Alocação de biomassa e a distribuição de raízes do híbrido interespecífico de palma de óleo (Elaeis o/e/fera x Elaeis guineensis).

No entanto, a produção em parte desta área tem sido limitada pela ocorrência da anomalia Amarelecimento Fatal (AF), cujo sintoma inicial é o apodrecimento de raízes de absorção. Esse sintoma pode ser mais severo em materiais genéticos de palma de óleo que apresentem sistema radicular reduzido. Avaliamos características morfológicas de raízes e parte aérea de mudas de três genótipos de palma de óleo com graus diferentes de resistência ao AF: Tenera
(sensível), Caiaué (resistente) e Flíbrido Interespecífico (resistente). O Híbrido Interespecífico apresentou valores de massa de raízes finas e muito finas superiores em relação ao Caiaué (155 e 99%, respectivamente) e Tenera (228 e 94%, respectivamente). O Híbrido Interespecífico também apresentou valores superiores de comprimento de raízes finas e muito finas em relação ao Caiaué (155 e 99% respectivamente) e Tenera (228 e 94% respectivamente). Em um outro estudo avaliamos a biomassa abaixo do solo em um plantio comercial de híbrido interespecífico de palma de óleo (Elaeis oleifera cortês x Elaeis guineensis jacq.), cultivar BRS Manicoré, no município de Moju, nordeste do estado do Pará. Amostras de raízes foram obtidas por meio de coleta de monólitos em trincheiras com 4 metros de comprimento, 50 cm de largura e 290 cm de profundidade. Foram coletados monólitos em 8 camadas (2 - 15, 2 - 30 e 4 de 50 cm de profundidade) e em cada camada, foram coletados 8 monólitos medindo 50 cm de comprimento, 30 de largura e a altura de acordo com a camada coletada. Em laboratório, as raízes foram classificadas de acordo com seu diâmetro em primárias (^5,0 mm), secundárias (1-4,9 mm), terciárias (0,5 - 0,9 mm) e quaternárias (0,25 - 0,49 mm). A profundidade de 15-30 cm concentra mais de 50% da DMS total (50,77% TP e 59,91 % TC), e destes, 81,49% da DMS é de raízes primárias. As terciárias representam 8,96% do total da DMS de TP e 4,85% de TC e a quantidade de raízes quaternárias na região entre plantas é 78,05% maior do que a quantidade encontrada na região de carreamento. O cultivo de palma de óleo com 8 anos de idade apresenta uma biomassa total de raízes superior à de culturas sazonais, vegetação secundária, cultivo Tradicional, cultivo de Maracujá e pimenta do
reino.Brazil has an extensive area with climate and soil suitable for oil palm planting. However, production in part of this area has been limited by the occurrence of the Fatal Yellowing (FA) anomaly, whose initial symptom is rotting of absorption roots. This symptom may be more severe in oil palm genetic materials that have a reduced root system. We evaluated morphological characteristics of roots and aboveground parts of seedlings of three oil palm genotypes with different degrees of resistance to AF: Tenera (sensitive), Caiaué (resistant) and Interspecific Hybrid (resistant). The Interspecific Hybrid showed higher fine and very fine root mass values compared to Caiaué (155 and 99%, respectively) and Tenera (228 and 94%, respectively). The Interspecific Hybrid also showed higher values of fine and very fine root length compared to Caiaue (155 and 99% respectively) and Tenera (228 and 94% respectively). In another study we evaluated the belowground biomass in a commercial plantation of interspecific oil palm hybrid (Elaeis oleifera cortês x Elaeis guineensis jacq.), cultivar BRS Manicoré, in the municipality of Moju, northeastern Pará state. Root samples were obtained by collecting monoliths in trenches 4 meters long, 50 cm wide and 290 cm deep. Monoliths were collected in 8 layers (2 - 15, 2 - 30 and 4 of 50 cm depth) and in each layer, 8 monoliths measuring 50 cm long, 30 wide and the height according to the layer collected were collected. In the laboratory, the roots were classified according to their diameter into primary (^5.0 mm), secondary (1-4.9 mm), tertiary (0.5 - 0.9 mm) and quaternary (0.25 - 0.49 mm). The depth of 15-30 cm concentrates more than 50% of the total DMS (50.77% TP and 59.91 % TC), and of these, 81.49% of the DMS is from primary roots. Portugal and Brazil: How much of ‘Our’ past is ‘Theirs’ too?
