    Assessing the impact of the 2011 EU Transport White Paper - a rail freight demand forecast up to 2050 for the EU27

    Purpose This paper presents a rail freight forecast for the EU27 for the period up to 2050. Background Rail freight’s market share of the transport sector in Europe has been falling or remained stagnant since 1970. In contrast, the share of road freight has been increasing. As rail freight transport is generally considered an environmentally friendly option, various measures have been implemented for more than two decades, at national and European level, to encourage a modal shift from road to rail. In the latest EU Transport White paper policy paper of 2011, an optimistic target is set for greater use of rail freight transport, in both the near and the longer term future. Specifically: a modal shift, from road to rail, and waterway transport, of 30 % by 2030 and 50 % by 2050 - for distances greater than 300 km. Methodology With the aforesaid policy objectives, the current research examines the possible effects of these aims in terms of future levels of rail freight demand. This research relies heavily upon the EU TRANS-TOOLS modelling tool and explores three scenarios. A Reference scenario - with no significant change to current rail freight policy, infrastructure and existing trends is considered alongside two White Paper scenarios (High and Low) which take more optimistic views of the white paper policy objectives. Results The study finds that the Reference and White Paper Low scenarios demonstrate similar results in terms of growth and modal split. In stark contrast, the White paper High scenario results show that demand for rail freight services almost doubles compared to the Reference values. Conclusions The rail sector is expected to attract new commodities from road transport such as foodstuffs and building and transport materials. To meet this demand, the rail industry - including operators, infrastructure managers and governments - will have to invest in technologies, infrastructure and terminals with a view to significantly increasing productivity against current levels

    Coupling virtual watersheds with ecosystem services assessment: A 21st century platform to support river research and management

    The demand for freshwater is projected to increase worldwide over the coming decades, resulting in severe water stress and threats to riverine biodiversity, ecosystem functioning and services. A major societal challenge is to determine where environmental changes will have the greatest impacts on riverine ecosystem services and where resilience can be incorporated into adaptive resource planning. Both water managers and scientists need new integrative tools to guide them towards the best solutions that meet the demands of a growing human population but also ensure riverine biodiversity and ecosystem integrity. Resource planners and scientists could better address a growing set of riverine management and risk mitigation issues by (1) using a “Virtual Watersheds” approach based on improved digital river networks and better connections to terrestrial systems; (2) integrating Virtual Watersheds with ecosystem services technology (ARtificial Intelligence for Ecosystem Services: ARIES), and (3) incorporating the role of riverine biotic interactions in shaping ecological responses. This integrative platform can support both interdisciplinary scientific analyses of pressing societal issues and effective dissemination of findings across river research and management communities. It should also provide new integrative tools to identify the best solutions and trade-offs to ensure the conservation of riverine biodiversity and ecosystem services

    G-CSF Prevents the Progression of Structural Disintegration of White Matter Tracts in Amyotrophic Lateral Sclerosis: A Pilot Trial

    Background: The hematopoietic protein Granulocyte-colony stimulating factor (G-CSF) has neuroprotective and regenerative properties. The G-CSF receptor is expressed by motoneurons, and G-CSF protects cultured motoneuronal cells from apoptosis. It therefore appears as an attractive and feasible drug candidate for the treatment of amyotrophic lateral sclerosis (ALS). The current pilot study was performed to determine whether treatment with G-CSF in ALS patients is feasible.Methods: Ten patients with definite ALS were entered into a double-blind, placebo-controlled, randomized trial. Patients received either 10 mu g/kg BW G-CSF or placebo subcutaneously for the first 10 days and from day 20 to 25 of the study. Clinical outcome was assessed by changes in the ALS functional rating scale (ALSFRS), a comprehensive neuropsychological test battery, and by examining hand activities of daily living over the course of the study (100 days). The total number of adverse events (AE) and treatment-related AEs, discontinuation due to treatment-related AEs, laboratory parameters including leukocyte, erythrocyte, and platelet count, as well as vital signs were examined as safety endpoints. Furthermore, we explored potential effects of G-CSF on structural cerebral abnormalities on the basis of voxel-wise statistics of Diffusion Tensor Imaging (DTI), brain volumetry, and voxel-based morphometry.Results: Treatment was well-tolerated. No significant differences were found between groups in clinical tests and brain volumetry from baseline to day 100. However, DTI analysis revealed significant reductions of fractional anisotropy (FA) encompassing diffuse areas of the brain when patients were compared to controls. On longitudinal analysis, the placebo group showed significant greater and more widespread decline in FA than the ALS patients treated with G-CSF.Conclusions: Subcutaneous G-CSF treatment in ALS patients appears as feasible approach. Although exploratory analysis of clinical data showed no significant effect, DTI measurements suggest that the widespread and progressive microstructural neural damage in ALS can be modulated by G-CSF treatment. These findings may carry significant implications for further clinical trials on ALS using growth factors

    The Farsi version of the Hypomania Check-List 32 (HCL-32): Applicability and indication of a four-factorial solution

