Possibilities and limits of questionaire based soft skills evaluation as admission criteria for medical schools
Mit der Neuregelung der Studienplatzverteilung von 2005 haben die Universitäten in Deutschland die Möglichkeit, bis zu 60% der Studienplätze nach universitätseigenen Kriterien zu vergeben. Implizit wird vom Gesetzgeber und der öffentlichen Meinung gefordert, nicht-leistungsbezogene Kriterien und Persönlichkeitsmerkmale verstärkt zur Bewerberauswahl einzusetzen (Motivation, Identifikation, Vermeidung von Fehlvorstellungen). Da in Anbetracht der Bewerberzahlen mündliche Auswahlgespräche als ungeeignet erscheinen, wurde vom Fachbereich Medizin der Johann Wolfgang Goethe Universität ein Fragebogen entworfen, um nichtschulische Leistungen zu erfassen. Dieser Fragebogen wurde am Beginn des Wintersemesters 2005/2006 von allen Studienanfängern der JWG-Universität Frankfurt und der Medizinischen Universität Innsbruck ausgefüllt. Entgegen der initialen Erwartungen der Verfasser gaben nur etwa 15% Prozent Medizin-spezifische berufliche Vorerfahrungen an (Rettungsdienst, Ausbildung als Krankenschwester/pfleger oder ähnliches); dagegen wurden von etwa 60% angegeben, mindestens ein Musikinstrument zu spielen oder länger sportlich aktiv gewesen zu sein. Die Zusammenstellung der Selbstangaben zeigt, dass Medizin-relevante Vorkenntnisse nur bei einem kleinen Anteil der Studienbewerber in größerem Umfang vorhanden sind. Aufgrund der großen Streuung in der Art und Dauer der angegebenen Vorleistungen sollte die Erhebung von Parametern zur Beurteilung von soft skills, z.B. durch Online-Fragebogen, als (Vor)Selektionsinstrument nur sehr vorsichtig eingesetzt werden.Due to a reorganization in the admission procedure for German medical students in 2005 Medical Faculties may admit up to 60% of their students according to specific university criteria. Both legislation and public opinion implicitly require nonacademic criteria and personality traits to be included into the selection criteria (motivation, identification, avoidance of misconceptions). Since interviews appear to be not suitable for the high number of applicants the Medical Faculty of the Johann Wolfgang Goethe University in Frankfurt/Main has developed a questionnaire covering extracurricular achievements. This questionnaire has be answered by first year students of the JWG university Frankfurt and the Medical University Innsbruck at the begin of the winter term 2005/2006. Contrary to initial expectations, only 15% of the students stated specific job experience in a medically related field (emergency service, nurse or similar occupations), whereas more than 60% indicated to play at least one instrument and/or being engaged in sports activities. The summary of these self experiences indicate that only a minority of admitted students have medically related experience to a larger degree. The wide variation in time and duration given for the activities indicate that (online) questionnaires should be treated carefully as a preselection method for medical studies admission
GLRB allelic variation associated with agoraphobic cognitions, increased startle response and fear network activation : a potential neurogenetic pathway to panic disorder
The molecular genetics of panic disorder (PD) with and without agoraphobia (AG) are still largely unknown and progress is hampered by small sample sizes. We therefore performed a genome-wide association study with a dimensional, PD/AG - related anxiety phenotype based on the Agoraphobia Cognition Questionnaire (ACQ) in a sample of 1,370 healthy German volunteers of the CRC TRR58 MEGA study wave 1. A genome-wide significant association was found between ACQ and single non-coding nucleotide variants of the GLRB gene (rs78726293, p=3.3x10-8; rs191260602, p=3.9x10-8). We followed up on this finding in a larger dimensional ACQ sample (N=2,547) and in independent samples with a dichotomous AG phenotype based on the Symptoms Checklist (SCL-90; N=3,845) and a case control sample with the categorical phenotype PD/AG (Ncombined =1,012) obtaining highly significant p-values also for GLRB single nucleotide variants rs17035816 (p=3.8x10-4) and rs7688285 (p=7.6x10-5). GLRB gene expression was found to be modulated by rs7688285 in brain tissue as well as cell culture. Analyses of intermediate PD/AG