acrylate, which was reduced to propionate. Thus, sulfate-reducing bacteria might be responsible for at least a part of the observed DMSP transformations in anoxic sediments. It was also shown that a well-known oxidation product of DMS, dimethyl sulfoxide, can function as an alternative electron acceptor in the metabolism of some marine sulfate reducers.     How a Diverse Research Ecosystem Has Generated New Rehabilitation Technologies: Review of NIDILRR’s Rehabilitation Engineering Research Centers

    Over 50 million United States citizens (1 in 6 people in the US) have a developmental, acquired, or degenerative disability. The average US citizen can expect to live 20% of his or her life with a disability. Rehabilitation technologies play a major role in improving the quality of life for people with a disability, yet widespread and highly challenging needs remain. Within the US, a major effort aimed at the creation and evaluation of rehabilitation technology has been the Rehabilitation Engineering Research Centers (RERCs) sponsored by the National Institute on Disability, Independent Living, and Rehabilitation Research. As envisioned at their conception by a panel of the National Academy of Science in 1970, these centers were intended to take a “total approach to rehabilitation”, combining medicine, engineering, and related science, to improve the quality of life of individuals with a disability. Here, we review the scope, achievements, and ongoing projects of an unbiased sample of 19 currently active or recently terminated RERCs. Specifically, for each center, we briefly explain the needs it targets, summarize key historical advances, identify emerging innovations, and consider future directions. Our assessment from this review is that the RERC program indeed involves a multidisciplinary approach, with 36 professional fields involved, although 70% of research and development staff are in engineering fields, 23% in clinical fields, and only 7% in basic science fields; significantly, 11% of the professional staff have a disability related to their research. We observe that the RERC program has substantially diversified the scope of its work since the 1970’s, addressing more types of disabilities using more technologies, and, in particular, often now focusing on information technologies. RERC work also now often views users as integrated into an interdependent society through technologies that both people with and without disabilities co-use (such as the internet, wireless communication, and architecture). In addition, RERC research has evolved to view users as able at improving outcomes through learning, exercise, and plasticity (rather than being static), which can be optimally timed. We provide examples of rehabilitation technology innovation produced by the RERCs that illustrate this increasingly diversifying scope and evolving perspective. We conclude by discussing growth opportunities and possible future directions of the RERC program

    A cardinal role for cathepsin D in co-ordinating the host-mediated apoptosis of macrophages and killing of pneumococci

    The bactericidal function of macrophages against pneumococci is enhanced by their apoptotic demise, which is controlled by the anti-apoptotic protein Mcl-1. Here, we show that lysosomal membrane permeabilization (LMP) and cytosolic translocation of activated cathepsin D occur prior to activation of a mitochondrial pathway of macrophage apoptosis. Pharmacological inhibition or knockout of cathepsin D during pneumococcal infection blocked macrophage apoptosis. As a result of cathepsin D activation, Mcl-1 interacted with its ubiquitin ligase Mule and expression declined. Inhibition of cathepsin D had no effect on early bacterial killing but inhibited the late phase of apoptosis-associated killing of pneumococci in vitro. Mice bearing a cathepsin D-/- hematopoietic system demonstrated reduced macrophage apoptosis in vivo, with decreased clearance of pneumococci and enhanced recruitment of neutrophils to control pulmonary infection. These findings establish an unexpected role for a cathepsin D-mediated lysosomal pathway of apoptosis in pulmonary host defense and underscore the importance of apoptosis-associated microbial killing to macrophage function

    The microaerophilic microbiota of de-novo paediatric inflammatory bowel disease: the BISCUIT study

    <p>Introduction: Children presenting for the first time with inflammatory bowel disease (IBD) offer a unique opportunity to study aetiological agents before the confounders of treatment. Microaerophilic bacteria can exploit the ecological niche of the intestinal epithelium; Helicobacter and Campylobacter are previously implicated in IBD pathogenesis. We set out to study these and other microaerophilic bacteria in de-novo paediatric IBD.</p> <p>Patients and Methods: 100 children undergoing colonoscopy were recruited including 44 treatment naïve de-novo IBD patients and 42 with normal colons. Colonic biopsies were subjected to microaerophilic culture with Gram-negative isolates then identified by sequencing. Biopsies were also PCR screened for the specific microaerophilic bacterial groups: Helicobacteraceae, Campylobacteraceae and Sutterella wadsworthensis.</p> <p>Results: 129 Gram-negative microaerophilic bacterial isolates were identified from 10 genera. The most frequently cultured was S. wadsworthensis (32 distinct isolates). Unusual Campylobacter were isolated from 8 subjects (including 3 C. concisus, 1 C. curvus, 1 C. lari, 1 C. rectus, 3 C. showae). No Helicobacter were cultured. When comparing IBD vs. normal colon control by PCR the prevalence figures were not significantly different (Helicobacter 11% vs. 12%, p = 1.00; Campylobacter 75% vs. 76%, p = 1.00; S. wadsworthensis 82% vs. 71%, p = 0.312).</p> <p>Conclusions: This study offers a comprehensive overview of the microaerophilic microbiota of the paediatric colon including at IBD onset. Campylobacter appear to be surprisingly common, are not more strongly associated with IBD and can be isolated from around 8% of paediatric colonic biopsies. S. wadsworthensis appears to be a common commensal. Helicobacter species are relatively rare in the paediatric colon.</p&gt

