    Improvement of Lamb Preweaning Performance by Combination of Superovulation of Ewes Prior to Mating and Temulawak Extract Plus Administration During Pregnancy

    Abstract. Superovulation is one of a reproductive technology to improve livestock productivity. The research was conducted to optimize the superovulation technology by combining it with administration of temulawak extract plus during pregnancy. Sixteen ewes were injected prostaglandin hormone at a dose of 10 mg/kg body weight intramuscularly twice, with eleven days interval, to synchronize estrous cycle. On the eleventh day, superovulation was  induced by injection of pregnant mare serum gonadotrophin  hormone at a dose of 200 IU/ewe intramuscularly. The ewes showing the estrous signs were mated naturally. Temulawak extract plus was administered weekly during pregnancy with a dose of 1 mg/kg body weight. Parameters measured in this study were lambs birth weight and preweaning growth. Superovulation increased average litter size. The result showed that, regardless of litter size, superovulation increased lambs birth weight by 15% as compared to controls and temulawak extract plus groups. Superovulation before mating and temulawak extract plus administration during pregnancy improved lambs growth in the first month and the third month postpartum. Superovulation prior to mating increased lambs birth weight and improved lambs growth performance before weaning. Keywords: superovulation, temulawak extract plus, lambs growth performance, ewes Abstrak.  Superovulasi merupakan salah satu teknologi reproduksi untuk meningkatkan produktivitas ternak. Penelitian ini dilakukan untuk mengoptimalkan teknologi superovulasi dengan menggabungkan pemberian ekstrak temulawak plus selama kebuntingan. Enam belas domba disuntik hormon prostaglandin dengan dosis 10 mg/kg bobot badan secara intramuskular sebanyak dua kali, dengan selang waktu sebelas hari, untuk menyesuaikan siklus estrus. Pada hari ke sebelas, superovulasi diinduksi dengan injeksi pregnant mare serum gonadotrophin hormon dengan dosis 200 IU/domba secara intramuskuler. Domba yang menunjukkan tanda-tanda estrus kemudian dikawinkan secara alami. Ekstrak temulawak plus diberikan setiap minggu selama kebuntingan dengan dosis 1 mg/kg bobot badan. Parameter yang diukur dalam penelitian ini adalah bobot lahir anak domba dan pertumbuhan prasapih. Superovulasi meningkatkan rata-rata litter size. Hasil penelitian menunjukkan bahwa, terlepas dari litter size, superovulasi meningkatkan bobot lahir anak domba sebesar 15% dibandingkan dengan kelompok ekstrak temulawak plus. Superovulasi sebelum kawin dan pemberian ekstrak temulawak plus selama kebuntingan  meningkatkan pertumbuhan anak domba di bulan pertama dan bulan ketiga paska melahirkan.     Prospects of hybrid renewable energy-based power system: A case study, post analysis of Chipendeke Micro-Hydro, Zimbabwe

    Fossil fuel-based energy sources are the major contributors to greenhouse gas (GHG) emission and thus the use of renewable energy (RE) is becoming the best alternative to cater for the increasing energy demand in both developing and developed nations. Chipendeke is a rural community in Zimbabwe, in which electricity demand is partially served by the only micro-hydro plant and hence, load shedding is a regular practice to keep essential services running. This study explored a suitable opportunity to identify a feasible system with different energy sources that can fulfill the current and projected future load demand of the community. A techno-economic feasibility study for a hybrid RE based power system (REPS) is examined considering various energy sources and cost functions. Six different system configurations have been designed with different sizing combinations to identify the most optimum solution for the locality considering techno-economic and environmental viability. The performance metrics considered to evaluate the best suitable model are; Net Present Cost (NPC), Cost of Energy (COE), Renewable Fraction (RF), excess energy and seasonal load variations. In-depth, sensitivity analyses have been performed to investigate the variations of the studied models with a little variation of input variables. Of the studied configurations, an off-grid hybrid Hydro/PV/DG/Battery system was found to be the most economically feasible compared to other configurations. This system had the lowest NPC and COE of 307,657and 307,657 and 0.165/kWh respectively and the highest RF of 87.5%. The proposed hybrid system could apply to any other remote areas in the region and anywhere worldwide

    Inhibiting ERK Activation with CI-1040 Leads to Compensatory Upregulation of Alternate MAPKs and Plasminogen Activator Inhibitor-1 following Subtotal Nephrectomy with No Impact on Kidney Fibrosis

