    Effect of connective tissue growth factor (CCN2/CTGF) on proliferation and differentiation of mouse periodontal ligament-derived cells

    Background: CCN2/CTGF is known to be involved in tooth germ development and periodontal tissue remodeling, as well as in mesenchymal tissue development and regeneration. In this present study, we investigated the roles of CCN2/CTGF in the proliferation and differentiation of periodontal ligament cells (murine periodontal ligament-derived cell line: MPL) in vitro. Results: In cell cultures of MPL, the mRNA expression of the CCN2/CTGF gene was stronger in sparse cultures than in confluent ones and was significantly enhanced by TGF-β. The addition of Recombinant CCN2/CTGF (rCCN2) to MPL cultures stimulated DNA synthesis and cell growth in a dose-dependent manner. Moreover, rCCN2 addition also enhanced the mRNA expression of alkaline phosphatase (ALPase), type I collagen, and periostin, the latter of which is considered to be a specific marker of the periosteum and periodontium; whereas it showed little effect on the mRNA expression of typical osteoblastic markers, e.g., osteopontin and osteocalcin. Finally, rCCN2/CTGF also stimulated ALPase activity and collagen synthesis. Conclusion: These results taken together suggest important roles of CCN2/CTGF in the development and regeneration of periodontal tissue including the periodontal ligament.</p

We introduce the incremental and cooperative version of the problem, IC-PHNDP for short, in which facilities are added incrementally to the network (one at a time), contributing to the service levels. We first develop a general non-linear model of this problem and then present a method to make it linear. As the problem is of a combinatorial nature, an efficient Variable Neighbourhood Search (VNS) algorithm is proposed to solve it. In order to gain insight into the problem, the computational studies were performed with randomly generated instances of different settings. Results clearly show that VNS performs well in solving IC-PHNDP with errors not more than 1.54%.Peer reviewe

    Histological and quantitative polymerase chain reaction-based analysis of Buruli ulcer using mapping biopsy method.

    Background:In Japan, Buruli ulcer cases are often advanced, requiring surgical treatment. However, extensive debridement is often difficult because of cosmetic and functional sequelae. Moreover, the lesions are complicated and composed of edematous erythema, necrotic ulcer, and erythematous skin lesions caused by a paradoxical reaction, which also make it difficult to perform adequate debridement.Methodology/principal findings:We performed quantitative polymerase chain reaction (PCR) analysis for IS2404 using 29 samples taken from mapping biopsy. We evaluated the relationship among mycobacterial burden, histopathological findings, and clinical outcomes using 83 tissue samples taken from mapping biopsy and debrided Buruli ulcer. On quantitative PCR, the Cp values of IS2404 amplification were substantially different in each site. The major histological findings could be divided into massive subcutaneous necrosis with scant inflammatory cell infiltration and dense inflammatory cell infiltration. Of the 84 sites, 34 were subjected to repeated histological evaluations. In these sites, histological necrosis did not disappear over time despite standard antibiotic treatment. In contrast, the ulcers were cured and no recurrences were observed without resecting the 11 biopsied sites that lacked histological necrosis. Although quantitative PCR revealed that a lower Cp value of IS2404 was associated with histological massive necrosis, sites that showed lower Cp values clinically did not always need debridement.Conclusion/significance:Our descriptive study revealed that the histological findings and amounts of mycobacterial DNA differed according to the sites despite being found in one lesion. Our results showed that the need for surgical debridement in each site was correlated with histological necrosis without inflammatory cell infiltration, as the inflammation is supposed to represent an active host immune response rather than mycobacterial burden. We suggest that the debridement of lesions with histological necrosis in mapping biopsy may be useful for Japanese cases with unsuccessful standard antibiotic treatment to achieve sufficient clinical improvement