phenotypes demonstrated increased startle reflex and increased fear network as well as general sensory activation by GLRB risk gene variants rs78726293, rs191260602, rs17035816 and rs7688285. Partial Glrb knockout-mice demonstrated an agoraphobic phenotype. In conjunction withthe clinical observation that rare coding GLRB gene mutations are associated with the neurological disorder hyperekplexia characterized by a generalized startle reaction and agoraphobic behavior, our data provide evidence that non-coding, though functional GLRB gene polymorphisms may predispose to PD by increasing startle response and agoraphobic cognitions.PostprintPeer reviewe
Linking the Epigenome to the Genome: Correlation of Different Features to DNA Methylation of CpG Islands
DNA methylation of CpG islands plays a crucial role in the regulation of gene expression. More than half of all human promoters contain CpG islands with a tissue-specific methylation pattern in differentiated cells. Still today, the whole process of how DNA methyltransferases determine which region should be methylated is not completely revealed. There are many hypotheses of which genomic features are correlated to the epigenome that have not yet been evaluated. Furthermore, many explorative approaches of measuring DNA methylation are limited to a subset of the genome and thus, cannot be employed, e.g., for genome-wide biomarker prediction methods. In this study, we evaluated the correlation of genetic, epigenetic and hypothesis-driven features to DNA methylation of CpG islands. To this end, various binary classifiers were trained and evaluated by cross-validation on a dataset comprising DNA methylation data for 190 CpG islands in HEPG2, HEK293, fibroblasts and leukocytes. We achieved an accuracy of up to 91% with an MCC of 0.8 using ten-fold cross-validation and ten repetitions. With these models, we extended the existing dataset to the whole genome and thus, predicted the methylation landscape for the given cell types. The method used for these predictions is also validated on another external whole-genome dataset. Our results reveal features correlated to DNA methylation and confirm or disprove various hypotheses of DNA methylation related features. This study confirms correlations between DNA methylation and histone modifications, DNA structure, DNA sequence, genomic attributes and CpG island properties. Furthermore, the method has been validated on a genome-wide dataset from the ENCODE consortium. 2: Geomagnetische Erkundung von Trockenmaaren im Vulkanfeld der Westeifel
Im Vulkanfeld der Westeifel wurden 11 weitere Maare geomagnetisch vermessen.
Hochwertige magnetische Störfelder deuten auf zusedimentierte oder wieder freierodierte
Schlackenkegel in den ehemaligen Kraterböden der Maare hin (finale Schlackenförderung).
Wenn keine Schlackenkegel oder keine Intrusionen in den Maardiafremen
auftrefen, dann weisen junge, mit mächtigen Kratersedimenten aufgefüllte Maare niedrige
Störfelder mit schwachen Randgradienten auf. Etwas ältere Maare, bei denen ein Großteil
der gering magnetisierten Kratersedimente bereits erodiert ist, zeigen höhere magnetische
Anomalien mit hohen Randgradienfen und deutlichen Randminima.Abstract: 11 maars of the Westeifel volcanic field were surveyed geomagnetically. High
magnetic anomalies indicate the existence of scoria cones covered by Sediment or already
revealed by erosion, within the former crater bottoms of the maars (final scoria production).
If scoria cones and intrusions are absent from the maar diatremes then young maars with a
thick pile of crater Sediments show small anomalies with low border gradients. Older maars
are characterized by high border gradients and significant border minima, because most of

1: Erkundung neuer Maare im Vulkanfeld der Eifel mit Hilfe geomagnetischer Untersuchungen
Mit Hilfe geomagnetischer Messungen in Kesseltälern im Vulkanfeld der
Westeifel konnten 15 bisher weitgehend unbekannte Maare nachgewiesen werden. Der Vergleich
mit magnetischen Anomalien von acht jungen Maaren (mit Kraterseen) aus dem
Vulkanfeld der Westeifel sowie mit der Anomalie des 1977 entstandenen Ukinrek-Ost-
Maares (Alaska) ergab, daß hochwertige magnetische Störfelder auf zusedimentierte
Schlackenkegel in den ehemaligen Kraterböden bzw. auf Randintrusionen im Schlot hinweisen.
