Plate bearing tests for working platforms
During piling and other construction works, a working platform is often constructed across the site. These platforms comprise aggregate material placed and compacted to a designed thickness. Satisfactory performance of the platform may be confirmed by a plate bearing test. Current guidance given on plate bearing testing of granular soils suggests that the plate be at least five times the nominal size of the coarsest material. For a working platform this may be large and the reaction load required from plant and resources to carry out the bearing test may become excessively high. The aim of the research presented in this paper was to investigate the effect of particle to plate size ratios to establish if the use of a smaller plate would still allow a reliable test to be performed on site. Plate bearing tests were carried out in a centrifuge using a large, coarse grained limestone. The limestone was graded to a scale representation of 6F2 material, a commonly specified particle size distribution for working platforms. The size of plate was varied and the load displacement response recorded. The measured bearing capacity was correlated with the ratio of particle to plate size

Centrifuge modelling to determine the influence of pile stiffness on pile capacity
The stiffness of piles relates to their ability to resist deformation in response to an applied force. The modulus of elasticity of an uncracked concrete piles typically varies between 30-40kN/m2. Under axial loading these high stiffness piles transfer the load through the pile to the base. This results in low mobilisation of shaft friction as the stiff pile displaces uniformly and therefore the magnitude of skin friction along the entire length of the pile is small. In addition, base resistance of deep piles is mobilised at very high loads which may exceed the working load of the pile shaft. The adhesion factor, α, for bored piles in London Clay can range between 0.45 and 0.6 suggesting that a significant proportion of the soil strength cannot be mobilised. This low mobilisation of shaft resistance means that the ultimate bearing capacity is much reduced. The research investigated the behaviour of a low stiffness pile under axial load and compared this with a conventional high stiffness pile. The results demonstrated that the low stiffness pile exhibited marginally greater capacity at working load and a noticeably improved capacity at ultimate load
Meta-analysis of IDH-mutant cancers identifies EBF1 as an interaction partner for TET2
Isocitrate dehydrogenase (IDH) genes 1 and 2 are frequently mutated in acute myeloid leukaemia (AML), low-grade glioma, cholangiocarcinoma (CC) and chondrosarcoma (CS). For AML, low-grade glioma and CC, mutant IDH status is associated with a DNA hypermethylation phenotype, implicating altered epigenome dynamics in the aetiology of these cancers. Here we show that the IDH variants in CS are also associated with a hypermethylation phenotype and display increased production of the oncometabolite 2-hydroxyglutarate, supporting the role of mutant IDH-produced 2-hydroxyglutarate as an inhibitor of TET-mediated DNA demethylation. Meta-analysis of the acute myeloid leukaemia, low-grade glioma, cholangiocarcinoma and CS methylation data identifies cancer-specific effectors within the retinoic acid receptor activation pathway among the hypermethylated targets. By analysing sequence motifs surrounding hypermethylated sites across the four cancer types, and using chromatin immunoprecipitation and western blotting, we identify the transcription factor EBF1 (early B-cell factor 1) as an interaction partner for TET2, suggesting a sequence-specific mechanism for regulating DNA methylation

Distinct H3F3A and H3F3B driver mutations define chondroblastoma and giant cell tumor of bone
It is recognized that some mutated cancer genes contribute to the development of many cancer types, whereas others are cancer type specific. For genes that are mutated in multiple cancer classes, mutations are usually similar in the different affected cancer types. Here, however, we report exquisite tumor type specificity for different histone H3.3 driver alterations. In 73 of 77 cases of chondroblastoma (95%), we found p.Lys36Met alterations predominantly encoded in H3F3B, which is one of two genes for histone H3.3. In contrast, in 92% (49/53) of giant cell tumors of bone, we found histone H3.3 alterations exclusively in H3F3A, leading to p.Gly34Trp or, in one case, p.Gly34Leu alterations. The mutations were restricted to the stromal cell population and were not detected in osteoclasts or their precursors. In the context of previously reported H3F3A mutations encoding p.Lys27Met and p.Gly34Arg or p.Gly34Val alterations in childhood brain tumors, a remarkable picture of tumor type specificity for histone H3.3 driver alterations emerges, indicating that histone H3.3 residues, mutations and genes have distinct functions
Dynamic reorganization of task-related network interactions in post-stroke aphasia recovery
Post-stroke aphasia is a network disorder characterized by language impairments and aberrant network activation. While patients with post-stroke aphasia recover over time, the dynamics of the underlying changes in the brain remain elusive. Neuroimaging work demonstrated that language recovery is a heterogeneous process, characterized by varying activation levels in several regions of the left-hemispheric language network and the domain-general bilateral multiple-demand network. Crucially, this activation seems to depend on the time elapsed since stroke and the lesion location. Yet, beyond task-related brain activation, the degree and nature of interactions between regions of the language and the multiple-demand network are not well understood. In this longitudinal functional neuroimaging study, we characterized task-related functional interactions between regions of the language and the multiple-demand network during language processing. We hypothesized that interactions between language regions and between language and multiple-demand regions should change over time and depend on lesion location. We compared changes in effective connectivity in patients with left-hemispheric frontal or temporo-parietal stroke (n=17 per group) and healthy controls (n=17) with Dynamic Causal Modelling. All patients repeatedly underwent an auditory sentence comprehension paradigm during functional neuroimaging in the acute (≤ 1 week), subacute (1-2 weeks), and chronic (> 6 months) phases after stroke. We found overall increased task-related connectivity from regions of the multiple-demand to the language network across patients, resembling the principal pattern of task-related interactions in controls. Early facilitation from multiple-demand to language regions correlated with later language improvement across both groups. Crucially, recruitment of specific connections from regions of the multiple-demand to language network depended on lesion location and changed over time. In the chronic phase, patients with frontal stroke showed facilitatory modulation from the right dorsolateral prefrontal cortex, while patients with temporo-parietal stroke integrated the supplementary motor area/dorsal anterior cingulate cortex. Besides this across-network reorganization, facilitatory connectivity between regions of the language network emerged in all patients in the subacute phase. Additionally, patients with frontal stroke showed facilitatory influences from the right lesion homologue to the remaining undamaged left inferior frontal cortex in the acute phase. Collectively, we provide first evidence that functional interactions of regions within and across the language and the multiple-demand network facilitate aphasia recovery. The identified dynamic reorganization principles over the time course of recovery may inform the future use of personalized treatment protocols with neurostimulation in aphasia rehabilitation. These protocols should be tailored to the individual lesion location and recovery phase

