Fermionic microstates within Painlev\'e-Gullstrand black hole
We consider the quantum vacuum of fermionic field in the presence of a
black-hole background as a possible candidate for the stabilized black hole.
The stable vacuum state (as well as thermal equilibrium states with arbitrary
temperature) can exist if we use the Painlev\'e-Gullstrand description of the
black hole, and the superluminal dispersion of the particle spectrum at high
energy, which is introduced in the free-falling frame. Such choice is inspired
by the analogy between the quantum vacuum and the ground state of quantum
liquid, in which the event horizon for the low-energy fermionic quasiparticles
also can arise. The quantum vacuum is characterized by the Fermi surface, which
appears behind the event horizon. We do not consider the back reaction, and
thus there is no guarantee that the stable black hole exists. But if it does
exist, the Fermi surface behind the horizon would be the necessary attribute of
its vacuum state. We also consider exact discrete spectrum of fermions inside
the horizon which allows us to discuss the problem of fermion zero modes.Comment: LaTeX, 20 pages, 2 figure
Different incidences of knee arthroplasty in the Nordic countries.

To make projections regarding future needs for primaries and revisions, additional knowledge is important. We analyzed and compared the incidences among 4 Nordic countries Patients and methods - Using Nordic Arthroplasty Register Association (NARA) data from 4 countries, we analyzed differences between age and sex groups. We included patients over 30 years of age who were operated with TKA or unicompartmental knee arthroplasty (UKA) during the period 1997-2012. The negative binomial regression model was used to analyze changes in general trends and in sex and age groups. Results - The average annual increase in the incidence of TKA was statistically significant in all countries. The incidence of TKA was higher in women than in men in all 4 countries. It was highest in Finland in patients aged 65 years or more. At the end of the study period in 2012, Finland's total incidence was double that of Norway, 1.3 times that of Sweden and 1.4 times that of Denmark. The incidence was lowest in the youngest age groups (< 65 years) in all 4 countries. The proportional increase in incidence was highest in patients who were younger than 65 years. Interpretation - The incidence of knee arthroplasty steadily increased in the 4 countries over the study period. The differences between the countries were considerable, with the highest incidence in Finland. Bundling up carbon nanotubes through Wigner defects
We show, using ab initio total energy density functional theory, that the
so-called Wigner defects, an interstitial carbon atom right besides a vacancy,
which are present in irradiated graphite can also exist in bundles of carbon
nanotubes. Due to the geometrical structure of a nanotube, however, this defect
has a rather low formation energy, lower than the vacancy itself, suggesting
that it may be one of the most important defects that are created after
electron or ion irradiation. Moreover, they form a strong link between the
nanotubes in bundles, increasing their shear modulus by a sizeable amount,
clearly indicating its importance for the mechanical properties of nanotube

