    Empleo de materiales secundarios como materia prima de nuevos tipos de cementos

    The present paper is a comparative study of some characteristics of new belite cements obtained from two kind of wastes, which were used as secondary raw materials: fly ash (FA), of low CaO content, from coal combustion, and ash from incineration of municipal solid waste (MSWIA). Cements were synthesised in a range of temperature between 700°C and 900°C from MSWIA and FA, which were previously activated by hydrothermal treatment at 200°C The evolution of cemented phases with the heating temperature was followed by X-ray diffraction (XRD). The results were compared with those obtained from heating the starting FA and MSWIA without the previous hydrothermal treatment. The degree of hydration was quantitatively evaluated by the combined water content, determined from thermogravimetric analyses, during a period of 28 days or 200 days from mixing depending of hydration kinetics of each cement.Este trabajo es un estudio comparativo de algunas de las características de nuevos cementos belíticos, obtenidos a partir de dos tipos de residuos, como materia prima secundaria: cenizas volantes (CV) de bajo contenido en cal, procedentes de la combustión del carbón y cenizas procedentes de la incineración de residuos sólidos urbanos (CIRSU). Los cementos fueron sintetizados en un rango de temperaturas comprendido entre 700°C y 900°C después de un tratamiento hidrotermal de la CV y CIRSU a 200°C La evolución de las fases cementicias, con la temperatura de calentamiento, fue estudiada por difracción de rayos X (DRX). Los resultados fueron comparados con aquellos obtenidos, directamente, por calentamiento de los residuos, sin un tratamiento hidrotermal previo de los mismos. El grado de hidratación fue cuantitativamente evaluado, por medio del análisis termogravimétrico, a partir del agua combinada de los cementos hidratados durante un período de 28 días o 200 días, dependiendo de la cinética de hidratación de cada cemento

    Influence of the type of rare-earth cation on electrical properties of Ba(Zr,Ti)O3 ceramics

    En este artículo es investigado el efecto del radio iónico de diferentes cationes de tierras raras sobre las propiedades eléctricas del compuesto Ba(Zr,Ti)O3. La cerámica BaZr0.09Ti0.9O3 fue dopada con iones de Gd3+, Pr3+ y La3+, sustituyendo un 5 % de contenido de cationes Ba2+. Los resultados muestran que el coeficiente de difusividad se incrementa con el aumento del radio iónico, mientras el carácter de la transición de fase permanece casi constante, de acuerdo con el modelo fenomenológico de Santos-Eiras. En todas las muestras se reporta un comportamiento no relajador. El proceso de conductividad iónica en el BaZr0.09Ti0.9O3 aparece a 200 ºC, valor inferior al observado en el material cerámico BaTiO3. El valor de energía de activación en el proceso de conductividad del BaZr0.09Ti0.9O3 (Ea= 0.81 eV) indica un mecanismo de compensación de carga en la estructura cristalina similar al BaTiO3, es decir vacancias de oxígeno simplemente ionizadas. Para el compuesto BaZr0.09Ti0.9O3 dopado con iones Gd3+, Pr3+ y La3+ aparece además un mecanismo de conducción de electrones, debido a la sustitución del Ba2+.Postprint (published version

    LmjF.22.0810 from Leishmania major Modulates the Th2-Type Immune Response and Is Involved in Leishmaniasis Outcome

    A novel serine/threonine protein kinase, LmjF.22.0810, was recently described in Leishmania major. After generating an L. major cell line overexpressing LmjF.22.0810 (named LmJ3OE), the ability of this novel protein to modulate the Th2-type immune response was analyzed. Our results suggest that the protein kinase LmjF.22.0810 might be involved in leishmaniasis outcomes. Indeed, our study outlined the LmJ3OE parasites infectivity in vitro and in vivo. Transgenic parasites displayed lower phagocytosis rates in vitro, and their promastigote forms exhibited lower expression levels of virulence factors compared to their counterparts in control parasites. In addition, LmJ3OE parasites developed significantly smaller footpad swelling in susceptible BALB/c mice. Hematoxylin–eosin staining allowed the observation of a lower inflammatory infiltrate in the footpad from LmJ3OE-infected mice compared to animals inoculated with control parasites. Gene expression of Th2-associated cytokines and effectors revealed a dramatically lower induction in interleukin (IL)-4, IL-10, and arginase 1 (ARG1) mRNA levels at the beginning of the swelling; no expression change was found in Th1-associated cytokines except forIL-12. Accordingly, such results were validated by immunohistochemistry studies, illustrating a weaker expression of ARG1 and a similar induction for inducible NO synthase (iNOS) in footpads from LmJ3OE-infected mice compared to control L. major infected animals. Furthermore, the parasite burden was lower in footpads from LmJ3OE-infected mice. Our analysis indicated that such significant smaller footpad swellings might be due to an impairment of the Th2 immune response that subsequently benefits Th1 prevalence. Altogether, these studies depict LmjF.22.0810 as a potential modulator of host immune responses to Leishmania. Finally, this promising target might be involved in the modulation of infection outcome

