Toward dynamic pain expressions in avatars : perceived realism and pain level of diffrent action unit orders

The facial expression of pain can be decomposed in three sets of Action Units (AUs), the smallest discriminating facial movements: Brow lowering (B), Nose wrinkling + Upper lip raising (N), and Orbit tightening + Eyelid closure (O). This study compared the perception of realism and pain level from different onset orders of AUs in avatars. Seven videos of facial expressions of pain were created with four different avatars (2 women): six sequential onsets combining the three sets of AUs and one synchronized onset. 45 healthy adults (22 women; aged 23.6 ± 5.2 years) rated the realism of facial movements, and the level of intensity and unpleasantness of perceived pain. A more realistic expression was associated with the onset of O before or at the same time as N, a more intense expression was associated when B occurred last, and a higher level of unpleasantness was associated with the onset of N before B. Therefore, the sequence ONB yielded the highest ratings on both measures of realism and pain levels. These findings describe the perceived content of different orders of facial movements that could contribute to the creation of realistic pain-expressing virtual agents designed to study human-computer interactions

This is the final version. Available from Nature Research via the DOI in this record. Data availability: All data used in this review is available from publicly available and herein referenced sources. A list of included studies is provided in Supplementary Data 1. All data generated or analyzed during this study are included in this published article and its supplementary information files. Source data for the figures are available as Supplementary Data 2.BACKGROUND: Monogenic insulin resistance (IR) includes lipodystrophy and disorders of insulin signalling. We sought to assess the effects of interventions in monogenic IR, stratified by genetic aetiology. METHODS: Systematic review using PubMed, MEDLINE and Embase (1 January 1987 to 23 June 2021). Studies reporting individual-level effects of pharmacologic and/or surgical interventions in monogenic IR were eligible. Individual data were extracted and duplicates were removed. Outcomes were analysed for each gene and intervention, and in aggregate for partial, generalised and all lipodystrophy. RESULTS: 10 non-randomised experimental studies, 8 case series, and 23 case reports meet inclusion criteria, all rated as having moderate or serious risk of bias. Metreleptin use is associated with the lowering of triglycerides and haemoglobin A1c (HbA1c) in all lipodystrophy (n = 111), partial (n = 71) and generalised lipodystrophy (n = 41), and in LMNA, PPARG, AGPAT2 or BSCL2 subgroups (n = 72,13,21 and 21 respectively). Body Mass Index (BMI) is lowered in partial and generalised lipodystrophy, and in LMNA or BSCL2, but not PPARG or AGPAT2 subgroups. Thiazolidinediones are associated with improved HbA1c and triglycerides in all lipodystrophy (n = 13), improved HbA1c in PPARG (n = 5), and improved triglycerides in LMNA (n = 7). In INSR-related IR, rhIGF-1, alone or with IGFBP3, is associated with improved HbA1c (n = 17). The small size or absence of other genotype-treatment combinations preclude firm conclusions. CONCLUSIONS: The evidence guiding genotype-specific treatment of monogenic IR is of low to very low quality. Metreleptin and Thiazolidinediones appear to improve metabolic markers in lipodystrophy, and rhIGF-1 appears to lower HbA1c in INSR-related IR. For other interventions, there is insufficient evidence to assess efficacy and risks in aggregated lipodystrophy or genetic subgroups.Wellcome TrustWellcome Trus

Proximity intervention is increasingly used in Quebec’s low-income housing (HLM) to reach the residents and to encourage them to use the services offered by the community. This article presents the results of a research project on local networks, proximity intervention and the impact of their action on the residents of six low-income housing projects. By interviewing local actors, social workers and tenants and through the actor network theory, it documents the deployment of these interventions, their challenges and their impacts

AXL confers cell migration and invasion by hijacking a PEAK1-regulated focal adhesion protein network

Aberrant expression of receptor tyrosine kinase AXL is linked to metastasis. AXL can be activated by its ligand GAS6 or by other kinases, but the signaling pathways conferring its metastatic activity are unknown. Here, we define the AXL-regulated phosphoproteome in breast cancer cells. We reveal that AXL stimulates the phosphorylation of a network of focal adhesion (FA) proteins, culminating in faster FA disassembly. Mechanistically, AXL phosphorylates NEDD9, leading to its binding to CRKII which in turn associates with and orchestrates the phosphorylation of the pseudo-kinase PEAK1. We find that PEAK1 is in complex with the tyrosine kinase CSK to mediate the phosphorylation of PAXILLIN. Uncoupling of PEAK1 from AXL signaling decreases metastasis in vivo, but not tumor growth. Our results uncover a contribution of AXL signaling to FA dynamics, reveal a long sought-after mechanism underlying AXL metastatic activity, and identify PEAK1 as a therapeutic target in AXL positive tumors.ISSN:2041-172

Spelling performance on the web and in the lab

International audienceSeveral dictionary websites are available on the web to access semantic, synonymous, or spelling information about a given word. During nine years, we systematically recorded all the entered letter sequences from a French web dictionary. A total of 200 million ortho-graphic forms were obtained allowing us to create a large-scale database of spelling errors that could inform psychological theories about spelling processes. To check the reliability of this big data methodology, we selected from this database a sample of 100 frequently misspelled words. A group of 100 French university students had to perform a spelling-to-dictation test on this list of words. The results showed a strong correlation between the two data sets on the frequencies of produced spellings (r = 0.82). Although the distributions of spelling errors were relatively consistent across the two databases, the proportion of correct responses revealed significant differences. Regression analyses allowed us to generate possible explanations for these differences in terms of task-dependent factors. We argue that comparing the results of these large-scale databases with those of standard and controlled experimental paradigms is certainly a good way to determine the conditions under which this big data methodology can be adequately used for informing psychological theories
