Breath analysis as a diagnostic tool in breast cancer patients
Diese Arbeit beschÀftigt sich mit der Analyse von volatile organic compounds
(VOCs), und ihrer verÀnderten Konzentrationen im Körper von
Mammakarzinompatientinnen. Das Ziel dieser Pilotstudie war der Nachweis eines
verÀnderten VOC-Spektrums im Atem von Patientinnen mit neu diagnostiziertem,
primÀr nicht metastasiertem Mammakarzinom. Dazu wurde ein System zur
Atemanalyse entwickelt und die Atemproben standardisiert entnommen.
Laborchemisch gelang die Probenaufgabe schonend und reproduzierbar mittels
Thermodesorption und Kryofokussierung. Die Trennung und der Nachweis der
Analyte erfolgten per Gaschromatographie und Massenspektrometrie, einer
zuverlÀssigen, nachweisstarken und reproduzierbaren Methode, die bereits in
der Forschung mit Atemanalysen etabliert wurde. Erstmalig wurde der Einsatz
der VOC Atemanalyse im klinischen Setting evaluiert. Die Ergebnisse unserer
Studie zeigen, dass die exspiratorische Atemluft eine Vielzahl
leichtflĂŒchtiger, organischer Verbindungen im Spurenbereich enthĂ€lt. Es
konnten insgesamt zwischen 83 und 100 verschiedene Analyten pro
Studienteilnehmerin identifiziert werden, zu denen Alkane, Alkene, Ketone,
Halogenkohlenwasserstoffe, Ester, UngesÀttigte Kohlenwasserstoffe, Terpene,
Siloxane, Alkohole, Aldehyde und Aromaten gehören. FĂŒr jede Substanz wurde der
alveolÀre Gradient durch Subtraktion der Raumluftkonzentration von der
Atemluftkonzentration gebildet. Bei den flĂŒchtigen organischen Substanzen
konnten nach qualitativer und quantitativer statistischer Analyse folgende
AbhÀngigkeiten nachgewiesen werden: Einen deutlichen Unterschied zwischen
Patientinnen der Kontroll- und Mammakarzinomgruppe zeigten einzelne Alkane,
Alkene, Terpene, Halogenkohlenwasserstoffe und aromatische Verbindungen. Ein
signifikanter Konzentrationsunterschied wurde fĂŒr 3-Methylhexan, Decen,
Caryophyllen, Naphthalin und Trichlorethylen gemessen. Die Konzentration von
3-Methylhexan, Decen, Caryophyllen und Naphthalin war im Atem von gesunden
Probandinnen signifikant höher als im Atem der erkrankten Frauen. Hingegen war
die Konzentration von Trichlorethylen im Atem der erkrankten Frauen
signifikant höher. Die Bedeutung dieser Ergebnisse liegt zunÀchst im Bereich
der medizinischen Grundlagenforschung und der Entwicklung einer Referenz-
Methodik zur weiteren Untersuchung exhalierter VOCs. Ein Teil der Befunde
bleibt deskriptiv, solange die pathophysiologische Rolle spezifischer VOCs
ungeklÀrt ist. Es ist anzunehmen, dass VerÀnderungen im Atem von
Mammakarzinompatientinnen schon sehr frĂŒhzeitig zu detektieren sind, da sich
bei malignen VerĂ€nderungen auf der Molekularebene schon frĂŒh ganze
Zellprozesse Àndern: Es kommt zur Induktion des Cytochrom-P450-Systems und
nachfolgend zu verĂ€nderten Stoffwechselprozessen. Diese VerĂ€nderungen fĂŒhren
zu einem signifikant differenten VOC-Spektrum im Atem. Daher könnte man die
Methode besonders bei frĂŒhen Karzinomen, die in der Bildgebung noch nicht zu
detektieren sind, heranziehen. Bei gröĂeren Tumoren werden im Atem auch höhere
VOC-Spektren gemessen, da es im Verlauf auch zur Produktion der VOCs kommt.
Diese Arbeit lÀsst den Schluss zu, dass chemische Analysen der Exhalationsluft
auf flĂŒchtige organische Substanzen im klinischen Alltag durchfĂŒhrbar sind und
ein hohes Potential in der Diagnose des Mammakarzinoms besitzen. Somit stellen
VOCs potentielle Marker fĂŒr Brustkrebs dar. Ihre diesbezĂŒgliche Eignung wurde
in dieser Studie mit der Quantifizierung organischer Spurenkomponenten unter
Einsatz der hochsensitiven Methoden der Gaschromatographie und der
Massenspektrometrie bestÀtigt. Validierte Messungen exhalierter VOCs könnten
zukĂŒnftig fĂŒr ein Bio-Monitoring auch auĂerhalb wissenschaftlicher
Untersuchungen zur VerfĂŒgung stehen. Der Aufforderung des Arztes: âBitte tief
durchatmen!â kĂ€me eine neue Bedeutung zu.Background: Carcinogenic products in the exhaled breath of cancer patients are
of growing medical interest as they can serve as noninvasive disease markers.