    Occupational and Environmental Risks from Mancozeb in Aviation Facilities in the Banana Producing Area of Teapa, Tabasco, Mexico

    Purpose: To identify occupational risks and practices in the four aerial fumigation facilities in the Sierra banana growing region and to evaluate environmental impacts from Mancozeb use.Methods: Occupational health risks were identified and practices in the four aerial fumigation facilities in the Sierra banana growing region were documented. Semi-structured interviews were conducted with installation managers. To evaluate environmental impact, samples of water and soil were collected and analysed in an acute toxicity bioassay with Vibrio fischeri, using uncontaminated soil and water to establish background levels.Results: Even in the largest and best-equipped facility, serious risks to workers’ health were encountered from inadequate use of protective equipment by workers. Furthermore, excessive use of pesticide, lack of re-use of unused product, poor facility design and poor use of wash-water were observed, resulting in  maximum toxicity of > 5 times background in discharged water. Parallel soil samples were > 20 times  more toxic than background, being most toxic at > 0.5 km from the discharge point.Conclusion: Areas of opportunity to improve worker health and the environment include proper and  consistent use of protective equipment, re-use of unused product, wastewater reduction and appropriate wastewater treatment.Keywords: Black Sigatoka, Ethylene thiourea, Fumigation, Pesticide, Mancozeb, Airports, Microtox, Vibrio fischeri, Occupational health risks, Toxicit

    Disconnecting Symmetry Breaking from Seeded Growth for the Reproducible Synthesis of High Quality Gold Nanorods

We demonstrate herein that the reproducibility and reliability of colloidal gold nanorod (AuNR) synthesis can be greatly improved by disconnecting the symmetry-breaking event from the seeded growth process. We have used a modified silver-assisted seeded growth method in the presence of the surfactant hexadecyltrimethylammonium bromide and n-decanol as a co-surfactant to prepare small AuNRs in high yield, which were then used as seeds for the growth of high quality AuNR colloids. Whereas the use of n-decanol provides a more-rigid micellar system, the growth on anisotropic seeds avoids sources of irreproducibility during the symmetry breaking step, yielding uniform AuNR colloids with narrow plasmon bands, ranging from 600 to 1270 nm, and allowing the fine-tuning of the final dimensions. This method provides a robust route for the preparation of high quality AuNR colloids with tunable morphology, size, and optical response in a reproducible and scalable manner

    General Microbiota of the Soft Tick Ornithodoros turicata Parasitizing the Bolson Tortoise (Gopherus flavomarginatus) in the Mapimi Biosphere Reserve, Mexico

