Regulated proteolysis of the alternative sigma factor SigX in Streptococcus mutans: implication in the escape from competence
BACKGROUND: SigX (σ(X)), the alternative sigma factor of Streptococcus mutans, is the key regulator for transcriptional activation of late competence genes essential for taking up exogenous DNA. Recent studies reveal that adaptor protein MecA and the protease ClpC act as negative regulators of competence by a mechanism that involves MecA-mediated proteolysis of SigX by the ClpC in S. mutans. However, the molecular detail how MecA and ClpC negatively regulate competence in this species remains to be determined. Here, we provide evidence that adaptor protein MecA targets SigX for degradation by the protease complex ClpC/ClpP when S. mutans is grown in a complex medium. RESULTS: By analyzing the cellular levels of SigX, we demonstrate that the synthesis of SigX is transiently induced by competence-stimulating peptide (CSP), but the SigX is rapidly degraded during the escape from competence. A deletion of MecA, ClpC or ClpP results in the cellular accumulation of SigX and a prolonged competence state, while an overexpression of MecA enhances proteolysis of SigX and accelerates the escape from competence. In vitro protein-protein interaction assays confirm that MecA interacts with SigX via its N-terminal domain (NTD(1–82)) and with ClpC via its C-terminal domain (CTD(123–240)). Such an interaction mediates the formation of a ternary SigX-MecA-ClpC complex, triggering the ATP-dependent degradation of SigX in the presence of ClpP. A deletion of the N-terminal or C-terminal domain of MecA abolishes its binding to SigX or ClpC. We have also found that MecA-regulated proteolysis of SigX appears to be ineffective when S. mutans is grown in a chemically defined medium (CDM), suggesting the possibility that an unknown mechanism may be involved in negative regulation of MecA-mediated proteolysis of SigX under this condition. CONCLUSION: Adaptor protein MecA in S. mutans plays a crucial role in recognizing and targeting SigX for degradation by the protease ClpC/ClpP. Thus, MecA actually acts as an anti-sigma factor to regulate the stability of SigX during competence development
A Study of the Problem Solving Behavior in Youth with Mental Retardation : In Comparison to Children without Mental Retardation on the Transformation Task
The present study was performed to evaluate the food safety of TT51-1, a new type of genetically modified rice that expresses the Cry1Ab/Ac protein (Bt toxin) and is highly resistant to most lepidopteran pests. Sixteen male and 16 female cynomolgus monkeys were randomly divided into four groups: conventional rice (non-genetically modified rice, non-GM rice), positive control, 17.5% genetically modified rice (GM rice) and 70% GM rice. Monkeys in the non-GM rice, positive control, and GM rice groups were fed on diets containing 70% non-GM rice, 17.5% GM rice or 70% GM rice, respectively, for 182 days, whereas animals in the positive group were intravenously injected with cyclophosphamide every other day for a total of four injections before the last treatment. Six months of treatment did not yield abnormal observations. Specifically, the following parameters did not significantly differ between the non-GM rice group and GM rice groups: body weight, food consumption, electrocardiogram, hematology, immuno-phenotyping of lymphocytes in the peripheral blood, mitogen-induced peripheral blood lymphocyte proliferation, splenocyte proliferation, KLH-T cell-dependent antibody response, organ weights and ratios, and histological appearance (p>0.05). Animals from the GM rice group differed from animals in the non-GM rice group (p<0.05) in several parameters: specifically, their body temperatures and serum alanine aminotransferase (ALT) levels were higher, whereas their levels of serum K+, Cl- and cytokines (IL-2, IL-4 and IL-5) were lower. Because dose- or time-dependent changes were not observed in this study and animals appeared histologically normal, the aforementioned differences were not considered to be adverse or related to the treatment with GM rice. In conclusion, a 6-month feeding study of TT51-1 did not show adverse immunotoxicological effects on cynomolgus monkeys

Construction of a map-based reference genome sequence for barley, Hordeum vulgare L.
Barley (Hordeum vulgare L.) is a cereal grass mainly used as animal fodder and raw material for the malting industry. The map-based reference genome sequence of barley cv. `Morex' was constructed by the International Barley Genome Sequencing Consortium (IBSC) using hierarchical shotgun sequencing. Here, we report the experimental and computational procedures to (i) sequence and assemble more than 80,000 bacterial artificial chromosome (BAC) clones along the minimum tiling path of a genome-wide physical map, (ii) find and validate overlaps between adjacent BACs, (iii) construct 4,265 non-redundant sequence scaffolds representing clusters of overlapping BACs, and (iv) order and orient these BAC clusters along the seven barley chromosomes using positional information provided by dense genetic maps, an optical map and chromosome conformation capture sequencing (Hi-C). Integrative access to these sequence and mapping resources is provided by the barley genome explorer (BARLEX).Peer reviewe
Physical Properties, Antioxidant and Antimicrobial Activity of Chitosan Films Containing Carvacrol and Pomegranate Peel Extract
Chitosan-based active films were developed by incorporation of carvacrol (10 g/L), pomegranate peel extract (PPE, 10 g/L) and carvacrol + PPE (10 g/L of each) and their physical, antioxidant and antimicrobial properties were investigated. Incorporation of carvacrol and carvacrol + PPE into the films significantly decreased the water vapor permeability, tensile strength and percentage of elongation at break. Incorporation of carvacrol, PPE and carvacrol + PPE into the films decreased the transparency, but significantly increased the total phenol content and antioxidant activity. All the films, with the exception of PPE-incorporated film, exhibited antibacterial activity against Escherichia coli and Staphylococcus aureus. In addition, the antibacterial activity against Staphylococcus aureus of the film incorporated with carvacrol + PPE was moderately higher than that incorporated with carvacrol or PPE alone, suggesting a synergistic action between carvacrol and PPE
AF-OSD: An Anchor-Free Oriented Ship Detector Based on Multi-Scale Dense-Point Rotation Gaussian Heatmap
Due to the complexity of airborne remote sensing scenes, strong background and noise interference, positive and negative sample imbalance, and multiple ship scales, ship detection is a critical and challenging task in remote sensing. This work proposes an end-to-end anchor-free oriented ship detector (AF-OSD) framework based on a multi-scale dense-point rotation Gaussian heatmap (MDP-RGH) to tackle these aforementioned challenges. First, to solve the sample imbalance problem and suppress the interference of negative samples such as background and noise, the oriented ship is modeled via the proposed MDP-RGH according to its shape and direction to generate ship labels with more accurate information, while the imbalance between positive and negative samples is adaptively learned for the ships with different scales. Then, the AF-OSD based on MDP-RGH is further devised to detect the multi-scale oriented ship, which is the accurate identification and information extraction for multi-scale vessels. Finally, a multi-task object size adaptive loss function is designed to guide the training process, improving its detection quality and performance for multi-scale oriented ships. Simulation results show that extensive experiments on HRSC2016 and DOTA ship datasets reveal that the proposed method achieves significantly outperforms the compared state-of-the-art methods