    Microscopic approach to current-driven domain wall dynamics

    This review describes in detail the essential techniques used in microscopic theories on spintronics. We have investigated the domain wall dynamics induced by electric current based on the ss-dd exchange model. The domain wall is treated as rigid and planar and is described by two collective coordinates: the position and angle of wall magnetization. The effect of conduction electrons on the domain wall dynamics is calculated in the case of slowly varying spin structure (close to the adiabatic limit) by use of a gauge transformation. The spin-transfer torque and force on the wall are expressed by Feynman diagrams and calculated systematically using non-equilibrium Green's functions, treating electrons fully quantum mechanically. The wall dynamics is discussed based on two coupled equations of motion derived for two collective coordinates. The force is related to electron transport properties, resistivity, and the Hall effect.     This study aimed to obtain useful suggestions for coaching, by clarifying the characteristics of changing the direction of A quick, by motion analysis. On applying motion analysis to a total of 6 middle blockers in university, or men`s V・ Challenge League specializing in quick attack, it was found that there was a difference in changing direction of A quick. This study obtained useful suggestions for coaching due to the above. To change the direction of A quick, there are three important methods: 1. changing direction by movement of the trunk rather than the shoulder joint, and shifting the impact point by half on the ball indirection in which the player wants to hit. 2. In case the player wants to hit the ball to cross, twisting the player’s body largely backward from takeoff to taking back and shifting the impact point to the right side, rather than hitting by turning, and stooping the trunk to transmit force to the ball for impact. 3. In case the player wants to hit the ball by turning, he does not twist his body backward from takeoff to taking back and shifting the impact point to the left side, rather than hitting to cross, without stooping the trunk in order to obtain space for swinging, and bending the trunk to the left, in order to transmit force to the ball

    This study was conducted to gain a compositive and holistic understanding of the blocking tricks employed by elite center position volleyball players to "discontinue the excessive use of \u27hands up\u27 blocking and employ arm swing jumps." In pursuit of this, we employed a sequential exploratory strategy in addition to using observational evaluation criteria to examine how players "discontinue the excessive use of \u27hands up\u27 blocking and employ arm swing jumps" from both quantitative and qualitative aspects. The results were as follows.- A qualitative (sensory) trick to "discontinue the excessive use of \u27hands up\u27 blocking and employ arm swing jumps" involved blocking by holding the hands up around the shoulders and face, while a quantitative (actual movement) trick also mostly involved blocking by holding the hands up around the shoulders and face. This supported the findings obtained using a qualitative examination method.- When Mr. Tomimatsu performed \u27hands down\u27 blocking, results suggested a blocking sensation that was difficult to extract quantitatively.- Tricks to "discontinue the excessive use of \u27hands up\u27 blocking and employ arm swing jumps" were found to manifest as different movements in actual volleyball matches based on an examination of two research participants

    Complications Associated With Spine Surgery in Patients Aged 80 Years or Older: Japan Association of Spine Surgeons with Ambition (JASA) Multicenter Study

    Study Design:Retrospective study of registry data.Objectives:Aging of society and recent advances in surgical techniques and general anesthesia have increased the demand for spinal surgery in elderly patients. Many complications have been described in elderly patients, but a multicenter study of perioperative complications in spinal surgery in patients aged 80 years or older has not been reported. Therefore, the goal of the study was to analyze complications associated with spine surgery in patients aged 80 years or older with cervical, thoracic, or lumbar lesions.Methods:A multicenter study was performed in patients aged 80 years or older who underwent 262 spinal surgeries at 35 facilities. The frequency and severity of complications were examined for perioperative complications, including intraoperative and postoperative complications, and for major postoperative complications that were potentially life threatening, required reoperation in the perioperative period, or left a permanent injury.Results:Perioperative complications occurred in 75 of the 262 surgeries (29%) and 33 were major complications (13%). In multivariate logistic regression, age over 85 years (hazard ratio [HR] = 1.007, P = 0.025) and estimated blood loss ≥500 g (HR = 3.076, P = .004) were significantly associated with perioperative complications, and an operative time ≥180 min (HR = 2.78, P = .007) was significantly associated with major complications.Conclusions:Elderly patients aged 80 years or older with comorbidities are at higher risk for complications. Increased surgical invasion, and particularly a long operative time, can cause serious complications that may be life threatening. Therefore, careful decisions are required with regard to the surgical indication and procedure in elderly patients