    The general bacterial microbiota of the soft tick Ornithodoros turicata found on Bolson tortoises (Gopherus flavomarginatus) were analyzed using next generation sequencing. The main aims of the study were to establish the relative abundance of bacterial taxa in the tick, and to document the presence of potentially pathogenic species for this tortoise, other animals, and humans. The study was carried-out in the Mapimi Biosphere Reserve in the northern-arid part of Mexico. Bolson tortoises (n = 45) were inspected for the presence of soft ticks, from which 11 tortoises (24.4%) had ticks in low loads (1–3 ticks per individual). Tick pools (five adult ticks each) were analyzed through 16S rRNA V3–V4 region amplification in a MiSeq Illumina, using EzBioCloud as a taxonomical reference. The operational taxonomic units (OTUs) revealed 28 phyla, 84 classes, 165 orders, 342 families, 1013 genera, and 1326 species. The high number of taxa registered for O. turicata may be the result of the variety of hosts that this tick parasitizes as they live inside G. flavomarginatus burrows. While the most abundant phyla were Proteobacteria, Actinobacteria, and Firmicutes, the most abundant species were two endosymbionts of ticks (Midichloria-like and Coxiella-like). Two bacteria documented as pathogenic to Gopherus spp. were registered (Mycoplasma spp. and Pasteurella testudinis). The bovine and ovine tick-borne pathogens A. marginale and A. ovis, respectively, were recorded, as well as the zoonotic bacteria A. phagocytophilum,Coxiella burnetii, and Neoehrlichia sp. Tortoises parasitized with O. turicata did not show evident signs of disease, which could indicate a possible ecological role as a reservoir that has yet to be demonstrated. In fact, the defense mechanisms of this tortoise against the microorganisms transmitted by ticks during their feeding process are still unknown. Future studies on soft ticks should expand our knowledge about what components of the microbiota are notable across multiple host–microbe dynamics. Likewise, studies are required to better understand the host competence of this tortoise, considered the largest terrestrial reptile in North America distributed throughout the Chihuahuan Desert since the late Pleistocene

    A novel HPV 16 L1-based chimeric virus-like particle containing E6 and E7 seroreactive epitopes permits highly specific detection of antibodies in patients with CIN 1 and HPV-16 infection

    <p>Abstract</p> <p>Background</p> <p>The presence of IgG antibodies to HPV-16 L1-virus like particles (VLPs) in serum has been reported as a result of persistent exposure to the virus and as a marker of disease progression. However, detection of VLP-specific antibodies in sera does not always indicate a malignant lesion as positive results may also be due to a nonmalignant viral infection. Furthermore, malignant lesions are associated with an increased antibody titer for E6 and E7 proteins. The aim of this study was to develop an ELISA using a novel chimeric virus-like particle (cVLP) encoding an L1 protein fused with a string of HPV-16 E6 and E7 seroreactive epitopes to its C-terminus to be used for detection of HPV-16 specific antibodies in patients with cervical intraepithelial lesion grade 1 (CIN 1).</p> <p>Results</p> <p>The sera of 30 patients with CIN 1 who also tested positive for HPV-16 DNA and of 30 age-matched normal donors negative for HPV infection were tested for the presence of IgG antibodies specific for either VLP-L1 (HPV-16 L1), gVLP (derived from Gardasil), or cVLP by ELISA. The cVLP-reactive sera yielded two distinct groups of results: (H) reactivity levels that presented very strong cVLP-specific titers, and (L) reactivity levels with significantly lower titers similar to those obtained with VLP-L1 and gVLP antigens. Additionally, the sera that presented the higher cVLP titers closely matched those that had significantly stronger reactivity to E6 and E7 epitopes. Interestingly, the samples with the highest titers corresponded to patients with the higher numbers of sexual partners and pregnancies. On the other hand only 4 out of the 12 sera that harbored antibodies with VLP neutralizing ability corresponded to the group with high cVLP antibody titers.</p> <p>Conclusion</p> <p>We report for the first time that chimeric particles containing HPV-16 L1 protein fused with E6 and E7 seroreactive epitopes enable much better detection of IgG antibodies in the sera of CIN 1 patients positive for HPV-16 infection than those obtained with VLPs containing only the HPV-16 L1 protein. We also found that the sera with higher cVLP antibody titers corresponded to patients with more sexual partners and pregnancies, and not always with to those with a high neutralizing activity. This novel assay could help in the development of a tool to evaluate cervical cancer risk.</p

    Iridoid esters from Valeriana pavonii Poepp. & Endl. as GABAA modulators: Structural insights in their binding mode and structure-activity relationship

