Second Generation of Antisense Oligonucleotides: From Nuclease Resistance to Biological Efficacy in Animals

From efforts to improve the biophysical properties of antisense oligonucleotides by incorporating backbone- or sugar-modified nucleoside analogs, 2'-O-methoxyethyl ribonucleosides 8b were identified as building blocks for a second generation of antisense oligonucleotides. Compounds containing these modifications were demonstrated to combine the benefit of a high binding affinity to the RNA complement with a large increase in nuclease resistance, allowing the use of regular phosphodiester linkages. Chimeric oligonucleotides with 2'-O-methoxyethyl ribonucleosides, 8b, in the wings and a central DNA-phosphorothioate window were shown to efficiently downregulate C-'raf' kinase and PKC-α messenger-RNA in tumor cell lines resulting in a profound inhibition of cell proliferation. The same compounds were able to effectively reduce the growth of tumors in animal models at low concentrations indicating the potential utility of these second generation antisense oligonucleotides for therapeutic applications

Seasonal pattern of apoplastic solute accumulation and loss of cell turgor during ripening of Vitis vinifera fruit under field conditions

Using a novel pressure membrane (PM) apparatus for the extraction of apoplastic fluid from field-grown grape (Vitis vinifera L.) berries, our hypothesis that significant apoplast solutes accumulate at the beginning of the ripening process (i.e. veraison), and that this accumulation might contribute to progressive berry softening due to a progressive loss of mesocarp cell turgor pressure (P) was tested. It was necessary to correct the solute potential (Ψs) of fluid collected with the PM for dilution due to the presence of a dead volume in the apparatus, but after correction, the Ψs obtained with the PM agreed with that obtained by low speed centrifugation. A clear decline in fruit apoplastic solute potential (ψSA) began approximately 10 d prior to fruit coloration, and it was found to be coincident with a decline in mesocarp cell P and fruit elasticity (E). By late in fruit development when berry growth ceased (90 d after anthesis), both apoplast and fruit Ψs reached almost –4 MPa. These results support the hypothesis that a decrease in ψSA is responsible for the observed loss in mesocarp cell P, and is the mechanistic cause of berry softening

The Panel on Food Additives and Nutrient Sources added to Food (ANS) provides a scientific statement presenting a conceptual framework for the risk assessment of certain food additives re-evaluated under Commission Regulation (EU) No 257/2010. This framework will be used in the evaluation made by the Panel, but the expert judgement of the scientific background, on a case-by-case basis, remains essential to reach a final conclusion. The outcome of the re-evaluation of food additives taking into account all available information is presented in the document, as well as the exposure assessment scenarios to be carried out by the Panel considering the use levels set in the legislation and the availability of adequate usage or analytical data