A unified model of post-stroke language deficits including discourse production and their neural correlates.
The clinical profiles of individuals with post-stroke aphasia demonstrate considerable variation in the presentation of symptoms. Recent aphasiological studies have attempted to account for this individual variability using a multivariate data-driven approach (principal component analysis) on an extensive neuropsychological and aphasiological battery, to identify fundamental domains of post-stroke aphasia. These domains mainly reflect phonology, semantics and fluency; however, these studies did not account for variability in response to different forms of connected speech, i.e. discourse genres. In the current study, we initially examined differences in the quantity, diversity and informativeness between three different discourse genres, including a simple descriptive genre and two naturalistic forms of connected speech (storytelling narrative, and procedural discourse). Subsequently, we provided the first quantitative investigation on the multidimensionality of connected speech production at both behavioural and neural levels. Connected speech samples across descriptive, narrative, and procedural discourse genres were collected from 46 patients with chronic post-stroke aphasia and 20 neurotypical adults. Content analyses conducted on all connected speech samples indicated that performance differed across discourse genres and between groups. Specifically, storytelling narratives provided higher quantities of content words and lexical diversity compared to composite picture description and procedural discourse. The analyses further revealed that, relative to neurotypical adults, patients with aphasia, both fluent and non-fluent, showed reduction in the quantity of verbal production, lexical diversity, and informativeness across all discourses. Given the differences across the discourses, we submitted the connected speech metrics to principal component analysis alongside an extensive neuropsychological/aphasiological battery that assesses a wide range of language and cognitive skills. In contrast to previous research, three unique orthogonal connected speech components were extracted in a unified model, reflecting verbal quantity, verbal quality, and motor speech, alongside four core language and cognitive components: phonological production, semantic processing, phonological recognition, and executive functions. Voxel-wise lesion-symptom mapping using these components provided evidence on the involvement of widespread cortical regions and their white matter connections. Specifically, left frontal regions and their underlying white matter tracts corresponding to the frontal aslant tract and the anterior segment of the arcuate fasciculus were particularly engaged with the quantity and quality of fluent connected speech production while controlling for other co-factors. The neural correlates associated with the other language domains align with existing models on the ventral and dorsal pathways for language processing

Genome-wide Association Study Identifies Two Susceptibility Loci for Osteosarcoma
Osteosarcoma is the most common primary bone malignancy of adolescents and young adults. In order to better understand the genetic etiology of osteosarcoma, we performed a multi-stage genome-wide association study (GWAS) consisting of 941 cases and 3,291 cancer-free adult controls of European ancestry. Two loci achieved genome-wide significance: rs1906953 at 6p21.3, in the glutamate receptor metabotropic 4 [GRM4] gene (P = 8.1 ×10-9), and rs7591996 and rs10208273 in a gene desert on 2p25.2 (P = 1.0 ×10-8 and 2.9 ×10-7). These two susceptibility loci warrant further exploration to uncover the biological mechanisms underlying susceptibility to osteosarcoma
Favouritism: exploring the 'uncontrolled' spaces of the leadership experience
In this paper, we argue that a focus on favouritism magnifies a central ethical ambiguity in leadership, both conceptually and in practice. The social process of favouritism can even go unnoticed, or misrecognised if it does not manifest in a form in which it can be either included or excluded from what is (collectively interpreted as) leadership. The leadership literature presents a tension between what is an embodied and relational account of the ethical, on the one hand, and a more dispassionate organisational ‘justice’ emphasis, on the other hand. We conducted 23 semi-structured interviews in eight consultancy companies, four multinationals and four internationals. There were ethical issues at play in the way interviewees thought about favouritism in leadership episodes. This emerged in the fact that they were concerned with visibility and conduct before engaging in favouritism. Our findings illustrate a bricolage of ethical justifications for favouritism, namely utilitarian, justice, and relational. Such findings suggest the ethical ambiguity that lies at the heart of leadership as a concept and a practice
