    Consistency of pacing and metabolic responses during 2000-m rowing ergometry

    PURPOSE: This study investigated the pacing strategy adopted and the consistency of performance and related physiological parameters across three 2000-m rowing-ergometer tests. METHODS: Fourteen male well-trained rowers took part in the study. Each participant performed three 2000-m rowing-ergometer tests interspersed by 3-7 d. Throughout the trials, respiratory exchange and heart rate were recorded and power output and stroke rate were analyzed over each 500 m of the test. At the completion of the trial, assessments of blood lactate and rating of perceived exertion were measured. RESULTS: Ergometer performance was unchanged across the 3 trials; however, pacing strategy changed from trial 1, which featured a higher starting power output and more progressive decrease in power, to trials 2 and 3, which were characterized by a more conservative start and an end spurt with increased power output during the final 500 m. Mean typical error (TE; %) across the three 2000-m trials was 2.4%, and variability was low to moderate for all assessed physiological variables (TE range = 1.4-5.1%) with the exception of peak lactate (TE = 11.5%). CONCLUSIONS: Performance and physiological responses during 2000-m rowing ergometry were found to be consistent over 3 trials. The variations observed in pacing strategy between trial 1 and trials 2 and 3 suggest that a habituation trial is required before an intervention study and that participants move from a positive to a reverse-J-shaped strategy, which may partly explain conflicting reports in the pacing strategy exhibited during 2000-m rowing-ergometer trials

    A discourse analysis of trainee teacher identity in online discussion forums

    Teacher education involves an identity transformation for trainees from being a student to being a teacher. This discourse analysis examined the online discussion board communications of a cohort of trainee teachers to better understand the situated identities of the trainees and how they were presented online. Their discussion board posts were the primary method of communication during placement periods and, as such, provided insight into how the trainees situated their identities in terms of being a student or being a teacher. During the analysis, the community boundaries, language and culture were explored along with the tutor's power and role in the identity transformation process. This involved looking at the lexis used by the students, the use of pronouns to refer to themselves and others such as teachers and pupils, the types of messages allowed in the community and the effect of the tutor's messages on their communication. The research found that the trainees felt comfortable with teaching but did not feel like teachers during the course. Tutors and school teachers need to develop an awareness of the dual nature of trainees' identities and help promote the transition from student to teacher. In the beginning of the course, trainees should be familiarised with teacher vocabulary and practical concepts in addition to pedagogical theory. Towards the end of the course, trainee identity as teachers could be promoted through the use of authentic assessments that mirror real teacher tasks and requirements

    Reading Videogames as (authorless) Literature

    This article presents the outcomes of research, funded by the Arts and Humanities Research Council in England and informed by work in the fields of new literacy research, gaming studies and the socio-cultural framing of education, for which the videogame L.A. Noire (Rockstar Games, 2011) was studied within the orthodox framing of the English Literature curriculum at A Level (pre-University) and Undergraduate (degree level). There is a plethora of published research into the kinds of literacy practices evident in videogame play, virtual world engagement and related forms of digital reading and writing (Gee, 2003; Juul, 2005; Merchant, Gillen, Marsh and Davies, 2012; Apperley and Walsh, 2012; Bazalgette and Buckingham, 2012) as well as the implications of such for home / school learning (Dowdall, 2006; Jenkins, 2006; Potter, 2012) and for teachers’ own digital lives (Graham, 2012). Such studies have tended to focus on younger children and this research is also distinct from such work in the field in its exploration of the potential for certain kinds of videogame to be understood as 'digital transformations' of conventional ‘schooled’ literature. The outcomes of this project raise implications of such a conception for a further implementation of a ‘reframed’ literacy (Marsh, 2007) within the contemporary curriculum of a traditional and conservative ‘subject’. A mixed methods approach was adopted. Firstly, students contributing to a gamplay blog requiring them to discuss their in-game experience through the ‘language game’ of English Literature, culminating in answering a question constructed with the idioms of the subject’s set text ‘final examination’. Secondly, students taught their teachers to play L.A. Noire, with free choice over the context for this collaboration. Thirdly, participants returned to traditional roles in order to work through a set of study materials provided, designed to reproduce the conventions of the ‘study guide’ for literature education. Interviews were conducted after each phase and the outcomes informed a redrafting of the study materials which are now available online for teachers – this being the ‘practical’ outcome of the research (Berger and McDougall, 2012). In the act of inserting the study of L.A. Noire into the English Literature curriculum as currently framed, this research moves, through a practical ‘implementation’ beyond longstanding debates around narratology and ludology (Frasca, 2003; Juul, 2005) in the field of game studies (Leaning, 2012) through a direct connection to new literacy studies and raises epistemological questions about ‘subject identity’, informed by Bernstein (1996) and Bourdieu (1986) and the implications for digital transformations of texts for both ideas about cultural value in schooled literacy (Kendall and McDougall, 2011) and the politics of ‘expertise’ in pedagogic relations (Ranciere, 2009, Bennett, Kendall and McDougall, 2012a)