    Background: Data from the Iranian population for hypomania core symptom clusters are lacking. The aim of the present study was therefore to apply the Farsi version of the Hypomania-Check-List 32 (HCL-32), and to explore its factorial structure.Methods: A total of 163 Iranian out-patients took part in the study; 61 suffered from Major Depressive Disorder (MDD), and 102 suffered from Bipolar Disorders (BP). Participants completed the Mood Disorder Questionnaire (MDQ) and the Hypomania Checklist (HCL-32). Exploratory factor analyses were used to examine the properties of the HCL-32. A ROC-curve analysis was performed to calculate sensitivity and specificity.Results: The HCL-32 differentiated between patients with MDD and with BP. Psychometric properties were satisfactory: sensitivity: 73; specificity: 91. MDQ and HCL-32 did correlate highly. No differences were found between patients suffering from BP I and BP II.Discussion: Instead of the two-factorial structure of the HCL-32 reported previously, the present pattern of factorial results suggest a distinction between four factors: two broadly positive dimensions of hypomania ("physically and mentally active"; "positive social interactions") and two rather negative dimensions ("risky behavior and substance use"; "difficulties in social interaction and impatience").Conclusion: The Farsi version of the HCL-32 proved to be applicable, and therefore easy to introduce within a clinical context.     A high fat diet increases mitochondrial fatty acid oxidation and uncoupling to decrease efficiency in rat heart

    Elevated levels of cardiac mitochondrial uncoupling protein 3 (UCP3) and decreased cardiac efficiency (hydraulic power/oxygen consumption) with abnormal cardiac function occur in obese, diabetic mice. To determine whether cardiac mitochondrial uncoupling occurs in non-genetic obesity, we fed rats a high fat diet (55% kcal from fat) or standard laboratory chow (7% kcal from fat) for 3 weeks, after which we measured cardiac function in vivo using cine MRI, efficiency in isolated working hearts and respiration rates and ADP/O ratios in isolated interfibrillar mitochondria; also, measured were medium chain acyl-CoA dehydrogenase (MCAD) and citrate synthase activities plus uncoupling protein 3 (UCP3), mitochondrial thioesterase 1 (MTE-1), adenine nucleotide translocase (ANT) and ATP synthase protein levels. We found that in vivo cardiac function was the same for all rats, yet oxygen consumption was 19% higher in high fat-fed rat hearts, therefore, efficiency was 21% lower than in controls. We found that mitochondrial fatty acid oxidation rates were 25% higher, and MCAD activity was 23% higher, in hearts from rats fed the high fat diet when compared with controls. Mitochondria from high fat-fed rat hearts had lower ADP/O ratios than controls, indicating increased respiratory uncoupling, which was ameliorated by GDP, a UCP3 inhibitor. Mitochondrial UCP3 and MTE-1 levels were both increased by 20% in high fat-fed rat hearts when compared with controls, with no significant change in ATP synthase or ANT levels, or citrate synthase activity. We conclude that increased cardiac oxygen utilisation, and thereby decreased cardiac efficiency, occurs in non-genetic obesity, which is associated with increased mitochondrial uncoupling due to elevated UCP3 and MTE-1 levels

    Tamarindus indica Extract Alters Release of Alpha Enolase, Apolipoprotein A-I, Transthyretin and Rab GDP Dissociation Inhibitor Beta from HepG2 Cells

    Background: The plasma cholesterol and triacylglycerol lowering effects of Tamarindus indica extract have been previously described. We have also shown that the methanol extract of T. indica fruit pulp altered the expression of lipid-associated genes including ABCG5 and APOAI in HepG2 cells. In the present study, effects of the same extract on the release of proteins from the cells were investigated using the proteomics approach. Methodology/Principal Findings: When culture media of HepG2 cells grown in the absence and presence of the methanol extract of T. indica fruit pulp were subjected to 2-dimensional gel electrophoresis, the expression of seven proteins was found to be significantly different (p<0.03125). Five of the spots were subsequently identified as alpha enolase (ENO1), transthyretin (TTR), apolipoprotein A-I (ApoA-I; two isoforms), and rab GDP dissociation inhibitor beta (GDI-2). A functional network of lipid metabolism, molecular transport and small molecule biochemistry that interconnects the three latter proteins with the interactomes was identified using the Ingenuity Pathways Analysis software. Conclusion/Significance: The methanol extract of T. indica fruit pulp altered the release of ENO1, ApoA-I, TTR and GDI-2 from HepG2 cells. Our results provide support on the effect of T. indica extract on cellular lipid metabolism, particularly that of cholesterol

    The design and protocol of acupuncture for migraine prophylaxis: A multicenter randomized controlled trial

    Background: Many studies have already reported encouraging results in the prophylactic therapy of migraine by acupuncture, but there seems to be a lack of high quality randomized controlled trials from China. We design and perform a randomized controlled clinical trial to evaluate the efficacy of acupuncture compared with flunarizine in the prophylactic therapy of patients with migraine without aura in China. Methods: This trial is a multicenter, prospective, randomized controlled clinical trial. The 140 migraine patients are randomly allocated to two different groups. The acupuncture groups (n = 70) is treated with acupuncture and placebo medicine; while the control group (n = 70) is treated with sham acupuncture and medicine (Flunarizine). Both Flunarizine and placebo are taken 10 mg once per night for the first 2 weeks and then 5 mg once per night for the next 2 weeks. Patients in both groups receive 12 sessions of verum/sham acupuncture in 4 weeks. Discussion: The study design and the long term clinical practice of acupuncturists guarantee a high external validity for the results. The results of our trial will be helpful to supply the evidence on the efficacy of acupuncture for migraine prophylaxis in China.
