    Influence of fault geometries and mechanical anisotropies on the growth and inversion of hanging-wall synclinal basins: insights from sandbox models and natural examples

    Salt is mechanically weaker than other sedimentary rocks in rift basins. It commonly acts as a strain localizer, and decouples supra- and sub-salt deformation. In the rift basins discussed in this paper, sub-salt faults commonly form wide and deep ramp synclines controlled by the thickness and strength of the overlying salt section, as well as by the shapes of the extensional faults, and the magnitudes and slip rates along the faults. Upon inversion of these rift basins, the inherited extensional architectures, and particularly the continuity of the salt section, significantly controls the later contractional deformation. This paper utilizes scaled sandbox models to analyse the interplay between sub-salt structures and supra-salt units during both extension and inversion. Series 1 experiments involved baseline models run using isotropic sand packs for simple and ramp-flat listric faults, as well as for simple planar and kinked planar faults. Series 2 experiments involved the same fault geometries but also included a pre-extension polymer layer to simulate salt in the stratigraphy. In these experiments, the polymer layer decoupled the extensional and contractional strains, and inhibited the upwards propagation of sub-polymer faults. In all Series 2 experiments, the extension produced a synclinal hanging-wall basin above the polymer layer as a result of polymer migration during the deformation. During inversion, the supra-polymer synclinal basin was uplifted, folded and detached above the polymer layer. Changes in thickness of the polymer layer during the inversion produced primary welds and these permitted the sub-polymer deformation to propagate upwards into the supra-salt layers. The experimental results are compared with examples from the Parentis Basin (Bay of Biscay), the Broad Fourteens Basin (southern North Sea), the Feda Graben (central North Sea) and the Cameros Basin (Iberian Range, Spain)

    Breast cancer cells adapt contractile forces to overcome steric hindrance

    Cell migration through the extracellular matrix is governed by the interplay between cell-generated propulsion forces, adhesion forces, and resisting forces arising from the steric hindrance of the matrix. Steric hindrance in turn depends on matrix porosity, matrix deformability, cell size, and cell deformability. In this study, we investigate how cells respond to changes in steric hindrance that arise from altered cell mechanical properties. Specifically, we measure traction forces, cell morphology, and invasiveness of MDA-MB 231 breast cancer cells in three-dimensional collagen gels. To modulate cell mechanical properties, we either decrease nuclear deformability by twofold overexpression of the nuclear protein lamin A or we introduce into the cells stiff polystyrene beads with a diameter larger than the average matrix pore size. Despite this increase of steric hindrance, we find that cell invasion is only marginally inhibited, as measured by the fraction of motile cells and the mean invasion depth. To compensate for increased steric hindrance, cells employ two alternative strategies. Cells with higher nuclear stiffness increase their force polarity, whereas cells with large beads increase their net contractility. Under both conditions, the collagen matrix surrounding the cells stiffens dramatically and carries increased strain energy, suggesting that increased force polarity and increased net contractility are functionally equivalent strategies for overcoming an increased steric hindrance

    FUT8-directed core fucosylation of N-glycans is regulated by the glycan structure and protein environment

    FUT8 is an essential a-1, 6-fucosyltransferase that fucosylates the innermost GlcNAc of N-glycans, a process called core fucosylation. In vitro, FUT8 exhibits substrate preference for the biantennary complex N-glycan oligosaccharide (G0), but the role of the underlying protein/peptide to which N-glycans are attached remains unclear. Here, we explored the FUT8 enzyme with a series of N-glycan oligosaccharides, N-glycopeptides, and an Asn-linked oligosaccharide. We found that the underlying peptide plays a role in fucosylation of paucimannose (low mannose) and high-mannose N-glycans but not for complex-type N-glycans. Using saturation transfer difference (STD) NMR spectroscopy, we demonstrate that FUT8 recognizes all sugar units of the G0 N-glycan and most of the amino acid residues (Asn-X-Thr) that serve as a recognition sequon for the oligosaccharyltransferase (OST). The largest STD signals were observed in the presence of GDP, suggesting that prior FUT8 binding to GDP-ß-l-fucose (GDP-Fuc) is required for an optimal recognition of N-glycans. We applied genetic engineering of glycosylation capacities in CHO cells to evaluate FUT8 core fucosylation of high-mannose and complex-type N-glycans in cells with a panel of well-characterized therapeutic N-glycoproteins. This confirmed that core fucosylation mainly occurs on complex-type N-glycans, although clearly only at selected glycosites. Eliminating the capacity for complex-type glycosylation in cells (KO mgat1) revealed that glycosites with complex-type N-glycans when converted to high mannose lost the core Fuc. Interestingly, however, for erythropoietin that is uncommon among the tested glycoproteins in efficiently acquiring tetra-antennary N-glycans, two out of three N-glycosites obtained Fuc on the high-mannose N-glycans. An examination of the N-glycosylation sites of several protein crystal structures indicates that core fucosylation is mostly affected by the accessibility and nature of the N-glycan and not by the nature of the underlying peptide sequence. These data have further elucidated the different FUT8 acceptor substrate specificities both in vitro and in vivo in cells, revealing different mechanisms for promoting core fucosylation.