    Local political leadership and the modernisation of local government

    Political leadership has been a key element of central government’s attempts to ‘modernise’ local government over the past decade, within a discourse that emphasised ‘strong’ and ‘visible’ leadership and the role of leaders and leadership in driving change within local authorities. In the context of such an approach, and also taking account of academic discourse, this article draws upon interviews with nearly thirty individuals in leadership positions in local authorities in England, Scotland and Wales to assess their experiences of leadership and their views of some aspects of the role and work of councils. It suggests that whilst there is broad convergence between the aspirations of government and the narratives that emerge from these leaders on some aspects of local political leadership, there are also differences, perhaps most notably over the relationship between changes to decision making structures and the loci of political power

    Contested perspectives on the social impacts of a residential fieldtrip

    Background: Research into outdoor learning reveals social benefits for trip participants, both individually and collectively. However, this is not universal and individual participants can experience increased isolation from the wider group. Purpose: This research investigated the underexplored negative experiences of an individual trip participant, in the context of a program promoting collectivist beliefs. Methodology/Approach: An ethnographic methodology was adopted, to include full immersion into all aspects of the week-long trip. Data were collected by observation and interviews and analyzed using a conceptual framework around individualism and collectivism. Findings/Conclusions: The teachers perceived the trip as successful, in meeting their collectivist goal of enhancing group cohesion, and data revealed the building of community feelings amongst the majority of participants, alongside feelings of belonging, togetherness and mutual support. However, one participant exhibited contrasting individualist perceptions and experiences around interpretations of freedom, privacy, adversity and cohesion. Implications: Trip leaders need to be mindful of participants as individuals, taking care not to seek nor project a blanket group identity over all. Within the confines of health and safety and duty of care boundaries, and commensurate with the age range of trip participants, accompanying teachers should understand and respect individual needs within collectivist group socialization agendas

    Experimental Testing and modeling of a Dual-Fired LiBr-H2O Absorption Chiller

    An LiBr-H2O chiller was modified to utilize heat sources from natural gas combustion and/or from hot fluid. This was achieved by replacing the original gas-fired generator with a dual-fired generator. Steam was used as the hot fluid. The generator of the chiller can be powered by each source separately or both sources simultaneously. Experimental investigation was performed to obtain capacity and coefficient of performance (COP) of the original chiller and the modified chiller. During the experiments, the modified chiller was powered solely by steam, natural gas, or both. There was a significant increase in COP and capacity when steam was used as a heat source. The tests using natural gas resulted in performance similar to the original chiller. The experimental conditions were closely modeled by a numerical program

    Species conservation profile and amended distribution of Cousinia knorringiae (Asteraceae), a narrow endemic of the Western Tian-Shan