Breath analysis can be used as an alternative or complementary diagnostic tool
in breast cancer patients which have a different pattern of chemical
composition in their breath. This study aims to verify the existence of
specific volatile organic compounds (VOCs) in the breath of breast cancer
patients. Methods: In this prospective study we included ten patients
suffering from breast cancer and ten healthy pair-matched women. Breath
samples were taken of each member of the two respective groups and scanned by
gas chromatography/mass spectometry for contained volatile organic compounds
such as alkanes, ketones, halogenated hydrocarbon, aldehydes, and esters.
Results: The spectrum of VOCs did significantly differ within the two groups.
Five specific VOCs could be identified as typical discriminatory markers in
the breath samples. Four VOCs were elevated in the healthy controls, one
specific VOC was found to be elevated in women affected by breast cancer.
Conclusions: This pilot study shows a specific VOC pattern using the gas
chromatography in the breath of breast cancer patients. Five specific breast
cancer- VOCs could be identified. At relatively low expenses the
identification of VOCs may be used to detect breast cancer
Are Explanatory Videos an Effective Tool for Learning in the Undergraduate Laboratory? Evaluation of Video Scaffolding for Experimental Problem Solving on the Basis of Self-Concept of Ability
Erkenntnisgewinnung ist in der universitÀren LehrkrÀftebildung noch nicht entsprechend ihrer hohen Bedeutung als zentraler Kompetenzbereich der Naturwissenschaften in der Schule reprÀsentiert. In der ChemielehrkrÀftebildung an der Technischen UniversitÀt Braunschweig erfolgte daher ihre explizite Vermittlung und Förderung mit experimentellen Problemlöseaufgaben in einem fachwissenschaftlichen, organisch-chemischen Laborpraktikum. ZusÀtzlich wurden ErklÀrvideos zu Erkenntnisgewinnung bzw. labormethodischen Techniken als kognitives Scaffolding in der Vorbereitungszeit der Studierenden eingesetzt. Im Beitrag werden die ErklÀrvideos als Lerntool vorgestellt und anhand ihres Einflusses auf die FÀhigkeitsselbsteinschÀtzungen der Studierenden evaluiert. WÀhrend signifikante ZuwÀchse in den FÀhigkeitsselbsteinschÀtzungen durch die Teilnahme am Praktikum gefunden wurden, zeigten sich keine Interaktionseffekte durch den Einsatz der ErklÀrvideos. Die Erkenntnisse zum Einsatz von ErklÀrvideos im universitÀren Laborpraktikum werden diskutiert.
 Given its high importance as a central competence in the natural sciences in schools, scientific inquiry is not yet adequately represented in preservice science teacher education at university level. In preservice chemistry teacher education at Technische UniversitĂ€t Braunschweig, scientific inquiry is taught explicitly and promoted with experimental problem solving in an organic chemistry laboratory. In addition, cognitive scaffolding is provided by explanatory videos on scientific reasoning and practical skills for studentsâ prelaboratory preparation. In the article, the explanatory videos are introduced as a tool for learning and evaluated regarding their influence on studentsâ self-concept of ability. While significant gains were found in the studentsâ self-concept of ability due to participation in the laboratory, there was no interaction of the exposure to the videos. Measuring and Fostering Preservice Chemistry Teachersâ Scientific Reasoning Competency
Developing scientific reasoning (SR) is a central goal of science-teacher education worldwide. On a fine-grained level, SR competency can be subdivided into at least six skills: formulating research questions, generating hypotheses, planning experiments, observing and measuring, preparing data for analysis, and drawing conclusions. In a study focusing on preservice chemistry teachers, an organic chemistry lab course was redesigned using problem-solving experiments and SR video lessons to foster SR skills. To evaluate the intervention, a self-assessment questionnaire was developed, and a performance-based instrument involving an experimental problem-solving task was adapted to the target group of undergraduates. The treatment was evaluated in a pre-post design with control group (cook-book experiments, no SR video lessons) and alternative treatment group (problem-solving experiments, unrelated video lessons). Interrater reliability was excellent (Ï from 0.915 to 1.000; ICC (A1)). Data analysis shows that the adapted instrument is suitable for university students. First insights from the pilot study indicate that the cook-book lab (control group) only fosters studentsâ skill in observing and measuring, while both treatment groups show an increase in generating hypotheses and planning experiments. No pretest-posttest differences were found in self-assessed SR skills in the treatment groups. Instruments and data are presented and discussed
Nonâviral cellular substrates for human immunodeficiency virus type 1 protease
A Computer search revealed 10 proteins with homology to the sequence we originally identified in vimentin as the site of cleavage by human immunodeficiency virus type 1 (HIVâ1) protease. Of these 10 proteins (actin, αâactinin, spectrin, tropomyosins, vinculin, dystrophin, MAPâ2, villin, TRKâ1 and lg ÎŒâchain), we show that 4 of the first 5 were cleaved in vitro by this protease, as arc MAPâ1 and â2 [(1990) J. Gen. Virol. 71, 1985â1991]. In these proteins, cleavage is not restricted to a single motif, but occurs at many sites. However, cleavage is not random, since 9 other proteins including the cytoskeletal proteins filamin and band 4.1 are not cleaved in the in vitro assay. Thus, the ability of HIVâ1 protease to cleave specific components or the cytoskeleton may be an important, although as yet unevaluated aspect of the life cycle of this retrovirus and/or may directly contribute to the pathogenesis observed during infectio
