The compositional HJ-Biplot—A new approach to identifying the links among bioactive compounds of tomatoes

    Get PDF
Tomatoes have been described as a functional food because of their particular composition of different bioactive compounds. In this study, the proximate composition, minerals and trace elements, and antioxidant compounds were determined in two tomato cultivars (Mariana and Dunkan) that were grown in Gran Canaria (Spain) either conventionally or hydroponically. Although compositional data of this type require being subjected to the specific statistical techniques of compositional analysis, this approach has not usually been considered in this context. In the present case, a compositional Mann–Whitney U test of the data showed significant differences for each factor (cultivar and cultivation system) in several of the compositional variables studied. For the differences between cultivars, these parameters were the protein, Mg, lycopene, ascorbic acid, citric acid, and fumaric acid contents. For the differences between cultivation systems, they were mainly those of the mineral and trace elements group. Although one-year data are insufficient to make clear relationship among compounds because more repetitions in several localities and years are necessary, the compositional HJ-biplot (in which the links provide estimates of the linear relationship among variables) results agreed with other scientific results about linear relationship among some compounds analyzed.peerReviewe

Expression of the p53 protein in oral squamous cell carcinomas associated with Epstein-Barr virus

The behaviour of the p53 protein has been investigated in some human carcinomas associated with Epstein-Barr virus (EBV) but not in EBV-positive oral squamous cell carcinomas (OSCC). The present study aimed to compare the p53 protein expression in EBV-positive OSCC with that in EBV-negative OSCC. The cases had been gathered in a study previously published. An immunohistochemical technique with BP53-12 monoclonal antibody was applied on 74 of the 107 OSCC from the earlier work. The nuclear or cytoplasmic expression of the p53 protein was classified as, absent (0% of neoplastic cells positive), mild (50% positive). The p53 protein was expressed by 60.8% of the OSCC. Out of the fourteen EBV-positive OSCC, 57.1% (8 cases) expressed p53, always in the nucleus and never in the cytoplasm. Of the 60 EBV-negative OSCC, 61.6% (37 cases) expressed the p53 protein. Of 37 cases 33 (89.1%) showed nuclear expression of p53 and nineteen cases (51.3%) revealed cytoplasmic expression. There was a statistically significant inverse correlation between cytoplasmic expression of the p53 protein and the presence of EBV DNA (p <0.01). Thus, the EBV-positive tumours less frequently expressed p53 in the cytoplasm. No evidence of an accumulation of the p53 protein in OSCC associated with EBV was recorded

Objective. To analyze the genetic uniformity of the MSXJ papaya hybrid vitroplants, obtained via direct organogenesis. Design/methodology/approach. The MSXJ papaya hybrid presents quality characteristics for national and export markets. The in vitro plant tissue culture represents a tool for its multiplication and conservation, but somaclonal variation can decrease its genetic and agronomic uniformity. In order to analyze the genetic uniformity of this hybrid vitroplants, 10 ISSR primers were used in micropropagated vitroplants during nine subcultures. DNA extraction was carried out with the CTAB method. The data analysis was performed with the PopGene v 1.3.1 program. Results. Eighty five loci of 200 to 2000 bp were generated, with 37 polymorphic loci. In the cluster analysis, three groups were observed that separated subculture one, subcultures two through eight, and subculture nine; the Gst value of 0.87 indicated genetic uniformity up to subculture eight. Limitations/implications. Papaya is one of the most important tropical fruits worldwide; however it is necessary to have healthy and genetically uniform plants that guarantee their quality. In vitro propagation allows to produce healthy and uniform plants, but it is necessary to study their genetic uniformity during their micropropagation. Findings/conclusions. The in vitro multiplication of the MSXJ papaya hybrid allowed to regenerate vigorous plants in 30 days. Molecular profiles indicated that up to subculture eight plantlets were genetically uniform, so it is recommended not to carry out more than eight subcultures during micropropagation. Objective: To analyze the genetic uniformity of MSXJ hybrid papaya in vitro plants, obtained by direct organogenesis.Design/Methodology/Approach: The MSXJ papaya hybrid demonstrates quality characteristics for the national and exports market. In vitro culture of plant tissues represents a useful tool for their multiplication and conservation, but somaclonal variation can diminish their genetic and agronomic uniformity. Study focus Medina del Campo groundwater body (MCGWB) is a multilayer semiconfined aquifer subject to intensive pumping since the 1970’s, where the current existence of spas where there used to be traditional baths could confirm the existence of deep groundwater flow paths. The old spring of Las Salinas (OSLS) is a saline anomaly in an aquifer with predominance of CaCO3H waters whose occurrence has not yet been formally explained. Long-term geological, geophysical, hydrogeological and hydrochemical records were integrated and complemented with field work to clarify its existence. New hydrological insights for the region Outcomes led to the conclusion that the hydrochemistry of the Olmedo and Palacio de las Salinas salt baths is associated with the existence of a major threshold in the impervious basement of the aquifer, which intercepted deep regional groundwater flow and caused upwelling to the surface under unperturbed conditions. These results allow for the development of a conceptual flow model at the regional scale that explains the changes in natural water chemistry that have been identified in recent decades. This research has received funding from the European Union H2020 Programme under Grant Agreement No. 730497 for the research project NAIAD-NAture Insurance value: Assessment and Demonstration.Peer reviewe

Uso y resultados del astegolimab en el manejo del asma severa: ¿qué se conoce?