    Background Cousinia knorringiae Bornm. (Asteraceae) belongs to C. sect. Subappendiculatae Tscherneva, a group of the species-rich and taxonomically difficult genus Cousinia Cass. This species is narrowly distributed in the Western Tian-Shan and has been known as endemic to Kyrgyzstan. It inhabits bare rocks and screes at elevations of 1200-1500 m above sea level. This species is of conservation interest because of its small population size and limited distribution. New information Cousinia knorringiae is reported for the first time from eastern Uzbekistan on the basis of specimens collected on Ungur-Tepa Mt., a south-western outlier of the Bozbu-Too Mts. (Western Tian-Shan). The conservation status of the species is assessed as Endangered (EN), based on criterion D (estimated population size 200-250 mature individuals), according to the IUCN Red List Categories and Criteria (version 3.1). A new distribution map and a line drawing for C. knorringiae are provided and its type locality is clarified. The new knowledge suggests that the species is endemic to the East Fergana botanical hotspot, which includes a transborder territory shared between Kyrgyzstan and Uzbekistan and should, therefore, be subjected to international conservation measures. The southern extension of Ungur-Tepa Mt. harbours important plant species, which cannot be found elsewhere in Uzbekistan and may, therefore, be proposed for legal protection.Peer reviewe

    Zinc transporter HKE4 as a new target in antihormone resistance of breast cancer

    Background Oestrogen receptor-positive breast cancers develop resistance to anti-oestrogens by utilising alternative growth factor pathways as observed in our tamoxifen-resistant cell line (TAMR). These include EGFR, IGF1-R and Src signalling as well as increased growth and invasion. Zinc is elevated in breast cancer tissue and has been demonstrated to activate certain growth factor signalling pathways. We have tested the expression level of members of the LIV-1 family of zinc influx transporters and discovered that HKE4 (SLC39A7, ZIP7), previously shown by us capable of increasing the intracellular zinc levels, has increased expression in TAMR. We have therefore investigated whether the development of the more aggressive phenotype observed in our TAMR cells, including activation of these signalling pathways as well as increased growth and invasion, is due to an increase of intracellular zinc and as a direct result of increased expression of HKE4. Methods All nine members of the LIV-1 subfamily of ZIP transporters were measured in our model of tamoxifen-resistant breast cancer using Affymetrix arrays. Zinc-induced activation of growth factor signalling pathway components was investigated by western blot and/or fluorescent microscopy. Short-term (15-min) treatments with 20 μM zinc included ionophore, whereas long-term (hours/days) did not. Recombinant LIV-1 family members with a V5 tag were expressed using pcDNA3.1/V5-His-TOPO vector, and siRNA (Dharmacon smartpools with relevant controls) was used to reduce endogenous expression. Results HKE4 (SLC39A7), a ZIP transporter from the LIV-1 subfamily, was discovered to be elevated in TAMR cells by Affymetrix analysis and confirmed by PCR and western blot. We have observed that our TAMR cells have a twofold increase in intracellular zinc compared with wild-type cells, using the zinc-specific fluorescent dye Newport Green. Short-term zinc treatment of TAMR cells activates the signalling pathways implicated in antihormone-resistant proliferation and is reduced by both the zinc chelator TPEN and the Src kinase inhibitor SU6556. The same effects are observed after longer term (6 days) zinc treatment with additional increases in cell growth and invasion through Matrigel. Since we have previously demonstrated that HKE4 is capable of increasing intracellular zinc in cells and, more recently, that these TAMR have elevated intracellular zinc levels, we have tested the hypothesis that elevated HKE4 expression is directly responsible for the aggressive phenotype observed in our TAMR cells. Reducing HKE4 levels by siRNA demonstrated a role for this molecule in driving the zinc-induced activation of multiple signalling pathways. In the presence of siRNA for HKE4, the previously observed zinc-induced activation of EGFR, Src, and IGF1-R was eradicated and the EGF-stimulated activation was also decreased. Additionally, we have demonstrated the converse by transfecting recombinant HKE4 into wild-type cells and/or treating them with zinc to observe the activation of these signalling pathways and increases in invasive capability. Interestingly, we have observed a similar role of HKE4 in our model of faslodex-resistant breast cancer. Conclusion The presented results propose that HKE4, a member of the LIV-1 subfamily of ZIP transporters, is directly involved in the activation of the aggressive phenotype observed with the development of antihormone resistance, and as such is a potential new target for the prevention of resistance to antihormones in breast cancer progression