Cumulative mutagenesis of the basic residues in the 201-218 region of insulin-like growth factor (IGF)-binding protein-5 results in progressive loss of both IGF-I binding and inhibition of IGF-I biological action
We have reported previously that mutation of two conserved nonbasic amino acids (G203 and Q209) within the highly basic 201–218 region in the C-terminal domain of IGF-binding protein-5 (IGFBP-5) decreases binding to IGFs. This study reveals that cumulative mutagenesis of the 10 basic residues in this region, to create the C-Term series of mutants, ultimately results in a 15-fold decrease in the affinity for IGF-I and a major loss in heparin binding. We examined the ability of mutants to inhibit IGF-mediated survival of MCF-7 cells and were able to demonstrate that this depended not only upon the affinity for IGF-I, but also the kinetics of this interaction, because IGFBP-5 mutants with similar affinity constants (KD) values, but with different association (Ka) and dissociation (Kd) rate values, had markedly different inhibitory properties. In contrast, the affinity for IGF-I provided no predictive value in terms of the ability of these mutants to enhance IGF action when bound to the substratum. Instead, these C-Term mutants appeared to enhance the actions of IGF-I by a combination of increased dissociation of IGF-IGFBP complexes from the substratum, together with dissociation of IGF-I from IGFBP-5 bound to the substratum. These effects of the IGFBPs were dependent upon binding to IGF-I, because a non-IGF binding mutant (N-Term) was unable to inhibit or enhance the actions of IGF-I. These results emphasize the importance of the kinetics of association/dissociation in determining the enhancing or inhibiting effects of IGFBP-5 and demonstrate the ability to generate an IGFBP-5 mutant with exclusively IGF-enhancing activity
Distinct pathways in the over-expression of matrix metalloproteinases in human fibroblasts by relaxation of mechanical tension.
peer reviewedaudience: researcherThe aim of the work was to analyze, on a comparative basis, the signaling pathways operating in the regulation of a panel of matrix metalloproteinases (MMP) expressed by human dermal fibroblasts submitted to mechanical stress relaxation by cytochalasin D (CD) and in a retracting collagen gel (RCG). The mRNA steady-state level of MMPs was measured by a quantitative RT-PCR procedure using a synthetic RNA as internal standard. In monolayer, most MMPs were barely detected, except MMP-2. Disruption of the actin stress fibers by CD induced a moderate increase of MMP-2 mRNA and a much larger stimulation of MMP-3, -9, -13 and -14 mRNAs. In RCG, a significant up-regulation of these MMPs was also observed although to a lower extent than in CD-treated monolayers. Among the investigated MMPs, the MMP-8 and -11 were not reproducibly detected. MMP-2 was processed to its active form both by CD and in RCG. The CD-induced up-regulation of gene expression was largely repressed by blocking protein synthesis by cycloheximide for all the MMPs, by inhibiting the tyrosine-kinases of the src family by herbimycin A for all MMPs, except MMP-2, and by inhibiting the TPA-inducible PKC isoforms by bisindoyl maleimide for all MMPs, except MMP-14. The up-regulation induced by stress relaxation in RCG was protein synthesis-dependent for MMP-2 and MMP-13, tyrosine kinases-dependent for MMP-3 and MMP-13, as previously described for MMP-1. Inhibiting TPA-inducible PKC did not affect any MMP in RCG except MMP-13, which was strongly induced. The processing of MMP-2 was tyrosine kinases-dependent but PKC-independent. Inhibitors of the ERK1,2 and p38 MAP kinases pathways diversely affected the MMPs expression. Inhibiting the Rho-kinase activity by Y-27632 was inactive. These results point to the potent regulation operated by the status of the cytoskeleton on the cell phenotype, and to distinct regulatory pathways involved in the control of different MMPs expression

Emerging from this prospect is the development of in vitro representations of organs for drug toxicity assessment. Due to the ever-increasing interest in ocular drug delivery as a route for administration as well as the rise of new ophthalmic therapeutics, there is a demand for physiologically accurate in vitro models of the eye to assess drug delivery and safety of new ocular medicines. This review summarizes current existing ocular models and highlights the important factors and limitations that need to be considered during their use.Peer reviewe
Bandsaw Safety Improvements
Boeing Canada Winnipeg (BCW) is one of the largest aerospace composite manufacturers in Canada [1], producing over 500 parts and assemblies for its parent company Boeing. BCW is currently faced with a safety issue related to the use of band saws during a core chamfering process. Band saw operators are at risk of cutting themselves on the band saw blade, as well as being exposed to hazardous core dust that is created during the chamfering operation. Team 24: Honeycomb Badgers Industries has been commissioned by BCW to propose a design to mitigate, or eliminate these risks. The two constraints placed on the project are that it must be completed by December 5, 2018, and that the design must fit inside the BCW building. BCW outlined four requirements that must be met by the proposed design:1.The design must protect the band saw operator from being cut on by the blade.2.The design must minimize the band saw operator’s exposure to core dust.3.The design must function for various dimensions of cores.4.The design must function for various chamfer angles. Through a concept generation and selection process, a blade guard with a pivoting head and a downdraft table were chosen to be developed. The blade guard features a two-part construction with a pivoting head that encloses the band saw blade. The blade guard moves vertically on a track, and rests on the core during the chamfering operation. The downdraft table is located underneath the band saw blade, and it vacuums the core dust into a sink. The deliverables requested by BCW are a 3D CAD model, preliminary assembly drawings, a detailed concept selection process, a detailed design process, and justification for the final design. The project was broken into three phases. The first phase was problem identification and creating a project management plan, the second phase was concept generation and selection, and the third phase was concept development, final deliverables, and closeout procedures.