Danach sind fast die Hälfte der untersuchten Maare nicht rein phreatomagmatisch
entstanden, sondern weisen während den Eruptionen oder am Ende der Eruptionstätigkeit
„normal" entgasenden Magmenaufstieg auf.Abstract: In the volcanic area of the Westeifel the results of geomagnetic surveys within
local crater-shaped enlargements of valleys prove the existence of 15 maars which have
been unknown so far. Taking into account the magnetic anomalies of eight young Westeifelian
maars (with crater lakes) and the anomaly of Eastern Ukinrek Maar, which was
formed in 1977, it can be inferred that strong magnetic anomalies are associated with sediment-
covered scoria cones on the former crater bottom and/or with marginal intrusions
within the diatremes. This leads to the conclusion that almost half of the investigated maars
do not have a solely phreatomagmatic origin, but exhibit normally' degassing rising magma

In der vulkanischen Westeifel wurden seit 1823 kraterförmige Erweiterungen
einiger Täler, sogenannte Kesseltäler, als vermutlich den Maaren nahestehend beschrieben;
ihre Genese blieb jedoch unklar. Die geomagnetische Vermessung, einige
Bohrungen und neue, bisher unbekannte Aufschlüsse weisen im Untergrund von 4 der 5
bearbeiteten Kesseltälern Pyroklastite nach. Diese Kesseltäler sind eindeutig als Maare
anzusehen, deren ursprünglich vorhandene, ausgeworfene Pyroklastite bereits überwiegend
erodiert wurden. Folglich werden die meisten Kesseltäler als alte Maare interpretiert.Abstract: In the volcanic field of the Western Eifel, Germany, local crater-shaped
enlargements of valleys, so-called “Kesseltäler", have been known since 1823. A genetic
relationship with the maar volcanoes was always assumed but owing to the lack of ejecta

Hochauflösende fluviolakustrine Sedimente des jüngeren Pleistozän aus dem Alfbachtal bei Gillenfeld (Westeifel) – erste Ergebnisse
Im Alfbachtal im quartären Vulkanfeld der Westeifel führte die Eruption
mehrerer Schlackenkegel während der Weichsel-Eiszeit zur Bildung einer Barriere,
vor der sich eine bis zu 51 m mächtige Abfolge aus Lavaströmen und Sedimenten bildete.
In einer Bohrung bei Gillenfeld besteht die Talfüllung über einer basalen 70 cm
mächtigen mafischen Tephra aus einer unteren Abfolge etwa 22 m mächtiger pelitischer
feingeschichteter Seesedimente und oberhalb einer Erosionsdiskordanz aus einer oberen
Abfolge etwa 10 m mächtiger fluviolakustriner Grobklastika, Pelite und Mudden.
Nach Radiokohlenstoff-Datierungen, Korrelation von Tephren und faziellen Korrelationen
mit Sedimenten der Eifelmaare wurden die oberen fluviolakustrinen Sedimente
zwischen etwa 14 600 und 11 800 BP und die unteren lakustrinen Pelite zwischen etwa
31 000 und 25 600 BP abgelagert. In dem Zeitraum von etwa 27500 bis 25700 BP eruptierten
mindestens 13 Maare im Südost-Teil in der Westeifel. Mafische Maartephren in
den Sedimenten des ehemaligen Alf-Stausees werden beschrieben und für einige dieser
Tephren werden mögliche Eruptionspunkte diskutiert.Abstract: In the Alf valley in the Quaternary Westeifel Volcanic Field the eruption of
several scoria cones during the Weichsel Glacial resulted in the formation of a natural
dam and an up to 51 m thick sequence of lava flows and sediments was formed
upstream. In a drilling near Gillenfeld the valley fill consists of a basal 70 cm thick
mafic tephra, a lower sequence of c. 22 m thick pelitic finely bedded lacustrine sediments
and resting upon an erosional discordance an upper sequence of c. 10 m thick fluviolacustrine
coarse clastics, pelites and organic lake sediments. According to radiocarbon
dating, tephra chronology and facies correlation with sediments of Eifel maars the upper
fluviolacustrine sediments were deposited between c. 14 600 and 11 800 BP and the lower
lacustrine pelites between c. 31 000 and 25 600 BP. In the time interval from c. 27500
to 25700 BP at least 13 maars erupted in the SE-part of the Westeifel. Mafic maar tephra
in the sediments of the former Alf Lake are described and for some of these tephra

Geochemistry of natural wetlands in former uranium milling sites (eastern Germany) and implications for uranium retention
Discharge from former uranium mining and milling areas is world wide a source of elevated uranium contents in wetlands. The efficiency of organic rich wetland environments for entrapment and accumulation of uranium was assessed in this work using hydrogeochemical field studies of natural small-sized wetlands in Thuringia and Saxony, Eastern Germany. The objective was to estimate, if artificial wetlands can be used in a similar way: as a sustainable 'passive' treatment methodology. World wide, as an experiment a first dozen of such systems is implemented for uranium bearing mine waters, primarily aiming at uranium reduction and precipitation.
Pore water and solid phase samples were collected from the upper decimetres of substrate profiles in minerotrophic volunteer wetlands and natural fen-type wetlands which facilitate uranium accumulation. Elemental analyses, correlation techniques and sequential chemical extraction were applied to evaluate retention mechanisms (e.g., reduction, mineral sorption). No dominant process was pronounced, but the bulk of uranium is retained in moderately labile forms, predominantly as operationally defined organically bound or acid soluble (‘specifically adsorbed’) phases. Macrophyte intracellular uranium accumulation (‘phytoaccumulation’) is not responsible for the high uranium concentrations in the wetland substrates. Although there is no evidence for stable U(IV) mineralisation via ‘reductive precipitation’, high accumulation efficiency of the wetlands is obvious, resulting from processes regarded as more labile.
