In vitro inhibition of linoleic acid peroxidation by primary S-nitrosothiols
Nitric oxide ( NO) is an effective chain-breaking antioxidant in the inhibition of lipid peroxidation and circulates in vivo mainly as primary S-nitrosothiols (RSNOs). In this work, the in vitro peroxidation of linoleic acid-SDS comicelles (LA-SDS) catalyzed by soybean lipoxygenase (SLO) and FeII ions was monitored in the presence and absence of three primary RSNOs: S-nitrosocysteine, S-nitroso-N-acetylcysteyne and S-nitrosoglutathione. Kinetic measurements based on the formation of conjugated double bonds and fluorescent oxidized LA-lysine adducts, showed that RSNOs are more potent antioxidants than their corresponding free thiols (RSHs) in equimolar conditions. These results are consistent with the blocking of LA-SDS peroxidation by RSNOs through the inactivation of peroxyl/alkoxyl (LOO /LO ) radicals, leading to nitrogen-containing products of oxidized LA, which release free NO. These results indicate that endogenous RSNOs may play a major role in the blocking of lipid peroxidation in vivo, through the primary inactivation of alkoxyl/peroxyl radicals and also of preformed lipid hydroperoxides.O óxido nítrico ( NO) é um antioxidante efetivo na inibição da peroxidação lipídica e circula in vivo principalmente como S-nitrosotióis primários (RSNOs). Neste trabalho, a peroxidação in vitro de comicelas do ácido linileico-SDS (LA-SDS) catalisada por lipoxigenase de soja (SLO) e íons FeII foi monitorada na presença e na ausência de três RSNOs: S-nitrosocisteína, S-nitroso-N-acetilcisteína e S-nitrosoglutationa. Medidas cinéticas baseadas na formação de duplas ligações conjugadas e adutos fluorescentes de lisina-La oxidado mostraram que os RSNOs são antioxidantes mais potentes que seus tióis livres (RSHs) correspondentes em condições equimolares. Esses resultados são consistentes com o bloqueio da peroxidação de LA-SDS por RSNOs através da inativação dos radicais peroxila/alcoxila (LOO /LO ), levando a produtos nitrogenados do LA oxidado, os quais liberam NO. Ants Sleep, Plants do not: The Variation in Species’ Activity Influences the Topology of Interaction Networks
The emergence of graph theory allowed using the complex network approach to aggregate detailed information about interactions between species. Although the use of the complex network approach has improved the understanding about community structuring, few aspects such as the temporal variation in the species’ activity pattern in the networks’ topology were explored so far. The current study used the ecological network approach to investigate ants interacting in the extrafloral nectary (EFN) of plants in order to test the hypothesis that the temporal variation in the foraging behavior of these animals affects the networks’ topology. In order to assess the temporal effect on the interaction networks, 24-hour collections divided in two 12-hour shifts (day and night) were performed in 20 plots, thus totaling 288 collection hours over 6 months. The ant-plant interaction networks presented similarity among the topological metrics assessed throughout the day. Different ant species presented distinct foraging times. Thus, two modules referring to the day and night shifts emerged from the network and presented specific species at each foraging shift. On the other hand, the plants kept on providing the resource (active EFNs) throughout the day. The results found in the current study have shown that ecological networks keep their structures constant; however, the ecological processes ruling these networks can better respond to the effects caused, for example, by the temporal variation in species’ activity. Several studies demonstrated its efficacy in the control of the amount and the timely delivery of drugs when it is incorporated in multilayer films. It has been demonstrated that oxide graphene layers are able to work as drug delivery or just to delay consecutive drug dosage, allowing the operation of time-controlled systems. This review presents the latest research developments of biomedical applications using graphene oxide as the main component of a drug delivery system, with focus on the production and characterization of films, in vitro and in vivo assays, main applications of graphene oxide biomedical devices, and its biocompatibility properties.publishersversionpublishe
Enhanced sweet taste perception in obesity: joint analysis of gustatory data from multiple studies

Notably, possible associations between sweet taste perception and measures of food reward remain undetermined.
Materials and methods: We conducted a cross-sectional analysis comparing 246 individuals with severe obesity and 174 healthy volunteers using a validated method for taste perception assessment. We included gustatory variables, namely intensity and pleasantness ratings of sour, salt, sweet, and bitter tastants, and taste thresholds assessed by electrogustometry. Reward-related feeding behavior, including hedonic hunger, food addiction, feeding behavior traits, and acceptance of foods and alcohol, was evaluated using self-rated scales for comparison with gustatory measures.
Result: In logistic regressions adjusted for age, gender, educational level, and research center, we found that a greater likelihood of belonging to the obesity group was associated with higher sweet intensity ratings (OR = 1.4, P = 0.01), hedonic hunger, food addiction symptoms, restrained and emotional eating (1.7 < OR ≤ 4.6, all P ≤ 0.001), and lower alcohol acceptance (OR = 0.6, P = 0.0002). Using principal component analysis, we found that while hedonic hunger, food addiction, and emotional eating were strongly interrelated, they were not associated with sweet intensity perception that, in turn, had a closer relationship with alcohol acceptance and restrained eating.
