    Growth and yield responses to amending the sugarcane monoculture: interactions between break history and nitrogen fertiliser

    Experiments were established in the Burdekin Irrigation Area in North Queensland, Australia, to measure whether yield improvements from breaking the sugarcane monoculture or fumigating the soil could be modified by the application of different rates of nitrogen (N) fertiliser. Experiments were conducted in consecutive crop cycles (phase 1, planted 1998; phase 2, planted 2001) using the variety Q117, with the interaction between N applications and rotation histories discussed for the two plant crops. Histories consisted of alternate crop, bare fallow or mixed grass-legume pastures for periods of 42-66 months, compared with continuous cane as plough-out replant without (PORP) or with (PORP-F) soil fumigation. The N strategies involved combinations of N rates (0-180kgN/ha) and application times (at planting, 90 days after planting (DAP) or split between these times) in phase 1 and N rates (0-300kgN/ha) in phase 2. Histories had differing effects on N available to the cane crop and hence on response to N fertiliser. Some combinations of history and N rate were N-limited and strong linear relationships between biomass production or cane yield and crop N content could be developed. Critical N contents for biomass production (R2≤0.93) and fresh-weight cane yield (R2≤0.88) were 1.42 and 0.57kgN/t, respectively. Application of N fertiliser was shown to have significant impacts on both tiller addition and the retention of tillers to produce harvestable stalks. However, the application of fertiliser N had limited (phase 1) or no (phase 2) capacity to provide the quantum of yield response in soil health benefits associated with breaking the sugarcane monoculture. Increasing N application rates above that required to optimise crop yield resulted in significant decreases in sugar content of cane and thus lower sugar yields. Yield increases solely from improved soil health (i.e. exclusive of N response) constituted advantages averaging 15% (phase 1) to 20% (phase 2) compared with PORP. These effects were manifest early in the establishment of primary shoots in the plant crops, although the longevity of these benefits was limited. Replanting cane after a 3-year crop cycle (plant, 1st and 2nd ratoon) on land that had been under pasture, crop, bare fallow or PORP-F histories (phase 2, cycle 2) showed carryover effects of histories on N availability and fertiliser N responsiveness, but limited yield impacts attributable to residual soil health benefits. These results reinforce the importance of crop rotation during breaks between sugarcane cycles to maintain soil health and improve crop productivity

    Antigen depot is not required for alum adjuvanticity

    Alum adjuvants have been in continuous clinical use for more than 80 yr. While the prevailing theory has been that depot formation and the associated slow release of antigen and/or inflammation are responsible for alum enhancement of antigen presentation and subsequent T- and B-cell responses, this has never been formally proven. To examine antigen persistence, we used the chimeric fluorescent protein EαGFP, which allows assessment of antigen presentation in situ, using the Y-Ae antibody. We demonstrate that alum and/or CpG adjuvants induced similar uptake of antigen, and in all cases, GFP signal did not persist beyond 24 h in draining lymph node antigen-presenting cells. Antigen presentation was first detectable on B cells within 6–12 h of antigen administration, followed by conventional dendritic cells (DCs) at 12–24 h, then finally plasmacytoid DCs at 48 h or later. Again, alum and/or CpG adjuvants did not have an effect on the magnitude or sequence of this response; furthermore, they induced similar antigen-specific T-cell activation in vivo. Notably, removal of the injection site and associated alum depot, as early as 2 h after administration, had no appreciable effect on antigen-specific T- and B-cell responses. This study clearly rules out a role for depot formation in alum adjuvant activity

    The development and evaluation of mini-GEMs: a short, focused, online e-learning videos in geriatric medicine

    Mini Geriatric E-Learning Modules (Mini-GEMs) are short, focused, e-learning videos on geriatric medicine topics, hosted on YouTube, which are targeted at junior doctors working with older people. This study aimed to explore how these resources are accessed and used. The authors analyzed the viewing data from 22 videos published over the first 18 months of the Mini-GEM project. We conducted a focus group of U.K. junior doctors considering their experiences with Mini-GEMS. The Mini-GEMs were viewed 10,291 times over 18 months, equating to 38,435 minutes of total viewing time. The average viewing time for each video was 3.85 minutes. Learners valued the brevity and focused nature of the Mini-GEMs and reported that they watched them in a variety of settings to supplement clinical experiences and consolidate learning. Watching the videos led to an increase in self-reported confidence in managing older patients. Mini-GEMs can effectively disseminate clinical teaching material to a wide audience. The videos are valued by junior doctors due to their accessibility and ease of use