    Context: Valeriana pavonii Poepp. & Endl. (Caprifoliaceae), is a plant used in traditional medicine as a tranquilizer in Colombia. Valerian extracts have been widely used since ancient times for their sedative and anxiolytic properties; however, the way its active metabolites, including iridoids, interact on their respective targets is not fully understood. Aims: To isolate and identificate active iridoid esters from V. pavonii. Perform in vitro inhibition assays and computational analyses to study their possible interaction on the benzodiazepine site of the GABAA receptor. Methods: Two compounds were obtained from dichloromethane and petroleum ether fractions of V. pavonii, respectively, by chromatographic techniques. The structural elucidation was performed by NMR and spectroscopic analyses. In vitro inhibition assays of the binding of 3H-flunitrazepam (3H-FNZ) for the benzodiazepine binding site of the GABAA receptor (BDZ-bs of the GABAA receptor) were carried out. Results: Two iridoid esters, hydrine-type valepotriates (compounds 1 and 2), were reported for the first time in V. pavonii. Both iridoids, 1 and 2, inhibited the binding of 3H-FNZ on the BDZ-bs of the GABAA receptor (40% at 300 µM). Docking studies and MMGBSA calculations revealed that these compounds exhibited molecular interactions with crucial residues of the benzodiazepine site, similar to those observed for drugs like flunitrazepam, diazepam, and flumazenil. Conclusions: These findings contribute to understanding the in vivo activity of extracts of Valeriana pavonni on the central nervous system, which showed promising effects, especially as anticonvulsants, sedative-hypnotics, and antidepressants, through the modulation of the GABAergic system by hydrine-type valepotriates and its derivatives

    Magnesium Inhibits Wnt/β-Catenin Activity and Reverses the Osteogenic Transformation of Vascular Smooth Muscle Cells

    Magnesium reduces vascular smooth muscle cell (VSMC) calcification in vitro but the mechanism has not been revealed so far. This work used only slightly increased magnesium levels and aimed at determining: a) whether inhibition of magnesium transport into the cell influences VSMC calcification, b) whether Wnt/β-catenin signaling, a key mediator of osteogenic differentiation, is modified by magnesium and c) whether magnesium can influence already established vascular calcification. Human VSMC incubated with high phosphate (3.3 mM) and moderately elevated magnesium (1.4 mM) significantly reduced VSMC calcification and expression of the osteogenic transcription factors Cbfa-1 and osterix, and up-regulated expression of the natural calcification inhibitors matrix Gla protein (MGP) and osteoprotegerin (OPG). The protective effects of magnesium on calcification and expression of osteogenic markers were no longer observed in VSMC cultured with an inhibitor of cellular magnesium transport (2-aminoethoxy-diphenylborate [2-APB]). High phosphate induced activation of Wnt/β-catenin pathway as demonstrated by the translocation of β-catenin into the nucleus, increased expression of the frizzled-3 gene, and downregulation of Dkk-1 gene, a specific antagonist of the Wnt/β-catenin signaling pathway. The addition of magnesium however inhibited phosphate-induced activation of Wnt/β-catenin signaling pathway. Furthermore, TRPM7 silencing using siRNA resulted in activation of Wnt/β-catenin signaling pathway. Additional experiments were performed to test the ability of magnesium to halt the progression of already established VSMC calcification in vitro. The delayed addition of magnesium decreased calcium content, down-regulated Cbfa-1 and osterix and up-regulated MGP and OPG, when compared with a control group. This effect was not observed when 2-APB was added. In conclusion, magnesium transport through the cell membrane is important to inhibit VSMC calcification in vitro. Inhibition of Wnt/β-catenin by magnesium is one potential intracellular mechanism by which this anti-calcifying effect is achieved

    The Gaia spectrophotometric standard stars survey - IV. Results of the absolute photometry campaign

    We present Johnson-Kron-Cousins BVRI photometry of 228 candidate spectrophotometric standard stars for the external (absolute) flux calibration of Gaia data. The data were gathered as part of a 10-yr observing campaign with the goal of building the external grid of flux standards for Gaia and we obtained absolute photometry, relative photometry for constancy monitoring, and spectrophotometry. Preliminary releases of the flux tables were used to calibrate the first two Gaia releases. This paper focuses on the imaging frames observed in good sky conditions (about 9100). The photometry will be used to validate the ground-based flux tables of the Gaia spectrophotometric standard stars and to correct the spectra obtained in non-perfectly photometric observing conditions for small zero-point variations. The absolute photometry presented here is tied to the Landolt standard stars system to ≃1 per cent or better, depending on the photometric band. Extensive comparisons with various literature sources show an overall ≃1 per cent agreement, which appears to be the current limit in the accuracy of flux calibrations across various samples and techniques in the literature.