    Extracellular-signal regulated kinase (ERK) activation by MEK plays a key role in many of the cellular processes that underlie progressive kidney fibrosis including cell proliferation, apoptosis and transforming growth factor β1-mediated epithelial to mesenchymal transition. We therefore assessed the therapeutic impact of ERK1/2 inhibition using a MEK inhibitor in the rat 5/6 subtotal nephrectomy (SNx) model of kidney fibrosis. There was a twentyfold upregulation in phospho-ERK1/2 expression in the kidney after SNx in Male Wistar rats. Rats undergoing SNx became hypertensive, proteinuric and developed progressive kidney failure with reduced creatinine clearance. Treatment with the MEK inhibitor, CI-1040 abolished phospho- ERK1/2 expression in kidney tissue and prevented phospho-ERK1/2 expression in peripheral lymphocytes during the entire course of therapy. CI-1040 had no impact on creatinine clearance, proteinuria, glomerular and tubular fibrosis, and α-smooth muscle actin expression. However, inhibition of ERK1/2 activation led to significant compensatory upregulation of the MAP kinases, p38 and JNK in kidney tissue. CI-1040 also increased the expression of plasminogen activator inhibitor-1 (PAI-1), a key inhibitor of plasmin-dependent matrix metalloproteinases. Thus inhibition of ERK1/2 activation has no therapeutic effect on kidney fibrosis in SNx possibly due to increased compensatory activation of the p38 and JNK signalling pathways with subsequent upregulation of PAI-1

    Assessment of Virally Vectored Autoimmunity as a Biocontrol Strategy for Cane Toads

    BACKGROUND: The cane toad, Bufo (Chaunus) marinus, is one of the most notorious vertebrate pests introduced into Australia over the last 200 years and, so far, efforts to identify a naturally occurring B. marinus-specific pathogen for use as a biological control agent have been unsuccessful. We explored an alternative approach that entailed genetically modifying a pathogen with broad host specificity so that it no longer caused disease, but carried a gene to disrupt the cane toad life cycle in a species specific manner. METHODOLOGY/PRINCIPAL FINDINGS: The adult beta globin gene was selected as the model gene for proof of concept of autoimmunity as a biocontrol method for cane toads. A previous report showed injection of bullfrog tadpoles with adult beta globin resulted in an alteration in the form of beta globin expressed in metamorphs as well as reduced survival. In B. marinus we established for the first time that the switch from tadpole to adult globin exists. The effect of injecting B. marinus tadpoles with purified recombinant adult globin protein was then assessed using behavioural (swim speed in tadpoles and jump length in metamorphs), developmental (time to metamorphosis, weight and length at various developmental stages, protein profile of adult globin) and genetic (adult globin mRNA levels) measures. However, we were unable to detect any differences between treated and control animals. Further, globin delivery using Bohle iridovirus, an Australian ranavirus isolate belonging to the Iridovirus family, did not reduce the survival of metamorphs or alter the form of beta globin expressed in metamorphs. CONCLUSIONS/SIGNIFICANCE: While we were able to show for the first time that the switch from tadpole to adult globin does occur in B. marinus, we were not able to induce autoimmunity and disrupt metamorphosis. The short development time of B. marinus tadpoles may preclude this approach

    Prevalence and determinants of asthma in adult male leather tannery workers in Karachi, Pakistan: A cross sectional study

    BACKGROUND: This study aimed to estimate the prevalence and to identify some risk factors of adult asthma in male leather tannery workers in Karachi, Pakistan. METHODS: A cross sectional study was conducted from August 2003 to March 2004 on leather tannery workers of Karachi, Pakistan. Data were collected from 641 workers engaged in 95 different tanneries in Korangi industrial area selected as sample of convenience. Face to face interviews were performed using a structured pre-tested questionnaire by trained data collectors. RESULTS: Prevalence of adult asthma was 10.8% (69/641) in this study population. The prevalence of perceived work-related asthma was 5.3% (34/641). Multivariable logistic regression model showed that after taking into account the age effect, the leather tannery worker were more likely to be asthmatic, if they were illiterate (adjusted OR = 2.13, 95% CI: 1.17–3.88), of Pathan ethnicity (adjusted OR = 2.69; 95% CI: 1.35–5.36), ever-smoked (adjusted OR = 2.22, 95% CI: 1.16–4.26), reportedly never used gloves during different tanning tasks (OR = 3.28; 95% CI : 1.72–6.26). Also, the final model showed a significant interaction between perceived allergy and duration of work. Those who perceived to have allergy were more likely to have asthma if their duration of work was 8 years (adjusted OR = 2.26; 95% CI: 1.19 – 4.29) and this relationship was even stronger if duration was 13 years (adjusted OR = 3.67; 95% CI: 1.98–6.79). CONCLUSION: Prevalence of asthma in leather tannery workers appears to be high and is associated with educational status, ethnicity, smoking, glove use, perceived to have allergy and duration of work