Political and cultural changes in the 1980s have had deep effects on Portuguese and Brazilian educational systems. In 1991, the Portuguese primary and secondary national curricula were defined and these statutory requirements are still in place. In Brazil, political open-mindedness in the 1990s allowed a curricular flexibility within national curricula parameters leading to a decision that primary and secondary curricula should have a common basis which would be complemented in each Brazilian state according to regional and local aims and features. As regards history, the curriculum documents recommended that teachers approach topics with a critical stance on ‘identity’ in order to make the old perception of the ‘us’ and the ‘others’ more inclusive, and to update a traditional perspective on history teaching. Some qualitative research studies carried out in Portugal and Brazil have been focusing on historical consciousness, aiming to understand students’ ideas on significance and empathy towards ‘the other’ in the context of ‘Discoveries’.info:eu-repo/semantics/publishedVersio

Processo dialógico no ensino de microbiologia na escola do campo
A escola do campo é um espaço que favorece a interlocução de saberes, contribuindo para a formação humanista e científica do jovem campesino através do ensino orientado para a sociedade. O estudo tem como objetivo identificar e discutir processos dialógicos utilizadas pelos professores no ensino de Microbiologia na escola do campo. Trata-se de um estudo descritivo, com abordagem quantitativa e qualitativa em razão do uso de elementos como: a fala cotidiana dos sujeitos, observação, registro e análise dos fatos, número, frequência e intensidade dos comportamentos dos indivíduos envolvidos. O instrumento de coleta foi o questionário utilizado junto aos alunos do 2º ano do ensino médio (234) e professores de Biologia (11) de 09 escolas do campo. A partir da perspectiva freiriana, os resultados apontam a prática pedagógica docente como uma dinâmica em construção, requerendo um aprofundamento teórico frente a realidade do campo, porém o professor destaca, no seu discurso, a importância da escuta, da discussão e da reflexão, possibilitando a transformação da realidade mediada pelo diálogo. Quanto ao ensino de Microbiologia há predomínio de aula expositiva e uso do livro didático. São pontuadas dificuldades estruturais, falta de insumos e apoio insuficiente da gestão para viabilizar atividades práticas, no entanto, alunos e professores, mesmo não tendo essa atividade, referem ser essa modalidade mais motivadora, dinâmica e significativa para o estudo dos microrganismos. Há inserção da universidade nesse espaço educacional, através de cooperação e apoio a formação do jovem no campo, seja através da partilha de espaços de aprendizagem para os alunos do ensino médio, seja na capacitação e atualização dos professores e na produção conjunta de materiais instrucionais que deem sentido ao conteúdo da Microbiologia no cotidiano do campo. Acreditamos que esse aceno, contribuirá para o processo de reflexão do ensinar e aprender nas escolas do campo.The rural school is a space that favors the interlocution of knowledge, contributing to the humanistic and scientific formation of the young peasant through the teaching oriented to the society. The objective of this study is to identify and discuss the dialogic processes used by teachers in the teaching of microbiology in the rural school. This is a descriptive study, with a quantitative and qualitative approach based on the use of elements such as: everyday speech of subjects, observation, recording and analysis of the facts, number, frequency and intensity of the behaviors of the individuals involved. The collection instrument was the questionnaire used with the students of the second year of high school (234) and teachers of Biology (11) of 09 schools in the field. From the perspective of Freire, the results point to the teaching pedagogical practice as a dynamic in construction, requiring a theoretical deepening of the reality of the field, but the teacher emphasizes in his speech the importance of listening, discussion and reflection, making possible the transformation of reality mediated by dialogue. Regarding the teaching of Microbiology there is a predominance of expository class and use of textbooks. Structural difficulties, lack of inputs and insufficient support of management to make practical activities feasible, however, students and teachers, even without this activity, point out that this modality is more motivating, dynamic and significant for the study of microorganisms. There is insertion of the university in this educational space, through cooperation and support the training of the young in the field, either through the sharing of learning spaces for high school students, or in the training and updating of teachers and joint production of instructional materials that the Microbiology content in the daily life of the field. We believe that this nod will contribute to the reflection process of teaching and learning in rural schools
