    TBL1 is required for the mesenchymal phenotype of transformed breast cancer cells

    The epithelial-to-mesenchymal transition (EMT) and its reversion (MET) are related to tumor cell dissemination and migration, tumor circulating cell generation, cancer stem cells, chemoresistance, and metastasis formation. To identify chromatin and epigenetic factors possibly involved in the process of EMT, we compare the levels of expression of epigenetic genes in a transformed human breast epithelial cell line (HMEC-RAS) versus a stable clone of the same cell line expressing the EMT master regulator ZEB1 (HMEC-RAS-ZEB1). One of the factors strongly induced in the HMEC-RAS-ZEB1 cells was Transducin beta-like 1 (TBL1), a component of the NCoR complex, which has both corepressor and coactivator activities. We show that TBL1 interacts with ZEB1 and that both factors cooperate to repress the promoter of the epithelial gene E-cadherin (CDH1) and to autoactivate the ZEB1 promoter. Consistent with its central role, TBL1 is required for mesenchymal phenotypes of transformed breast epithelial and breast cancer cell lines of the claudin-low subtype. Importantly, a high expression of the TBL1 gene correlates with poor prognosis and increased proportion of metastasis in breast cancer patients, indicating that the level of TBL1 expression can be used as a prognostic marker.Ministerio de Economía y Competitividad BFU2014-53543-P, BFU2017-85420-RJunta de Andalucía BIO-32

Rowling. Se exponen fragmentos de sus novelas en los que se hace referencia a las diferentes especialidades médicas, así como a sus ejecutantes. La medicina es un elemento indispensable en la construcción de mundos fi cticios; la personalidad del médico que se muestra en las novelas de Harry Potter es apenas distinta de la de los profesionales en la vida real. Abstract: Medicine has a significant influence in fiction literature. This essay emphasizes the association of the contemporary practice of medicine and the narrative work of Joanne K. Rowling´s novels. It shows passages of her novels in which she talks about the different medical specialties and its performers. Medicine is an indispensable element for the construction of fictitious worlds, and the physician’s personality shown in Harry Potter´s novels is almost the same that professionals perform in real life

    La sistematización de los procesos de educación Artística que adelantan la corporación Autónoma Forjar (CAF) y el Centro de Promoción y Cultura (CPC), visibiliza los aportes que la experiencia artística hace a la educación popular. Estas organizaciones desarrollan pedagogías alternativas en la zona suroccidental de Kennedy [en Bogotá] para transformar las dinámicas socioculturales [derivadas de las causas históricas que alimentan el conflicto armado interno] que se viven en este sector. Se propone interpretar estos procesos a través de una lectura sujeto(a)-sujeto(a), per sé una actitud autocrítica periódica, con el fin de aportar a los objetivos que se trazan estas organizaciones, que han logrado pervivir en el tiempo a través de la fórmula escogida -la educación artística- que les ha permitido oxigenarse y reinventarse. Es aquí donde se hace hincapié en que su praxis de educación artística tiene unas apuestas políticas y éticas particulares, que difieren de la de los organismos institucionales que tienen como misión la promoción de mercados culturales. La distancia política de ambas apuestas, permiten involucrar variables conceptuales que dejan entrever los tránsitos y matices de la gestión entre ambas fórmulas de las organizaciones populares. Esto se debe a los contextos de emergencia de ambos procesos organizativos, del desarrollo de capacidades (infraestructura, financiación, relevos generacionales etc.) Y la lectura de realidad, punto de partida de las acciones transformadoras en sus respectivos territorios

    Merotelic attachments and non-homologous end joining are the basis of chromosomal instability

    Although the large majority of solid tumors show a combination of mitotic spindle defects and chromosomal instability, little is known about the mechanisms that govern the initial steps in tumorigenesis. The recent report of spindle-induced DNA damage provides evidence for a single mechanism responsible for the most prominent genetic defects in chromosomal instability. Spindle-induced DNA damage is brought about by uncorrected merotelic attachments, which cause kinetochore distortion, chromosome breakage at the centromere, and possible activation of DNA damage repair pathways. Although merotelic attachments are common early in mitosis, some escape detection by the kinetochore pathway. As a consequence, a proportion of merotelic attachments gives rise to chromosome breakage in normal cells and in carcinomas. An intrinsic chromosome segregation defect might thus form the basis of tumor initiation. We propose a hypothesis in which merotelic attachments and chromosome breakage establish a feedback loop that results in relaxation of the spindle checkpoint and suppression of anti-proliferative pathways, thereby promoting carcinogenesis

