    Multisite Evaluation and Validation of a Sensitive Diagnostic and Screening System for Spinal Muscular Atrophy that Reports SMN1 and SMN2 Copy Number, along with Disease Modifier and Gene Duplication Variants

    Spinal muscular atrophy is a severe autosomal recessive disease caused by disruptions in the SMN1 gene. The nearly identical SMN2 gene copy number is associated with disease severity. SMN1 duplication markers, such as c.*3+80T>G and c.*211_*212del, can assess residual carrier risk. An SMN2 disease modifier (c.859G>C) can help inform prognostic outcomes. The emergence of multiple precision gene therapies for spinal muscular atrophy requires accurate and rapid detection of SMN1 and SMN2 copy numbers to enable early treatment and optimal patient outcomes. We developed and evaluated a singletube PCR/capillary electrophoresis assay system that quantifies SMN1/2 copy numbers and genotypes three additional clinically relevant variants. Analytical validation was performed with human cell lines and whole blood representing varying SMN1/2 copies on four capillary electrophoresis instrument models. In addition, four independent laboratories used the assay to test 468 residual clinical genomic DNA samples. The results were >98.3% concordant with consensus SMN1/2 exon 7 copy numbers, determined using multiplex ligation-dependent probe amplification and droplet digital PCR, and were 100% concordant with Sanger sequencing for the three variants.     Mental state language and quality of conversational experience in deaf and hearing children

    Deaf children from hearing parents show a protracted delay in their performance on standard ‘theory of mind’ measures that concern their knowledge of false beliefs and other reality incongruent mental states. Considerable evidence indicates that children’s early experience of adults’ mental state talk predicts their later social cognitive development. However, no previous study has analyzed access to conversation about mental states in very young deaf children. We compared the conversational input of hearing parents to young deaf and hearing children aged 17 to 35 months in the UK and Sweden. Parents of hearing children used far more cognitive mental state language with their infants and their conversations were characterized by more communicatively effective turn-taking than parents of deaf children. These findings indicate that conversational input about mental states to very young deaf children differs significantly in those areas of interaction thought to be crucial for later social cognitive development and this difference is robust across two different cultures

    Observing the Evolution of the Universe

    How did the universe evolve? The fine angular scale (l>1000) temperature and polarization anisotropies in the CMB are a Rosetta stone for understanding the evolution of the universe. Through detailed measurements one may address everything from the physics of the birth of the universe to the history of star formation and the process by which galaxies formed. One may in addition track the evolution of the dark energy and discover the net neutrino mass. We are at the dawn of a new era in which hundreds of square degrees of sky can be mapped with arcminute resolution and sensitivities measured in microKelvin. Acquiring these data requires the use of special purpose telescopes such as the Atacama Cosmology Telescope (ACT), located in Chile, and the South Pole Telescope (SPT). These new telescopes are outfitted with a new generation of custom mm-wave kilo-pixel arrays.     Effects of antiplatelet therapy on stroke risk by brain imaging features of intracerebral haemorrhage and cerebral small vessel diseases: subgroup analyses of the RESTART randomised, open-label trial

    Background Findings from the RESTART trial suggest that starting antiplatelet therapy might reduce the risk of recurrent symptomatic intracerebral haemorrhage compared with avoiding antiplatelet therapy. Brain imaging features of intracerebral haemorrhage and cerebral small vessel diseases (such as cerebral microbleeds) are associated with greater risks of recurrent intracerebral haemorrhage. We did subgroup analyses of the RESTART trial to explore whether these brain imaging features modify the effects of antiplatelet therapy

    The need for focused, hard X-ray investigations of the Sun

    Understanding the nature of energetic particles in the solar atmosphere is one of the most important outstanding problems in heliophysics. Flare-accelerated particles compose a huge fraction of the flare energy budget; they have large influences on how events develop; they are an important source of high-energy particles found in the heliosphere; and they are the single most important corollary to other areas of high-energy astrophysics. Despite the importance of this area of study, this topic has in the past decade received only a small fraction of the resources necessary for a full investigation. For example, NASA has selected no new Explorer-class instrument in the past two decades that is capable of examining this topic. The advances that are currently being made in understanding flare-accelerated electrons are largely undertaken with data from EOVSA (NSF), STIX (ESA), and NuSTAR (NASA Astrophysics). This is despite the inclusion in the previous Heliophysics decadal survey of the FOXSI concept as part of the SEE2020 mission, and also despite NASA's having invested heavily in readying the technology for such an instrument via four flights of the FOXSI sounding rocket experiment. Due to that investment, the instrumentation stands ready to implement a hard X-ray mission to investigate flare-accelerated electrons.
