    Correlative Light and Electron Microscopy of Platelet Adhesion and Fibrinogen Receptor Expression using Colloidal-Gold Labeling

    Differences in the shape change responses of platelets to various polymers may determine the thrombotic potential of these materials. Substrate-dependent variation in the expression and motility of the platelet fibrinogen receptor may underlie these differences due to this ligand\u27s essential role in platelet aggregation. In this study we examine platelet activation on polyetherurethaneureas (PEUUs) presently being evaulated for vascular prosthetic applications. These polymers are prepared as 50-100nm thin films suitable as substrates for consecutive light microscopy, high voltage electron microscopy (HVEM), and SEM. 18nm colloidal gold coupled to fibrinogen permits visualization of that receptor\u27s motility in living cells by video-enhanced light microscopy. Subsequent HVEM and SEM of identified cells provides correlative ultrastructure and surface morphology. The use of these novel support films coupled with the multiple modes of microscopy and colloidal gold labeled ligands permits in depth study of the molecular biology of cell adhesion to materials with varied, and known, surface properties. The motility of the platelet fibrinogen receptor was related to the extent of cytoskeletal reorganization, which, in turn, was influenced by polymer surface energetics. Platelets adherent to more hydrophobic PEUUs had greater receptor mobility and receptor redistribution than platelets adherent to more hydrophilic PEUUs. The most extensive receptor motility and redistribution was observed on Formvar, a non-PEUU with low surface-water energy, suggesting that additional surface properties are of importance in determining platelet spreading and fibrinogen receptor motility

    The dialogics of satire : foci and faultlines in George Orwell's Animal Farm and Nineteen Eighty-Four

    Dissertation (PhD)--Stellenbosch University, 2000.ENGLISH ABSTRACT: This thesis uses Mikhail Bakhtin's theory of dialogism, as well as postmodernism, to open up faultlines in satire, and to explore and challenge various perceptions and discourses surrounding and related to it. Both dialogism and postmodernism are used to suggest fresh approaches to satire, by repositioning it in relation to other discourses and reframing it as a complex dynamic, rather than a closed and inflexible system. Chapter 1 of the thesis opens with an historical survey of the beginnings and subsequent development of satire. It also contains a general discussion of the nature of satiric strategies and opens the door for the incorporation of postmodern perspectives into the argument. Chapter 2 contrasts the issues of morality and re-presentation in satire, arguing that satirists do not simply invite their audience to condemn, but offer them an opportunity to discover alternative worlds. The affinity between satire and postmodernism is emphasised by the postmodern predilection for modes highly favoured by satire: allegory, parody and fantasy. In Chapter 3 the issue of language and its referents is explored, starting with Saussure's theory of how the signifier and the signified function. It is argued that satire has never respected this fixed relationship, and that it is in this respect similar to deconstruction. The last part of the chapter is devoted to examining four key socio-political discourses - psychoanalysis, ideology, propaganda and political myth - in relation to satire. These four discourses are, like satire, intent on influencing the perceptions which people have of the world. The intention in juxtaposing these discourses is to create a dialogic process which will throw a fresh light on all of them, including satire itself. The four socio-political discourses named above play an important part in Animal Farm and Nineteen Eighty-Four, and are relevant to the subsequent discussion of these novels. Chapter 4 consists of a detailed discussion of Animal Farm, in which the various layers comprising the work are examined. The satirical aspects of the novel are closely related to the fabular and fairy tale elements which are an important part of its constitution. These elements or levels are juxtaposed with the historical details alluded to continuously in Animal Farm and indicate its close concern with the world outside the novel. Chapter 5 consists of a detailed exploration of Nineteen Eighty-Four, which is illuminated by a process of dialogism between the modernist ideology from which the novel springs and the postmodern perspective introduced into the thesis, as well as the four socio-political discourses mentioned earlier. The main postmodern theories used in this chapter are those of Foucault. The last section of the thesis demonstrates how Orwell's personal experience drives his satire, and relates this specifically to a discussion of utopia / dystopia in satire.AFRIKAANSE OPSOMMING: Die Dialogiek van Satire: Fokuspunte en Breuke in Orwell se Animal Farm en Nineteen Eighty-Four: Hierdie proefskrif maak gebruik van Mikhail Bakhtin se teorie van dialogisme, sowel as die postmodernisme, om die breuke in satire bloot te le, en om die verskillende persepsies en diskoerse wat verband hou met die satire te ondersoek en te bevraagteken. Beide die dialogisme en die postmodernisme word gebruik om nuwe perspektiewe op satire te open, deur dit te herposisioneer in verhouding tot ander diskoerse en dit voor te stel in terme van 'n komplekse dinamika eerder as 'n geslote en onbuigsame sisteem. Die eerste hoofstuk van die proefskrif begin met 'n historiese oorsig van die oorspronge en daaropvolgende ontwikkeling van satire.     The Proportional Odds Model for Assessing Rater Agreement with Multiple Modalities