In-depth analysis of the Quercus suber metabolome under drought stress and recovery reveals potential key metabolic players
Cork oak (Quercus suber L.) is a species of ecological, social and economic importance in the Mediterranean region. Given its xerophytic adaptability, the study of cork oak's response to drought stress conditions may provide important data in the global scenario of climate change. The mechanisms behind cork oak's adaptation to drought conditions can inform the design and development of tools to better manage this species under the changing climate patterns. Metabolomics is one of the most promising omics layers to capture a snapshot of a particular physiological state and to identify putative biomarkers of stress tolerance. Drastic changes were observed in the leaf metabolome of Q. suber between the different experimental conditions, namely at the beginning of the drought stress treatment, after one month under drought and post rehydration. All experimental treatments were analyzed through sPLS to inspect for global changes and stress and rehydration responses were analyzed independently for specific alterations. This allowed a more in-depth study and a search for biomarkers specific to a given hydric treatment. The metabolome analyses showed changes in both primary and secondary metabolism, but highlighted the role of secondary metabolism. In addition, a compound-specific response was observed in stress and rehydration. Key compounds such as L-phenylalanine and epigallocatechin 3-gallate were identified in relation to early drought response, terpenoid leonuridine and the flavonoid glycoside (-)-epicatechin-3'-O-glucuronide in long-term drought response, and flavone isoscoparine was identified in relation to the recovery process. The results here obtained provide novel insights into the biology of cork oak, highlighting pathways and metabolites potentially involved in the response of this species during drought and recovery that may be essential for its adaptation to long periods of drought. It is expected that this knowledge can encourage further functional studies in order to validate potential biomarkers of drought and recovery that maybe used to support decision-making in cork oak breeding programs.publishe
Regeneração de plantas de Eucalyptus globulus por embriogénese somática
A embriogénese somática (ES) foi investigada em Eucalyptus globulus com
vista a desenvolver um protocolo padrão de micropropagação, desde a
indução à aclimatização.
A propagação clonal de E. globulus recorrendo a ES tem o potencial de ir ao
encontro das necessidades industriais de material de alta qualidade e
uniforme, e de rapidamente capturar os benefícios dos programas de
melhoramento, sendo encarada como um método efectivo de conseguir
ganhos genéticos num curto espaço de tempo.
O Capítulo I faz uma revisão da importância de E. globulus, assim como de
alguns aspectos mais importantes relativos à micropropagação via
embriogénese somática, e dedica uma secção ao conhecimento actual sobre o
uso da embriogénese somática no género Eucalyptus. Finalmente, os
objectivos de investigação desta Tese são apresentados.
O Capítulo II centra-se no processo de ES primário, sobretudo nas condições
que influenciam a indução do potencial embriogénico. Este Capítulo está
dividido em 5 grandes estudos: No Capítulo II.1 o tipo de explante, a fonte de
carbohidratos e a exposição a reguladores de crescimento na embriogénese
somática é realçada. Neste Capítulo publica-se, pela primeira vez, a
regeneração de plantas por embriogénese somática nesta espécie. No
Capítulo II.2 é discutida a influência da composição salina dos meios e a
adição de antioxidantes. No Capítulo II.3 é efectuada uma comparação
preliminar entre a composição salina do meio de cultura e o conteúdo mineral
dos explantes (embriões zigóticos). No Capítulo II.4 são apresentados estudos
histocitológicos e de acumulação de reservas em E. globulus durante a
indução, expressão e diferentes estádios de desenvolvimento do embrião
somático primário. Esta informação sobre as alterações observadas contribui
para uma melhor compreensão do processo de ES primária. Finalmente, e
com base nos resultados obtidos no Capítulo II.1 e II.2, está agora disponível
um protocolo padrão (standard) para induzir ES primária e regeneração de
plantas a partir de embriões zigóticos que permitiu os estudos de controlo
genético apresentados no Capítulo II. 5. Neste estudo a frequência de ES foi
estudada em 13 famílias de polinização aberta durante três anos consecutivos
e a indução de ES provou ser uma característica variável entre as 13 famílias e
entre os anos de produção de semente testados. Baseado nestes resultados,
os estudos de controlo genético foram conduzidos recorrendo a um
cruzamento dialélico com cinco árvores parentais; os resultados obtidos
sugeriram que a indução de ES está sobre controlo de efeitos genéticos
aditivos.