    Analysis of Learning Records to Detect Student Cheating on Online Exams: Case Study during COVID-19 Pandemic

    In March 2020, due to the Covid19 pandemic, higher education had to switch from face-to-face to exclusively virtual mode overnight. In this unexpected scenario, supervisors also had to adapt the assessment procedures, including the exams. This caused a significant controversy, as, according to many students, supervisors were more concerned about how to prevent students from cheating, than actually measuring their learning. This paper introduces an experience that implemented several of the students' requests in an online exam and conducts a comprehensive analysis of students’ behavior according to the virtual learning environment records. Different existing software tools are used for the analysis, complemented with a Python application ad-hoc developed. The objective indicators gathered provide evidence that some students cheated and invite focusing on evidence-based assessment

    Electronic excitation of furfural as probed by high-resolution vacuum ultraviolet spectroscopy, electron energy loss spectroscopy, and ab initio calculations

The electronic spectroscopy of isolated furfural (2-furaldehyde) in the gas phase has been investigated using high-resolution photoabsorption spectroscopy in the 3.5-10.8 eV energy-range, with absolute cross section measurements derived. Electron energy loss spectra are also measured over a range of kinematical conditions. Those energy loss spectra are used to derive differential cross sections and in turn generalised oscillator strengths. These experiments are supported by ab initio calculations in order to assign the excited states of the neutral molecule. The good agreement between the theoretical results and the measurements allows us to provide the first quantitative assignment of the electronic state spectroscopy of furfural over an extended energy range.F.F.S. and P.L.V. acknowledge the Portuguese Foundation for Science and Technology (FCT-MEC) through Grant Nos.     Different resource allocation strategies result from selection for litter size at weaning in rabbit does

    This study examined the effect of long-term selection of a maternal rabbit line, solely for a reproductive criterion, on the ability of female rabbits to deal with constrained environmental conditions. Female rabbits from generations 16 and 36 (n = 72 and 79, respectively) of a line founded and selected to increase litter size at weaning were compared simultaneously. Female rabbits were subjected to normal (NC), nutritional (NF) or heat (HC) challenging conditions from 1st to 3rd parturition. Animals in NC and NF were housed at normal room temperatures (18°C to 25°C) and respectively fed with control (11.6 MJ digestible energy (DE)/kg dry matter (DM), 126 g digestible protein (DP)/kg DM, and 168 g of ADF/kg DM) or low-energy fibrous diets (9.1 MJ DE/kg DM, 104 g DP/kg DM and 266 g ADF/kg DM), whereas those housed in HC were subjected to high room temperatures (25°C to 35°C) and the control diet. The litter size was lower for female rabbits housed in both NF and HC environments, but the extent and timing where this reduction took place differed between generations. In challenging conditions (NF and HC), the average reduction in the reproductive performance of female rabbits from generation 16, compared with NC, was &#8722;2.26 (P<0.05) and &#8722;0.51 kits born alive at 2nd and 3rd parturition, respectively. However, under these challenging conditions, the reproductive performance of female rabbits from generation 36 was less affected at 2nd parturition (&#8722;1.25 kits born alive), but showed a greater reduction at the 3rd parturition (&#8722;3.53 kits born alive; P<0.05) compared with NC. The results also showed differences between generations in digestible energy intake, milk yield and accretion, and use of body reserves throughout lactation in NC, HC and NF, which together indicate that there were different resource allocation strategies in the animals from the different generations. Selection to increase litter size at weaning led to increased reproductive robustness at the onset of an environmental constraint, but failure to sustain the reproductive liability when the challenge was maintained in the long term.     Breakup reaction models for two- and three-cluster projectiles

