    Bounding film drainage in common thin films

    A review of thin film drainage models is presented in which the predictions of thinning velocities and drainage times are compared to reported values on foam and emulsion films found in the literature. Free standing films with tangentially immobile interfaces and suppressed electrostatic repulsion are considered, such as those studied in capillary cells. The experimental thinning velocities and drainage times of foams and emulsions are shown to be bounded by predictions from the Reynolds and the theoretical MTsR equations. The semi-empirical MTsR and the surface wave equations were the most consistently accurate with all of the films considered. These results are used in an accompanying paper to develop scaling laws that bound the critical film thickness of foam and emulsion films

    Uncoupling of p97 ATPase activity has a dominant negative effect on protein extraction

    p97 is a highly abundant, homohexameric AAA+ ATPase that performs a variety of essential cellular functions. Characterized as a ubiquitin-selective chaperone, p97 recognizes proteins conjugated to K48-linked polyubiquitin chains and promotes their removal from chromatin and other molecular complexes. Changes in p97 expression or activity are associated with the development of cancer and several related neurodegenerative disorders. Although pathogenic p97 mutations cluster in and around p97's ATPase domains, mutant proteins display normal or elevated ATPase activity. Here, we show that one of the most common p97 mutations (R155C) retains ATPase activity, but is functionally defective. p97-R155C can be recruited to ubiquitinated substrates on chromatin, but is unable to promote substrate removal. As a result, p97-R155C acts as a dominant negative, blocking protein extraction by a similar mechanism to that observed when p97's ATPase activity is inhibited or inactivated. However, unlike ATPase-deficient proteins, p97-R155C consumes excess ATP, which can hinder high-energy processes. Together, our results shed new insight into how pathogenic mutations in p97 alter its cellular function, with implications for understanding the etiology and treatment of p97-associated diseases

    An informed thought experiment exploring the potential for a paradigm shift in aquatic food production

    The Neolithic Revolution began c. 10000 years ago and is characterised by the ultimate, near complete transition from hunting and gathering to agricultural food production on land. The Neolithic Revolution is thought to have been catalysed by a combination of local population pressure, cultural diffusion, property rights and climate change. We undertake a thought experiment that examines trends in these key hypothesised catalysts and patters of today to explore whether society could be on a path towards another paradigm shift in food production: away from hunting of wild fish towards a transition to mostly fish farming. We find similar environmental and cultural pressures have driven the rapid rise of aquaculture, during a period that has now been coined the Blue Revolution, providing impetus for such a transition in coming decades to centuries. We also highlight the interacting and often mutually reinforcing impacts of 1)technological and scientific advancement, 2)environmental awareness and collective action and 3)globalisation and trade influencing the trajectory and momentum of the Blue Revolution. We present two qualitative narratives that broadly fall within two future trajectories: 1)a ubiquitous aquaculture transition and 20commercial aquaculture and fisheries coexistence. This scenarios approach aims to encourage logical, forward thinking, and innovative solutions to complex systems dynamics. Scenario-based thought experiments are useful to explore large scale questions, increase the accessibility to a wider readership and ideally catalyse discussion around proactive governance mechanisms. We argue the future is not fixed and society now has greater foresight and capacity to choose the workable balance between fisheries sand aquaculture that supports economic, environmental, cultural and social objectives through combined planning, policies and management

    Macromolecular interactions during gelatinisation and retrogradation in starch-whey systems as studied by rapid visco-analyser