    In this paper, we develop a model for evaluating an ordinal rating systems where we assume that the true underlying disease state is continuous in nature. Our approach in motivated by a dataset with 35 microscopic slides with 35 representative duct lesions of the pancreas. Each of the slides was evaluated by eight raters using two novel rating systems (PanIN illustrations and PanIN nomenclature),where each rater used each systems to rate the slide with slide identity masked between evaluations. We find that the two methods perform equally well but that differentiation of higher grade lesions is more consistent across raters than differentiation across raters for lower grade lesions. A proportional odds model is assumed, which allows us to estimate rater-specific thresholds for comparing agreement. In this situation where we have two methods of rating, we can determine whether the two methods have the same thresholds and whether or not raters perform equivalently across methods. Unlike some other model-based approaches for measuring agreement, we focus on the interpretation of the model parameters and their scientific relevance. We compare posterior estimates of rater-specific parameters across raters to see if they are implementing the intended rating system in the same manner. Estimated standard deviation distributions are used to make inferences as to whether raters are consistent and whether there are differences in rating behaviors in the two rating systems under comparison

    Design optimization of a high-power transformer for three-phase dual active bridge DC-DC converter for MVDC grids

    High-power DC-DC converter will be one of the essential technologies for the future DC grids. Especially, a three-phase dual-active bridge DC-DC (3DAB) Converter is highly suitable for high-power DC systems. Key component within this converter is the high power transformer operated at a medium frequency (MF) range. The design and optimization of this key component is presented in this paper. The transformer provides galvanic isolation from low voltage level to medium level or high voltage level and provides stepping up or down of the output (secondary) voltage. At first, a design of three-phase medium frequency transformer is developed. The designed transformer is then validated using two-dimensional (2D) transient finite element analysis (FEA) and performance under the non-sinusoidal excitation is determined. After that, the optimization is carried out, for the set of selected design variables, in order to enhance the power density and efficiency of the targeted transformer. Finally, the performance of the full system, 3DAB converter, is determined using the parameters of optimized transformer. Also, two different rated input voltages for 4MW/1kHz three-phase transformer are considered and analysed in this paper

    Microbial ecology of Rum Jungle II: environmental study of two flooded opencuts and smaller, associated water bodies.

    The microbial status of the flooded Intermediate and White's opencuts of the abandoned uranium mine at Rum Jungle was investigated by sampling the water column and sediments of several areas in each opencut. Smaller water bodies associated with the experimental heap-leach pile were also investigated. Several groups of bacteria were identified and population sizes were estimated using selective media techniques. Various physicochemical parameters of each sample were determined and correlated with the occurence of bacteria. Both opencuts although behaving differently were found to be heavily polluted by sulphuric acid and heavy metals White's more so than Intermediate. White's opencut was found to be stratified into an aerobic zone about five metres deep and a microaerophilic zone below this. Large populations of Thiobacillus ferrooxidans and autotrophic sulphur-oxidising bacteria indicated that degradation of sulphidic minerals in the walls and floors of the opencuts was still occurring. The isolation of T. ferrooxidans from sediments also containing anaerobic bacterial species suggested that T. ferrooxidans was degrading sulphidic minerals either anaerobically or microaerophilically. The smaller water bodies also were found to be heavily polluted by acid and heavy metals from drainage and seepage from the sulphidic heap-leach pile

    Preliminary Survey of Chemical Contaminants in Water, Sediment, and Aquatic Biota at Selected Sites in Northeastern Florida Bay and Canal C-111

    Several actions are under way to alter water management capabilities and practices in south Florida in order to restore a more natural hydroperiod for the Everglades. Because relatively little research has been conducted on contaminants entering Florida Bay, we undertook a preliminary study in June 1995 to determine contaminant concentrations in surface water, sediment, and biota prior to major changes in water management. The areas studied were the C-111 canal (five sites) beginning just above water control structure S-197 and extending to Manatee Bay (part of the Biscayne Bay system), Shell Creek (three sites), the mouth of Taylor River, Trout Creek (two sites), and a site near the Key Largo Ranger Station. Hydrographic observations were made at each site, and samples of water and sediment were collected at each of the five areas except the Key Largo site. Bioresidues were determined for indigenous oysters collected from the C-111 canal and Shell Creek and for transplanted oysters exposed at the Key Largo site, the C-111 canal, Shell Creek, Taylor River, and Trout Creek for 5-29 days. Water samples were analyzed for selected organochlorine pesticides and metals; sediments were analyzed for the same compounds and for polycyclic aromatic hydrocarbons (PAHs). Fish filets and shucked oysters were analyzed for selected organochlorine pesticides, polychlorinated biphenyl (PCB) congeners, and metals. Other than low dissolved oxygen at some C-111 canal sites, water quality was generally good. Most contaminants were below water and sediment quality guidelines designed to protect aquatic life, and contaminant concentrations in oysters were low. Threshold effect levels were exceeded for some organochlorine pesticides and PAHs in some sediment samples, but all values measured were below probable effect levels. Low concentrations of organochlorine pesticides were detected in sediments from the C-111 canal, Shell Creek, Taylor River, and Trout Creek; except for Taylor River, low concentrations of PCB congeners and PAHs were detected from the same areas. The only organochlorine pesticide detected in filets from seven fish was 3.2 v-g DDE/kg (wet weight) in a sea catfish. Total mercury concentrations in filets of four species of fishes ranged from 0.53 to 1.3 μg/g, wet weight, which falls within a limited consumption advisory by the State of Florida