    Work-related allergy in medical doctors: atopy, exposure to domestic animals, eczema induced by common chemicals and membership of the surgical profession as potential risk factors

    Purpose To investigate the risk factors associated with work-related allergy-like symptoms in medical doctors. Methods Self-administered questionnaire survey and CAP test were conducted among medical school students in the 4th grade of their 6-year medical course in 1993–1996 and 1999–2001. Follow-up questionnaires were sent in 2004 to the graduates. These questionnaires enquired into personal and family history of allergic diseases, lifestyle, history of allergy-like symptoms including work-relatedness and occupational history as medical doctors. Relationships between allergy-like symptoms and relevant factors were evaluated by multivariate logistic regression analysis. Results Of 261 respondents at the follow-up survey, 139 (53.3%) and 54 (20.7%) had a history of any allergy-like symptoms and any work-related allergy-like symptoms, respectively. Female gender and family history of allergic diseases were signiWcantly associated with any allergy-like symptoms. Personal history of allergic disease, exposure to domestic animals, eczema caused by rubber gloves, metallic accessories, or cosmetics during schooling days, and membership of the surgical profession were signiW- cant risk factors for work-related allergy-like symptoms. On the contrary, to work-related allergy-like symptoms, gender, age, and smoking status were not signiWcantly related, and consumption of prepared foods was inversely related. Conclusions Personal history of atopy and eczema induced by common goods and the history of keeping domestic animals may be predictors of work-related allergy-like symptoms in doctors. After graduation from medical school, physicians start with exposure to various allergens and irritants at work, which relate to work-related allergy-like symptoms, especially for surgeons

    Macrophages undergo metabolic changes during activation that are coupled to functional responses. The gram negative bacterial product lipopolysaccharide (LPS) is especially potent at driving metabolic reprogramming, enhancing glycolysis and altering the Krebs cycle. Here we describe a role for the citrate-derived metabolite malonyl-CoA in the effect of LPS in macrophages. Malonylation of a wide variety of proteins occurs in response to LPS. We focused on one of these, glyceraldehyde-3-phosphate dehydrogenase (GAPDH). In resting macrophages, GAPDH binds to and suppresses translation of several inflammatory mRNAs, including that encoding TNFα. Upon LPS stimulation, GAPDH undergoes malonylation on lysine 213, leading to its dissociation from TNFα mRNA, promoting translation. We therefore identify for the first time malonylation as a signal, regulating GAPDH mRNA binding to promote inflammation

    Individuals with Le(a+b−) Blood Group Have Increased Susceptibility to Symptomatic Vibrio cholerae O1 Infection

    Cholera remains a severe diarrheal disease, capable of causing extensive outbreaks and high mortality. Blood group is one of the genetic factors determining predisposition to disease, including infectious diseases. Expression of different Lewis or ABO blood group types has been shown to be associated with risk of different enteric infections. For example, individuals of blood group O have a higher risk of severe illness due to V. cholerae compared to those with non-blood group O antigens. In this study, we have determined the relationship of the Lewis blood group antigen phenotypes with the risk of symptomatic cholera as well as the severity of disease and immune responses following infection. We show that individuals expressing the Le(a+b−) phenotype were more susceptible to symptomatic cholera, while Le(a–b+) expressing individuals were less susceptible. Individuals with the Le(a–b−) blood group had a longer duration of diarrhea when infected, required more intravenous fluid replacement, and had lower plasma IgA antibody responses to V. cholerae LPS on day 7 following infection. We conclude that there is an association between the Lewis blood group and the risk of cholera, and that this risk may affect the outcome of infection as well as possibly the efficacy of vaccination