    Breakup reactions are one of the main tools for the study of exotic nuclei, and in particular of their continuum. In order to get valuable information from measurements, a precise reaction model coupled to a fair description of the projectile is needed. We assume that the projectile initially possesses a cluster structure, which is revealed by the dissociation process. This structure is described by a few-body Hamiltonian involving effective forces between the clusters. Within this assumption, we review various reaction models. In semiclassical models, the projectile-target relative motion is described by a classical trajectory and the reaction properties are deduced by solving a time-dependent Schroedinger equation. We then describe the principle and variants of the eikonal approximation: the dynamical eikonal approximation, the standard eikonal approximation, and a corrected version avoiding Coulomb divergence. Finally, we present the continuum-discretized coupled-channel method (CDCC), in which the Schroedinger equation is solved with the projectile continuum approximated by square-integrable states. These models are first illustrated by applications to two-cluster projectiles for studies of nuclei far from stability and of reactions useful in astrophysics. Recent extensions to three-cluster projectiles, like two-neutron halo nuclei, are then presented and discussed.     Abnormalities on 1q and 7q are associated with poor outcome in sporadic Burkitt's lymphoma. A cytogenetic and comparative genomic hybridization study

    Comparative genomic hybridization (CGH) studies have demonstrated a high incidence of chromosomal imbalances in non-Hodgkin's lymphoma. However, the information on the genomic imbalances in Burkitt's Lymphoma (BL) is scanty. Conventional cytogenetics was performed in 34 cases, and long-distance PCR for t(8;14) was performed in 18 cases. A total of 170 changes were present with a median of four changes per case (range 1-22). Gains of chromosomal material (143) were more frequent than amplifications (5) or losses (22). The most frequent aberrations were gains on chromosomes 12q (26%), Xq (22%), 22q (20%), 20q (17%) and 9q (15%). Losses predominantly involved chromosomes 13q (17%) and 4q (9%). High-level amplifications were present in the regions 1q23-31 (three cases), 6p12-p25 and 8p22-p23. Upon comparing BL vs Burkitt's cell leukemia (BCL), the latter had more changes (mean 4.3 +/- 2.2) than BL (mean 2.7 +/- 3.2). In addition, BCL cases showed more frequently gains on 8q, 9q, 14q, 20q, and 20q, 9q, 8q and 14q, as well as losses on 13q and 4q. Concerning outcome, the presence of abnormalities on 1q (ascertained either by cytogenetics or by CGH), and imbalances on 7q (P=0.01) were associated with a short survival

    Modelizando el control de las evaporitas y la geometría de falla en desarrollo de cuencas sinclinales y en su posterior inversión: Aplicación a cuencas ibéricas

    En los sistemas de rift con evaporitas, la sal juega un papel clave durante la extensión desacoplando la deformación de los materiales infra- y suprayacentes. Si posteriormente estas cuencas son invertidas, la compresión inicial está fuertemente condicionada por la arquitectura extensiva heredada así como por la continuidad del nivel evaporítico. A partir de una serie de modelos analógicos esta investigación se centra en el papel que juega la sal en sistemas de rift y en su posterior inversión. Se analizan también otros parámetros clave en la deformación como la geometría de la falla principal, la potencia del nivel evaporítico y la tasa de extensión/inversión.Los resultados experimentales muestran que independientemente de la geometría de la falla principal, la presencia de un nivel dúctil potente o una tasa de deformación lenta favorecen el desacople entre los materiales infra- y supra-silicona desarrollándose dos estilos estructurales claramente diferentes.Mientras la formación de fallas es común en los materiales infrayacentes al nivel dúctil, la cobertera mimetiza la geometría de estos mediante amplios pliegues. Estos resultados se comparan con diversas cuencas sinclinales ibéricas con diferentes grados de inversión

    Arrangement of Type IV Collagen and Laminin on Substrates with Controlled Density of ¿OH Groups

We studied the adsorption pattern of these proteins on five model surfaces with tailored density of-OH groups obtained by copolymerization of different ratios ethyl acrylate (EA) and hydroxyl EA (HEA): X OH=0, X OH=0.3, X OH=0.5, X OH=0.7, and X OH=1 (where X refers the ratio of HEA). Atomic force microscopy revealed substratum-specific adsorption patterns of Col IV and Lam, ranging from single molecules deposition on more hydrophilic substrata to the formation of complex networks on hydrophobic ones. Human umbilical endothelial cells were used to study the biological performance of adsorbed proteins, following the overall cell morphology, the quantities for cell adhesion and spreading, and the development of focal adhesion complexes and actin cytoskeleton. Surprisingly, two optima in the cellular interaction were observed-one on the most hydrophilic X OH=1 and other on the relatively hydrophobic X OH=0.3 substrate-valid for both Col IV and Lam. When the proteins were adsorbed consecutively, a hydrophobic shift to X OH=0 substratum was obtained.
