    Proteomic analysis of mononuclear cells of patients with minimal-change nephrotic syndrome of childhood

    Background/Aims. Recently, peripheral blood mononuclear cell transcriptome analysis has identified genes that are upregulated in relapsing minimal-change nephrotic syndrome (MCNS). In order to investigate protein expression in peripheral blood mononuclear cells (PBMC) from relapsing MCNS patients, we performed proteomic comparisons of PBMC from patients with MCNS in relapse and controls. Methods. PBMC from a total of 20 patients were analysed. PBMC were taken from five patients with relapsing MCNS, four in remission, five patients with other glomerular diseases and six controls. Two dimensional electrophoresis was performed and proteome patterns were compared. Results. Automatic heuristic clustering analysis allowed us to pool correctly the gels from the MCNS patients in the relapse and in the control groups. Using hierarchical population matching, nine spots were found to be increased in PBMC from MCNS patients in relapse. Four spots were identified by mass spectrometry. Three of the four proteins identified (l-plastin, α-tropomyosin and annexin III) were cytoskeletal-associated proteins. Using western blot and immunochemistry, l-plastin and α-tropomyosin 3 concentrations were found to be enhanced in PBMC from MCNS patients in relapse. Conclusions. These data indicate that a specific proteomic profile characterizes PBMC from MCNS patients in relapse. Proteins involved in PBMC cytoskeletal rearrangement are increased in relapsing MCNS. We hypothesize that T-cell cytoskeletal rearrangement may play a role in the pathogenesis of MCNS by altering the expression of cell surface receptors and by modifying the interaction of these cells with glomerular cell

    Presence of Palaemon macrodactylus in a Europe estuary: evidence for a successful invasion of the Gironde (SW France)

    Palaemon macrodactylus, an exotic shrimp native to the northwest Pacific Ocean, has been recorded in many estuaries along the Atlantic coast of Europe since the late 1990’s. In this study, a regular monthly survey, held since 1992 of the middle section of the Gironde estuary, revealed this species’ rapid and full colonization of the system since its appearance in samples during summer 1998. In the Gironde, the population of P. macrodactylus is self-sustaining and the species is now established there. Our study highlights two important elements that might explain its successful colonization: its ability to quickly invade a niche under-exploited by the similar native species and its greater reproductive output. The species’spatio-temporal distribution in relation to environmental variables was studied and compared with that of the native species P. longirostris. The exotic species was shown to mainly inhabit the polyhaline and mesohaline section of the estuary, which are sections generally under-exploited by females of the native species. Furthermore, fecundity analysis and larval abundance survey results revealed a higher potential fecundity and a greater proportion of larvae and juvenile stages for P. macrodactylus in the natural environment

    Nods, Nalps and Naip: intracellular regulators of bacterial-induced inflammation

    The innate immune system is the most ancestral and ubiquitous system of defence against microbial infection. The microbial sensing proteins involved in innate immunity recognize conserved and often structural components of microorganisms. One class of these pattern-recognition molecules, the Toll-like receptors (TLRs), are involved in detection of microbes in the extracellular compartment whereas a newly discovered family of proteins, the NBS-LRR proteins (for nucleotide-binding site and leucine-rich repeat), are involved in intracellular recognition of microbes and their products. NBS-LRR proteins are characterized by three structural domains: a C-terminal leucine-rich repeat (LRR) domain able to sense a microbial motif, an intermediary nucleotide binding site (NBS) essential for the oligomerization of the molecule that is necessary for the signal transduction induced by different N-terminal effector motifs, such as a pyrin domain (PYD), a caspase-activating and recruitment domain (CARD) or a baculovirus inhibitor of apoptosis protein repeat (BIR) domain. Two of these family members, Nod1 and Nod2, play a role in the regulation of pro-inflammatory pathways through NF-κB induced by bacterial ligands. Recently, it was shown that Nod2 recognizes a specific peptidoglycan motif from bacteria, muramyl dipeptide (MDP). A surprising number of human genetic disorders have been linked to NBS-LRR proteins. For example, mutations in Nod2, which render the molecule insensitive to MDP and unable to induce NF-κB activation when stimulated, are associated with susceptibility to a chronic intestinal inflammatory disorder, Crohn's disease. Conversely, mutations in the NBS region of Nod2 induce a constitutive activation of NF-κB and are responsible for Blau syndrome, another auto-inflammatory disease. Nalp3, which is an NBS-LRR protein with an N-terminal Pyrin domain, is also implicated in rare auto-inflammatory disorders. In conclusion, NBS-LRR molecules appear as a new family of intracellular receptors of innate immunity able to detect specific bacterial compounds and induce inflammatory response; the dysregulation of these processes due to mutations in the genes encoding these proteins is involved in numerous auto-inflammatory disorders.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/75732/1/j.1462-5822.2003.00304.x.pd

    Present dynamics of the savanna-forest boundary in the congolese Mayombe : a pedological, botanical and isotopic (13C and 14C) study

    Isolated savannas enclosed by forest are especially abundant in the eastern part of the Congolese Mayombe. They are about 3000 years old, and were more extensive some centuries ago. The boundary between forest and savanna is very abrupt, as a consequence of the numerous savanna fires lit by hunters. Floristic composition and vegetation structure data, organic carbon ratios, delta 14C and delta 13C measurements presented here show that forest is spreading over savanna at the present time and suggest that the rate of forest encroachment is currently between 14 and 75 m per century, and more probably about 20-50 m per century. As most savannas are less than 1 km across, such rates mean, assuming there are no changes in environmental conditions, that enclosed savannas could completely disappear in the Mayombe in about 1000-2000 years.     Cutting tool tracking and recognition based on infrared and visual imaging systems using principal component analysis (PCA) and discrete wavelet transform (DWT) combined with neural networks

    The implementation of computerised condition monitoring systems for the detection cutting tools’ correct installation and fault diagnosis is of a high importance in modern manufacturing industries. The primary function of a condition monitoring system is to check the existence of the tool before starting any machining process and ensure its health during operation. The aim of this study is to assess the detection of the existence of the tool in the spindle and its health (i.e. normal or broken) using infrared and vision systems as a non-contact methodology. The application of Principal Component Analysis (PCA) and Discrete Wavelet Transform (DWT) combined with neural networks are investigated using both types of data in order to establish an effective and reliable novel software program for tool tracking and health recognition. Infrared and visual cameras are used to locate and track the cutting tool during the machining process using a suitable analysis and image processing algorithms. The capabilities of PCA and Discrete Wavelet Transform (DWT) combined with neural networks are investigated in recognising the tool’s condition by comparing the characteristics of the tool to those of known conditions in the training set. The experimental results have shown high performance when using the infrared data in comparison to visual images for the selected image and signal processing algorithms

    Paleofire reconstruction based on an ensemble-member strategy applied to sedimentary charcoal

    Paleofire events obtained from the statistical treatment of sedimentary charcoal records rely on a number of assumptions and user's choices, increasing the uncertainty of reconstructio\ns. Among the assumptions made when analyzing charcoal series is the choice of a filtering method for raw Charcoal Accumulation Rate (CHARraw). As there is no ultimate CHAR raw filtering method, we propose an ensemble-member approach to reconstruct fire events. We modified the commonly used procedure by including a routine replicating the analysis of a charcoal record using custom smoothing parameters. Dates of robust fire events, uncertainties in fire-return intervals and fire frequencies are derived from members' distributions. An application of the method is used to quantify uncertainties due to data treatment in two CHARraw sequences from two different biomes, subalpine and boreal
