    The Birth of Endowiki, An Italian Online Platform For Continuous Medical Education in Endocrinology

    Background Conventional tools for medical education are burdened by many drawbacks. Textbooks become rapidly outdated, meeting attendance is expensive, and results reported in journals are not easily suitable for clinical practice. Uptodate and Endotext are excellent online tools, but they have been developed for a clinical context far different from that of our country. Methodology Italian Association of Clinical Endocrinologists (AME) charged a task force aiming to develop an educational tool specifically tailored for Italian clinical endocrinologists. Required characteristics were clinical approach, modularity, continuous updating, full online availability (even by mobile devices), open sections and sections for registered members only, opportunity for individualization, indexing and search engine to facilitate browsing. Starting from an open-source platform, Joomla, several changes were implemented. Results Three editors, 2 central editorial secretaries, 30 section coordinators and over 350 authors have been involved in the writing of 21 different sections covering all fields of endocrine and metabolic diseases. The access to Endowiki is free for AME members and is fully open in sections for patients. All readers are requested to serve as referee, pointing to mistakes and need for revision. The system is attended daily by a mean of 250 individuals. Conclusions Endowiki stands alone as an opportunity for medical education in Italy. The big challenges will be the continuous updating and the link to the national certified system for CME

    A Multimodal Perception Framework for Users Emotional State Assessment in Social Robotics

    In this work, we present an unobtrusive and non-invasive perception framework based on the synergy between two main acquisition systems: the Touch-Me Pad, consisting of two electronic patches for physiological signal extraction and processing; and the Scene Analyzer, a visual-auditory perception system specifically designed for the detection of social and emotional cues. It will be explained how the information extracted by this specific kind of framework is particularly suitable for social robotics applications and how the system has been conceived in order to be used in human-robot interaction scenarios

    Gaia's Cepheids and RR Lyrae Stars and Luminosity Calibrations Based on Tycho-Gaia Astrometric Solution

    Gaia Data Release 1 contains parallaxes for more than 700 Galactic Cepheids and RR Lyrae stars, computed as part of the Tycho-Gaia Astrometric Solution (TGAS). We have used TGAS parallaxes, along with literature (V,I,J,Ks,W1V, I, J, {K_\mathrm{s}}, W_1) photometry and spectroscopy, to calibrate the zero point of the Period-Luminosity and Period-Wesenheit relations of classical and type II Cepheids, and the near-infrared Period-Luminosity, Period-Luminosity-Metallicity and optical Luminosity-Metallicity relations of RR Lyrae stars.     Dynamical Generation of Elementary Fermion Mass: First Lattice Evidence

    Using lattice simulations we demonstrate from first principles the existence of a non-perturbative mechanism for elementary particle mass generation in models with gauge fields, fermions and scalars, if an exact invariance forbids power divergent fermion masses and fermionic chiral symmetries broken at UV scale are maximally restored. We show that in the Nambu-Goldstone phase a fermion mass term, unrelated to the Yukawa operator, is dynamically generated. In models with electro-weak interactions weak boson masses are also generated opening new scenarios for beyond the Standard Model physics.Comment: 24 pages, 17 figure

    Confirmation of a New Metal-poor Globular Cluster in the Galactic Bulge

All rights reserved.We use deep near-IR photometry of the VISTA Variables in the V'a L'ctea (VVV) Survey and deep DECam Plane Survey (DECaPS) optical photometry to confirm the physical reality of the candidate globular cluster (GC) Minni 22, which is located in the Galactic bulge. This object, which was detected as a high density region in our maps of bulge red giants, is now confirmed as a real GC based on the optical and near-IR color'magnitude diagrams. We also recover three known fundamental mode (ab type) RR Lyrae stars within 2 arcmin of the cluster center. The presence of RR Lyrae stars also seems to confirm Minni 22 as a bonafide old and metal-poor GC. We estimate a cluster reddening E(J - Ks) = 0.6 mag and determine its heliocentric distance D = 7.4 ± 0.3 kpc. The optical and near-IR color'magnitude diagrams reveal well-defined red giant branches in all cases, including a red giant branch bump at Ks = 13.30 ± 0.05 mag. The comparison with theoretical isochrones yields a mean metallicity of [Fe/H] = -1.3 ± 0.3 dex, and age of t ∼ 11.2 Gyr. This is a good example of a new low-luminosity (MV = -6.2 mag) GC found in the central bulge of the Milky Way. After discussing the different ways to confirm the existence of bulge GC candidates, we find that one of the best methods is to use the CMDs from the combination of the DECaPS + VVV photometries.Peer reviewe