No capítulo III, os estudos apresentados direccionaram-se para a ES
secundária. O estabelecimento da ES secundária só foi possível depois de se
obter a ES primária nesta espécie. Tal como é descrito no Capítulo III.1, este
processo repetitivo de ES mostrou-se reprodutível levando à obtenção de
grande quantidade de material embriogénico. Os resultados obtidos,
relativamente ao nível de ploidia e conteúdo de DNA deste material
embriogénico, comprovaram que o protocolo utilizado na proliferação de
embriões somáticos secundários não afectou estes parâmetros. De forma a
optimizar a manutenção/ multiplicação e conversão destes embriões em
plantas, alguns factores como a composição do meio basal, reguladores de
crescimento e intensidade luminosa foram investigados no Capítulo III.2. No
capítulo III.3, é estudado o último e crucial passo do processo ES, a
aclimatização. A aclimatização de plantas derivadas de ES é acompanhado
por estudos histocitológicos e por análises de variação de ploidia.
Finalmente, no Capítulo IV são apresentadas as conclusões da Tese de
Doutoramento, onde alguns aspectos inovadores deste processo de ES nesta
espécie recalcitrante são realçados.
ABSTRACT: Somatic embryogenesis (SE) was investigated in Eucalyptus globulus in order
to develop a standard plant micropropagation protocol from induction to plant
acclimatization. Clonal propagation of E. globulus through somatic
embryogenesis has the potential to meet the increasing industrial demands for
high quality uniform materials and to rapidly capture the benefits of breeding
programs, being regarded as an effective method for achieving higher genetic
gains in a shorter time.
Chapter I revises the importance of E. globulus, as well as some of the most
important aspects of SE process, and gives particular emphasis on the state of
the art of SE in Eucalyptus genus. Finally, the research objectives of this
Thesis are presented.
Chapter II is focused on primary SE process, mostly on the conditions
influencing induction of SE potential. This chapter is divided in five main
studies. In Chapter II.1, the roles of explants type, carbohydrate source and
exposure to exogenous plant growth regulators on SE are particularly
emphasized. This was the first report on plant regeneration by primary SE in
this species. In Chapter II.2 the influence of several salt medium composition
and the addition of antioxidants on SE potential are discussed. In Chapter II.3 a
preliminary comparison of salt medium composition and the mineral content of
the explants (zygotic embryos) are performed. In Chapter II.4 histocytological
and reserve accumulation studies are presented in order to describe changes
observed in E. globulus SE, during induction, expression and different
developmental stages of primary somatic embryos, providing valuable
information on the primary SE process. Finally, based on the results obtained in
Chapter II.1 and II.2, a standard protocol to induce primary SE from zygotic
embryos with plant regeneration is now available allowing studies concerning
genetic control presented in Chapter II.5. In this study the frequency of SE
induction was investigated among 13 open pollinated families during three
consecutive years and SE induction proved to be a variable character among
families and the years of seed production tested. Based on these results
genetic control was studied using a diallel mating design with five parent trees
and results suggest that SE induction is under control of genetic additive
effects.
In Chapter III, the studies presented are focused on secondary SE. The
establishment of secondary SE was possible after attaining primary SE in this
species. As described in Chapter III.1, this repetitive SE process has shown to
be reproducible leading to large amounts of embryogenic material and showed
to be, for ploidy and DNA content, a true-to-type SE process. In order to
optimize the multiplication and conversion of these somatic embryos into
plantlets, some factors such as basal medium composition, growth regulators
and light intensity are investigated in Chapter III.2. In Chapter III.3, plant
acclimatization (the last step of any SE protocol) was investigated. The
acclimatization of SE- derived plants was followed by histo-cytological studies
before ploidy analysis.
Finally, in Chapter IV the general conclusions of the present PhD Thesis are
presented, where the innovative aspects of the developed SE process in this
recalcitrant species are highlighted
