    A interação e a brincadeira como orientação para o desenvolvimento infantil / Interaction and play as guidelines for child development

    O presente artigo é decorrente do trabalho desenvolvido por um grupo de cinco acadêmicos na disciplina de Estágio Supervisionado sob a forma de Prática de Ensino III, do quarto ano do curso de Pedagogia da Universidade Estadual do Oeste do Paraná – UNIOESTE, campus de Cascavel - PR, como forma de divulgar o trabalho desenvolvido junto a uma escola campo de estágio de Educação Infantil e proporcionar uma compreensão sobre o cotidiano escolar e suas especificidades na fase inicial dessa modalidade de ensino. Para alcançar o objetivo proposto, que consiste na apreensão de como ocorre o desenvolvimento infantil e na busca de encaminhamentos teóricos e metodológicos para conduzir o processo de ensino e aprendizagem, realizamos, na escola, observações em salas de aula, encontros com a equipe pedagógica e professores, regências em uma turma de Educação Infantil I e em uma turma de Educação Infantil III, bem como desenvolvemos um projeto que integrou as crianças ao participarem da apreciação de uma peça teatral organizada e apresentada pelos acadêmicos. Nessa perspectiva, evidenciamos, neste texto, a importância da interação e do brincar no desenvolvimento infantil, além de tecermos algumas considerações a partir do trabalho pedagógico desenvolvido nas duas turmas mencionadas. Teoricamente, o estudo é pautado, sobretudo, em Arce (2013), Piorski (2016); Vigotski (2002) e Zinchenko (2012 apud ARCE, 2013).

    DIRAC [DIRAC] [TSA-08] is a software framework for building distributed computing systems. It was primarily designed forthe needs of the LHCb [LHCb] Collaboration, and is now used by many other communities within EGI [EGI] as a primary wayof accessing grid resources. In France, dedicated instances of the service have been deployed in different locations toanswer specific needs. Building upon this existing expertise, France Grilles [FG] initiated last year a project to deploy anational, multi-community instance in order to share expertise and provide a consistent high-quality service. After describingDIRAC main aims and functionalities, this paper presents the motivations for such a project, as well as the wholeorganizational and technical process that led to the establishment of a production instance that already serves 13communities: astro.vo.eu-egee.org, biomed, esr, euasia, gilda, glast.org, prod.vo.eu-eela.eu, superbvo.org,vo.formation.idgrilles.fr, vo.france-asia.org, vo.france-grilles.fr, vo.msfg.fr and vo.mcia.fr

    Reflexões acerca do estágio supervisionado sob a forma de prática de ensino nos anos iniciais do ensino fundamental / Reflections on supervised internship in the form of teaching practice in the early years of elementary education

    Temos por intencionalidade refletir e analisar a inserção do acadêmico de pedagogia no ambiente escolar, por meio da realização do Estágio Supervisionado sob a forma de Prática de ensino II, o qual possibilitou o conhecimento do cotidiano escolar, com ênfase ao trabalho do coordenador pedagógico, considerando os pressupostos da Pedagogia Histórico-Crítica e da Psicologia Histórico-Cultural que embasam o Currículo para a Rede Pública Municipal de Ensino de Cascavel – Ensino Fundamental – Anos Iniciais (CASCAVEL, 2008). Como metodologia, realizamos observações no espaço escolar e na sala de aula, entrevistas com a equipe pedagógica, elaboração de planos de aula e realização das respectivas regências em séries iniciais inseridas no Ciclo Básico de Alfabetização. Desenvolvemos, também, juntamente com a diretora, a equipe de coordenadores pedagógicos e os professores da escola pública do município de Cascavel, que foi o campo do estágio mencionado, um projeto de extensão que consistiu na análise dos descritores de Língua Portuguesa da Prova Brasil. Por meio dessa experiência, obtivemos a percepção das características da sala de aula e da relação professor-aluno, entendendo que ela pode resultar em um ambiente agradável e proveitoso ou em um ambiente autoritário que, por consequência, limita a participação do aluno e a qualidade na apropriação do conhecimento. Constatamos, ainda, que os resultados quanto a essa apropriação também são decorrentes dos encaminhamentos teóricos e metodológicos utilizados pelo professor para explorar os conteúdos, o que pode ser enriquecido pela respectiva contextualização, mas com a certeza de que nosso compromisso maior é oportunizar o conhecimento científico ao aluno. 

    Paisajes de barro: conociendo los usos tradicionales de la arcilla en Antofagasta de la Sierra (Catamarca)

    The Research and Dissemination Project “Paisajes de barro” (Clay Landscapes) proposes a cross-disciplinary and participative approach to the use of earth as raw material in the Antofagasta de la Sierra Department (Catamarca). In terms of research, our goal is to study the use of this material during recent history and its current status. Thus, together with local communities, our group of students and graduates is surveying the cultural heritage linked to the use of clay, in both their material (architectural structures, pottery) and intangible (knowledge, uses, traditions) manifestations. For the last three years we have taken part in diverse activities that involve exchange of knowledge about the different uses of earth in these Puna villages. However, the work is not restricted to research; such activities are also aimed at documenting and disseminating the objects, structures and know-how of clay technologies, as well as contributing to their joint preservation and valorization. Thus, our project recovers ancient practices, uses and traditions while reflecting upon the advantages of these technologies for modern societies at Antofagasta de la Sierra, both from a cultural perspective and as sustainable economy and good living.El Proyecto de Extensión e Investigación “Paisajes de barro” propone un acercamiento al uso del barro como materia prima en el Departamento de Antofagasta de la Sierra (Catamarca) desde una perspectiva multidisciplinar y participativa.     Primary hepatocytes as an useful bioassay to characterize metabolism and bioactivity of illicit steroids in cattle.