    Non-perturbative generation of elementary fermion masses: a numerical study

    In this talk we present a numerical lattice study of an SU(3) gauge model where an SU(2) doublet of non-Abelian strongly interacting fermions is coupled to a complex scalar field doublet via a Yukawa and a Wilson-like term. The model enjoys an exact symmetry, acting on all fields, which prevents UV power divergent fermion mass corrections, despite the presence of these two chiral breaking operators in the Lagrangian. In the phase where the scalar potential is non-degenerate and fermions are massless, the bare Yukawa coupling can be set at a critical value at which chiral fermion transformations become symmetries of the theory. Numerical simulations in the Nambu-Goldstone phase of the critical theory, for which the renormalized Yukawa coupling by construction vanishes, give evidence for non-perturbative generation of a UV finite fermion mass term in the effective action.Comment: 7 pages, 8 figures, Proceedings of The 36th Annual International Symposium on Lattice Field Theory, July 22-28, 2018, East Lansing, Michigan, US

    Dexmedetomidine-Tiletamine-Zolazepam Followed by Inhalant Anesthesia in Spectacled Bears (Tremarctos ornatus)

    Background: The spectacled bear (Tremarctos ornatus) is the only bear species inhabiting South America and is classified as vulnerable according to the International Union for Conservation of Nature (IUCN) Red List. Among the few publications on the use of general anesthesia and advanced monitoring of ursids in veterinary hospital settings, little is described regarding chemical restraint, general anesthesia and monitoring of spectacled bears. This case series describes the use of a dexmedetomidine-tiletamine-zolazepam chemical restraint combination and its effects on cardiorespiratory variables and arterial blood gases observed in two spectacled bears submitted to isoflurane anesthesia for imaging and/or surgical procedures.Cases: Two female, one adult and one senile, all-term captive spectacled bears were referred to the Veterinary Medical Teaching Hospital at the Universidade Estadual Paulista - Unesp, Botucatu campus, both with a presumable history of recent trauma. After immobilization with an intramuscular (IM) administration of tiletamine-zolazepam (3.8 - 4.3 mg/kg) and dexmedetomidine (6.4 - 7.6 µg/kg), anesthesia induction was achieved by means of intravenous (IV) propofol (1 - 2 mg/kg). The patients then underwent isoflurane inhalant anesthesia and were submitted to intermittent positive-pressure ventilation through the remainder of the procedures. Initial settings of inspiratory flow rate were adjusted to obtain Ppeak of 10 cmH2O and tidal volumes (Vt) of 10 mL/kg, as well as respiratory rates (ƒR)and inspiration-to-expiration (I:E) ratio of 10 breaths/min and 1:2, respectively, and were then adjusted throughout anesthesia to maintain normocapnia (end-tidal carbon dioxide concentrations between35 and 45 mmHg). One of the individuals was chemically restrained (6.4 mg/kg of tiletamine-zolazepam and 7.7 µg/kg of dexmedetomidine) on a second anesthetic event for imaging procedures. Arterial blood gas analysis were performed with the subjects breathing room air and oxygen-enriched air. Both animals exhibited severe hypoxemia (partial pressure of oxygen [PaO2] < 60 mmHg) while breathing room air (inspired oxygen fraction [FiO2] ≅ 0.21). An impaired blood oxygenation (PaO2/FiO2 < 400) was still observed despite mechanical ventilation and the provision of 1.0 FiO2. Alveolar recruitment maneuvers (3 sequential mechanical sights with peak airway pressure at 20 - 30 cmH2O during 15 - 30 s each) were then performed, which resulted in improved PaO2/FiO2 ratios. All other blood gas, electrolytes and acid-base variables did not appear to be importantly altered by chemical restraint and general anesthesia.Discussion: Dexmedetomidine-tiletamine-zolazepam resulted in reliable chemical restraints and is a feasible option for immobilizing spectacled bears, though severe hypoxemia may proceed. Hypoxemia is the most commonly described complication in bear anesthesia, and was also evidenced in the current report. However, low PaO2/FiO2 ratios tend to be accompanied by hypercapnia and therefore counteracted by oxygen supplementation in bears, which was not observed in the present report. In fact, blood oxygenation only reached regular values after alveolar recruitment maneuvers, which is compatible to an atelectasis-related hypoxemia. Therefore, either inhalant anesthesia or field chemical restraint should be accompanied by advanced monitoring (cardiorespiratory variables and blood gas analysis) in until further studies address the management of hypoxemia in spectacled bear. since Advanced monitoring was of major importance for a safe outcome and an uneventful recovery in this species.Keywords: balanced anesthesia, dexmedetomidine, general anesthesia, spectacled bear, Tremarctos ornatus, wildlife