    Gelatinisation and retrogradation of starch-whey mixtures were studied in water (pH 7) using the Rapid Visco-Analyser (RVA). The starch:whey ratios ranged from 0:100 - 100:0. Wheat starch, and whey protein concentrate (about 80% solids basis) and isolate (about 96% solids basis) were used. Mixtures with whey isolates were generally more viscous than those with whey concentrates, and this was attributed to fewer non-protein milk components in the former. Whey protein concentrates and isolates reduced the peak, trough and final viscosities of the mixtures, but the breakdown and setback ratios of the mixtures were increased. The gelatinisation temperature increased with whey substitutions indicating that whey protein delayed starch gelatinisation. The temperature of fastest viscosity development decreased as the amount of whey was increased. Whey protein isolate generally exercised a lesser effect than the concentrate. At between 40 - 50% whey substitutions, the dominant phase changed from starch to protein irrespective of the source of the whey protein. An additive law poorly defined selected RVA parameters. Both macromolecules interacted to define the viscosity of the mixture, and an exponential model predicted the viscosity better than the additive law. The results obtained in this study are discussed to assist the understanding of extrusion processing of starch-whey systems as models for whey-fortified snack and ready-to-eat foods.     Late Quaternary uplift along the North America-Caribbean plate boundary: Evidence from the sea level record of Guantanamo Bay, Cuba

    The tectonic setting of the North America-Caribbean plate boundary has been studied intensively, but some aspects are still poorly understood, particularly along the Oriente fault zone. Guantanamo Bay, southern Cuba, is considered to be on a coastline that is under a transpressive tectonic regime along this zone, and is hypothesized to have a low uplift rate. We tested this by studying emergent reef terrace deposits around the bay. Reef elevations in the protected, inner part of the bay are ~11e12 m and outercoast, wave-cut benches are as high as ~14 m. Uranium-series analyses of corals yield ages ranging from ~133 ka to ~119 ka, correlating this reef to the peak of the last interglacial period, marine isotope stage (MIS) 5.5. Assuming a span of possible paleo-sea levels at the time of the last interglacial period yields long-term tectonic uplift rates of 0.02e0.11 m/ka, supporting the hypothesis that the tectonic uplift rate is low. Nevertheless, on the eastern and southern coasts of Cuba, east and west of Guantanamo Bay, there are flights of multiple marine terraces, at higher elevations, that could record a higher rate of uplift, implying that Guantanamo Bay may be anomalous. Southern Cuba is considered to have experienced a measurable but modest effect from glacial isostatic adjustment (GIA) processes. Thus, with a low uplift rate, Guantanamo Bay should show no evidence of emergent marine terraces dating to the ~100 ka (MIS 5.3) or ~80 ka (MIS 5.1) sea stands and results of the present study support this

    Is hypoxia’s influence restricted to the deep? Evaluation of nearshore community composition in Hood Canal, Washington, a seasonally hypoxic estuary

    Hypoxia [dissolved oxygen (DO) \u3c 2 mg L-1] has been identified as a key threat to the Puget Sound ecosystem, particularly in Hood Canal. Hood Canal is subject to seasonal hypoxia in its southern reaches, and prior work has demonstrated avoidance patterns of demersal species from the deep, offshore hypoxia-impacted waters. However, the non-lethal impact of low DO conditions on the nearshore community is not well understood, despite its importance to the estuary (e.g., nursery habitat). We evaluated the nature and extent of the sub-lethal influence of hypoxia on the nearshore community using underwater video monitoring techniques. Within two regions of Hood Canal, a southern highly impacted region and a northern reference region, we recorded weekly underwater video of the benthos via transects at three depths (10, 20, 30m) to measure species density and composition. Weekly monitoring of water quality revealed strong differences in DO over time and space, with the vertical extent of low DO waters increasing markedly at the end of summer in the south. While we were unable to detect acute shifts in nearshore densities, the community composition was significantly different between the two study regions; the south was primarily composed of hypoxia tolerant invertebrates and fewer fish species compared to the north. Moreover, the tolerant invertebrates displayed a three-fold increase in presence below a specific DO threshold (mean threshold ± SE = 3.95 mg L-1 ± 0.22), while the more sensitive species (e.g., fish) declined. Post-hoc comparisons of our findings to long-term DO trends in Hood Canal revealed the potential for a more persistent low DO state in the southern reaches. As a result, this study provides further insight into the complex regional differences in community structure and potential sensitivity of the nearshore community to other perturbations in Hood Canal