    Cattle hepatocytes have already been used in veterinary in vitro toxicology, but their usefulness as a multi-parametric screening bioassay has never been investigated so far. In this study, cattle hepatocytes were incubated with illicit steroids/prohormones (boldenone, BOLD; its precursor boldione, ADD; dehydroepiandrosterone, DHEA; an association of ADD:BOLD), to characterize their transcriptional effects on drug metabolizing enzymes (DMEs) and related nuclear receptors (NRs), on cytochrome P450 3A (CYP3A) apoprotein and catalytic activity as well as to determine ADD and BOLD metabolite profiling. DHEa-exposed cells showed an up-regulation (higher than 2.5-fold-changes) of three out of six NRs, CYP2B22 and CYP2C87; likewise, ADD:BOLD increased CYP4A11 mRNA levels. In contrast, a reduction of CYP1A1 and CYP2E1 mRNAs (lower than 2.5 121-fold-changes) was noticed in ADD- and DHEA-incubated cells. No effect was noticed on CYP3A gene and protein expression, though an inhibition of 6\u3b2-, 2\u3b2- and 16\u3b2-hydroxylation of testosterone (higher than 60% of control cells) was observed in ADD- and BOLD-exposed cells. Finally, 17\u3b1-BOLD was the main metabolite extracted from hepatocyte media incubated with ADD and BOLD, but several mono-hydroxylated BOLD and ADD derivatives were detected, too. Collectively, cattle hepatocytes can represent a complementary screening bioassay, useful to characterize growth promoters metabolite profiling and their effects upon DMEs expression, regulation and function

    Introduction. Cattle HPCs have been successfully used to investigate the metabolism and bioactivity of illicit steroids and prohormones (ISAPs). Nevertheless, their usefulness as a multi-parametric screening bioassay has never been investigated so far. Thus, cattle HPCs were incubated with known ISAPs and submitted to investigations aiming to characterize: (a) their effects on mRNA levels of drug metabolizing enzymes (DMEs) and related transcription factors (TFs); (b) on CYP3A28 apoprotein and catalytic activity; (c) the ISAPs metabolite profiling. Materials and Methods. HPCs were incubated with 100 \u3bcM boldenone (BOLD), its precursor androsta-l,4-diene-3,17-dione (ADD), dehydroepiandrosterone (DHEA) or an association of BOLD:ADD (90:10 \ub5M). ISAPs effects on 16 DMEs and 5 TFs mRNAs were measured, after 6 hrs of incubation, by qPCR; their effect on CYP3A28 apoprotein and catalytic activity, after 24 hrs of incubation, by immunoblotting and a testosterone (TST) HPLC assay. Finally, the time-dependent (0, 3, 6 and 24 hrs) metabolite profiling of ADD, BOLD and ADD:BOLD was characterized by using a LC-HRMS technique. Results. Cells exposed to DHEA showed a significant up-regulation of CYP2C9, GSTA1, DHEA-ST, 17\u3b2HSDII, CAR, PXR, RXR\u3b1 and PPAR\u3b1 mRNAs. In contrast, ISAPs did not alter mRNA levels in BOLD-, ADD- or BOLD:ADD-exposed cells, except for a significant reduction of CYP1A1 mRNA in ADD-incubated hepatocytes. No effect of ISAPs was ever noticed on CYP3A28 gene and protein expression, while a significant inhibition of 6\u3b2-, 2\u3b2 and 16\u3b2-OHTSTase was observed in cells exposed to ADD and BOLD. Finally, the production of ADD and \u3b1-BOLD from BOLD as well as of \u3b1-BOLD and \u3b2-BOLD from ADD as main metabolites was demonstrated. Furthermore, several OH-ADD derivatives were detected, and 16-OH-ADD epimers were more abundant than 6-OH-ADD ones. As observed with other anabolic steroids, four mono-hydroxylated BOLD derivatives were isolated and tentatively identified. Conclusions. Cattle HPCs represent an useful in vitro model to study species-specific differences in xenobiotic metabolism and molecular mechanisms involved in DMEs expression and regulation, as well as a complementary tool for the screening of ISAPs abuse in cattle. If liver tissue collected at the slaughterhouse was useful to obtain viable HPCs, this in vitro model might reduce the use of experimental animals, according to the 3R\u2019s philosophy

    Effects of Illicit Dexamethasone upon Hepatic Drug Metabolizing Enzymes and Related Transcription Factors mRNAs and Their Potential Use As Biomarkers in Cattle

    In cattle fattening, the illicit use of growth promoters (GPs) represents a major problem. The synthetic corticosteroid dexamethasone (DEX) is the GP mostly used, alone or in combination with other steroids or beta-agonists. Recently, GPs were shown to disrupt some cattle cytochromes P450 (CYPs) at the post-transcriptional level; therefore, the effects of two illicit protocols containing DEX (alone or together with 17beta-estradiol, 17betaE) upon main cattle liver drug metabolizing enzymes (DMEs) mRNAs and related transcription factors were investigated by quantitative real time RT-PCR. Eleven genes, out of the 18 considered, were significantly modulated by GPs. Corticosteroid-responsive genes did not respond univocally, whereas retinoic X receptor alpha (RXRalpha) and estrogen receptor alpha (ERalpha) were upregulated depending on the illicit protocol used. Nowadays, an increasing interest has been noticed toward the detection of biomarkers of response (BMRs) to be used in the screening of GPs misuse in cattle farming. In the present study, CYP2B6-like, CYP2E1, glutathione S-transferase A1- and sulfotransferase A1-like (GSTA1- and SULT1A1-like) mRNAs were significantly modulated regardless of the GP, the illicit protocol, and the animal breed, representing promising BMRs.