    Expression of an IKKγ Splice Variant Determines IRF3 and Canonical NF-κB Pathway Utilization in ssRNA Virus Infection

    Single stranded RNA (ssRNA) virus infection activates the retinoic acid inducible gene I (RIG-I)- mitochondrial antiviral signaling (MAVS) complex, a complex that coordinates the host innate immune response via the NF-kappaB and IRF3 pathways. Recent work has shown that the IkappaB kinase (IKK)gamma scaffolding protein is the final common adapter protein required by RIG-I.MAVS to activate divergent rate-limiting kinases downstream controlling the NF-kappaB and IRF3 pathways. Previously we discovered a ubiquitous IKKgamma splice-variant, IKKgammaDelta, that exhibits distinct signaling properties.We examined the regulation and function of IKKgamma splice forms in response to ssRNA virus infection, a condition that preferentially induces full length IKKgamma-WT mRNA expression. In IKKgammaDelta-expressing cells, we found increased viral translation and cytopathic effect compared to those expressing full length IKKgamma-WT. IKKgammaDelta fails to support viral-induced IRF3 activation in response to ssRNA infections; consequently type I IFN production and the induction of anti-viral interferon stimulated genes (ISGs) are significantly attenuated. By contrast, ectopic RIG-I.MAVS or TNFalpha-induced canonical NF-kappaB activation is preserved in IKKgammaDelta expressing cells. Increasing relative levels of IKKgamma-WT to IKKgammaDelta (while keeping total IKKgamma constant) results in increased type I IFN expression. Conversely, overexpressing IKKgammaDelta (in a background of constant IKKgamma-WT expression) shows IKKgammaDelta functions as a dominant-negative IRF3 signaling inhibitor. IKKgammaDelta binds both IKK-alpha and beta, but not TANK and IKKepsilon, indicating that exon 5 encodes an essential TANK binding domain. Finally, IKKgammaDelta displaces IKKgammaWT from MAVS explaining its domainant negative effect.Relative endogenous IKKgammaDelta expression affects cellular selection of inflammatory/anti-viral pathway responses to ssRNA viral infection

    Paramyxovirus Infection Regulates T Cell Responses by BDCA-1 + and BDCA-3 + Myeloid Dendritic Cells

    Abstract Respiratory syncytial virus (RSV) and human Metapneumovirus (hMPV), viruses belonging to the family Paramyxoviridae, are the most important causes of lower respiratory tract infection in young children. Infections with RSV and hMPV are clinically indistinguishable, and both RSV and hMPV infection have been associated with aberrant adaptive immune responses. Myeloid Dendritic cells (mDCs) play a pivotal role in shaping adaptive immune responses during infection; however, few studies have examined how interactions of RSV and hMPV with individual mDC subsets (BDCA-1 + and BDCA-3 + mDCs) affect the outcome of anti-viral responses. To determine whether RSV and hMPV induce virus-specific responses from each subset, we examined co-stimulatory molecules and cytokines expressed by BDCA-1 + and BDCA-3 + mDCs isolated from peripheral blood after infection with hMPV and RSV, and examined their ability to stimulate T cell proliferation and differentiation. Our data show that RSV and hMPV induce virus-specific and subset-specific patterns of co-stimulatory molecule and cytokine expression. RSV, but not hMPV, impaired the capacity of infected mDCs to stimulate T cell proliferation. Whereas hMPVinfected BDCA-1 + and BDCA-3 + mDCs induced expansion of Th17 cells, in response to RSV, BDCA-1 + mDCs induced expansion of Th1 cells and BDCA-3 + mDCs induced expansion of Th2 cells and Tregs. These results demonstrate a virusspecific and subset-specific effect of RSV and hMPV infection on mDC function, suggesting that these viruses may induce different adaptive immune responses