Conclusion: We found that individuals with obesity report higher sweet taste intensity ratings than healthy controls. Reward-related gustatory and psychometric predictors of weight loss following bariatric surgery: a multicenter cohort study

Objectives: We aimed to determine whether gustatory and psychometric measures of reward-related feeding are predictors of bariatric-induced weight loss.
Methods: A multicenter longitudinal cohort study was conducted in patients scheduled for bariatric surgery (surgical group), assessed at baseline and 2 follow-up assessments. Predictions of % weight loss from baseline (%WL) according to baseline gustatory measures, including intensity and pleasantness ratings of sweet and other tastants, and psychometric measures of reward-related feeding behavior, including hedonic hunger scores, were assessed with multivariable linear regression. Exploratory analyses were conducted to test for associations between %WL and changes in gustatory and psychophysical measures, as well as for comparisons with data from patients on the surgery waiting list (control group).
Results: We included 212 patients, of whom 96 in the surgical group and 50 in the control group were prospectively assessed. The groups were similar at baseline and, as expected, bariatric surgery resulted in higher %WL (BTreatment-Time = 2.4; 95% CI: 2.1-2.8; P < 0.0001). While variation in gustatory measures did not differ between groups, in the surgery group baseline sweet intensity predicted %WL at the primary endpoint (11 to 18 months postoperatively; β = 0.2; B = 0.2, 95% CI: 0.02 to 0.3; P = 0.02), as did hedonic hunger scores (β = -0.2; B = -2.0, 95% CI: -3.8 to -0.3; P = 0.02). Furthermore, at this endpoint, postsurgical reduction of sweet taste intensity and acceptance of sweet foods were associated with %WL (β = -0.3; B = -3.5, 95% CI: -5.8 to -1.3; P = 0.003, and β = -0.2; B = -4.7, 95% CI: -8.5 to -0.8; P = 0.02, respectively). The use of sweet intensity as a predictor of weight change was confirmed in another bariatric cohort.
Conclusions: Sweet intensity ratings and hedonic hunger scores predict %WL after surgery. Effects of treatments for drooling on caries risk in children and adolescents with cerebral palsy
Neuromuscular impairment makes individuals with cerebral palsy (CP) more prone to drooling. Among the treatment options, there are procedures that interfere with saliva production. It is imperative to evaluate the effect of the different modalities since the reduction in salivary flow rate/production may exacerbate the risk of dental caries. The aim of this study was to compare the effects of different treatments for drooling on caries risk and salivary parameters in children and adolescents with CP. A total of 142 children and adolescents with CP, aged 6 to 18 years, were assigned to groups based on the different treatments they had received for drooling: G1?anticholinergic drugs (n = 18), G2?botulinum toxin injection (n = 16), G3?salivary glands surgery (n = 16), G4?no treatment (n = 42), and G5?non-drooling subjects (n = 50). All participants were evaluated on the Simplified Oral Hygiene Index, and for the prevalence of dental caries (decayed, missing, and filled teeth index and white spot lesions). Unstimulated whole saliva was collected, and salivary flow rate and osmolality were measured. Chi-square, ANOVA and Poisson regression were calculated. Prevalence ratios and their respective 95 % confidence intervals were obtained. The significance level was fixed at 5%. No differences were found in the decayed, missing, and filled teeth index (p = 0.128) and Simplified Oral Hygiene Index (p = 0.674) among the different groups. G3 presented significantly higher percentages of WSL (p<0.001), lower values of salivary flow rate (p<0.001), and higher values of osmolality (p<0.001). The white spot lesion prevalence ratio was higher only for G3 (Prevalence ratio = 14.36; IC 95% = 4.64-44.40; p<0.001). Children and adolescents with CP who had received surgical treatment for drooling exhibited higher number of white spot lesions because of the reduced salivary flow rate and higher salivary osmolality
Objective assessment of the cryoprotective effects of dimethylformamide for freezing goat semen
AbstractThe aim of this work was to assess the cryoprotective effects of dimethylformamide (DMF) for freezing goat semen, using an objective analysis by computer-assisted sperm analysis (CASA). Twenty-one ejaculates (seven per animal) were collected from three stud bucks with the aid of an artificial vagina and immediately evaluated for gross and microscopic characteristics. The semen was diluted in two steps with a Tris–egg yolk extender containing 6% glycerol or 6% DMF, frozen in 0.50-mL straws, and stored in liquid nitrogen. Samples were accessed for sperm morphology, sperm membrane structural and functional integrity, and by CASA, immediately after thawing. There were differences (P<0.05) between glycerol and DMF with regard to subjective progressive motility (23.9±2.2% vs. 16.6±2.0%), objective progressive motility (3.5±0.4% vs. 1.8±0.3%), linearity (53.9±1.6% vs. 48.1±1.4%) and amplitude of lateral head (2.3±0.1 vs. 2.9±0.1mm), which confirmed the efficiency of glycerol. In conclusion, dimethylformamide could be used as an alternative cryoprotectant for goat semen freezing. However it was showed that no benefits were derived by using dimethylformamide to replace glycerol at an equal 6% concentration