    Cochrane Qualitative and Implementation Methods Group guidance paper 6:Methods for question formulation, searching, and protocol development for qualitative evidence synthesis

    This paper updates previous Cochrane guidance on question formulation, searching, and protocol development, reflecting recent developments in methods for conducting qualitative evidence syntheses to inform Cochrane intervention reviews. Examples are used to illustrate how decisions about boundaries for a review are formed via an iterative process of constructing lines of inquiry and mapping the available information to ascertain whether evidence exists to answer questions related to effectiveness, implementation, feasibility, appropriateness, economic evidence, and equity. The process of question formulation allows reviewers to situate the topic in relation to how it informs and explains effectiveness, using the criterion of meaningfulness, appropriateness, feasibility, and implementation. Questions related to complex questions and interventions can be structured by drawing on an increasingly wide range of question frameworks. Logic models and theoretical frameworks are useful tools for conceptually mapping the literature to illustrate the complexity of the phenomenon of interest. Furthermore, protocol development may require iterative question formulation and searching. Consequently, the final protocol may function as a guide rather than a prescriptive route map, particularly in qualitative reviews that ask more exploratory and open-ended questions

    Bursting out of our bubble: using creative techniques to communicate within the systematic review process and beyond

Available on open access from BMC via the DOI in this recordBACKGROUND: Increasing pressure to publicise research findings and generate impact, alongside an expectation from funding bodies to go beyond publication within academic journals, has generated interest in alternative methods of science communication. Our aim is to describe our experience of using a variety of creative communication tools, reflect on their use in different situations, enhance learning and generate discussion within the systematic review community. METHODS: Over the last 5 years, we have explored several creative communication tools within the systematic review process and beyond to extend dissemination beyond traditional academic mechanisms. Central to our approach is the co-production of a communication plan with potential evidence users which facilitates (i) the identification of key messages for different audiences, (ii) discussion of appropriate tools to communicate key messages and (iii) exploration of avenues to share them. We aim to involve evidence users in the production of a variety of outputs for each research project cognisant of the many ways in which individuals engage with information. RESULTS: Our experience has allowed us to develop an understanding of the benefits and challenges of a wide range of creative communication tools. For example, board games can be a fun way of learning, may flatten power hierarchies between researchers and research users and enable sharing of large amounts of complex information in a thought provoking way, but they are time and resource intensive both to produce and to engage with. Conversely, social media shareable content can be quick and easy to produce and to engage with but limited in the depth and complexity of shareable information. DISCUSSION: It is widely recognised that most stakeholders do not have time to invest in reading large, complex documents; creative communication tools can be a used to improve accessibility of key messages. Furthermore, our experience has highlighted a range of additional benefits of embedding these techniques within our project processes e.g. opening up two-way conversations with end-users of research to discuss the implications of findings.National Institute for Health Research (NIHR

    A comparison of sexual behaviour and attitudes of healthy adolescents in a Danish high school in 1982, 1996, and 2001

    AIM: To assess changes in sexual behaviour among students at a high school in Denmark from 1982 to 2001. METHODS: An anonymous self-administered questionnaire was used to compare data from three identical cross-sectional surveys performed in 1982, 1996, and 2001. RESULTS: Girls: More girls reported their first sexual intercourse before their 16th birthday in 2001 (42%) than in 1996 (29%) In 1982 it was also 42% (Chi-square for trend: p = 0.003). Fewer girls with no regular partner used condoms for their personal protection in 2001 (2%) than in 1996 (9%) and 1982 (0%) (Chi-square for trend p = 0.016). The proportion of girls with no regular partner who considered protection from sexually transmitted disease important for their choice of contraception was 39% in 2001 compared with 71% in 1996 and only 10% in 1982 (Chi-square for trend: p < 0.0001). Boys: More boys reported sexual debut before their 16th birthday in 2001 (40%) than in 1996 (37%) and 1982 (24%) (Chi-square for trend: p = 0.023). For boys with no regular partner, condom was preferred for personal protection by 85% in 2001, 91% in 1996 and 61% in 1982 (Chi-square for trend p = 0.007). Protection against sexually transmitted infection declined, especially among boys with no regular partner, from 51% in 2001 to 72% in 1996 and 21% in 1982 Chi-square for trend: p < 0.0001). The tendency towards earlier sexual debut and less use of safe sex practices to protect against sexually transmitted infections (STI) was accompanied by a rise in the number of detected STIs during this period. CONCLUSIONS: The period from 1982 to 1996 during which sexual attitudes were directed toward safer sex seems to have given way to a reverse trend in the period from 1996 to 2001. These findings may have significant implications for health care authorities organising preventive strategies for healthy adolescents