Asthma is a chronic inflammatory disease of the respiratory tract which causes high health costs, substantially affects the quality of life and, depending on certain associated risk factors, reduces the functional capacity of the sufferer. By 2019, asthma affected 262 million people (4.3 % of the world’s population) and caused 461,000 deaths. It is estimated that therewill be an additional 100 million people with asthma by 2025. Severe asthma is a phenotype resistant to corticosteroids which causes a greater number of exacerbations and substantially affects the quality of life and functional capacity of the affected person. Its management was initially aimed at suppressing the symptoms and then evolved to understand, although not completely, the intrinsic systems of its generation. Thus, new ways of influencing its management have been studied by modulating the immune response and the inflammatory cascade with the generation of biological drugs. As a result of the study and identification of various endotypes and phenotypes, drugs with different mechanisms of action have been designed and have demonstrated to be considerably useful in recent years. However, there is evidence that resistance even to these drugs has occurred, being necessary to continue researching new therapeutic targets. Astegolimab is a novel human IgG2 monoclonal antibody that blocks IL-33 signaling by targeting ST2, its receptor, thus controlling the inflammatory response in severe asthma. A phase 2b clinical trial is currently undergoing, although previous results have found positive and significant results regarding immunomodulation, pulmonary function, symptomatology and quality of life. At present, there is almost no literature that has analyzed the potential of astegolimab in severe asthma, and practically only trials that have evaluated it and some reviews that have shared its pharmacokinetics and pharmacodynamics are available. Based on the above, the aim of this review is to synthesize evidence related to the results of the use of astegolimab insevere asthma and discuss epidemiological and pathophysiological aspects that highlight the need for the development of a safe, effective and efficient drug.

    Get PDF
Objectives. This article estimates the frequency of polyautoimmunity and associated factors in a large retrospective cohort of patients with SLE. Methods. RELESSER (Spanish Society of Rheumatology Lupus Registry) is a nationwide multicentre, hospital-based registry of SLE patients. This is a cross-sectional study. The main variable was polyautoimmunity, which was defined as the co-occurrence of SLE and another autoimmune disease, such as autoimmune thyroiditis, RA, scleroderma, inflammatory myopathy and MCTD. We also recorded the presence of multiple autoimmune syndrome, secondary SS, secondary APS and a family history of autoimmune disease. Multiple logistic regression analysis was performed to investigate possible risk factors for polyautoimmunity. Results. Of the 3679 patients who fulfilled the criteria for SLE, 502 (13.6%) had polyautoimmunity. The most frequent types were autoimmune thyroiditis (7.9%), other systemic autoimmune diseases (6.2%), secondary SS (14.1%) and secondary APS (13.7%). Multiple autoimmune syndrome accounted for 10.2% of all cases of polyautoimmunity. A family history was recorded in 11.8%. According to the multivariate analysis, the factors associated with polyautoimmunity were female sex [odds ratio (95% CI), 1.72 (1.07, 2.72)], RP [1.63 (1.29, 2.05)], interstitial lung disease [3.35 (1.84, 6.01)], Jaccoud arthropathy [1.92 (1.40, 2.63)], anti-Ro/SSA and/or anti-La/SSB autoantibodies [2.03 (1.55, 2.67)], anti-RNP antibodies [1.48 (1.16, 1.90)], MTX [1.67 (1.26, 2.18)] and antimalarial drugs [0.50 (0.38, 0.67)]. Conclusion. Patients with SLE frequently present polyautoimmunity. We observed clinical and analytical characteristics associated with polyautoimmunity. Our finding that antimalarial drugs protected against polyautoimmunity should be verified in future studies

Congreso realizado en Málaga en junio de 2022.[EN] Karst aquifers and, especially, their caves and springs, are singular sites of the geoheritage due to their scientific, touristic and use (water resources) values. Besides, they are recognized as habitats of special protection by the Habitat Directive (92/43/CEE) and would be used for sustainable development in UNESCO Global Geoparks (UGGp). However, their management continues to be a challenge at the present day since karst aquifers and their associated springs and caves are vulnerable environments used by many actors and affected by land uses and changes in surface hydrology and groundwater. Our work aims to implement of a suitable management of aquifers and springs in the Courel Mountains UGGp based on hydrogeological studies. The studies include mainly the characterization of aquifers’ functioning, the elaboration of the spring inventory, physicochemical analyses and geoconservation actions. This work is funded by the Courel Mountains UGGp and the project IGCP-730 Hydrogeological significance of Mediterranean geoparks, and supported by the International Geosciences Program of UNESCO.Peer reviewe
