respectivamente. Os efeitos de meio foram significantes (
Wild Roman chamomile extracts and phenolic compounds: enzymatic assays and molecular modelling studies with VEGFR-2 tyrosine kinase
Angiogenesis is a process by which new blood vessels are formed from the pre-existing vasculature, and it is a key process that leads to tumour development. Some studies have recognized phenolic compounds as chemopreventive agents; flavonoids, in particular, seem to suppress the growth of tumor cells modifying the cell cycle. Herein, the antiangiogenic activity of Roman chamomile (Chamaemelum nobile L.) extracts (methanolic extract and infusion) and the main phenolic compounds present (apigenin, apigenin-7-O-glucoside, caffeic acid, chlorogenic acid, luteolin, and luteolin-7-O-glucoside) was evaluated through enzymatic assays using the tyrosine kinase intracellular domain of the Vascular Endothelium Growth Factor Receptor-2 (VEGFR-2), which is a transmembrane receptor expressed fundamentally in endothelial cells involved in angiogenesis, and molecular modelling studies. The methanolic extract showed a lower IC50 value (concentration that provided 50% of VEGFR-2 inhibition) than the infusion, 269 and 301 μg mL(-1), respectively. Regarding phenolic compounds, luteolin and apigenin showed the highest capacity to inhibit the phosphorylation of VEGFR-2, leading us to believe that these compounds are involved in the activity revealed by the methanolic extract. The authors are grateful to strategic projects PEst-OE/AGR/UI0690/2014 and PEst-C/QUI/UI0686/2013-2014 for financial support to the research centres. Generation and characterization of a recombinant chimeric protein (rCpLi) consisting of B-cell epitopes of a dermonecrotic protein from Loxosceles intermedia spider venom
AbstractA chimeric protein was constructed expressing three epitopes of LiD1, a dermonecrotic toxin from the venom of Loxosceles intermedia spider. This species is responsible for a large number of accidents involving spiders in Brazil. We demonstrated that the chimeric protein (rCpLi) generated is atoxic and that antibodies previously developed in rabbits against synthetic epitopes reactive with rCpLi in ELISA and immunoblot assays. The antibody response in rabbits against the rCpLi was evaluated by ELISA and we have detected an antibody response in all immunized animals. Overlapping peptides covering the amino acid sequence of the rCpLi were synthesized on a cellulose membrane, and their recognition by rabbit anti-rCpLi serum assessed. Three different antigenic regions were identified. The percentage of inhibition of the dermonecrotic, hemorrhagic and edematogenic activities caused by the recombinant protein LiD1r in naïve rabbits was assessed by pre-incubation with anti-rCpLi antibodies. Anti-rCpLi induced good dermonecrotic and hemorrhagic protection. The levels of protection were similar to the antiboides anti-LiD1r. In summary, we have developed a polyepitope recombinant chimeric protein capable of inducing multiple responses of neutralizing antibodies in a rabbit model. This engineered protein may be a promising candidate for therapeutic serum development or vaccination
N-terminal-pro-B type natriuretic peptide as a useful tool to evaluate pulmonary hypertension and cardiac function in CDH infants
Objective: In congenital diaphragmatic hernia (CDH) the severity of pulmonary hypertension (PH) is considered, by several authors, determinant of clinical outcome. Plasmatic N-terminal-pro-B type natriuretic peptide (NT-proBNP) might be useful in diagnosis and management of PH in newborns, although its interest in CDH infants remains to be defined. Early NT-proBNP levels were assessed in CDH infants and correlated with cardiovascular echocardiographic parameters. Patients and Methods: 28 newborns, CDH and age-matched controls were enrolled in a prospective study. Clinical condition, NT-proBNP plasmatic levels, echo parameters of PH and biventricular function were assessed at 24 h after delivery as well as survival outcome. Results: Estimated mean pulmonary pressure and NT-proBNP were significantly higher in CDH than control infants. NT-proBNP significantly correlated with estimated pulmonary artery pressure, right ventricular Tei index, and tricuspid E/A ratio. Additionally, we found that CDH infants with NT-proBNP >11,500 pg/ml experienced a worse prognosis. Conclusions: We demonstrated that PH is associated with NT-proBNP elevation and diastolic impairment in CDH infants. Early elevations in NT-proBNP levels seem to alert for a subset of CDH infants with worse prognosis. Copyrigh
Dilatonic current-carrying cosmic strings
We investigate the nature of ordinary cosmic vortices in some scalar-tensor
extensions of gravity. We find solutions for which the dilaton field condenses
inside the vortex core. These solutions can be interpreted as raising the
degeneracy between the eigenvalues of the effective stress-energy tensor,
namely the energy per unit length U and the tension T, by picking a privileged
spacelike or timelike coordinate direction; in the latter case, a phase
frequency threshold occurs that is similar to what is found in ordinary neutral
current-carrying cosmic strings. We find that the dilaton contribution for the
equation of state, once averaged along the string worldsheet, vanishes, leading
to an effective Nambu-Goto behavior of such a string network in cosmology, i.e.
on very large scales. It is found also that on small scales, the energy per
unit length and tension depend on the string internal coordinates in such a way
as to permit the existence of centrifugally supported equilibrium
configuration, also known as vortons, whose stability, depending on the very
short distance (unknown) physics, can lead to catastrophic consequences on the
evolution of the Universe.Comment: 10 pages, ReVTeX, 2 figures, minor typos corrected. This version to

One-neutron removal reactions on neutron-rich psd-shell nuclei
A systematic study of high energy, one-neutron removal reactions on 23
neutron-rich, psd--shell nuclei (Z=5-9, A=12-25) has been carried out. The
longitudinal momentum distributions of the core fragments and corresponding
single-neutron removal cross sections are reported for reactions on a carbon
target. Extended Glauber model calculations, weighted by the spectroscopic
factors obtained from shell model calculations, are compared to the
experimental results. Conclusions are drawn regarding the use of such reactions
as a spectroscopic tool and spin-parity assignments are proposed for 15B, 17C,
19-21N, 21,23O, 23-25F. The nature of the weakly bound systems 14B and 15,17C
is discussed.Comment: 11 pages + 2 figure