    On the ability of spectroscopic SZ effect measurements to determine the temperature structure of galaxy clusters

    (abridged) We explore in this paper the ability of spatially resolved spectroscopic measurements of the SZ effect (SZE) to determine the temperature profile of galaxy clusters. We derive a general formalism for the thermal SZE in galaxy clusters with a non-uniform temperature profile that can be applied to both cool-core clusters and non-cool core cluster with an isothermal or non-isothermal temperature structure. We derive an inversion technique through which the electron distribution function can be extracted from spectroscopic SZE observations over a wide frequency range. We study the fitting procedure to extract the cluster temperature from a set of simulated spatially resolved spectroscopic SZE observations in different bands of the spectrum, from 100 to 450 GHz. The results of our analysis for three different cluster prototypes (A2199 with a low-temperature cool core, Perseus with a relatively high-temperature cool core, Ophiuchus with an isothermal temperature distribution) provide both the required precision of the SZE observations and the optimal frequency bands for a determination of the cluster temperature similar or better than that obtainable from X-ray observations. The precision of SZE-derived temperature is also discussed for the outer regions of clusters. We also study the possibility to extract, from our method, the parameters characterizing the non-thermal SZE spectrum of the relativistic plasma contained in the lobes of radio galaxies as well as the spectrum of relativistic electrons co-spatially distributed with the thermal plasma in clusters with non-thermal phenomena.     Testing the feasibility of the Dignity Therapy interview: adaptation for the Danish culture

    <p>Abstract</p> <p>Background</p> <p><b>'</b>Dignity Therapy' (DT) is a brief, flexible intervention, which allows patients to complete an interview and create a document regarding their life, identity and what they want to leave in writing for their loved ones. DT is based on the DT Question Protocol. Developed and tested in English speaking settings, DT has proven to be a feasible and effective way to enhance patient dignity, while diminishing suffering and depression. The aim of this study was to test the acceptability and feasibility of the DT Question Protocol among Danish health professionals and cancer patients, and to obtain preliminary estimates of patient uptake for DT. These results will be used to inform a larger evaluation study.</p> <p>Method</p> <p>Ten professionals were interviewed about their perception of DT and the Question Protocol. It was then tested with 20 patients at two palliative care sites and one gynecologic oncology department. Data was analyzed using content analysis techniques to evaluate the protocol for relevance, acceptability and comprehension. The interest and relevance of the intervention was also determined by examining the preliminary participation rate.</p> <p>Results</p> <p>Overall, DT was perceived to be comprehensible and relevant. Professionals highlighted six concerns that might warrant modification. These issues were examined using patient data. Some of their concerns overlapped with those raised by the professionals (e.g. <it>'unacceptable self-praise' </it>and '<it>interference with the lives of others'</it>). Tailoring DT to Danish culture required easily accommodated adjustments to the procedures and the DT Question Protocol. Some concerns expressed by health professionals may have reflected protectiveness toward the patients. While the intervention was relevant and manageable for patients admitted to palliative care, DT was less easily implemented at the gynecologic oncology department.</p> <p>Conclusion</p> <p>Based on patients' and professionals' reaction to the DT Question Protocol, and based on the preliminary proportion of participants accepting DT, the DT question protocol - with minor adaptations - appears to be a manageable, acceptable and relevant intervention for Danish patients admitted to palliative care.</p

    Impact of environmental moisture on C(3)A polymorphs in the absence and presence of CaSO4 center dot 0.5 H2O

    The phenomenon of water vapour sorption by anhydrous C3A polymorphs both in the absence and in the presence of CaSO4·0·5 H2O was studied utilising dynamic and static sorption methods. It was found that orthorhombic C3A starts to sorb water at 55% relative humidity (RH) and cubic C3A at 80% RH. Also, C3Ao sorbs a higher amount of water which is predominantly physically bound, whereas C3Ac preferentially interacts with water by chemical reaction. In the presence of calcium sulfate hemihydrate, ettringite was observed as the predominant pre-hydration product for both C3A modifications: that is, ion transport had occurred between C3A and sulfate. Environmental scanning electron microscopic imaging revealed that in a moist atmosphere, a liquid water film condenses on the surface of the phases as a consequence of capillary condensation between the particles. C3A and sulfate can then dissolve and react with each other. Seemingly, pre-hydration is mainly facilitated through capillary condensation and less through surface interaction with gaseous water molecules
