    The Enigmatic Genetic Landscape of Hereditary Hearing Loss: A Multistep Diagnostic Strategy in the Italian Population

    Hearing loss is the most frequent sensorineural disorder, affecting approximately 1:1000 newborns. Hereditary forms (HHL) represent 50–60% of cases, highlighting the relevance of genetic testing in deaf patients. HHL is classified as non-syndromic (NSHL—70% of cases) or syndromic (SHL—30% of cases). In this study, a multistep and integrative approach aimed at identifying the molecular cause of HHL in 102 patients, whose GJB2 analysis already showed a negative result, is described. In NSHL patients, multiplex ligation probe amplification and long-range PCR analyses of the STRC gene solved 13 cases, while whole exome sequencing (WES) identified the genetic diagnosis in 26 additional ones, with a total detection rate of 47.6%. Concerning SHL, WES detected the molecular cause in 55% of cases. Peculiar findings are represented by the identification of four subjects displaying a dual molecular diagnosis and eight affected by non-syndromic mimics, five of them presenting Usher syndrome type 2. Overall, this study provides a detailed characterisation of the genetic causes of HHL in the Italian population. Furthermore, we highlighted the frequency of Usher syndrome type 2 carriers in the Italian population to pave the way for a more effective implementation of diagnostic and follow-up strategies for this disease

    Genomic Studies in a Large Cohort of Hearing Impaired Italian Patients Revealed Several New Alleles, a Rare Case of Uniparental Disomy (UPD) and the Importance to Search for Copy Number Variations

    Hereditary hearing loss (HHL) is a common disorder characterized by a huge genetic heterogeneity. The definition of a correct molecular diagnosis is essential for proper genetic counseling, recurrence risk estimation, and therapeutic options. From 20 to 40% of patients carry mutations in GJB2 gene, thus, in more than half of cases it is necessary to look for causative variants in the other genes so far identified (~100). In this light, the use of next-generation sequencing technologies has proved to be the best solution for mutational screening, even though it is not always conclusive. Here we describe a combined approach, based on targeted re-sequencing (TRS) of 96 HHL genes followed by high-density SNP arrays, aimed at the identification of the molecular causes of non-syndromic HHL (NSHL). This strategy has been applied to study 103 Italian unrelated cases, negative for mutations in GJB2, and led to the characterization of 31% of them (i.e., 37% of familial and 26.3% of sporadic cases). In particular, TRS revealed TECTA and ACTG1 genes as major players in the Italian population. Furthermore, two de novo missense variants in ACTG1 have been identified and investigated through protein modeling and molecular dynamics simulations, confirming their likely pathogenic effect. Among the selected patients analyzed by SNP arrays (negative to TRS, or with a single variant in a recessive gene) a molecular diagnosis was reached in ~36% of cases, highlighting the importance to look for large insertions/deletions. Moreover, copy number variants analysis led to the identification of the first case of uniparental disomy involving LOXHD1 gene. Overall, taking into account the contribution of GJB2, plus the results from TRS and SNP arrays, it was possible to reach a molecular diagnosis in ~51% of NSHL cases. These data proved the usefulness of a combined approach for the analysis of NSHL and for the definition of the epidemiological picture of HHL in the Italian population

    Extensive soot compaction by cloud processing from laboratory and field observations

    Soot particles form during combustion of carbonaceous materials and impact climate and air quality. When freshly emitted, they are typically fractal-like aggregates. After atmospheric aging, they can act as cloud condensation nuclei, and water condensation or evaporation restructure them to more compact aggregates, affecting their optical, aerodynamic, and surface properties. Here we survey the morphology of ambient soot particles from various locations and different environmental and aging conditions. We used electron microscopy and show extensive soot compaction after cloud processing. We further performed laboratory experiments to simulate atmospheric cloud processing under controlled conditions. We find that soot particles sampled after evaporating the cloud droplets, are significantly more compact than freshly emitted and interstitial soot, confirming that cloud processing, not just exposure to high humidity, compacts soot. Our findings have implications for how the radiative, surface, and aerodynamic properties, and the fate of soot particles are represented in numerical models

    Guidelines for the management of neuroendocrine tumours by the Brazilian gastrointestinal tumour group

    Neuroendocrine tumours are a heterogeneous group of diseases with a significant variety of diagnostic tests and treatment modalities. Guidelines were developed by North American and European groups to recommend their best management. However, local particularities and relativisms found worldwide led us to create Brazilian guidelines. Our consensus considered the best feasible strategies in an environment involving more limited resources.     Toward Efficient and Printable Organic Solar Cells (Naar efficiënte en printbare organische zonnecellen)

    This thesis addresses a few of the most crucial topics in the development of polymer based organic solar cells.The first topic involves the study and characterization of novel organic polymers, with focus on low-bandgap materials able to harvest a broader portion of the solar spectrum. This is a crucial subject of research, since the efficiency of organic solar cells is strictly related to the properties of the active layer. The first part of the thesis discusses the recent progresses in the synthesis of novel polymers and highlights the fundamental properties that make them suitable for high-efficiency organic solar cells. The first chapter is dedicated to the detailed characterization of a series of soluble poly thienylene vinylene derivatives.The second topic concerns alternative deposition techniques to spin coating, the most used technique in laboratories for the production of polymer based organic solar cells. The promise of these devices, indeed, lies in their low-cost high-throughput manufacturability, but this low cost aspect can only be fully realized if the layers are deposited by in-line compatible methods.Among several techniques, ink-jet printing and spray coating are studied in depth in the second part of the thesis for the deposition of the active layer. A thorough investigation on the behavior of spray coating is reported, together with the optimization of the deposition of polymer based layers. The development of two-solvent systems extended the uniformity of the sprayed films owing to the spreading properties induced by outward Marangoni flows in the liquid, as explained in the central part of the thesis. The benefits of the enhanced uniformity are clear from the closing of the performance gap between spin coated devices and spray coated ones.The technique is particularly advantageous since the deposition is obtained as a superposition of micron-sized droplets, which either merge into a full wet layer or dry upon impact with substrate. In the latter case, the damage of the solvent on the previously deposited layers is reduced to minimum, so that spray coating can be used to deposit even a metal top contact from a metal nanoparticle based ink, as described in the last two chapters of the thesis.     Polymer based organic solar cells using ink-jet printed active layers

    Ink-jet printing is used to deposit polymer:fullerene blends suitable as active layer for organic solar cells. We show that merging of separately deposited ink droplets into a continuous, pinhole-free organic thin film results from a balance between ink viscosity and surface wetting, whereas for certain of the studied solutions clear coffee drop effect occurs for single droplets; this can be minimized for larger printed areas, yielding smooth layers with minimal surface roughness. Resulting organic films are used as active layer for solar cells with power conversion efficiency of 1.4% under simulated AM1.5 solar illumination.     Nanoparticle-based, spray-coated silver top contacts for efficient polymer solar cells

    We demonstrate a solution-processed top electrode for large area organic electronic devices. A Ag nanoparticle solution is spray-coated directly on top of an inverted bulk-heterojunction organic solar cell through a shadow mask. After sintering the Ag nanoparticle film at 150 degrees C, a temperature which is compatible with processes on flexible substrates, cells show performances comparable to those of reference devices with evaporated top-contacts.