    Quantification of Boron Compound Concentration for BNCT Using Positron Emission Tomography

    Boron neutron capture therapy requires a 2 mM 10B concentration in the tumor. The well-known BNCT patient treatment method using boronophenylalanine (BPA) as a boron-carrying agent utilizes [18F]fluoroBPA ([18F]FBPA) as an agent to qualify for treatment. Precisely, [ 18F]FBPA must have at least a 3:1 tumor to background tissue ratio to qualify the patient for BNCT treatment. Normal, hyperplasia, and cancer thyroids capture iodine and several other large ions, including BF4 −, through a sodium-iodine symporter (NIS) expressed on the cell surface in normal conditions. In cancer, NIS is also expressed within the thyroid cell and is not functional. Methods: To visualize the thyroids and NIS, we have used a [18F]NaBF4 positron emission tomography (PET) tracer. It was injected into the tail veins of rats. The [18F]NaBF4 PET tracer was produced from NaBF4 by the isotopic exchange of natural 19F with radioactive 18F. Rats were subject to hyperplasia and tumor-inducing treatment. The NIS in thyroids was visualized by immunofluorescence staining. The boron concentration was calculated from Standard Uptake Values (SUV) in the PET/CT images and from the production data. Results: 41 MBq, 0.64 pmol of [18F]NaBF4 PET tracer that contained 0.351 mM, 53 nmol of NaBF4 was injected into the tail vein. After 17 min, the peak activity in the thyroid reached 2.3 MBq/mL (9 SUVmax). The natB concentration in the thyroid with hyperplasia reached 381 nM. Conclusions: Such an incorporation would require an additional 110 mg/kg dose of [ 10B]NaBF4 to reach the necessary 2 mM 10B concentration in the tumor. For future BNCT treatments of thyroid cancer, contrary to the 131I used now, there is no post-treatment radioactive decay, the patient can be immediately discharged from hospital, and there is no six-month moratorium for pregnancy. This method can be used for BNCT treatment compounds of the type R-BFn, where 1 <= n <= 3, labeled with 18F relatively easily, as in our example. A patient may undergo injection of a mixture of nonradioactive R-BFn to reach the necessary 10B concentration for BNCT treatment in the tumor together, with [18F]R-BFn for boron mapping.Spanish RTI2018-098117-B-C21 projec

    Human activities and inundation risks in the plain of the Guadalquivir River (in the reach between Palam del Río and Seville)

    Se presenta la ocupación antrópica de la llanura de inundación del Guadalquivir desde época histórica. Con ello se pone de manifiesto la falta de una planificación integrada frente al riesgo de inundación de la misma desde mediados del siglo XX hasta la actualidad. Esta situación genera cambios geomorfológicos irreversibles en el cauce y un comportamiento diferenciado en el riesgo potencial de inundación. En la ciudad de Sevilla, las intervenciones estructurales unidas a la incisión del cauce permiten evacuar las crecidas para magnitudes inferiores a 3.500 m3/s. Aguas arriba de Cantillana, la colmatación del cauce vinculada a la presa, la convergencia de afluentes regulados y la expansión urbana, provocan en la actualidad riesgos de inundación en zonas históricamente no inundables para estos caudales.We present the anthropic occupation of the flood plains of the Guadalquivir River from historic times. With it, we illustrate the lack of an integrated plan that would defend it from the risk of a flood from the middle of 20th century until today. This situation creates irreversible geomorphological changes in the riverbed and a differentiated behavior in the potential risk of flooding. In Seville, the structural works together with the incision of the riverbed permit the draining of the flooding of magnitudes lower than 3.500 m3/s. Up the river from Cantillana, the filling of the riverbed linked to the damn, the confluence of the regulated tributaries and the urban spreading, provoke today risks of floods in historically non-floodable areas for these water flows

The objective of this study is to determine the stability region taking into account the electrical and control signal of the generator. The generator uses the concept of the electrostatic machine for the model of the generator.
